Hypoxia-induced GP73 promotes the Warburg effect in hepatocellular carcinoma via HIF-1α.

BACKGROUND: Transarterial chemoembolization (TACE)-induced hypoxia is a key driver of hepatocellular carcinoma (HCC) progression. This study investigated whether Golgi protein 73 (GP73) drives the Warburg effect and epithelial-mesenchymal transition (EMT) via hypoxia-inducible factor-1α (HIF-1α) under hypoxia, thereby promoting HCC malignancy. METHODS: HepG2 cells were exposed to hypoxia (1% O₂). Lentivirus-mediated GP73 overexpression (LV-GP73) and siRNA-mediated HIF-1α knockdown (si-HIF-1α) were established. Four groups were compared: H + LV-NC (negative control), H + LV-GP73, H + LV-GP73 +

Crucial Role of c-Myc/Monocarboxylate Transporter 4 Signaling in Capsaicin Induced Apoptotic and Anti-Warburg Effects in Hepatocellular Carcinoma.

Though Capsaicin from chili peppers was known to have antitumor effects in several cancers, the underlying antitumor pathogenesis of Capsaicin is not clear to date. Thus, the antitumor mechanism of Capsaicin was explored in Hep3B and Huh7 hepatocellular carcinoma (HCC) cells in relation to c-Myc/monocarboxylate transporter 4 (MCT4) signaling. To elucidate the antitumor mechanism of capsaicin, cytotoxicity assay, cell cycle analysis, Western blotting, RT-qPCR, RNA interference, ELISA, immunoprecipitation, and mouse xenograft model were used in this work. Capsaicin increased the cytotoxicity, su

Antagonistic Pleiotropy at the HCAR1 Locus Reveals a Genetic Basis for Solid Tumor Resistance in Schizophrenia.

Historic epidemiological studies have long observed a paradoxical resistance to solid tumors in cohorts with severe psychiatric conditions, such as schizophrenia (SCZ), despite higher prevalences of lifestyle risk factors. Concurrently, oncology has established that solid tumors utilize Warburg glycolysis to generate a "lactate shield," binding the HCAR1 receptor on infiltrating macrophages to suppress immune proliferation. Here, we identify the shared genomic etiology resolving this paradox. Utilizing cross-trait genome-wide association studies (GWAS), we demonstrate that solid tumors exhibit

Reversing the Warburg effect: Discovery of potent pyruvate dehydrogenase kinase inhibitors for inhibiting tumor growth.

Pyruvate dehydrogenase kinase 1 (PDK1) is overexpressed in tumors, driving cancer progression by promoting the Warburg effect. Consequently, PDK1 inhibition represents a promising anticancer strategy, though potent low-toxicity inhibitors remain scarce. In this study, 20 new derivatives were designed and synthesized through further structural optimization of the dichloroacetate (DCA) derivatives, and their inhibitory effects on PDK1 and anticancer activities were systematically evaluated. Compound D16 emerged as the lead candidate, demonstrating potent PDK1 inhibition (53.20 % at 1 μM), signif

Glycolysis-related gene expression profiling serves as a novel prognosis risk predictor for human hepatocellular carcinoma.

Metabolic pattern reconstruction is an important factor in tumor progression. Metabolism of tumor cells is characterized by abnormal increase in anaerobic glycolysis, regardless of high oxygen concentration, resulting in a significant accumulation of energy from glucose sources. These changes promotes rapid cell proliferation and tumor growth, which is further referenced a process known as the Warburg effect. The current study reconstructed the metabolic pattern in progression of cancer to identify genetic changes specific in cancer cells. A total of 12 common types of solid tumors were includ

Whole-transcriptome Analysis of Fully Viable Energy Efficient Glycolytic-null Cancer Cells Established by Double Genetic Knockout of Lactate Dehydrogenase A/B or Glucose-6-Phosphate Isomerase.

BACKGROUND/AIM: Nearly all mammalian tumors of diverse tissues are believed to be dependent on fermentative glycolysis, marked by elevated production of lactic acid and expression of glycolytic enzymes, most notably lactic acid dehydrogenase (LDH). Therefore, there has been significant interest in developing chemotherapy drugs that selectively target various isoforms of the LDH enzyme. However, considerable questions remain as to the consequences of biological ablation of LDH or upstream targeting of the glycolytic pathway. MATERIALS AND METHODS: In this study, we explore the biochemical and

Sublethal heat stress-induced O-GlcNAcylation coordinates the Warburg effect to promote hepatocellular carcinoma recurrence and metastasis after thermal ablation.

The malignant transformation of residual hepatocellular carcinoma (HCC) cells after thermal ablation is considered as the main factor promoting postoperative HCC progression, which greatly limits the improvement of long-term survival, and at present there is no effective targeted therapeutic strategies. The Warburg effect is a metabolic feature correlated highly with malignant transformation (e.g. epithelial-to-mesenchymal transition [EMT]). Here, we showed that sublethal heat stress triggered a stronger Warburg effect of HCC cells, which contributed to the thermotolerance and invasion of HCC

LncRNA YIYA drives pancreatic cancer proliferation under high-glucose conditions by reinforcing a RAS-PKM2-dependent Warburg phenotype.

Hyperglycaemia is an independent risk factor for pancreatic cancer (PC). Here, we show that both chronic and intermittent high glucose cause a robust upregulation of the novel lncRNA LINC00538 (YIYA) in pancreatic ductal adenocarcinoma (PDAC) cells. YIYA enhances PDAC cell proliferation in both 2D and 3D culture systems, and its expression shows a positive correlation with poor patient survival. Importantly, knockdown of YIYA attenuates the proliferative effect. Mechanistically, YIYA drives the Warburg phenotype in a KRAS-dependent manner. YIYA physically interacts with KRAS and the glycolytic

ETS1 drives EGF-induced glycolytic shift and metastasis of epithelial ovarian cancer cells.

Epithelial ovarian cancer (EOC), a leading cause of gynecological cancer-related morbidity and mortality and the most common type of ovarian cancer (OC), is widely characterized by alterations in the Epidermal Growth Factor (EGF) signaling pathways. The phenomenon of metastasis is largely held accountable for the majority of EOC-associated deaths. Existing literature reports substantiate evidence on the indispensable role of metabolic reprogramming, particularly the phenomenon of the 'Warburg effect' or aerobic glycolysis in priming the cancer cells towards Epithelial to Mesenchymal transition

ALKBH2 promotes the Warburg effect and bladder cancer progression under hypoxic conditions via the PI3K/AKT pathway.

Emerging evidence links DNA repair enzymes to metabolic reprogramming in cancer. ALKBH2 safeguards genomic integrity by reversing alkylation damage, yet its potential role beyond repair-specifically in modulating the Warburg effect-remains unexplored in bladder cancer. While implicated in digestive tract malignancies, ALKBH2's function in urothelial carcinoma is unknown. Investigating this gap may reveal ALKBH2 as a novel metabolic driver in bladder cancer progression. ALKBH2 expression was analyzed in bladder cancer tissues and matched adjacent normal tissues by qPCR, Western blotting, and im

NF-kappa B interacting long noncoding RNA enhances the Warburg effect and angiogenesis and is associated with decreased survival of patients with gliomas.

In various malignant tumors, NF-kappa B interacting long noncoding RNA (NKILA) displays antitumor activity by inhibiting the NF-kappa B pathway. However, the role of NKILA in gliomas remains unclear. Surprisingly, this study showed that NKILA is significantly upregulated in gliomas, and the increased levels of NKILA were correlated with a decrease in patient survival time. NKILA increased the expression level of hypoxia-inducible factor-1α, and the activity of the hypoxia pathway in gliomas. Furthermore, we demonstrated that NKILA enhances the Warburg effect and angiogenesis in gliomas both in

Recent progress in the development of small molecule pyruvate kinase M2 inhibitors: 2020-2025.

Pyruvate kinase M2 (PKM2) is a central regulator of glycolysis and anabolic metabolism, playing a pivotal role in cancer cell proliferation. Its multifunctional nature and involvement in various disease pathways make it an attractive therapeutic target, especially in oncology and inflammation. This review summarizes research over the past five years on small molecule PKM2 inhibitors. Activators of PKM2 promote the tetrameric form of PKM2, enhancing oxidative phosphorylation and reversing the Warburg effect. In contrast, inhibitors like micheliolide (MCL) and isoselenazolium compounds disrupt P

m6 A transferase METTL3-induced lncRNA ABHD11-AS1 promotes the Warburg effect of non-small-cell lung cancer.

N6 -methyladenosine (m6 A) and long noncoding RNAs (lncRNAs) are both crucial regulators in non-small-cell lung cancer (NSCLC) tumorigenesis. However, the pathological roles of m6 A and lncRNAs in NSCLC progression are still limited and undefined. Here, lncRNA ABHD11-AS1 was upregulated in NSCLC tissue specimens and cells and the ectopic overexpression was closely correlated with unfavorable prognosis of NSCLC patients. Functionally, ABHD11-AS1 promoted the proliferation and Warburg effect of NSCLC. Mechanistically, m6 A profile was analyzed by methylated RNA immunoprecipitation sequencing (Me

IL-10RA promotes lung cancer cell proliferation by increasing fatty acid oxidation via STAT3 signaling pathway.

Metabolic reprogramming in tumor cells plays a crucial role in promoting cell proliferation and metastasis, and is currently recognized as a significant marker of tumor progression. Interleukin-10 receptor subunit alpha (IL-10RA), a member of the type II cytokine receptor family, is predominantly expressed on macrophages and T cells and plays a crucial role in regulating immune cell metabolism and immune response. However, its role in the energy metabolic pathways of tumor cells remains unclear. In this study, we found increased expression of IL-10RA in human non-small cell lung cancer (NSCLC)

Nanomedicine-enabled disruption of glucose metabolism and synergistic antitumor therapy.

Tumor cells exploit glucose metabolic reprogramming, most notably the Warburg effect of aerobic glycolysis to fuel rapid proliferation, biosynthetic precursor production, and immunosuppressive microenvironment formation. While this creates therapeutic vulnerabilities, conventional metabolic inhibitors face critical limitations in systemic toxicity, metabolic heterogeneity, and inadequate tumor accumulation. Hence, this review details how nanomedicines overcome these limitations through active targeting, enhanced tumor accumulation, stimuli-responsive drug release, and co-delivering multiple dr
