Hot alkaline lysis gDNA extraction from formalin-fixed archival tissues.

Formalin fixation of natural history specimens and histopathological material has historically been viewed as an impediment to successful genomic analysis. However, the development of extraction methods specifically tailored to contend with heavily crosslinked archival tissues, re-contextualises millions of previously overlooked specimens as viable molecular assets. Here, we present an easy-to-follow protocol for screening archival wet specimens for molecular viability and subsequent genomic DNA extraction suitable for sequencing. The protocol begins with non-destructive assessment of specimen

Diagnosed and subjectively perceived long-term effects of COVID-19 infection on olfactory function assessed by supervised machine learning.

Loss of olfactory function is a typical acute coronavirus disease 2019 (COVID-19) symptom, at least in early variants of SARS-CoV2. The time that has elapsed since the emergence of COVID-19 now allows for assessing the long-term prognosis of its olfactory impact. Participants (n = 722) of whom n = 464 reported having had COVID-19 dating back with a mode of 174 days were approached in a museum as a relatively unbiased environment. Olfactory function was diagnosed by assessing odor threshold and odor identification performance. Subjects also rated their actual olfactory function on an 11-point n

Comprehensive two-dimensional gas chromatography-high resolution mass spectrometry with complementary ionization methods in the study of 5000-year-old mummy.

RATIONALE: Mummification is one of the defining customs of ancient Egypt. The nuances of the embalming procedure and the composition of the embalming mixtures have attracted the attention of scientists and laypeople for a long time. Modern analytical tools make mummy studies more efficient. METHODS: Comprehensive two-dimensional gas chromatography-high resolution mass spectrometry (GCxGC/HRMS) with complementary ionization methods (electron ionization, positive chemical ionization, and electron capture negative ionization [ECNI]) with a Pegasus GC-HRT+4D instrument was used to identify embalm

Effects of preservation methodology on stable isotope compositions of sea stars.

RATIONALE: Stable isotope analysis is used to investigate the trophic ecology of organisms and, in order to use samples from archived collections, it is important to know whether preservation methods alter the results. This study investigates the long-term effects of four preservation methods on isotopic compositions and isotopic niche parameters of sea stars. METHODS: We assessed the effects of preservation method (freezing, drying, formaldehyde, ethanol) and duration (0, 1, 3, 6, 9, 12, 24 months) on the stable isotope ratios of carbon, nitrogen and sulfur of sea star tissues. Isotopic rati

Features of the Femoral Proximocaudal Joint Capsule Insertion Among Canids.

This observational study was conducted to evaluate the anatomic relationship between the proximocaudal femoral joint capsule insertion and the femoral caudolateral curvilinear osteophyte (CCO), across ancient and modern domestic and non-domestic canids. Museum specimens of proximal femora were screened for presence of remnant enthesophytes of the caudal joint capsule insertion (first inclusion criterion) and then for the CCO (second inclusion criterion). The initially screened population included 267 dry bone specimens: Six Canis species, hybrid coyote × domestic dog, and five vulpines (three

Heritability of obsessive-compulsive trait dimensions in youth from the general population.

Obsessive-compulsive disorder (OCD) is a heritable childhood-onset psychiatric disorder that may represent the extreme of obsessive-compulsive (OC) traits that are widespread in the general population. We report the heritability of the Toronto Obsessive-Compulsive Scale (TOCS), a new measure designed to assess the complete range of OC traits in youth. We also examined the dimensional nature of the TOCS and the degree to which genetic effects are unique or shared between dimensions. OC traits were measured using the TOCS in 16,718 youth (6-18 years) at a science museum. We conducted a factor an

Leaping lizards landing on leaves: escape-induced jumps in the rainforest canopy challenge the adhesive limits of geckos.

The remarkable adhesive capabilities of geckos have garnered attention from scientists and the public for centuries. Geckos are known to have an adhesive load-bearing capacity far in excess (by 100-fold or more) of that required to support their body mass or accommodate the loading imparted during maximal locomotor acceleration. Few studies, however, have investigated the ecological contexts in which geckos use their adhesive system and how this may influence its properties. Here we develop a modelling framework to assess whether their prodigious adhesive capacity ever comes under selective ch

Jakob Erdheim (1874-1937): father of hypophyseal-duct tumors (craniopharyngiomas).

Jakob Erdheim (1874-1937) was a Viennese pathologist who identified and defined a category of pituitary tumors known as craniopharyngiomas. He named these lesions "hypophyseal duct tumors" (Hypophysenganggeschwülste), a term denoting their presumed origin from cell remnants of the hypophyseal duct, the embryological structure through which Rathke's pouch migrates to form part of the pituitary gland. He described the two histological varieties of these lesions as the adamantinomatous and the squamous-papillary types. He also classified the different topographies of craniopharyngiomas along the

Each flying fox on its own branch: a phylogenetic tree for Pteropus and related genera (Chiroptera: Pteropodidae).

Pteropodidae is a diverse Old World family of non-echolocating, frugivorous and nectarivorous bats that includes the flying foxes (genus Pteropus) and allied genera. The subfamily Pteropodinae includes the largest living bats and is distributed across an immense geographic range from islands in East Africa to the Cook Islands of Polynesia. These bats are keystone species in their ecosystems and some carry zoonotic diseases that are increasingly a focus of interest in biomedical research. Here we present a comprehensive phylogeny for pteropodines focused on Pteropus. The analyses included 50 of

Is lumbar stenosis associated with thoracic stenosis? A study of 1,072 human cadaveric specimens.

BACKGROUND CONTEXT: Tandem stenosis of the cervical and lumbar spine is known to occur in 5% to 25% of individuals with symptomatic neural compression in one region. However, the prevalence of concurrent lumbar and thoracic stenosis is not known. Whether this relationship is because of an increased risk of degenerative diseases in these individuals or because of the tandem presence of stenosis in lumbar and thoracic canal is unknown. PURPOSE: To determine the prevalence of concurrent lumbar and thoracic stenosis, and whether the presence of stenosis in the lumbar spine is associated with sten

Bovicola tibialis (Phthiraptera:Trichodectidae): occurrence of an exotic chewing louse on cervids in North America.

Through a recent (2003-2007) survey of ectoparasites on hoofed mammals in western North America, a literature review, and examination of archived museum specimens, we found that the exotic deer-chewing louse, Bovicola tibialis (Piaget), is a long-term, widespread resident in the region. The earliest known collection was from Salt Spring Island, Canada, in 1941. We found these lice on the typical host, that is, introduced European fallow deer (Dama dama L.), and on Asian chital (Axis axis [Erxleben] ), native Columbian black-tailed deer (Odocoileus hemionus columbianus [Richardson] ), and Rocky

Tandem stenosis: a cadaveric study in osseous morphology.

BACKGROUND: Tandem stenosis is the occurrence of concurrent cervical and lumbar stenosis. The prevalence has been estimated to be from 5% to 25%. Symptomatic tandem stenosis can present with a confusing scenario of both neurogenic claudication and myelopathy symptoms. PURPOSE: The purpose of this study was to determine 1) the prevalence of anatomic tandem stenosis in a cadaveric population, 2) if there was an associative relationship between lumbar and cervical stenosis, and 3) the positive predictive values of stenosis in one area for stenosis in the other. STUDY DESIGN: We obtained 440 ske

De-extinction of the Northern white rhinoceros (Ceratotherium simum cottoni).

The last survivors of the northern white rhinoceros (NWR, Ceratotherium simum cottoni), once widespread across Central and East Africa, are two non-reproductive females under continuous human protection. In order to prevent the extinction of this white rhinoceros subspecies, the BioRescue consortium founded in 2019 has developed an innovative reproductive biotechnology program. This holistic rescue strategy implements (i) advanced assisted reproductive technologies (aART) including in vitro fertilization (IVF) and embryo transfer, (ii) stem cell associated techniques (SCAT) for establishing in
