This article describes the reaction of vindoline with formaldehyde and trimethyl orthoformate to prepare vindolicine, tris-vindolicinyl methane and higher molecular weight homologues. The synthesis of 10-formyl vindoline as an intermediate allowed further exploration of its chemistry, in particular the reaction with acetone which yielded a symmetrical dimer, which was further reacted with vindoline to give molecules containing three and four vindoline units. These molecules were characterized by NMR and for some of them (vindolicine, 10-formyl vindoline, 10-(1'-(but-1'-en-3'-one))-vindoline) b

For peripheral neuropathy, standards of care are based on established evidence-based practice. Peripheral neuropathy is disease or dysfunction of one or more peripheral nerves (i.e., motor, sensory, or autonomic), resulting in numbness or weakness. Peripheral neuropathy occurs outside of the brain and spinal cord and is caused by cancer, treatment, or both. Chemotherapeutic agents that can cause peripheral neuropathy include epothilones, platinum analogs, taxanes, and vinca alkaloids, as well as immunomodulating drugs and proteasome inhibitors.

Five consecutive patients with progressive extracutaneous stage IV mycosis fungoides (MF) were treated with VM 25, 100 mg/m2 i.v., for at least 3 cycles. All patients had been extensively pretreated and in particular with vinca alkaloids. Two partial responses of 5 and 9+ months duration and 1 minimal response of 5 months duration were obtained. Transient myelosuppression was encountered in all patients. These preliminary results should encourage further exploration of VM 26, even in less advanced MF.

Vincristine, a cornerstone vinca alkaloid in pediatric oncology, has historically been regarded as a low-risk agent for drug-induced acute pancreatitis (DIAP). We report the first documented pediatric case of vincristine monotherapy-associated acute pancreatitis, challenging existing toxicity paradigms. A 3.25-year-old boy with stage IV Wilms tumor developed acute-onset fever and localized periumbilical pain 48 h after vincristine infusion (0.9 mg), administered within standard dosing. Laboratory testing confirmed marked pancreatic hyperenzymemia (serum α-amylase 2,022 U/L, 12.5 × ULN; lipase

Liver fibrosis is associated with increased mortality and morbidity. However, there is not effective treatment so far. Vinpocetine (Vinpo) is a synthetic derivative of vinca alkaloid vincamine. Limited previous reports have shown some beneficial effects of Vinpo in different organ fibrosis, but the ability of Vinpo to inhibit liver fibrosis induced by thioacetamide (TAA) has not been reported, that is why we investigate the potential ability of this vinca alkaloid derivative to attenuate liver fibrosis. Hepatic fibrosis was induced in male Sprague Dawley rats by TAA (200 mg/kg; ip; 3 times/wee

UNLABELLED: Vincristine, a vinca alkaloid, is widely used in many hematologic disorders and pediatric cancers, and acts by binding to the tubulin protein, to inhibit effective cell division. Vincristine-induced anemia has been observed, but its mechanism is not well understood. We describe a case involving serious vincristine-induced anemia in a patient with congenital spherocytosis and provide the explanation to the underlying mechanism. This report demonstrates that vincristine administration to patients with a red cell disorder may require additional clinical interventions to compensate for

Vinorelbine (NVB) is a novel vinca alkaloid FDA approved for use in some advanced carcinomas. However, its role in non-Hodgkin's lymphoma (NHL) is still not well defined. NVB is an antimicrotubule agent, but as yet, it is not known whether it induces apoptosis. By flow cytometry using nuclear staining (propidium iodide) and annexin V, we demonstrated that NVB and vincristine (VCR) induced both mitotic arrest and apoptosis in leukemia and lymphoma cells, in a drug exposure time dependent manner. Cell cycle kinetics in 3 different cell lines varied during vinca alkaloid treatment. The annexin V

16 adult idiopathic thrombocytopenic purpura (ITP) patients, with mean age 51 years, were treated by 5 weekly slow intravenous infusions of vinblastine (0.1 mg/kg). When a complete or partial response (CR or PR) was obtained, slow infusions were repeated at 2-weekly then monthly intervals, as maintenance therapy. In 4 cases with ITP of recent onset, but presenting contraindication or failure to prednisone and high dose intravenous immunoglobulins, CR was obtained in all patients after 4 to 15 days (mean 9.5). Only one of these patients relapsed. In 12 cases of refractory chronic ITP (of whom 1

The antitumour effect of vindesine (desacetyl vinvlastine amide sulfate) in disseminated malignant melanoma has been investigated in our oncology service in a phase-II study. Of 25 patients exposed to the drug, 23 were evaluable, and seven of these (30%) were considered objective responders. The observed response of malignant effusions is of particular interest. Side effects related to vindesine are similar to those of the other vinca alkaloids, although alopecia may be more pronounced with the doses used in this study.

BACKGROUND: Charcot Marie Tooth (CMT) disease is the most common form of hereditary neuropathy. Due to the high prevalence of mild and undiagnosed forms, patients with CMT disease may be exposed to severe neurotoxicity following the administration of neurotoxic chemotherapies. The aim of this report is to alert oncologists to the potential to precipitate severe irreversible peripheral neuropathies when administering neurotoxic compounds to undiagnosed CMT patients.
MATERIAL AND METHODS: A retrospective research in the OncoNeuroTox database was performed (2010-2016), searching for patients wit

Three separate in vitro admixture experiments were accomplished with bleomycin and other common parenteral medications. Bleomycin was assayed by a radioimmunoassay procedure. Bleomycin was found to be compatible with a majority of drugs including the aminoglycoside antibiotics, the vinca alkaloid antineoplastics, the antimetabolite fluorouracil, several phosphate salt corticosteroids, diphenhydramine and the anticoagulant heparin sulfate. Incompatible admixtures include the acidic cephalosporin and penicillin-derived antibiotics, the antineoplastics, methotrexate, and mitomycin C, the sodium s

Liver dysfunction in patients with lymphoma presents an important clinical dilemma because it limits the use of anthracyclines and vinca alkaloids for this potentially curable malignancy. Patients are typically treated with corticosteroids, rituximab and the nitrogen mustard mechlorethamine, and then transitioned to definitive therapy after improvement in liver function. Between 2010 and 2012 mechlorethamine was not available and other strategies were required. In this case series, we describe seven cases of patients with lymphoma and liver dysfunction successfully treated in this time period

One of the new agents intensively studied in the last decade for the treatment of metastatic breast cancer is vinorelbine (Navelbine; Burroughs Wellcome Co, Research Triangle Park, NC; Pierre Fabre Médicament, Paris, France). This vinca alkaloid produces response rates comparable to other single agents in women with metastatic breast cancer and is associated with a nonhematologic toxicity profile that makes it particularly advantageous for palliative treatment. In vitro studies provide evidence of the decreased neurotoxicity of vinorelbine compared with other vinca alkaloids and clinical studi

Catharanthus roseus is a medicinal plant widely known for producing monoterpenoid indole alkaloids (MIAs), including therapeutic compounds such as vinblastine and vincristine, which are crucial for cancer treatment. However, the naturally low concentration of these alkaloids in plant tissues poses a significant challenge for large-scale production. This study explores the application of siderophore-producing bacteria for seed bacterization of Catharanthus roseus to enhance the production of MIAs, including vindoline, catharanthine, and vinblastine. Utilizing High-Performance Liquid Chromatogra

Results of different types of treatment in 416 adults with chronic idiopathic thrombocytopenic purpura (ITP) were analyzed. Of 368 patients treated with corticosteroids, 278 (77%) achieved complete response (CR ie, normalization of platelet count). However, only 18% of patients had continued complete response (CCR). Results of splenectomy was analyzed in 126 patients, 49% of them achieved CCR and 76% of them had beneficial effects from the operation. Emergency splenectomy was a life saving procedure in 8 of 11 patients with life-threatening hemorrhage. Vinca alkaloids gave a 50% transient CR.

Ethyl apovincaminate (EGH-4405, Cavinton) treatment has been used in various sensorineural impairments of hearing as well as in ear diseases associated with vertigo. As shown by experinces, presbyacusis, vascular impairment of hearing and in certain cases toxic hypacusia are improved by Cavinton. Besides audiometric improvement of pure tone, significant improvement was noted in the speech-audiogram in all three groups of disease. Certain affections of the ear accompanied by vertigo responded to Cavinton. In Menière's disease it has proved to be superior to the vasodilators used up to now.

Immunological involvement of peripheral nervous system in non-Hodgkin lymphoma (NHL) is very rare and it may be difficult to differentiate it from vincristine-induced neuropathy. We report clinical and electrophysiological findings of an 8-year-old male with NHL who developed acute onset fulminant motor sensory autonomic neuropathy during induction chemotherapy which included vincristine. Characteristic clinical picture and nerve conduction studies favored Guillain-Barré syndrome. The patient improved rapidly with intravenous immunoglobulin and supportive care. It is possible that an immune me

Vinzolidine is a new, orally active, semisynthetic vinca alkaloid which shows broad anti-tumor activity against murine tumor test systems. This phase I study established a 1 day every 2 week schedule of 35 mg/m2 in good-risk patients and of 30 mg/m2 in poor-risk patients. Maximal tolerated dose was 45 mg/m2 with severe neutropenia, syndrome of inappropriate antidiuretic hormone, and paralytic ileus. Significant antitumor responses were seen in two patients with lymphoma and in one with squamous cell cancer of the lung.
