Ivermectin treatment was evaluated for efficacy and side effects in 40 patients in South India who had microfilaremia and bancroftian filariasis. Ivermectin was administered once orally at four dose levels (range, 25 to 200 micrograms/kg), and at each it was found to be completely effective in clearing blood microfilariae within five to 12 days. In most patients, microfilariae reappeared by three months; by six months the levels averaged 14% to 32% of pretreatment values in the four study groups, and all groups showed equivalent efficacy. Detailed monitoring identified some side effects in alm

A survey for Wuchereria bancrofti in Limbe, Haiti (est. pop. = 10,500) revealed that 17% (231/1,450) had a patent infection. Nearly half of those surveyed harbored fewer than 10 microfilariae (mf) per 20 mm3 of finger-prick blood; the median mf density for females and males was 12.4 and 9.5, respectively. Parasitemias occurred as early as age 4. Antibody titers greater than or equal to 1:20 against adult D. viteae antigen were observed in 38% of microfilaremic individuals and in 29% of amicrofilaremic individuals. Peak antibody responsiveness (40%) was observed between 5 and 9 years of age. In

A single dose of levamisole of 3 mg/kg had a marked microfilaricidal effect in some persons harbouring Wuchereria bancrofti, but there was considerable variation in the response. In some cases blood examinations for microfilariae remained negative or at a low level for several months. Persons with high pretreatment microfilaraemias responded less to therapy than did those with low counts. The main side-reaction to levamisole was fever, which began six to eight hours after treatment. Combined DEC-levamisole therapy appears to have no advantage over the use of DEC alone. Amodiaquine in a total d

The authors have examined the ultrastructure of the rectum of infective-stage Wuchereria bancrofti by transmission electron microscopy. Our observations show that the rectum is divided into anterior and posterior segments. The cells of the anterior rectum appear to be derived from the microfilarial R (rectal) cells described by other authors. In both stages, these cells show voluminous nuclei, abundant mitochondria, and small cytoplasmic processes which contain fibrillar components. Amorphous material associated with these processes appears throughout the larval rectum and may protrude from th

Indigenous cases of bancroftian filariasis have recently been identified in the country for the first time. The diagnosis of such infections depends on demonstrating the characteristic sheathed microfilariae of Wuchereria bancrofti in the blood. Giemsa, a stain recommended by many authors, was found to be unsuitable as the microfilarial sheath, an important distinguishing feature, does not take up the stain. In contrast, the sheath stains clearly in haematoxylin. The differences in the staining reaction of the microfilariae in the two stains is demonstrated. The need to establish the presence

The high biting rate of Culex pipiens fatigans in Rangoon, combined with a low prevalence of microfilaraemia due to Wuchereria bancrofti, suggested a poor efficiency of transmission. Data obtained by the WHO Filariasis Research Unit in Rangoon were analysed, and the efficiency of the parasite from Stage III larva in the mosquito to the production of microfilariae was estimated as 6.04-6.71 x 10(-5), indicating that an average of around 15 500 bites by "infective" mosquitos is necessary to produce 1 case of microfilaraemia. This surprising result prompted a step-by-step analysis of the success

In this paper the author has compiled an up-to-date list of the principal natural vectors of human Wuchereria infections, arranged by zoogeographical regions and countries, and gives data on natural and experimental filarial infection rates in vectors in different parts of the world. As an aid to workers in areas where Wuchereria and Plasmodium infections co-exist, he also provides a list of anophelines as filarial vectors, including in this list the results of natural and experimental studies of Wuchereria infection both alone and in combination with malaria. Finally, the author suggests a num

BACKGROUND: Lymphatic filariasis (LF) is a neglected tropical disease caused by the filarial nematodes Wuchereria bancrofti, Brugia malayi and Brugia timori. The Global Program to Eliminate LF uses mass drug administration (MDA) of anti-filarial drugs that clear microfilariae (Mf) from blood to interrupt transmission by mosquitos. New diagnostic tools are needed to assess the impact of MDA on bancroftian filariasis, because available serologic tests can remain positive after successful treatment.
METHODOLOGY/PRINCIPAL FINDINGS: We identified Wb-bhp-1, which encodes a W. bancrofti homologue of

The draft genome assembly of the Wolbachia endosymbiont of Wuchereria bancrofti (wWb) consists of 1060 850 bp in 100 contigs and contains 961 ORFs, with a single copy of the 5S rRNA, 16S rRNA and 23S rRNA and each of the 34 tRNA genes. Phylogenetic core genome analyses show wWb to cluster with other strains in supergroup D of the Wolbachia phylogeny, while being most closely related to the Wolbachia endosymbiont of Brugia malayi strain TRS (wBm). The wWb and wBm genomes share 779 orthologous clusters with wWb having 101 unclustered genes and wBm having 23 unclustered genes. The higher number o

INTRODUCTION: Since the launch of the Global Programme to Eliminate Lymphatic Filariasis, more than 70% of the endemic countries have implemented mass drug administration (MDA) to interrupt disease transmission. The monitoring of filarial infection in sentinel populations, particularly schoolchildren, is recommended to assess the impact of MDA. A key issue is choosing the appropriate tools for these initial assessments (to define the best intervention) and for monitoring transmission.
METHODS: This study compared the pre-MDA performance of five diagnostic methods, namely, thick film test, Kno

Wuchereria bancrofti (Wb) is the most widely distributed of the three nematodes known to cause lymphatic filariasis (LF), the other two being Brugia malayi and Brugia timori. Current tools available to monitor LF are limited to diagnostic tests targeting DNA repeats, filarial antigens, and anti-filarial antibodies. While these tools are useful for detection and surveillance, elimination programs have yet to take full advantage of molecular typing for inferring infection history, strain fingerprinting, and evolution. To date, molecular typing approaches have included whole mitochondrial genomes

OBJECTIVE: To evaluate the efficacy of doxycycline as a macrofilaricidal agent against Wuchereria bancrofti.
METHOD: In the Western Region of Ghana, 18 patients infected with W. bancrofti were recruited and treated with 200 mg doxycycline per day for 4 weeks. Seven untreated patients served as controls. Four months after doxycycline treatment, all patients received 150 mug/kg ivermectin. Patients were monitored for Wolbachia and microfilaria loads, antigenaemia and filarial dance sign (FDS).
RESULTS: Four months after doxycycline treatment, cases had a significantly lower Wolbachia load than

The polymorphism of the 18S rRNA gene in Wuchereria bancrofti microfilariae (mf) collected from three different zones in India was analyzed by polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP). The RFLPs of the amplified products obtained after digestion with restriction enzymes Ssp I, Msp I and Hha I showed no difference in the banding patterns among the mf isolates from different endemic zones. Further the sequencing of PCR products did not show any difference in the nucleotide sequence either. The phylogenetic analysis of the sequences of W. bancrofti mf is

The immunoscreening of a microfilarial cDNA library of Wuchereria bancrofti with microfilaraemic sera revealed many positive clones expressing filarial antigens. One immunoreactive clone, designated PMR1, was shown to encode a protein of 114 amino acid residues. The cDNA fragment was subcloned into an expression vector, Pinpoint XaT. The resulting recombinant (r)PMR1-biotin fusion protein was expressed in Escherichia coli (BL21 [DE3] pLys) and was affinity purified on avidin resin. Analysis of sera of different groups for filarial antibodies against rPMR1 showed it to be highly reactive with m

Wuchereria bancrofti is the major cause of lymphatic filariasis in humans. Although it is responsible for this immensely morbid and debilitating disease, very little is known of the basic molecular biology of this parasite, and there is a vast lack of knowledge on its gene organisation. In this study, the actin gene of W. bancrofti has been characterised by sequencing a clone isolated from a genomic DNA library of this parasite. The 5' flanking region had a potential TATA box and a putative mRNA initiation site. The gene had five exons encoding 376 amino acids, and four introns ranging in size

Wuchereria bancrofti is a mosquito-borne filarial nematode that commonly invades lymphatic vessels. Common clinical manifestations include elephantiasis, orchitis, epididymitis, and chyluria. This report concerns an Egyptian man who developed superior vena cava syndrome secondary to a mediastinal mass that was found to contain a filarial nematode consistent with W. bancrofti. This is the first case, to our knowledge, of this parasite causing fibrosing mediastinitis.

The exsheathment of ingested microfilariae (mf) of Wuchereria bancrofti in their vector Culex. (Cx.) pipiens and in a refractory mosquito species Aedes (Ae.) caspius was assessed. No exsheathed mf were encountered in the mid gut of tested mosquitoes, while they were observed in abdomen and thoracic muscles. All exsheathed mf migrated from the mid gut of both tested species. The lowest percentage of ingested mf was recorded in the mid gut of Cx. pipiens and the thoracic muscles of Ae. caspius one day post feeding. On the other hand, 94.2% out of the exsheathed mf reached the thoracic region of

Antigen detection assays serve as a useful adjunct to blood examinations for studies of filariasis, in terms of the diagnostic and epidemiologic information provided. We examined the utility of the Og4C3 antigen detection enzyme-linked immunosorbent assay for field studies and analyzed the distribution of Wuchereria bancrofti antigenemia in a Haitian population. Using serum samples collected following venipuncture, antigenemia levels were correlated with microfilaremia (P < 0.001). The microfilariae had a pronounced nocturnal periodicity while the sensitivity of the antigen assay was the same
