Hippocampal subregional texture features associated with Alzheimer's disease severity and cognition.

Alzheimer's disease (AD) exhibits selective vulnerability in hippocampal subfields, where microstructural alterations precede overt atrophy. Conventional morphometric measures, such as volume or thickness, are limited in their ability to capture subtle textural heterogeneity that reflects underlying cytoarchitectural disorganization, particularly perforant path fibres within the subiculum. The objective of this study is to determine whether hippocampal subregional texture-based radiomic features can serve as sensitive markers of disease severity and cognitive impairment along the AD continuum.

CT1812 Attenuates Surgery-Induced Cognitive Impairment and Modulates TMEM97- and PGRMC1-Associated Hippocampal Stress Responses in Aged Mice.

Postoperative neurocognitive disorders (PND) are common complications in older surgical patients, but druggable mechanisms linking surgical trauma to hippocampal synaptic and cognitive vulnerability remain poorly defined. The σ-2 receptor (σ-2R), encoded by transmembrane protein 97 (TMEM97), and progesterone receptor membrane component 1 (PGRMC1) intersect membrane lipid biology and synaptic stress signaling; CT1812, a pharmacological σ-2R complex modulator, has shown target engagement in early Alzheimer's disease trials, but whether this pathway participates in postoperative hippocampal injur

Xijiao Dihuang decoction attenuates microglia/macrophage-mediated inflammatory response in cerebral injury by modulating the PI3K/Akt/HIF-1α/NLRP3 axis.

ETHNOPHARMACOLOGICAL RELEVANCE: Xijiao Dihuang Decoction (XJDH), originating from Beiji Qianjin Yaofang (Thousand Gold Prescriptions for Emergencies) compiled by Sun Simiao during the Tang Dynasty (652 AD), is a classical Traditional Chinese Medicine (TCM) formula comprising Cornu Bubali (substituted for Rhinoceros horn), Radix Rehmanniae, Paeoniae Radix Rubra, and Cortex Moutan. In TCM clinical practice, XJDH has traditionally been prescribed for "Xue-Re" (Blood-Heat) syndrome characterized by febrile diseases with bleeding tendency, blood stasis and impaired consciousness, a clinical pattern

MIND Diet and Hippocampal Sclerosis Among Community-Based Older Adults.

IMPORTANCE: Hippocampal sclerosis (HS), defined as severe neuronal loss and astrogliosis in coronal sections of the midhippocampus cornu ammonis 1 or subiculum, is an important pathology associated with limbic-predominant age-related transactive response DNA-binding protein 43 encephalopathy neuropathological change (LATE-NC), Alzheimer disease, and dementia. The association of diet with HS or HS with LATE-NC in humans remains underexplored. OBJECTIVE: To investigate the association of the Mediterranean-Dietary Approaches to Stop Hypertension Intervention for Neurodegenerative Delay (MIND) di

Age-related and amyloid-beta-independent tau deposition and its downstream effects.

Amyloid-β (Aβ) is hypothesized to facilitate the spread of tau pathology beyond the medial temporal lobe. However, there is evidence that, independently of Aβ, age-related tau pathology might be present outside of the medial temporal lobe. We therefore aimed to study age-related Aβ-independent tau deposition outside the medial temporal lobe in two large cohorts and to investigate potential downstream effects of this on cognition and structural measures. We included 545 cognitively unimpaired adults (40-92 years) from the BioFINDER-2 study (in vivo) and 639 (64-108 years) from the Rush Alzheime

Therapeutic advances for treating memory impairments in perinatal brain injuries with implications for cerebral palsy: a systematic review and meta-analysis of preclinical studies.

Cerebral palsy (CP) is a neurodevelopmental disorder caused by damage to the immature brain. CP is considered the main cause of physical disability in childhood. Studies have shown that memory function and emotional behaviour are significantly impaired in CP. Current thought is that interventions for neuromotor damaged play a prominent role, but neglects the memory acquisition problems that affect the functioning and quality of life of these children. This systematic review aims to map and analyse pre-clinical interventions used to treat memory formation problems resulting from CP. For this, a

Prenatal development of the human entorhinal cortex.

Little is known about the development of the human entorhinal cortex (EC), a major hub in a widespread network for learning and memory, spatial navigation, high-order processing of object information, multimodal integration, attention and awareness, emotion, motivation, and perception of time. We analyzed a series of 20 fetal and two adult human brains using Nissl stain, acetylcholinesterase (AChE) histochemistry, and immunocytochemistry for myelin basic protein (MBP), neuronal nuclei antigen (NeuN), a pan-axonal neurofilament marker, and synaptophysin, as well as postmortem 3T MRI. Circuit- and Diagnosis-Specific DNA Methylation Changes at γ-Aminobutyric Acid-Related Genes in Postmortem Human Hippocampus in Schizophrenia and Bipolar Disorder.

IMPORTANCE: Dysfunction related to γ-aminobutyric acid (GABA)-ergic neurotransmission in the pathophysiology of major psychosis has been well established by the work of multiple groups across several decades, including the widely replicated downregulation of GAD1. Prior gene expression and network analyses within the human hippocampus implicate a broader network of genes, termed the GAD1 regulatory network, in regulation of GAD1 expression. Several genes within this GAD1 regulatory network show diagnosis- and sector-specific expression changes within the circuitry of the hippocampus, influenci

First effects of rising amyloid-β in transgenic mouse brain: synaptic transmission and gene expression.

Detecting and treating Alzheimer's disease, before cognitive deficits occur, has become the health challenge of our time. The earliest known event in Alzheimer's disease is rising amyloid-β. Previous studies have suggested that effects on synaptic transmission may precede plaque deposition. Here we report how relative levels of different soluble amyloid-β peptides in hippocampus, preceding plaque deposition, relate to synaptic and genomic changes. Immunoprecipitation-mass spectrometry was used to measure the early rise of different amyloid-β peptides in a mouse model of increasing amyloid-β ('

Longitudinal analysis of hippocampal subfield atrophy and network centrality associated with cognitive decline in Alzheimer's disease progression.

BACKGROUND: Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by structural degeneration of the hippocampus. Previous studies have demonstrated that specific hippocampal subfields, such as the cornu ammonis (CA)1 and the subiculum, are susceptible to AD-related atrophy. However, most previous studies have focused on cross-sectional volumetric changes without investigating the interrelationships between subfields or their network-level functions throughout the disease progression. PURPOSE: To examine the longitudinal volumetric changes in the hippocampal subfie

Hippocampal CA1 cell loss in a non-human primate model of transient global ischemia: a pilot study.

We exposed adult Rhesus (Macaca mulatta) to a transient global ischemia, which was induced by clipping the innominate and subclavian arteries that originated from the aortic arch. NHP1 received 20-min, while NHP2 and NHP3, were exposed to a 15-min transient global ischemia and were euthanized at day 1 (NHP1), day 5 (NHP2) or day 30 (NHP3) after ischemia, respectively. NHP1 displayed severe paralysis and rigidity, and intermittent convulsions over the next 24 h. Although histological examination of the brain revealed no detectable gross brain damage (i.e., swelling) and only minimal cell loss i
