Present in various subcellular compartments, cysteine is the major source of reduced sulfur and thus represents a key metabolite for various biosynthetic pathways as well as for redox homeostasis as a component of glutathione. As photosynthetic organisms assimilate inorganic sulfate and reduce it into sulfide before its incorporation into cysteine, there are strong relationships between cysteine homeostasis and all pathways involved in its synthesis and utilization. Over the last decade, cysteine degradation leading to hydrogen sulfide release has been linked to different physiological respons

Autophagy, a conserved intracellular recycling pathway, operates both non-selectively and through targeted degradation. In this study, we assess its role in Arabidopsis thaliana seed biology by examining loss-of-function mutants of ATG5 and ATG7 and their interaction with ABA signaling. Both atg5 and atg7 seeds display delayed germination relative to Col-0, an effect that is exacerbated by ABA. Histochemical staining reveals altered organization of lipid droplet and protein storage vacuole (PSV) organization in the mutants, and ATG8 fail to localize to PSVs when autophagy is impaired. Root-exuded organic acids are crucial in mitigating iron (Fe) and phosphorus (P) deficiencies, and their biosynthesis and secretion require significant metabolic investment. Recent studies have shown that roots can also uptake exudates. We hypothesized that citric acid uptake increases under Fe and P deficiencies, declines over time, and contributes to primary metabolism. We investigated citric acid uptake, translocation, and metabolization in Fe- and P-deficient tomato plants grown hydroponically using 13C-labelling with bulk stable-isotope and compound-specific stable-isotope analysis. During plant development and in response to stress conditions, autophagy contributes to the intracellular degradation of cellular components and subsequent nutrient recycling. As this process is highly connected to the nutrient status of the plant, autophagy also contributes to the mobilisation of sulfur from source to sink tissues as well as the maintenance of primary sulfate assimilation. In turn, sulfur signals regulate autophagy, with sulfide (an intermediate of primary sulfate assimilation) exerting a repressive effect and sulfur deficiency having a stimulatory effect. In addition to a su

The clubroot pathogen Plasmodiophora brassicae has an effector protein that can methylate the plant defense compound salicylic acid (SA), and the protein sequence is highly conserved in different isolates worldwide. Sequencing the full-length cDNA of P. brassicae Benzoic acid/Salicylic acid Methyltransferase (PbBSMT) from a small collection of clubs indicated the occurrence of a natural variation that differs by 18 amino acids, which was also found within the worldwide data collection for P. brassicae isolates. A search for more possible substrates including defense-inducing compounds revealed

Heat-stress driven by global warming alters rice (Oryza sativa) water-use dynamics. To assess adaptive water-use strategies under elevated air temperature, we conducted a three-year whole-season experiment in natural light climate chambers with ambient, +1 °C, +2 °C, and +5 °C warming treatments. Stomatal conductance (gs), transpiration rate (E), net photosynthetic rate (An) and leaf developmental dynamics were continuously measured across growth stages. Above the photosynthetic optimum leaf temperature (Tleaf-opt-An = 30.8 °C), An declined while gs increased. The increase in gs accelerated be

PREMISE: Medullary bundles, which are vascular bundles in the pith, have evolved multiple times. However, their functional significance remains poorly understood. Here, we investigated how medullary bundles are involved in the vascular architecture and hydraulic function in a tropical root climber, Adelobotrys scandens (Melastomataceae) to understand the functional significance of this vascular architecture in a wet-adapted root climber species as compared to what is known about medullary bundles in the lineages in hydraulically limited arid/saline environments.

As a master bZIP transcription factor, ELONGATED HYPOCOTYL 5 (HY5) coordinates plant growth with environmental acclimation across developmental stages. Beyond its role in photomorphogenesis, HY5 integrates diverse environmental and endogenous signals, linking environmental perception with developmental and metabolic programs to ensure coordinated physiological adaptation. This review synthesizes current understanding of the multilayered regulatory networks controlling HY5, ranging from constitutive photomorphogenic 1/suppressor of phytochrome A (COP1/SPA)-mediated ubiquitin-dependent degradati

PREMISE: Bamboos have extremely long internodes, making it possible to measure tensile properties and anatomical features of discrete nodes and internodes and understand how the plants endure mechanical stress and how the tissues function in commercial products. We compared upper, middle, and lower parts of culms in black bamboo, Phyllostachys nigra, to determine whether the nodes comply with the Niklas spring-like joint model.
METHODS: We measured flexural stiffness, modulus of elasticity (E), load at failure (Fmax), modulus of rupture (MOR), and density and fiber cap size of vascular bundle

The growing prevalence of gluten-related disorders in humans has driven the development of wheat varieties with reduced immunogenic gluten. This study aimed to integrate RNA interference (RNAi) and CRISPR genome editing within a doubled haploid (DH) platform to overcome challenges of gene redundancy and polyploidy in wheat gliadins. We generated DH lines from crosses between RNAi and CRISPR lines and elite wheat cultivars, enabling stable fixation of multiple genetic modifications in a single generation. Deep sequencing analysis of α-gliadin amplicons was conducted using a custom bioinformatic

Vitamin B1 is a vital co-factor in cellular metabolism, but it must be obtained through the diet in humans. Polished (white) rice, a dietary staple for much of the global population, contains very low levels of vitamin B1, and this contributes to widespread thiamin deficiency in regions that rely heavily on rice. To address this issue, we engineered rice to express the Saccharomyces cerevisiae thiamin transporter gene THI7 under the control of the rice endosperm-specific GLUTELIN1 (GT1) promoter. In transgenic lines with moderate-to-high THI7 expression, we found significantly increased thiami

PREMISE: Climate refugia endemics have persisted through past environmental shifts, but their responses to modern severe weather remain poorly understood. Torreya taxifolia (Taxaceae), a critically endangered conifer endemic to steephead ravines of Florida and Georgia, has declined for decades due to fungal canker diseases. Hurricane Michael, a Category 5 storm in 2018, caused extensive forest damage in its refugial habitat. We investigated how natural variation in disturbance was linked to tree health and microclimatic conditions relevant to disease.
METHODS: We monitored the health and habi

In the last two decades, the transcription factor phosphate starvation response 1 (PHR1) and its homologues in Arabidopsis and other plant species have emerged as undisputed master regulators of phosphorus starvation response (PSR). The role of Suppressor of Yeast Gpa1 (SYG1)/PHOsphate metabolism 81 (PHO81)/Xenotropic and Polytropic Retrovirus receptor 1 (XPR1) (SPX) proteins as negative regulators of PHR1 activity, and binding of this transcription factor to the P1BS element of the target phosphate starvation-inducible genes during the regulation of PSR, is well established. Given the central

Demand for rice (Oryza sativa L.) is rapidly increasing in sub-Saharan Africa; however, its productivity is constrained by nutrient-poor paddy soils, particularly those deficient in N and P. Improving root system architecture offers potential to enhance nutrient uptake efficiency and increase yield under such conditions. This study aimed to identify the most effective root phenotype for enhancing grain yield under the nutrient-deficient soil conditions in Madagascar. The co-evolutionary arms race between crops and their parasites requires continuous identification of new resistance mechanisms. Broomrape (Orobanche cumana), a root parasitic plant, poses a severe threat to sunflower (Helianthus annuus) production, yet the genetic architecture underlying host resistance remains poorly understood. To address this, we established a high-throughput phenotyping platform to quantify root infestation across a diverse sunflower association mapping (SAM) population. Combining this phenotypic resource with a dual genome-wide association study (GWAS) strategy based on

Structural failure of cereal stems during late-season climate extremes is a critical determinant of yield stability. In barley, breakage of the stem below the spike, known as head loss, leads to major yield losses, particularly in hot and dry regions where the crop is widely grown. Despite a predicted increase in head loss risk due to global warming, current understanding of the genetic, physiological, anatomical, and environmental factors that control head loss remains limited. Overcoming these knowledge gaps is essential to providing a systems-level strategy for barley breeders to develop cl

Tree growth is central to both terrestrial ecology and the forestry industry. Biotechnology for inducing the overproduction of phytohormones such as gibberellins (GAs) is a powerful tool for improving tree yield and wood properties. Augumentation of GAs in trees in temperate and boreal regions, however, prevents the induction of vegetative dormancy, which results in reduced yield and tree loss over time. Hybrid Populus trees (Populus tremula × P. tremuloides; Ptt) use an internal circadian clock to synchronise their daily metabolism and seasonal growth with predictable light and temperature ch

Throughout colonial history, botanical exploration expanded Western scientific knowledge while reinforcing systems of control, particularly through the erasure of plant knowledge regarding human reproduction issues. Abortifacients, long used by Black and Indigenous communities for fertility regulation and abortion, was among the areas for which traditional knowledge was most frequently silenced. These plants enabled reproductive autonomy and disrupted the colonial agenda of controlling laboring bodies. This synthesis draws heavily from Mexican ethnobotanical practices, and pays equal attention

PREMISE: The southwestern United States is at the forefront of nonnative Phragmites australis subsp. australis expansion. Native Phragmites australis subsp. americanus, likely in decline in North America, is ecologically and culturally important and of conservation interest. In the greater Grand Canyon region, the Glen Canyon Dam led to loss of habitat upstream and increases in Phragmites downstream. Recent increases correspond with changes to dam operations and the arrival of nonnative Phragmites. Determining the native status of Phragmites stands along the Colorado River and around Lake Powe

Resupination refers to the developmental orientation changes of flowers through ~180°, leaving them effectively upside-down. It is a widespread trait present in 14 angiosperm families, including the Orchidaceae, where it is a gravitropic phenomenon actively controlled by auxins. Here, we demonstrate that the passive gravitational pull on flower parts can have an additional influence on resupination. We studied a lady's slipper orchid in which some flowers naturally fail to resupinate. We conducted a manipulative experiment removing floral parts and showed that both the probability of complete
