Modulating the spatiotemporal dynamics of starch metabolism in postharvest sweetpotato using high voltage alternating electric field.

Although postharvest starch degradation was extensively studied, its initiation site and distribution patterns remain unclear. This study investigated the spatiotemporal dynamics of starch metabolism in sweetpotato tubers along vertical (top, middle, bottom) and radial directions (phloem, outer xylem, inner xylem) under 4 kV·m-1 high voltage alternating electric field (HVAEF) treatment stored at 13 °C for 8 weeks. Results illustrated that starch degradation was first detected in the middle region during early storage, and extended to the bottom and top, whereas HVAEF's inhibitory effect concen

Neutron Imaging to Reveal Phloem Function in Trees.

Understanding phloem function is key to understanding the regulation of plant function. The phloem distributes the main building blocks that support plant function and growth from the production sites to sites of consumption and storage. It also distributes important signaling molecules and interacts with the adjacent water-transport tissue, the xylem, to support hydraulic integrity of plants. Yet, studying phloem function is challenging because of the sensitivity of this tissue to mechanical stimulation that can lead to blockage of flow. Furthermore, its small size, adjacent location, and int

Water-carbon coordination in Taxodium distichum but not T. ascendens  under prolonged off-season submergence in the Three Gorges Reservoir riparian zone.

Prolonged winter submergence induced by reservoir impoundment poses severe physiological challenges to riparian trees, yet post-submergence recovery dynamics remain poorly understood. In this study, we investigated hydraulic function, xylem anatomy and non-structural carbohydrate (NSC) dynamics in two congeneric species, Taxodium ascendens and T. distichum, following a winter impoundment in the Three Gorges Reservoir. During the initial post-submergence exposure phase, we comparatively analysed stem xylem micro-anatomy, percentage loss of conductivity (PLC) and NSC allocation across the xylem

Diversity and Plasticity of Hydraulic Traits Across a Relative Moisture Supply Gradient: Results From Multiple Common Gardens.

Maintaining cellular hydration is essential for plant metabolic activity and carbon gain. Plants typically maintain hydration through efficiency of water transport networks, the ability to avoid network disruption, and retention of water via accumulation of osmotica. Given their global domain and their enduring presence across geological eras, plants have evolved with large variations in moisture supply across geographic and temporal space, notably via trait adaptation and plasticity. We quantified plastic and genetically fixed variation in 6 key hydraulic traits across 10 Eucalyptus species i

Cytoskeletal Regulation of Wood Formation and Stress Adaptation in Woody Plants.

The cytoskeleton regulates cell division, intracellular transport and cell-wall patterning, but its roles in secondary growth and long-term stress responses in woody plants remain poorly understood. We classify individual claims as direct functional evidence from woody species (Level I), correlative or functionally non-specific evidence from woody systems (Level II), mechanisms demonstrated only in non-woody models (Level III), or relationships untested in woody plants (Hypothesis). Direct functional evidence remains concentrated in Populus. In hybrid poplar, PagPCaP1a promotes salt-induced mi

Trehalose increases tomato drought tolerance, induces defenses, and increases resistance to bacterial wilt disease.

Ralstonia solanacearum causes bacterial wilt disease, leading to severe crop losses. Xylem sap from R. solanacearum-infected tomato is enriched in the disaccharide trehalose. Water-stressed plants also accumulate trehalose, which increases drought tolerance via abscisic acid (ABA) signaling. Because R. solanacearum-infected plants suffer reduced water flow, we hypothesized that bacterial wilt physiologically mimics drought stress, which trehalose could mitigate. We found that R. solanacearum-infected plants differentially expressed drought-associated genes, including those involved in ABA and

Functional ratios diverge across climates but converge within environments in 55 woody canopy plants.

Functional coordination among leaf, xylem, phloem and ray parenchyma governs the carbon-water economy of woody plants, yet the allocation principles and climatic drivers remain poorly explored. We hypothesized that species at lower altitudes and/or latitudes invest more in xylem and phloem per unit leaf area to compensate for higher transpirational losses, whereas species from higher altitudes and/or latitudes allocate proportionally more to ray parenchyma to enhance storage. Using canopy cranes across four Chinese forests, we quantified how leaf area, branch tissue cross-sectional areas, cons

Stiffness signatures along early stages of Xylella fastidiosa biofilm formation.

The pathogenicity of Xylella fastidiosa is associated with its systematic colonization of the plant xylem, forming bacterial biofilms. Mechanisms of bacterial transport among different xylem vessels, however, are not completely understood yet, but are strongly influenced by the presence of extracellular polymeric substances (EPS), which surrounds the assembly of cells forming the biofilm. In this work, we show quantitative measurements on the elastic properties of the system composed by EPS and bacterial cell. In order to investigate the mechanical properties of this system, force spectroscopy

Loss of conductance between mesophyll symplasm and intercellular air spaces explains nonstomatal control of transpiration.

The conventional assumption is that stomatal conductance ([Formula: see text]) dominates the regulation of water and carbon dioxide fluxes between leaves and the atmosphere. Here, a nanoreporter of water status at the mesophyll cell surface and local xylem within intact maize leaves documents significant undersaturation of water vapor in the outside-xylem zone (OXZ) and a large loss of conductance of this zone ([Formula: see text]) at moderate xylem water stress, without stomatal closure or turgor loss. The ratio of the resistances [Formula: see text] serves as a predictive phenotype of unders

Ectopic Vascular Induction in Arabidopsis Cotyledons for Sequential Analysis of Phloem Differentiation.

Vascular system is vital for the transport of water and nutrients as well as for providing mechanical support in many land plants. Plant transcription factors play a central role in regulating vascular development downstream of hormones and peptide signaling pathways. Particularly, cell culture systems have contributed to isolating such key transcription factors for xylem differentiation. However, there had been no efficient systems that can mimic phloem differentiation in the model plant Arabidopsis, preventing the identification of phloem-related transcription factors. We have recently estab

A tale of two pumps: Blue light and abscisic acid alter Arabidopsis leaf hydraulics via bundle sheath cell H+-ATPases.

The bundle sheath cell (BSC) layer tightly enveloping the xylem throughout the leaf is recognized as a major signal-perceiving "valve" in series with stomata, regulating leaf hydraulic conductance (Kleaf) and thereby radial water flow via the transpiring leaf. The BSC blue light (BL) signaling pathway increases Kleaf and the underlying BSC water permeability. Here, we explored the hypothesis that BSCs also harbor a Kleaf-downregulating signaling pathway related to the stress phytohormone abscisic acid (ABA). We employed fluorescence imaging of xylem sap in detached leaves and BSC protoplasts f

Mind the bubbles: achieving stable measurements of maximum hydraulic conductivity through woody plant samples.

The maximum specific hydraulic conductivity (k(max)) of a plant sample is a measure of the ability of a plants' vascular system to transport water and dissolved nutrients under optimum conditions. Precise measurements of k(max) are needed in comparative studies of hydraulic conductivity, as well as for measuring the formation and repair of xylem embolisms. Unstable measurements of k(max) are a common problem when measuring woody plant samples and it is commonly observed that k(max) declines from initially high values, especially when positive water pressure is used to flush out embolisms. This

Plant Acoustics for Evaluating Vase Life in Chrysanthemums: A Novel and Noninvasive Method.

Vase life is a key determinant of cut flower quality and market value. Conventional vase life assessment relies on visual inspection and physiological monitoring over several days to weeks, making it labor- and time-intensive. This study introduces a rapid and noninvasive approach using plant acoustics to assess postharvest vase-life-related variation in cut chrysanthemum flowers. Six chrysanthemum cultivars were grown under two supplemental lighting treatments (Hybrid and LED) and two planting densities (54 and 74 plants m-2). Acoustic monitoring was compared with optical microscopy for the a

Ralstonia solanacearum Alters Root Developmental Programmes in Auxin-Dependent and -Independent Manners.

Microbial pathogens and other parasites can modify the development of their hosts, either as a target or a side effect of their virulence activities. The plant-pathogenic bacterium Ralstonia solanacearum, causal agent of the devastating bacterial wilt disease, is a soilborne microbe that invades host plants through their roots and later proliferates in xylem vessels. In this work, we studied the early stages of R. solanacearum infection in the model plant Arabidopsis thaliana, using an in vitro infection system. In addition to the previously reported inhibition of primary root length and incre

Latex-less opium poppy: cause for less latex and reduced peduncle strength.

A genotype 'Sujata' developed earlier at CSIR-CIMAP from its parent 'Sampada' is considered to be the latex-less variety of Papaver somniferum. These two genotypes are contrasting in terms of latex and stem strength. Earlier we have carried out microarray analysis to identify differentially expressing genes from the capsules of the two genotypes. In this study, the peduncles of the two genotypes were compared for the anatomy revealing less number of laticifers in the cortex and vascular bundles. One of the important cell wall-related genes (for laccase) from the microarray analysis showing sig

Lignin macromolecular structural stability drives sapwood-heartwood color homogeneity in Populus deltoides.

Radial xylem maturation in woody plants typically involves programmed cell death (PCD) and accumulation of colored secondary metabolites, leading to distinct color convergent between sapwood and heartwood. In fast-growing P. deltoides, however, the xylem exhibits remarkable color convergence along the radial gradient. Through the integration of anatomical observations, chemical analyses, and advanced spectroscopy (FTIR, Raman, 2D-HSQC NMR), color convergence was analyzed from two coordinated regulatory features: (1) a non-condensed lignin structure characterized with an elevated syringyl/guaia

Wine-red lignin revisited, dirigent proteins, and native lignin macromolecular configuration.

The wine-red coloration of xylem in antisense cinnamyl alcohol dehydrogenase (CAD) down-regulated tobacco (Nicotiana tabacum) has long been reported as due to wine-red colored lignin. To investigate validity of the wine-red lignin report, both wild type (WT) and anti-sense CAD down-regulated tobacco stem xylem tissues were examined, with the red pigment found to be facilely removed by treatment with an 0.5 % HCl in MeOH solution at 4 °C. The red pigment in the xylem tissues was rapidly reconstituted in both pre-treated antisense CAD and WT xylem tissues by incubation with sinapyl aldehyde in M

Transcriptomic and metabolomic approaches elucidate the systemic response of wheat plants under waterlogging.

Extreme weather conditions lead to significant imbalances in crop productivity, which in turn affect food security. Flooding events cause serious problems for many crop species such as wheat. Although metabolic readjustments under flooding are important for plant regeneration, underlying processes remain poorly understood. Here, we investigated the systemic response of wheat to waterlogging using metabolomics and transcriptomics. A 12 d exposure to excess water triggered nutritional imbalances and disruption of metabolite synthesis and translocation, reflected by reductions in plant biomass an

Interplay of auxin, KANADI and Class III HD-ZIP transcription factors in vascular tissue formation.

Class III HD-ZIP and KANADI gene family members have complementary expression patterns in the vasculature and their gain-of-function and loss-of-function mutants have complementary vascular phenotypes. This suggests that members of the two gene families are involved in the establishment of the spatial arrangement of phloem, cambium and xylem. In this study, we have investigated the role of these two gene families in vascular tissue differentiation, in particular their interactions with the plant hormone auxin. We have analyzed the vasculature of plants that have altered expression levels of Cl

PalKCH1 and PalKCH2 are required for tension wood formation and gelatinous layer deposition in Populus.

Tension wood (TW) is a graviresponsive xylem tissue exclusive to angiosperm trees that counteracts gravitational stress. Its hallmark is a gelatinous layer (G-layer) in fiber secondary walls, rich in cellulose and low in lignin. Although kinesins mediate microtubule-microfilament crosstalk, their role in G-layer biogenesis during gravistimulation is unclear in woody angiosperms. We studied two kinesin paralogs, PalKCH1 and PalKCH2, in Populus alba var. pyramidalis during TW development. Both belong to the Group I KCH clade, possessing a kinesin motor domain and a calponin-homology (CH) domain.
