Mitochondrial proton-translocating NADH-dehydrogenase (complex I) is one of the largest and most complicated membrane bound protein complexes. Despite its central role in eukaryotic oxidative phosphorylation and its involvement in a broad range of human disorders, little is known about its structure and function. Therefore, we have started to use the powerful genetic tools available for the strictly aerobic yeast Yarrowia lipolytica to study this respiratory chain enzyme. To establish Y. lipolytica as a model system for complex I, we purified and characterized the multisubunit enzyme from Y li

Peanut meal (PM) is rich in proteins and fatty acids, which enables it to be a valuable substrate for microbial growth. This study aimed to investigate the effect of PM on the growth and erythritol production of Yarrowia lipolytica CA20 and to establish a procedure for low-cost erythritol production using PM. We found that PM could be used as the only medium component, except for glucose, to support cell growth and erythritol production during submerged fermentation. After optimization of medium compositions and growth conditions, erythritol of 125.25 ± 4.8 g/L was obtained after fermentation

Growth and TAG production by Yarrowia lipolytica were compared for cells cultured in nitrogen-complete medium containing waste glycerol derived from biodiesel production, as well as pure glycerol, dextrose, or canola oil as the carbon sources. Growth and TAG production were also analyzed for Y. lipolytica cells cultured in nitrogen-limited media containing either pure glycerol or glycerol plus dextrose. Significantly greater amounts of TAGs were synthesized by Y. lipolytica cultured in minimal media compared to rich media (approximately 3-fold on dry weight basis when grown on glycerol). Cultu

Yarrowia lipolytica is a dimorphic oleaginous non-conventional yeast widely used as a powerful host for expressing heterologous proteins, as well as a promising source of engineered cell factories for various applications. This microorganism has a documented use in Feed and Food and a GRAS (generally recognized as safe) status. Moreover, in vivo studies demonstrated a beneficial effect of this yeast on animal health. However, despite the focus on Y. lipolytica for the industrial manufacturing of heterologous proteins and for probiotic effects, its potential for oral delivery of recombinant the

We studied the steric course of the reaction catalyzed by the N-ethylmaleimide (NEM) reductase of Yarrowia (Candida) lipolytica (Y. lipolytica), using 4R-[4-2H1]NADPH and 4S-[4-2H1]NADPH as cofactors and N-ethylcitraconimide as substrate. Active substrates and inhibitors of NEM reductase and its subcellular distribution were also investigated to clarify the biochemical properties of this enzyme. NEM reductase catalyzes the reduction of N-ethylmaleimide to N-ethylsuccinimide with NAD(P)H as the cofactor. Several maleimide and cyclopentenone derivatives tested were also active substrates for NEM

Yarrowia lipolytica, also known as Candida lipolytica, is an emerging opportunistic "rare pathogenic yeast". Due to the limited data on its antifungal susceptibility, the clinical treatments become challenging. Based on the China Hospital Invasive Fungal Surveillance Network (2009-2022), we conducted a comprehensive multi-method study on clinical isolates from various central hospitals. This study is currently the largest study carried out to assess the antifungal susceptibility of Y. lipolytica. It is also the first to establish local epidemiological cut-off values (L-ECOFFs), identify its ER

The oleaginous yeast Yarrowia lipolytica possesses six acyl-CoA oxidase (Aox) isoenzymes encoded by genes POX1-POX6. The respective roles of these multiple Aox isoenzymes were studied in recombinant Y. lipolytica strains that express heterologous polyhydroxyalkanoate (PHA) synthase (phaC) of Pseudomonas aeruginosa in varying POX genetic backgrounds, thus allowing assessment of the impact of specific Aox enzymes on the routing of carbon flow to β-oxidation or to PHA biosynthesis. Analysis of PHA production yields during growth on fatty acids with different chain lengths has revealed that the PO

The yeast-to-filament transition is important for dimorphic fungi to adapt to different environmental conditions. Transcriptional activators that activate downstream target genes in response to environmental stimulations are particularly important for the control of this morphological transition. However, only a limited number of key activators of filamentation have been identified. Here, we report YlAaf1 and YlAaf2, a novel pair of SANT domain-containing transcription factors that promote filamentation in the dimorphic yeast Yarrowia lipolytica. We show that both deletion and co-overexpressio

Yarrowia lipolytica is a food-compatible yeast with strong potential for sustainable B-vitamin enrichment of functional food ingredients. This study evaluated the effects of glucose, xylose, mixed, and lignocellulosic sugars, as well as diverse nitrogen sources, on yeast growth and B-vitamin yields in controlled fermentations. Glucose with yeast extract led to the highest biomass and intracellular accumulation of thiamine (B1), niacin (B3), and pyridoxine (B6), while mixed sugars supported maximal intracellular folate (B9) content. Intracellular B-vitamin levels were consistently higher with y

Controllable regulatory elements, like inducible, titratable promoters, are highly desired in synthetic biology toolboxes. A set of previously developed erythritol-inducible promoters along with an engineered Yarrowia lipolytica host strain were shown to be a very potent expression platform. In this study, we push the previously encountered limits of the synthetic promoters' titratability (by the number of upstream motifs) by using a compatible transcription factor, Euf1, as the promoter titrator. Overexpression of spliced EUF1 turned out to be very efficient in promoting expression from the c

A comprehensive understanding of the growth-production trade-off under heat stress is essential for developing high-yield thermotolerant chassis. Here, the effects of elevated temperature on growth and erythritol biosynthesis were systematically investigated in YL (A high-erythritol-producing industrial Yarrowia lipolytica strain). Notably, a heat-induced abnormal growth-surge effect (AGSE) was observed and identified, in which high dissolved oxygen conditions were associated with altered ATP-related metabolic activity, oxidative stress responses, and increased accumulation of stress-protectiv

The hydrolysis of cellulose generates inhibitors like acetate, suppressing fermentation performance. Here, 25SrRNA methyltransferase CgBMT5 from stress-tolerant yeast Candida glycerinogenes was used as an anti-stress gene element in Saccharomyces cerevisiae and Yarrowia lipolytica. Expression of CgBMT5 in S. cerevisiae increased cell tolerance to acetate, high osmolarity, and heat stress and rescued the delay in cell growth under acetate stress. Ethanol productivity was improved from 0.52 g·(L/h) to 0.69 g·(L/h). CgBMT5 improved GFP expression. The transcription factor ARG81 binds to the promo

Drug resistance in economic animals to pathogens is a matter of widespread concern due to abuse of antibiotics. In order to develop a safe and economical immunopotentiator to raise the immunity and antibacterial response as a replacement for antibiotics, a recombinant yeast co-expressing pig interleukin-17 (IL-17) and IL-22 was constructed and designated as Po1h-pINA1297-IL-17/22. To evaluate the immunoregulator activities of Po1h-pINA1297-IL-17/22, two experiment groups (oral inoculation with Po1h-pINA1297 or Po1h-pINA1297-IL-17/22) and a negative control group (PBS) were set up using 4-week-

Fungal spoilage in cultured dairy products causes consumer dissatisfaction, food waste, and financial losses. This study aimed to identify and characterize fungi responsible for spoilage in cultured dairy products by amplifying the ITS1-5.8S-ITS2 ribosomal DNA (rDNA) region. A total of 200 isolates collected from 154 spoiled dairy products, including yogurt, cottage cheese, and sour cream. The most common genera identified were Penicillium (46.5% of isolates), Mucor (15% of isolates), and Yarrowia (12.5% of isolates). Penicillium remained the most frequently isolated fungal genus regardless of

In the present work, two different ways of lipids extraction from Yarrowia lipolytica yeast were investigated in order to maximize the extraction yield. Firstly, various modern techniques of extraction including ultrasound, microwave, and bead milling were tested to intensify the efficiency of lipid recovery. Secondly, several pretreatments such as freezing/defrosting, cold drying, bead milling, and microwave prior two washing of mixture solvent of chloroform:methanol (1:2, v/v) were study to evaluate the impact on lipid recovery. All these treatments were compared to conventional maceration,

BACKGROUND & AIMS: Pancreatic exocrine insufficiency (PEI) reduces pancreatic secretion of digestive enzymes, including lipases. Oral pancreatic enzyme replacement therapy (PERT) with pancreatin produces unsatisfactory results. The lipase 2 produced by the yeast Yarrowia lipolytica (YLLIP2; GenBank: AJ012632) might be used in PERT. We investigated its ability to digest triglycerides in a test meal and its efficacy in reducing fecal fat in an animal model of PEI.
METHODS: YLLIP2 was produced by genetically engineered Y lipolytica and purified from culture media. YLLIP2 or other gastric (LIPF)

In this study, the liquid lipase Yarrowia lipolytica lipase 2 (YLLip2) expressed by Pichia pastoris GS115 was used to produce biodiesel from waste oil. Four signal peptides were compared to express YLLip2 in P. pastoris, among which SP23 exhibited greater secretion performance. In a 1.3-L bioreactor with FM22 medium for 7 days incubation, the maximum YLLip2 activity and total protein content reached 895.44 ± 27.31 U/mL and 3.83 ± 0.31 g/L, respectively. Under the optimal reaction conditions of 30 °C, 20% moisture content, 50 U/g oil of enzyme dosage, and distributed methanol addition, the reac

In this study, isocitrate dehydrogenase (IDH) was employed exclusively as a structural protein template for the synthesis of gold nanoparticles, rather than relying on its enzymatic catalytic function. The reduction of Au³⁺ to Au⁰ is accomplished through the intrinsic reducing capability of amino acid residues (particularly tryptophan and tyrosine) within the protein scaffold, which simultaneously acts as a capping and stabilizing agent. This protein-templating strategy provides a straightforward, biocompatible, and cofactor-free approach for developing AuNP-based biosensing platforms. So, the
