Toxicity of free radicals to mesothelial cells and peritoneal membrane.

We studied the toxicity of free radicals to human mesothelial cells in vitro and to the peritoneal membrane of rats during peritoneal dialysis. Free radicals cause damage to mesothelial cells as measured by release of cytosolic markers such as 86Rb and lactate dehydrogenase. Vitamin E neutralized the toxic effect of free radicals in vitro. Human mesothelial cells exposed over 6 h to a mixture of essential and nonessential amino acids in medium are more vulnerable to the cytotoxic effect of free radicals than control cells exposed to medium alone. Cells exposed previously to glucose or glycerol

Mechanisms of inactivation of molybdoenzymes by cyanide.

The reduced forms of xanthine oxidase, xanthine dehydrogenase, aldehyde oxidase, and sulfite oxidase are inactivated by cyanide. Following gel filtration to remove excess of reductant and cyanide, the isolated enzymes remain inactive. Thiocyanate, a product of inactivation of the oxidized forms of the xanthine- and aldehyde-oxidizing enzymes by cyanide, is not released during inactivation of the reduced enzymes. Studies with [14C]cyanide show that, while stoichiometric binding is required for the onset of inactivation, its continued binding is not essential to maintenance of the inactivated st

Chronic kidney disease and reduced renal COX2 expression in xanthinuria: a case-control study.

BACKGROUND: Xanthinuria is a rare inherited metabolic disorder caused by xanthine dehydrogenase (XDH) deficiency, leading to excessive urinary xanthine excretion. It is associated with kidney stones and, in exceptional cases, renal failure. To date, no kidney biopsy findings have been reported in cases of xanthinuria. We present the first documented biopsy from such a patient, revealing a reduction in cyclooxygenase-2 (COX2) expression. CASE PRESENTATION: A 38-year-old man of Afghan origin presented with flank pain and a staghorn calculus in the right kidney. Laboratory tests revealed impaire

[Role of metabolism pathway interaction of formaldehyde and nitric oxide in the mechanism of their toxic effect. 2. Toxic effect of nitric oxide].

Toxic properties of NO in organism are realized under its hyperproduction and inhibition of the system of anti-oxidant protection as a result of complex chemical transformations, the transient metals, oxygen, superoxide and other radicals being their main participant. Here direct paths (through formation of nitrosil complexes with the gem and nongem iron) of the toxic action of NO and the path mediated by active forms of nitrogen are found, which disturb various biomolecules and subcellular component through the reactions of S- and N-nitrozation, nitration, oxidation, desamination and other re

Peroxisomes in wild-type and rosy mutant Drosophila melanogaster.

This study shows that peroxisomes are abundant in the Malpighian tubule and gut of wild-type Oregon R Drosophila melanogaster and that the peroxisomal population of the rosy-506 eye-color mutant differs from that of the wild type. Catalase activity in wild-type flies is demonstrable in bodies of appearance and centrifugal behavior comparable to the peroxisomes of vertebrate tissues. Xanthine oxidase (xanthine:oxygen oxidoreductase, EC 1.1.3.22) activity of the Malpighian tubule of wild-type flies is demonstrable cytochemically in bodies like those containing catalase. The rosy-506 mutant flies

Paralogous guanine deaminases acquired from bacteria by horizontal gene transfer promote purine homeostasis in Caenorhabditis elegans.

Disruptions in purine metabolism contribute to a range of human diseases, from rare genetic disorders such as Lesch-Nyhan syndrome and xanthinuria to common conditions including gout and cancer. To better understand the metabolic networks that regulate purine homeostasis, we developed a Caenorhabditis elegans model of xanthine dehydrogenase ( xdh-1 ) deficiency. Remarkably, xdh-1 mutant animals form rare xanthine stones, recapitulating a hallmark of human xanthinuria. To uncover genetic regulators of purine homeostasis, we performed a forward genetic screen for mutations that exacerbate xanthi

Reactive oxygen species in human inner ear perilymph.

CONCLUSIONS: The results reported here provide the first evidence of the production of superoxide, a biologically relevant reactive oxygen species (ROS), in human inner ear perilymph (hIP) in pathological conditions, by the activity of the xanthine dehydrogenase/xanthine oxidase (XA/XO) enzyme system. OBJECTIVE: To investigate the presence of ROS in hIP. METHODS: Since damage and apoptosis of inner ear hair cells may occur as a result of ROS-mediated injury, we investigated the presence and production of ROS in 105 hIP samples; 98 collected from patients affected by profound sensorineural he

Injury to rat hearts produced by an exogenous free radical generating system. Study into the role of arachidonic acid and eicosanoids.

This study was designed to evaluate the effect of an exogenous free radical generating system consisting of purine plus xanthine oxidase on the isolated rat heart and in particular to assess the possible contribution of arachidonic acid or its metabolites to toxicity produced by this drug combination. Purine plus xanthine oxidase produced a time-dependent depression in cardiac contractility which was associated with stimulated release of lactate dehydrogenase (LDH). Electron microscopic analysis revealed a distinct separation of the glycocalyx from the sarcolemmal membrane with no apparent int

Purine and pyrimidine metabolites in children's urine.

Various enzyme defects in the metabolic pathways of purines and pyrimidines are known, which result in different diseases occurring in children. They mainly affect kidney function, central nervous system, immunological and blood system. For example, complete deficiency of HPRT (hypoxanthine-guanine-phosphoribosyl-transferase) causes the Lesch Nyhan syndrome, which is characterized by hyperuricemia, mental retardation, choreoathetosis and compulsive self-mutilation. XDH deficiency (xanthine-dehydrogenase) causes in arthropathia and myopathia. For screening for these and other enzyme defects, ur

Discovery of a Gut Bacterial Pathway for Ergothioneine Catabolism.

Ergothioneine is a diet-derived micronutrient for humans. However, enzymes involved in the catabolism of ergothioneine in human gut bacteria have not yet been identified. Herein, we characterize a sulfidogenic pathway for gut bacterial catabolism of this micronutrient, which involves an unprecedented reductive desulfurization reaction catalyzed by members of the xanthine oxidoreductase family (XOR), a class of molybdenum-containing flavoproteins. Notably, this is the first C-S bond cleavage reaction known to be catalyzed by XORs. Evidence for operation of this pathway was gained through in vit

Elucidating the efficacy and mechanism of Ginkgo biloba extract in treating hyperuricemic nephropathy using network pharmacology, metabolomics, gut microbiota analysis, and experimental validation.

BACKGROUND: Although Ginkgo biloba extract (GBE) has been used to treat nephropathy, its effects and underlying mechanisms in hyperuricemic nephropathy (HN) remain unclear. METHODS: Based on biochemical parameters, inflammatory markers, and renal histopathological morphology, this study employed a rat HN model to examine the impact of GBE. Subsequently, an integrated strategy-combining network pharmacology, renal metabolomics, and gut microbiota profiling-was implemented to probe the mechanistic basis. Finally, Molecular dynamics (MD), molecular dynamic simulations, polymerase chain reaction

Goose astrovirus infection affects uric acid production and excretion in goslings.

In 2018, a new goose astrovirus (GAstrV) was reported in China, which causes 2 to 20% deaths in 4- to 16-day-old goslings causing great damages to the livestock industry. Gout is the typical feature of GAstrV infection in goslings. However, the mechanism of gout formation remains unclear. In the present study, 2-day-old goslings were infected intramuscularly with GAstrV for 14 D. One quarter of the infected goslings died, and typical gout pathological changes were found in the dead infected goslings. Pathological changes were observed in the morphology of the kidney and liver, such as degenera

Bucillamine prevents myocardial reperfusion injury.

Injury during reperfusion can partially offset the benefit of relief of ischemia in myocardial infarctions rapidly treated with thrombolytic drugs or angioplasty. We assessed whether bucillamine (N-[2-mercapto-2-methylpropionyl]-L-cysteine) is potentially useful to treat myocardial reperfusion injury. Bucillamine is a potent sulfhydryl donor not previously tested as a treatment of reperfusion injury. Cardiac myocytes were exposed to hydrogen peroxide or a xanthine/xanthine oxidase system resulting in injury-induced release of lactate dehydrogenase. Bucillamine (125-500 microM) prevented lactat

Discovery of novel 1,2,4-triazole derivatives as xanthine oxidoreductase inhibitors with hypouricemic effects.

Xanthine oxidoreductase (XOR) is a clinically validated target for the treatment of hyperuricemia and gout. A series of novel 1,2,4-triazoles were identified as potent XO inhibitors via a fused-pharmacophore strategy based on the interaction modes of febuxostat and topiroxostat. Among them, compound 7i showed an IC50 value of 0.20 nM against XOR, which was superior to febuxostat and topiroxostat. Furthermore, 7i exhibited significant hypouricemic and serum XOR inhibitory effects in potassium oxonate induced hyperuricemia mouse models. A single-dose toxicity assessment of 7i showed no noticeabl

A sensitive method to assay the xanthine oxidase activity in primary cultures of cerebellar granule cells.

Since xanthine oxidase (XO, Xanthine:oxidoreductase, E.C.1.2.3.22) is a key enzyme in reactive oxygen specie formation which plays a major role in cell oxidative stress, the availability of a sensitive and simple assay useful to detect its activity in monolayer cell cultures is worthwhile. In order to achieve this, we developed a method in which the conversion of pterine into isoxanthopterin is monitored fluorimetrically. Temperature assay was 50 degrees C. The activity of XO was detected in cerebellar granule cells exposed to glutamate. Since XO is formed from protease-dependent xanthine dehy

Antioxidant system of Litomosoides carinii and Setaria cervi: effect of a macrofilaricidal agent.

Filarial parasites, Litomosoides carinii and Setaria cervi, showed great susceptibility to the oxidants generated in vitro by the xanthine/xanthine-oxidase system. In order to counteract such injurious effects, both the filariids possessed an active antioxidant enzymes system. Superoxide dismutase, catalase and glutathione peroxidase were detected in appreciable amounts but glutathione reductase and glucose-6-phosphate dehydrogenase in very low quantities. The former three enzymes were also found to be released by the parasites into the ambient medium. The released enzymes may be responsible f

Cardiac injury and purine metabolic remodeling in Carassius auratus under environmentally relevant carbonate alkalinity exposure.

With the intensification of global warming and human activities, inland freshwaters are increasingly salinized and alkalized, often with carbonate alkalinity predominating. Such conditions disrupt acid-base and ion homeostasis in freshwater fish and may induce systemic energy metabolic reprogramming. However, the effects of carbonate alkalinity stress on the heart, an organ with high energy demands, remain poorly understood. Therefore, Carassius auratus (C. auratus), which exhibits relatively high tolerance to carbonate alkalinity, was selected as the experimental species in the present study.
