Xanthine oxidase converts nitric oxide to nitroxyl that inactivates the enzyme.

Xanthine oxidase (XO) was found to convert nitric oxide (NO* ) released from spermine-NONOate to nitroxyl (HNO), the one-electron reduction product of NO*, in the presence of its substrate hypoxanthine under anaerobic conditions. Under these conditions, XO lost its activity. Upon aerobic incubation of XO with its substrate, neither conversion of NO* to HNO nor inactivation of the enzyme was observed. Angeli's salt (an HNO generator) or synthetic peroxynitrite inactivated XO at low concentrations, whereas high concentrations of diethylamine-NONOate (an NO* donor) and SIN-1 (which generates pero

Gastric mucosal injury in the rat. Role of iron and xanthine oxidase.

Recent studies have implicated oxygen free radicals in ischemia-reperfusion injury to the gastric mucosa. The aims of the present study were to test the hypothesis that the enzyme xanthine oxidase is the source of the oxygen radicals in the ischemic stomach and determine the importance of the iron-catalyzed Haber-Weiss reaction in generating the cytotoxic oxygen radicals. Gastric mucosal clearance of 51Cr-labeled red blood cells was measured during a 30-min control period, a 30-min ischemic period (hemorrhage to 25 mmHg arterial pressure), and a 60-80-min reperfusion period (reinfusion of shed

Inhibition of xanthine oxidase protects against heatstroke-related acute kidney injury.

BACKGROUND: Heatstroke-induced acute kidney injury (HS-AKI) is a critical type of visceral injury. This study aimed to explore whether xanthine oxidase (XOD) promotes the progression of HS-AKI and whether febuxostat (Feb) attenuates HS-AKI through the inhibition of oxidative stress. MATERIALS AND METHODS: Both in vitro (HK-2 cells) and in vivo (a mouse model) approaches were used to evaluate the role of XOD in HS-AKI. In vitro, genetic gain-/loss-of-function (mediated by an XDH overexpression plasmid/siRNA) and pharmacologic intervention (febuxostat) were performed in HK-2 cells exposed to hy

Xanthine oxidase mediates chronic stress-induced cerebrovascular dysfunction and cognitive impairment.

Xanthine oxidase (XO) mediates vascular function. Chronic stress impairs cerebrovascular function and increases the risk of stroke and cognitive decline. Our study determined the role of XO on stress-induced cerebrovascular dysfunction and cognitive decline. We measured middle cerebral artery (MCA) function, free radical formation, and working memory in 6-month-old C57BL/6 mice who underwent 8 weeks of control conditions or unpredictable chronic mild stress (UCMS) with or without febuxostat (50 mg/L), a XO inhibitor. UCMS mice had an impaired MCA dilation to acetylcholine vs. controls (p < 0.0

Xanthine oxidase activity in thiopurine curative Chinese inflammatory bowel disease patients.

Xanthine oxidase (XO) competes with thiopurine S-methyltransferase (TPMT) and hypoxanthine guanine phosphoribosyltransferase (HPRT) to metabolize azathioprine (AZA)/6-mercaptopurine (6-MP) in vivo. A retrospective investigation was performed to detect the activity of XO in thiopurine curative Chinese inflammatory bowel disease (IBD) patients. We also evaluated whether a relationship between XO activity and incidence of thiopurine-induced adverse effects (AEs) existed. Clinical data and blood samples were collected from 140 IBD patients before receiving AZA/6-MP therapy, and the erythrocyte XO

Bis-thiobarbiturates as Promising Xanthine Oxidase Inhibitors: Synthesis and Biological Evaluation.

Xanthine oxidase (XO) is the enzyme responsible for the conversion of endogenous purines into uric acid. Therefore, this enzyme has been associated with pathological conditions caused by hyperuricemia, such as the disease commonly known as gout. Barbiturates and their congeners thiobarbiturates represent a class of heterocyclic drugs capable of influencing neurotransmission. However, in recent years a very large group of potential pharmaceutical and medicinal applications have been related to their structure. This great diversity of biological activities is directly linked to the enormous oppo

Schiff base transition metal complexes as novel inhibitors of xanthine oxidase.

Twenty transition metal complexes with Schiff bases were evaluated for their inhibitory activities on xanthine oxidase (XO), of which 11 were newly synthesized and characterized by X-ray single crystal diffraction. It was found that 9 of the 20 complexes showed potent inhibitory activities against XO near to the standard inhibitor allopurinol. The cadmium(II) complex (8) had the most potent inhibitory activity with the IC(50) value of 2.16 microM. Relationships between the structures and the activities showed that the ligands and the metal ions influenced the inhibitory activities. The XO inhi

Exploring the role of xanthine oxidase and aldehyde oxidase in metabolic dysfunction-associated steatotic liver disease (MASLD).

Globally, metabolic dysfunction-associated steatotic liver disease (MASLD), formerly termed as non-alcoholic fatty liver disease (NAFLD), has been affecting millions of people. It includes a spectrum of liver conditions, ranging from simple fat accumulation (steatosis) to advanced stages such as fibrosis, cirrhosis and hepatocellular carcinoma (HCC). Oxidative stress has been identified as a major contributing factor in driving disease progression. Enzymes such as xanthine oxidase (XO) and aldehyde oxidase (AO) are significant contributors of reactive oxygen species (ROS), which in turn promot

Effects of catechins, resveratrol, silymarin components and some of their conjugates on xanthine oxidase-catalyzed xanthine and 6-mercaptopurine oxidation.

BACKGROUND: Over the past two decades, the global incidence of gout has markedly increased, affecting people worldwide. Considering the side effects of xanthine oxidase (XO) inhibitor drugs (e.g. allopurinol and febuxostat) used in the treatment of hyperuricemia and gout, the potential application of phytochemicals has been widely studied. In addition, XO also takes part in the elimination of certain drugs, including 6-mercaptopurine. In the current explorative study, we aimed to examine the potential effects of tea catechins, resveratrol, silymarin flavonolignans and some of their conjugated

Inhibition of xanthine oxidase-catalyzed xanthine and 6-mercaptopurine oxidation by luteolin, naringenin, myricetin, ampelopsin and their conjugated metabolites.

Luteolin, naringenin, myricetin, and ampelopsin are abundant flavonoids in nature, and several dietary supplements also contain them at very high doses. After the peroral intake, flavonoids go through extensive presystemic biotransformation; therefore, typically their sulfate/glucuronic acid conjugates reach high concentrations in the circulation. Xanthine oxidase (XO) enzyme is involved in uric acid production, and it also takes part in the elimination of certain drugs (e.g., 6-mercaptopurine). The inhibitory effects of flavonoid aglycones on XO have been widely studied; however, only limited

Anatomo-pathological findings in a case of combined deficiency of sulphite oxidase and xanthine oxidase with a defect of molybdenum cofactor.

A case of combined deficiency of sulphite-oxidase and xanthine-oxidase with a defect of the molybdenum cofactor, which is vital to the activity of sulphite-, xanthine- and aldehyde-oxidase, is reported here. Seven cases of combined deficiencies have been described with regard to both clinical and laboratory findings. The clinical, laboratory and anatomo-pathological features and, in particular, the central nervous system lesions of the present case correspond exactly to those in the case described Rosenblum in which an isolated deficiency in sulphite-oxidase was present. As the cerebral altera

Bovine xanthine oxidase electrocatalysis: substrate oxidation and its role in nitrite reduction.

Xanthine oxidase plays a central role in purine catabolism in animals, plants and bacteria where it oxidises hypoxanthine to xanthine and xanthine to uric acid. In this work we have immobilised bovine xanthine oxidase on a glassy carbon electrode within a glutaraldehyde film and demonstrated hypoxanthine and xanthine oxidase electrocatalysis using mediator methylene blue as a mediator. The role of xanthine oxidase in the reduction of nitrite to nitric oxide has been probed in two ways. Using the electrochemically reduced mediator methyl viologen, nitrite reduction by xanthine oxidase is compli

Xanthine oxidase inhibitory and anti-hyperuricemic properties of sulfonate derivatives of naringenin.

Xanthine oxidase (XO) facilitates oxidation of hypoxanthine to xanthine and then to uric acid, potentially leading to hyperuricemia when production of excessive uric acid. In the present study, naringenin-3'-sulfonate sodium and naringenin-3',6-disulfonate sodium were synthesized by chemical modification of naringenin, and evaluated for XO inhibitory activities in vitro. The results indicated that naringenin-3'-sulfonate sodium exhibited significantly inhibitory activities compared to naringenin and naringenin-3',6-disulfonate sodium. Further inhibitory kinetics, fluorescence spectra, circular

Advances in Xanthine Oxidase Inhibition: A Review of Potential Bioactive Synthetic Compounds.

Hyperuricemia and gout are conditions that continue to present significant therapeutic challenges, mainly due to limitations of current treatments which may cause adverse effects such as hypersensitivity, hepatotoxicity, and cardiovascular risks. Additionally, the development of new drugs faces obstacles including toxicity, low bioavailability, and potential drug interactions. In this context, the search for new synthetic xanthine oxidase inhibitors is essential to improve treatment efficacy, selectivity, and safety. New molecules can not only prolong the duration of therapeutic effects but al

Acute cardiovascular events after discontinuation of xanthine oxidase inhibitors: a cohort study.

BACKGROUND: Xanthine oxidase inhibitors (XOis) are commonly used to treat gout and hyperuricemia. Beyond urate-lowering effects, XOis may influence cardiovascular outcomes via oxidative stress pathways. Prior evidence, including post hoc analyses of the CARES trial, suggests increased mortality after XOi discontinuation, raising concern for a potential "withdrawal syndrome." However, evidence from real-world outpatient populations is limited. OBJECTIVE: This study aims to evaluate whether the recent discontinuation of XOi therapy is associated with an increased risk of acute cardiovascular ev
