Pathogenomic Insights into Xanthomonas oryzae pv. oryzae's Resistome, Virulome, and Diversity for Improved Rice Blight Management.

Oryza sativa (rice) is a major staple food targeted for increased production to achieve food security. However, increased production is threatened by several biotic and abiotic factors, of which bacterial blight disease caused by Xanthomonas oryzae pathovar oryzae is severe. Developing effective control strategies requires an up-to-date understanding of its pathogenomics. This study analyzes the genomes of 30 X. oryzae strains collected from rice-producing regions across five continents to identify genetic elements critical for its pathogenicity and adaptability and for an intraspecific divers

Risks Posed by Intercrops and Weeds as Alternative Hosts to Xanthomonas campestris pv. musacearum in Banana Fields.

Alternative host plants are important in the survival and perpetuation of several crop pathogens and have been suspected to play a role in the survival of Xanthomonas campestris pv. musacearum (Xcm) and perpetuation of Xanthomonas wilt (XW) disease of banana and enset. This study determined the potential risk posed by two weeds (Canna spp. and wild sorghum) and common banana intercrops (maize, millet, sorghum, taro, and sugarcane) as alternative hosts to Xcm. The study employed screenhouse experiments, laboratory procedures and diagnosis of banana fields in XW-affected landscapes. Typical XW s

Screening and characterization of Xanthomonas oryzae pv. oryzae strains with resistance to pheazine-1-carboxylic acid.

Xanthomonas oryzae pv. oryzae (Xoo) causes bacterial leaf blight (BLB) and can be effectively controlled by phenazine-1-carboxylic acid (PCA), an antibiotic secreted by Pseudomonas spp. PCA resistance in Xoo was investigated in this research. Only four PCA-resistant strains were obtained by extensive screening, and the resistance was genetically stable in only one of them (P4). P4 was also resistant to phenazine and 1-hydroxyphezine but not to captan, bismerthiazol, or streptomycin. The following were reduced in P4 relative to the parental wild type: growth, virulence, EPS production, extracel

Tandem receiver domain response regulator DrdR modulates swarming by controlling PilB/PilT activity in Xanthomonas campestris.

Like many other bacterial organisms, Xanthomonas campestris pv. campestris (Xcc) employs two motility appendages, a single polar flagellum and type IV pili, for sensing environmental cues and traversing liquids and solid surfaces. However, compared to the flagellum, the molecular mechanisms underlying the function and regulation of the pilus in Xcc and other xanthomonads are not fully understood. In this study, we demonstrated that a special conserved orphan response regulator DrdR, which contains double receiver (REC) domains but lacks an output domain, modulates the activities of motor prote

Isolation of a complementary bacteriophage from a phage-resistant mutant expands control strategies against Xanthomonas campestris.

Bacterial resistance to bacteriophages represents a major limitation for the durable use of phage-based biocontrol strategies in agriculture. Here, we used a phage-resistance derivative of Xanthomonas campestris pv. campestris (Xcc) as a selective host to isolate an additional phage with activity against resistant bacteria. A spontaneous mutant, designated Xcc 8004R, was isolated following exposure of Xcc 8004 to phage X1. Xcc 8004R exhibited a growth profile comparable to that of the wild-type strain, but differed in colony appearance, X1 adsorption efficiency, and motility phenotypes. Whole-

Xanthomonas phaseoli pv. Manihotis effector XopAG impairs cassava immunity by hijacking MeHSP26 to suppress flavonoid biosynthesis.

Cassava bacterial blight (CBB), caused by Xanthomonas phaseoli pv. manihotis (Xpm), is a major disease constraining cassava production. Although overexpression of the Xpm effector XopAG enhances Arabidopsis susceptibility to Pseudomonas syringae pv. tomato DC3000, its virulence mechanism in cassava remains unclear. The study showed that deletion of XopAG did not affect morphology or extracellular enzyme production of Xpm but significantly attenuated its virulence on cassava. Quantitative proteomic analysis revealed that 267 differentially accumulated proteins (DAPs) at 6 h post-inoculation (hp

Stringent response regulators (p)ppGpp and DksA positively regulate virulence and host adaptation of Xanthomonas citri.

The bacterial stringent response is a response to nutrition deprivation and other stress conditions. In Gram-negative bacteria, this process is mediated by the small signal molecules guanosine pentaphosphate pppGpp and guanosine tetraphosphate ppGpp (collectively referred to as (p)ppGpp), and the RNA polymerase-binding transcription factor DksA. The (p)ppGpp synthetase RelA and the bifunctional (p)ppGpp synthase/hydrolase SpoT are responsible for cellular (p)ppGpp levels. Here, we investigated the roles of DksA and (p)ppGpp in the virulence traits of Xanthomonas citri subsp. citri (Xcc), the c

Copper chloride induces antioxidant gene expression but reduces ability to mediate H2O2 toxicity in Xanthomonas campestris.

Copper (Cu)-based biocides are currently used as control measures for both fungal and bacterial diseases in agricultural fields. In this communication, we show that exposure of the bacterial plant pathogen Xanthomonas campestris to nonlethal concentrations of Cu(2+) ions (75 µM) enhanced expression of genes in OxyR, OhrR and IscR regulons. High levels of catalase, Ohr peroxidase and superoxide dismutase diminished Cu(2+)-induced gene expression, suggesting that the production of hydrogen peroxide (H2O2) and organic hydroperoxides is responsible for Cu(2+)-induced gene expression. Despite high

First Report in Mali of Xanthomonas citri pv. mangiferaeindicae Causing Mango Bacterial Canker on Mangifera indica.

Bacterial canker (or black spot) of mango caused by Xanthomonas citri pv. mangiferaeindicae is an important disease in tropical and subtropical areas (1). X. citri pv. mangiferaeindicae can cause severe infection in a wide range of mango cultivars and induces raised, angular, black leaf lesions, sometimes with a chlorotic halo. Severe leaf infection may result in abscission. Fruit symptoms appear as small, water-soaked spots on the lenticels that later become star shaped, erumpent, and exude an infectious gum. Often, a "tear stain" infection pattern is observed on the fruit. Severe fruit infec

Septicemia due to Xanthomonas species and non-aeruginosa Pseudomonas species: increasing incidence of catheter-related infections.

We reviewed 149 episodes of septicemia caused by X. maltophilia and Pseudomonas spp. occurring over a 15-year period. The incidence of septicemia caused by these organisms increased in recent years and was most frequently associated with central venous catheterization. These infections were occasionally complicated by pneumonia or endocarditis, which was often fatal. Although the survival rate was superior to that seen with septicemia caused by other gram-negative organisms, recurrence of infection was significantly more frequent. Removal of central venous catheters is an essential component o

Design, synthesis, and multitarget anti-Xanthomonas oryzae pv. oryzae mechanism of indole-3-substituted aryl urea derivatives.

BACKGROUND: Bacterial leaf blight (BLB), caused by Xanthomonas oryzae pv. oryzae (Xoo), severely threatens rice production and global food security. Current commercial bactericides such as thiadiazole copper (TC) and bismerthiazol (BT) are losing efficacy due to widespread Xoo resistance, while research on structural modification and activity optimization of indole derivatives for Xoo control remains scarce, highlighting an urgent need for novel, safe, and effective anti-Xoo agents and filling this research gap. RESULTS: Twenty-nine indole derivatives (A1-A29) were synthesized from 3-carboxyi

Antimicrobial Activity of LysX and LysP Endolysins Against Pseudomonas syringae pv. syringae and Xanthomonas arboricola pv. juglandis.

Pseudomonas syringae pv. syringae and Xanthomonas arboricola pv. juglandis are the causal agents of bacterial canker in cherry and walnut blight, respectively, which cause significant production losses worldwide. These diseases have traditionally been controlled by copper-based agrochemicals and, more recently, antibiotics. However, the prolonged use of these compounds has led to the emergence of resistant bacterial strains. The search for new, efficient, and environmentally friendly biocontrol alternatives has intensified. Phages are promising candidates due to their ability to specifically i

The Xanthomonas axonopodis pv. glycines Cellulase EngXCA Governs Dual Roles in Virulence and Innate Immunity in Soybean.

The Xanthomonas axonopodis pv. glycines (Xag) cellulase EngXCA exhibits a dual functionality in the soybean pathosystem. Deletion of engXCA significantly attenuated bacterial growth and symptom severity, a phenotype that was fully restored by genetic complementation. Plants infected with ΔengXCA exhibited attenuated defense responses, including reduced H2O2 accumulation, callose deposition, and PCD, indicating that EngXCA acts as an immunity elicitor. Soybeans infected with ΔengXCAΔhrcC elicited weaker immune responses than those infected with ΔhrcC alone, confirming that EngXCA functions inde

New Axially Chiral Molecular Scaffolds with Antibacterial Activities against Xanthomonas oryzae pv. oryzae for Protection of Rice.

A new class of axially chiral thiazine molecules were constructed and showed promising antibacterial activities against the plant pathogen Xanthomonas oryzae pv. oryzae (Xoo). The axial chiralities of these compounds (R- or S-atropisomer) showed clear impacts on the in vitro inhibitory activities against Xoo. An optimal molecule of this class with the (S)-axially chiral configuration was identified to exhibit inhibitory activity against Xoo with an EC50 value of 4.18 μg/mL. This inhibition efficiency is superior to that of two commercial antibacterial agrochemicals, thiodiazole-copper and bism

Metabolic Response of Strawberry (Fragaria x ananassa) Leaves Exposed to the Angular Leaf Spot Bacterium (Xanthomonas fragariae).

Plants have evolved various defense mechanisms against biotic stress. The most common mechanism involves the production of metabolites that act as defense compounds. Bacterial angular leaf spot disease (Xanthomonas fragariae) of the strawberry (Fragaria x ananassa) has become increasingly destructive to strawberry leaves and plant production. In this study, we examined metabolic changes associated with the establishment of long-term bacterial disease stress using UPLC-QTOF mass spectrometry. Infected leaves showed decreased levels of gallic acid derivatives and ellagitannins, which are related

Opposite functions of a rice mitogen-activated protein kinase during the process of resistance against Xanthomonas oryzae.

The pathogen-induced plant defense signaling network consists of multiple components, although only some of them are characterized. Most of the known components function either as activators or repressors in host-pathogen interactions. Here we report that a mitogen-activated protein kinase, OsMPK6, functions both as an activator and a repressor in rice resistance against Xanthomonas oryzae pv. oryzae (Xoo), the causal organism of bacterial blight disease. Activation of OsMPK6 resulted in the formation of lesion mimics and local resistance to Xoo, accompanied by the accumulation of salicylic ac

A-to-I mRNA Editing in a Ferric Siderophore Receptor Improves Competition for Iron in Xanthomonas oryzae pv. oryzicola.

Iron is an essential element for the growth and survival of pathogenic bacteria; however, it is not fully understood how bacteria sense and respond to iron deficiency or excess. In this study, we show that xfeA in Xanthomonas oryzae pv. oryzicola senses extracytoplasmic iron and changes the hydrogen bonding network of ligand channel domains by adenosine-to-inosine (A-to-I) RNA editing. The frequency of A-to-I RNA editing during iron-deficient conditions increased by 76.87%, which facilitated the passage of iron through the XfeA outer membrane channel. When bacteria were subjected to high iron
