Withania coagulans extract attenuates oxidative stress-mediated apoptosis of cerebellar purkinje neurons after ischemia/reperfusion injury.

Cerebral ischemia/reperfusion (I/R) is known to increase reactive oxygen species (ROS) generation, consequences of oxidative stress (OS), and neuronal death in the susceptible brain areas including the cerebellum. Newly, remarkable attention has been paid to a natural diet with the capability to scavenge ROS. Withania coagulans root extract (WCE) is rich in components with antioxidants properties. Therefore, this study aimed to evaluate the effect of WCE on cerebellar Purkinje cells (PCs) against OS-mediated apoptosis after I/R injury. In this experimental study 64 male adult Wistar rats were

Root exudates of Withania somnifera prevent the risk of transmission of soil-dwelling Enterobacterales to nearby plants.

Soil-dwelling enteric pathogens are indicators of sewage or fecal matter contamination. When such organisms enter agricultural fields, they colonize crops and cause pre-harvest contamination of food products. Consumption of such contaminated products can lead to disease outbreaks. Thus, preventing pre-harvest contamination of food products by enteric bacteria dwelling in soil is essential. The Cooch Behar district of India receives > 3000 mm rainfall annually causing an anaerobic condition in soil, creating an ambient condition for the survival of enteric bacteria. While screening for native E

Withania somnifera Regulates Mitochondrial Biogenesis and Energetics in Rat Cortical Neurons: Role of BDNF and SIRT1.

Withania somnifera, a psychoactive plant with putative neuroprotective actions, is used in Indian traditional medicine for the treatment of neuropsychiatric and neurodegenerative disorders. However, the key mechanisms underlying the pleiotropic actions of Withania somnifera on the nervous system remain poorly understood. Given converging evidence suggests a critical role for mitochondrial dysfunction in the pathophysiology of neuropsychiatric and neurodegenerative diseases, we hypothesized that Withania somnifera may exert pleiotropic effects via targeting mitochondria. Treatment with Withania

Simultaneous quantification of withanolides in Withania somnifera by a validated high-performance thin-layer chromatographic method.

This paper describes a sensitive, selective, specific, robust, and validated densitometric high-performance thin-layer chromatographic (HPTLC) method for the simultaneous determination of 3 key withanolides, namely, withaferin-A, 12-deoxywithastramonolide, and withanolide-A, in Ashwagandha (Withania somnifera) plant samples. The separation was performed on aluminum-backed silica gel 60F254 HPTLC plates using dichloromethane-methanol-acetone-diethyl ether (15 + 1 + 1 + 1, v/v/v/v) as the mobile phase. The withanolides were quantified by densitometry in the reflection/absorption mode at 230 nm.

Withania Somnifera Extract Mitigates Experimental Acute Graft versus Host Disease Without Abrogating Graft Versus Leukemia Effect.

Acute graft versus host disease (aGvHD) is the major contributor of nonrelapse mortality in alloHSCT. It is associated with an inflammatory immune response manifesting as cytokine storm with ensuing damage to target organs such as liver, gut, and skin. Prevention of aGvHD while retaining the beneficial graft versus leukemia (GvL) effect remains a major challenge. Withania somnifera extract (WSE) is known for its anti-inflammatory, immune-modulatory, and anticancer properties, which are appealing in the context of aGvHD. Herein, we demonstrated that prophylactic and therapeutic use of WSE in ex

Chitosan activates a sequential Ca 2+ -ROS-NO-jasmonate signaling cascade governing withanolide accumulation in Withania somnifera.

Chitosan promotes withanolide accumulation in W. somnifera; however, the temporal organization of the underlying signaling network remains unresolved. We examined whether calcium (Ca2⁺), reactive oxygen species (ROS), nitric oxide (NO), and jasmone form an ordered cascade linking elicitor perception to specialized metabolism. Sixty-day-old greenhouse-grown plants were treated with 100 mg L-1 foliar chitosan alone or after pretreatment with LaCl₃, diphenyleneiodonium, cPTIO, or diethyldithiocarbamate, with inhibitor-only controls. Cytosolic Ca2+ -dependent fluorescence, NADPH oxidase activity,

Whole-genome sequencing shows modulation of neurodegenerative genes by Withania somnifera in human SK-N-SH cells.

BACKGROUND: Aging is driven by several primary and secondary hallmarks that manifest with age, of which neurodegenerative diseases are important manifestations. The ability to decelerate or reverse aging, and promote healthy aging, has garnered great interest in recent times. In traditional medicine, Withania somnifera (WS) or Ashwagandha has been recognized for its adaptogenic and rejuvenative effects. METHODS: To investigate WS-modulated global gene expression profiles, we performed whole-genome sequencing of WS-treated human neuroblastoma SK-N-SH cells at different doses (50 and 100 μg/mL)
