Respiratory and olfactory cytotoxicity of inhaled 2,3-pentanedione in Sprague-Dawley rats.

Flavorings-related lung disease is a potentially disabling disease of food industry workers associated with exposure to the α-diketone butter flavoring, diacetyl (2,3-butanedione). To investigate the hypothesis that another α-diketone flavoring, 2,3-pentanedione, would cause airway damage, rats that inhaled air, 2,3-pentanedione (112, 241, 318, or 354 ppm), or diacetyl (240 ppm) for 6 hours were sacrificed the following day. Rats inhaling 2,3-pentanedione developed necrotizing rhinitis, tracheitis, and bronchitis comparable to diacetyl-induced injury. To investigate delayed toxicity, additiona

Natural products for treatment of Plasmodium falciparum malaria: An integrated computational approach.

Malaria is a life-threatening infectious disease with an estimated 229 million cases in the year 2019 worldwide. Plasmodium falciparum 1-deoxy-d-xylulose-5-phosphate reductoisomerase (PfDXR) is one of the key enzymes in the biosynthetic pathway of isoprenoid, (required for parasite growth and survival) and considered as an attractive target for designing anti-malarial drugs. Fosmidomycin is an effective DXR inhibitor and has been proven effective and safe against P. falciparum in clinical trials. However, due to low bioavailability and inappropriate drug attributes, it is not a preferred optio

Top-Down Proteomics in the Assessment of Kidney Donor Quality: a novel approach to increased organ utilization.

Kidney transplantation faces a critical paradox: while thousands await organs, approximately 30% of potential deceased donor kidneys are discarded for various reasons, including subjective assessments due to the lack of an objective molecular biomarker of preservation quality. Here, we applied novel "top-down" proteoform imaging mass spectrometry across living donor (LD), deceased donor (brain death or cardiac death), and discarded human kidneys to quantify proteoforms correlating with post-transplant kidney function. This approach preserves post-translational modifications and splice variants

Low-dose l-carnitine supplementation attenuates hepatic lipid accumulation in chow-fed mice: Insights from integrated multi-omics analysis.

Most studies on l-carnitine and hepatic lipid metabolism have focused on deficiency or HFD-associated metabolic disorders, whereas its low-dose nutritional effects under standard chow-fed conditions remain unclear. In this study, standard chow-fed C57BL/6 J mice were orally administered low doses of l-carnitine for 35 days, and hepatic responses were evaluated using phenotypic, histological, metabolomic, transcriptomic, and proteomic analyses. l-carnitine supplementation reduced liver weight and hepatic lipid accumulation in a dose-associated manner, accompanied by decreased hepatocyte area an

Integrated transcriptomic and metabolomic analysis reveal the mechanism of citral production in Camphora officinarum Nees ex Wall leaves.

Camphora officinarum Nees ex Wall (C. officinarum), a citral-rich aromatic plant, is recognized as an unrivalled natural source of citral for spice production. Its large-scale cultivation in China via the sustainable coppice-rotation system has markedly revitalized rural economies. Nevertheless, the genetic basis driving its prolific citral accumulation remains elusive. We conducted an integrated transcriptomic and metabolomic analysis comparing three citral type accessions (C1, C2, C3) with a non-citral type control (C0). Metabolomic profiling identified 904 leaf metabolites, with terpenoids

β-Substitution and prodrug derivation leading to identification of fosmidomycin analogs with improved herbicidal activity.

BACKGROUND: The rapid evolution and spread of herbicide resistance poses a great challenge to the sustainability of modern agriculture and finding new herbicides based on novel modes of action (MOA) has become an urgent need. The enzyme 1-deoxy-d-xylulose-5-phosphate reductoisomerase (DXR) in the 2-C-methyl-d-erythritol 4-phosphate (MEP) pathway is considered an ideal novel target for developing new herbicides and therefore its natural inhibitor fosmidomycin (FOS) is an excellent starting point from which to design new herbicidal candidates. RESULTS: This work presents the design and synthesi

Soil functional microbial pathways related to GHGs emissions of coastal degraded mangroves-rice ecologies in Sundarban, India: A soil metagenomic approach.

Mangroves and their adjacent rice ecologies play a critical role in regulating greenhouse gas (GHGs) emissions, particularly in tropical coastal wetlands. This study assessed the impact of mangrove degradation on methane (CH4) and nitrous oxide (N2O) emissions by comparing two distinct ecosystems; degraded mangroves, and adjacent rice system in the Sundarban, India. Annual CH4 and N2O emissions were quantified and linked to soil microbial energy metabolism pathways; methanogenesis, methanotrophy, sulphur (S), and nitrogen (N) metabolism by using whole-genome metagenomic analysis. GWAS identifies a molecular marker cluster associated with monoterpenoids in grapes.

Monoterpenoids are vital compounds that impart a distinctive floral flavor. They exist in both glycosidic and free forms in grapes. The breeding of improved monoterpenoid varieties has consistently been a topic of interest, yet only a limited number of molecular markers have been documented. This study employed a genome-wide association study (GWAS) on an F1 population crossed between a typical muscat variety ('Muscat of Alexandria') and a non-aromatic variety ('Christmas Rose'), conducted over two consecutive years. A total of 4089 significant single nucleotide polymorphism sites (sigSNPs) an

Study of the Pollen Grain Metabolome under Deposition of Nitrogen and Phosphorus in Taxus baccata L. and Juniperus communis L.

Nitrogen plays an important role in both quantitative and qualitative aspects of plant reproduction, including pollen grain compounds and seed production. Recent studies have pointed out that pollen grains produced by male plants of T. baccata and J. communis subjected to a long period of fertilizer supplementation have lower in vitro germination ability and higher nitrogen content. To gain molecular insights into these observations, we conducted GC-MS analysis of both species to characterize the metabolomes of dry, mature pollen grains, which allowed for the identification and quantification

Metabolic engineering of Komagataella phaffii for the efficient utilization of methanol.

BACKGROUND: Komagataella phaffii, a type of methanotrophic yeast, can use methanol, a favorable non-sugar substrate in eco-friendly bio-manufacturing. The dissimilation pathway in K. phaffii leads to the loss of carbon atoms in the form of CO2. However, the ΔFLD strain, engineered to lack formaldehyde dehydrogenase-an essential enzyme in the dissimilation pathway-displayed growth defects when exposed to a methanol-containing medium. RESULTS: Inhibiting the dissimilation pathway triggers an excessive accumulation of formaldehyde and a decline in the intracellular NAD+/NADH ratio. Here, we desi

Thiamine pyrophosphate-dependent enzymes in Toxoplasma gondii and functional significance of an apicoplast-localized pyruvate dehydrogenase.

Thiamine pyrophosphate (TPP)-dependent enzymes are central to carbon metabolism and metabolic plasticity in intracellular protozoan parasites. Toxoplasma gondii harbors at least five TPP-dependent proteins, namely pyruvate dehydrogenase (PDH) and 1-deoxy-D-xylulose-5-phosphate synthase (DXPS) in the apicoplast (a plastid relict), branched-chain α-ketoacid dehydrogenase (BCKDH) and α-ketoglutarate dehydrogenase (α-KGDH) in the mitochondrion, and transketolase (TKT) in the cytosol. These enzymes mutually regulate metabolic flux via glycolysis, tricarboxylic acid (TCA) cycle, pentose phosphate pa

Metabolic Pathway Engineering Improves Dendrobine Production in Dendrobium catenatum.

The sesquiterpene alkaloid dendrobine, widely recognized as the main active compound and a quality control standard of medicinal orchids in the Chinese Pharmacopoeia, demonstrates diverse biological functions. In this study, we engineered Dendrobium catenatum as a chassis plant for the production of dendrobine through the screening and pyramiding of key biosynthesis genes. Initially, previously predicted upstream key genes in the methyl-D-erythritol 4-phosphate (MEP) pathway for dendrobine synthesis, including 4-(Cytidine 5'-Diphospho)-2-C-Methyl-d-Erythritol Kinase (CMK), 1-Deoxy-d-Xylulose 5

Regulatory Effects and Mechanisms of Dissolved Oxygen on Xylitol Biosynthesis from Glucose by Zygoascus hellenicus B11.

To overcome the bottleneck of high pollution associated with using xylose as a raw material for xylitol biosynthesis, this study focused on a yeast strain, Zygoascus hellenicus B11, which can directly utilize glucose as a carbon source. We investigated the regulatory effects and underlying mechanisms of different oxygen conditions on xylitol biosynthesis. Through systematic scale-up studies from shake flasks to bioreactors, we characterized the dynamic profiles of cell growth, substrate consumption, and xylitol synthesis under various agitation speeds and aeration conditions. Furthermore, by c

Fosmidomycin, an inhibitor of isoprenoid synthesis, induces persistence in Chlamydia by inhibiting peptidoglycan assembly.

The antibiotic, fosmidomycin (FSM) targets the methylerythritol phosphate (MEP) pathway of isoprenoid synthesis by inhibiting the essential enzyme, 1-deoxy-D-xylulose 5-phosphate reductoisomerase (Dxr) and is lethal to intracellular parasites and bacteria. The obligate intracellular bacterial pathogen, Chlamydia trachomatis, alternates between two developmental forms: the extracellular, infectious elementary body (EB), and the intracellular, replicative form called the reticulate body (RB). Several stressful growth conditions including iron deprivation halt chlamydial cell division and cause d

Metabolism of Malus halliana Roots Provides Insights into Iron Deficiency Tolerance Mechanisms.

Iron (Fe) deficiency is one of the most common micronutrient imbalances limiting plant growth globally, especially in arid and saline alkali regions due to the decreased availability of Fe in alkaline soils. Malus halliana grows well in arid regions and is tolerant of Fe deficiency. Here, a physiological and metabolomic approach was used to analyze the short-term molecular response of M. halliana roots to Fe deficiency. On the one hand, physiological data show that the root activity first increased and then decreased with the prolongation of the stress time, but the change trend of root pH was

Structure-Activity Relationships of the MEPicides: N-Acyl and O-Linked Analogs of FR900098 as Inhibitors of Dxr from Mycobacterium tuberculosis and Yersinia pestis.

Despite continued research efforts, the threat of drug resistance from a variety of bacteria continues to plague clinical communities. Discovery and validation of novel biochemical targets will facilitate development of new drugs to combat these organisms. The methylerythritol phosphate (MEP) pathway to make isoprene units is a biosynthetic pathway essential to many bacteria. We and others have explored inhibitors of the MEP pathway as novel antibacterial agents. Mycobacterium tuberculosis, the causative agent of tuberculosis, and Yersinia pestis, resulting in the plague or "black death", both

Metabolic Engineering of Lachancea kluyveri with Enhanced Glucose and Xylose Co-utilization for Lignocellulosic Biofuels.

Lignocellulose biorefinery provides a sustainable supply of fuels and value-added compounds. However, the major limitation of its application is the inefficient co-utilization of glucose and xylose by the model yeast. Here, we report for the first time on the metabolic engineering of non-model industrial yeast Lachancea kluyveri for co-utilization of glucose and xylose sustainably using lignocellulosic feedstock. Co-utilization in L. kluyveri was achieved by heterologous expression of XYLl, XYL2, and XYL3 and overexpression of TAL1 and TKL1, which eliminated the imbalanced cofactor specificity
