Detection of Wuchereria bancrofti specific antigens in the serum of endemic residents.

A sandwich enzyme-linked immunosorbent assay (s-ELISA) is developed for detecting circulatory antigens in individuals infected with Wuchereria bancrofti in an endemic area using antibody (Ig) against excretory-secretory-metabolic antigens of the microfilariae raised in rabbit (anti-mf-ESM) and labelled with alkaline phosphatase (ESM-Ig-conjugate). An optical density reading of a sample greater than 0.075 (after subtracting the background) was taken as positive in the s-ELISA. When homologous (WbmfESM) and heterologous (sonicated antigens of human and model intestinal helminths-Ascaris lumbrico

Efficacy of co-administered diethylcarbamazine and albendazole against adult Wuchereria bancrofti.

Although diethylcarbamazine (DEC) and albendazole are recommended to interrupt transmission of Wuchereria bancrofti, little is known about the macrofilaricidal effect of this drug combination. Forty-seven men with W. bancrofti infection were randomly assigned to receive a single dose of either DEC alone (6 mg/kg) (n=25) or a combination of DEC (6 mg/kg) and albendazole (400 mg) (n=22). Physical examinations for scrotal nodules (resulting from worm death) and ultrasound examinations (to detect living adult worms) were performed before treatment and 7, 14, 30, 45, 60, 90, 180, 270 and 360 days a

Factors Associated with Wuchereria bancrofti Microfilaremia in an Endemic Area of Mali.

Although Wuchereria bancrofti (Wb), the causative agent of lymphatic filariasis, is endemic throughout Mali, the prevalence of Wb microfilaremia (Mf) can vary widely between villages despite similar prevalence of infection as assessed by circulating antigen. To examine this variation, cross-sectional data obtained during screening prior to an interventional study in two neighboring villages in Mali were analyzed. The overall prevalence of Wb, as assessed by Wb CAg (circulating antigen), was 50.3% among 373 participants, aged 14-65. Wuchereria bancrofti Mf-positive and negative individuals appe

Identification of Wb123 as an early and specific marker of Wuchereria bancrofti infection.

BACKGROUND: The current antibody tests used for monitoring in lymphatic filariasis (LF) elimination programs suffer from poor specificity because of the considerable geographical overlap with other filarial infections such as Loa loa (Ll), Onchocerca volvulus (Ov), and Mansonella perstans (Mp). METHODS: Using bioinformatics to assemble into contigs 2048 expressed sequence tags (ESTs) from the L3 infective larvae of W. bancrofti (Wb), these were next assessed for homology to known proteins and nucleotides and to similar assemblies of L3 larval ESTs of B. malayi (Bm - n = 5068), Ov (n = 4166),

Evaluation of Wuchereria bancrofti GST as a vaccine candidate for lymphatic filariasis.

BACKGROUND: Lymphatic filarial parasites survive within the lymphatic vessels for years despite the complex immune environment surrounding them. Parasites possibly accomplish this by adopting various immunomodulatory strategies, which include release of glutathione-S-transferases (GSTs) that counteract the oxidative free radicals produced by the host. Since GSTs produced by parasites appear to be critical for the survival of parasites in the host, several studies evaluated the potential of parasite GSTs as vaccine candidates especially against schistosomiasis, fascioliasis and Seteria cervi. Concomitant infections of Plasmodium falciparum and Wuchereria bancrofti on the Kenyan coast.

BACKGROUND: Anopheles gambiae s.l. and An. funestus are important vectors of malaria and bancroftian filariasis, which occur as co-endemic infections along the Kenyan Coast. However, little is known about the occurrence and prevalence of concomitant infections of the two diseases in mosquito and human populations in these areas. This study reports the prevalence of concomitant infections of Plasmodium falciparum and Wuchereria bancrofti in mosquito and human populations in Jilore and Shakahola villages in Malindi, Kenya. METHODS: Mosquitoes were sampled inside houses by pyrethrum spray sheet

The relationship between infection and disease in Wuchereria bancrofti infection in Ghana.

The relationship between infection and clinical disease in Wuchereria bancrofti infection was investigated in a community-based study in different endemic areas in Ghana. At the individual level, there was no association between acute adenolymphangitis and infection (microfilaraemia) status. There was a negative association between infection status and lymphoedema/elephantiasis, but a positive association with hydrocele; however, the intensity of infection was negatively associated with both elephantiasis and hydrocele. The community prevalence of infection was strongly associated with the pre

Clearance of Wuchereria bancrofti antigen after treatment with diethylcarbamazine or ivermectin.

The present study was undertaken to assess the relationship between microfilarial clearance and clearance of circulating filarial antigen from the blood of Wuchereria bancrofti-infected persons following chemotherapy with either diethylcarbamazine or ivermectin. Patients received either 12 weekly doses of 6 mg/kg of diethylcarbamazine (DEC), a single dose of 6 mg/kg of DEC, a single dose of 420 microg/kg of ivermectin, or 20 microg/kg of ivermectin, followed by 6 mg/kg of DEC five days later. Microfilarial clearance was marked in all groups, but was significantly less in the single-dose DEC. I

Microfilariae of Wuchereria bancrofti in a lymph node aspirate. A case report.

Microfilariae of Wuchereria bancrofti were detected by fine needle aspiration biopsy (FNAB) in an axillary lymph node from a 27-year-old female. The patient also had intestinal tuberculosis. A routine hemogram did not show any eosinophilia, and three consecutive night blood samples also were negative for microfilariae. This was an early case of filariasis without microfilaremia and was detected by FNAB of the lymph node. The association of filariasis and tuberculosis was purely coincidental.

Release of prostaglandin E2 by microfilariae of Wuchereria bancrofti and Brugia malayi.

To elucidate the local release of immunomodulatory prostaglandins by intravascular filarial parasites, the formation of prostaglandin E2 (PGE2) was examined in individual microfilariae of Wuchereria bancrofti and Brugia malayi. Following incubation of living microfilariae immobilized in an agar matrix, prostaglandins released by the parasites were fixed by carbodiimide and localized by indirect immunofluorescence. Prostaglandin E2 was specifically detected around the entire surface of microfilariae with anti-PGE2 antiserum, but not with control nonimmune or PGE2 affinity-immunoadsorbed antiser

[Transplacental passage of Wuchereria bancrofti microfilariae in newborns of microfilaremic mothers].

The occurrence of circulating Wuchereria bancrofti microfilariae was investigated in 304 pregnant women from the "Unidade Mista Pof. Barros Lima" and "Hospital das Clínicas/UFPE". The filtration technique was employed to detect microfilaraemia during pregnancy, and 13 women were found to be positive (4.2%). The investigation of microfilariae in the cord blood of their children was negative (6/13), and also in venous blood samples collected up to 72 hours after birth and with 6 months of life. Milk samples from these mothers also had no microfilariae. The authors suggest that the occurrence of

Ivermectin for the treatment of Wuchereria bancrofti filariasis. Efficacy and adverse reactions.

Ivermectin treatment was evaluated for efficacy and side effects in 40 patients in South India who had microfilaremia and bancroftian filariasis. Ivermectin was administered once orally at four dose levels (range, 25 to 200 micrograms/kg), and at each it was found to be completely effective in clearing blood microfilariae within five to 12 days. In most patients, microfilariae reappeared by three months; by six months the levels averaged 14% to 32% of pretreatment values in the four study groups, and all groups showed equivalent efficacy. Detailed monitoring identified some side effects in alm

Parasitological, serological, and clinical studies of Wuchereria bancrofti in Limbe, Haiti.

A survey for Wuchereria bancrofti in Limbe, Haiti (est. pop. = 10,500) revealed that 17% (231/1,450) had a patent infection. Nearly half of those surveyed harbored fewer than 10 microfilariae (mf) per 20 mm3 of finger-prick blood; the median mf density for females and males was 12.4 and 9.5, respectively. Parasitemias occurred as early as age 4. Antibody titers greater than or equal to 1:20 against adult D. viteae antigen were observed in 38% of microfilaremic individuals and in 29% of amicrofilaremic individuals. Peak antibody responsiveness (40%) was observed between 5 and 9 years of age. In

Preliminary screening of antifilarial activity of levamisole and amodiaquine on Wuchereria bancrofti.

A single dose of levamisole of 3 mg/kg had a marked microfilaricidal effect in some persons harbouring Wuchereria bancrofti, but there was considerable variation in the response. In some cases blood examinations for microfilariae remained negative or at a low level for several months. Persons with high pretreatment microfilaraemias responded less to therapy than did those with low counts. The main side-reaction to levamisole was fever, which began six to eight hours after treatment. Combined DEC-levamisole therapy appears to have no advantage over the use of DEC alone. Ultrastructure of the rectum of infective-stage Wuchereria bancrofti (Nematoda: Filarioidea).

The authors have examined the ultrastructure of the rectum of infective-stage Wuchereria bancrofti by transmission electron microscopy. Our observations show that the rectum is divided into anterior and posterior segments. The cells of the anterior rectum appear to be derived from the microfilarial R (rectal) cells described by other authors. In both stages, these cells show voluminous nuclei, abundant mitochondria, and small cytoplasmic processes which contain fibrillar components. Amorphous material associated with these processes appears throughout the larval rectum and may protrude from th
