The patterns of gene expression for hepatocyte growth factor (HGF) and its receptor, c-Met, were revealed in the tooth germ of rat mandibular molars using RT-PCR. In situ hybridization demonstrated that the HGF gene was expressed only in the cells of the dental papilla of the tooth germ in vivo. The characteristic temporospatial distribution of HGF and c-Met during germ development was revealed using immunohistochemical studies in vivo. In order to demonstrate the functional role played by HGF in tooth development, HGF translation arrest by antisense phosphorothioate oligodeoxynucleotide (ODN)

Teeth develop from composite organ rudiments that are formed through the interaction of oral epithelium and mesenchyme of the first branchial arch; cells of the former differentiate into enamel-secreting ameloblasts whereas those of the latter differentiate into dentine-secreting odontoblasts. Experimental analysis of odontogenic tissue interactions in mammalian embryos has focused on the late developmental stages of morphogenesis and cytodifferentiation; little is known about initial pattern-forming events, during which presumptive tooth-forming cells are specified and the sites of tooth init

The Runx2 gene is a master transcription factor of bone and plays a role in all stages of bone formation. It is essential for the initial commitment of mesenchymal cells to the osteoblastic lineage and also controls the proliferation, differentiation, and maintenance of these cells. Control is complex, with involvement of a multitude of factors, thereby regulating the expression and activity of this gene both temporally and spatially. The use of multiple promoters and alternative splicing of exons further extends its diversity of actions. RUNX2 is also essential for the later stages of tooth f

OBJECTIVE: Recent studies have demonstrated that acid etching of vital dentin and pulpal tissue does not retard pulpal healing, odontoblastoid cell differentiation, or dentinal bridge formation when the pulp is capped with adhesive resins. The purpose of this study was to evaluate the pulpal response in nonexposed and exposed monkey pulps to treatment with the Clearfil Liner Bond 2 and Clearfil AP-X system.
METHOD AND MATERIALS: Class V and Class I cavities in nonexposed and exposed pulps were observed at 7 or 8, 27, and 97 days.
RESULTS: There were no differences in pulpal inflammation betw

To elucidate the defective dentin formation in osteogenesis imperfecta (OI), we analyzed the expression of selected fibronectin (FN) isoforms in the dentin matrix of a patient with dentinogenesis imperfecta (DI) associated with OI, and in normal teeth. Frozen tooth sections were immunostained with three monoclonal antibodies (MAbs). The MAb recognizing the major cell-binding region (f-33), shared by plasma FN (pFN) and cellular FN (cFN), stained the pulp of normal adult permanent teeth intensely, while no reactivity was present in predentin, (demineralized) dentin, or dental cementum. Dental pulp stem cells (DPSCs) possess multilineage differentiation potential and contribute to dental pulp regeneration through odontoblast replacement and neovascularization. Although mechanosensitive ion channels such as PIEZO1 are known to regulate stem cell fate, their role in DPSC-mediated angiogenesis under mechanical stress remains poorly understood. In this study, we investigated the involvement of PIEZO1 in regulating angiogenic gene expression and endothelial differentiation of immortalized human DPSCs (hDPSC-K4DT). Compressive force was applied using an in vitro weight-loading mode

Dentin hypersensitivity is characterized by transient, sharp pain resulting from dentin exposure due to enamel or cementum loss. Current treatments, which primarily focus on superficial tubule occlusion or nerve desensitization, provide only temporary relief. Copine7 (CPNE7) induces odontoblast differentiation and physiologic dentin regeneration, enabling biological dentin sealing and presenting a novel therapeutic approach. This study reports the first-in-human results of a randomized, double-blind, dose-escalation phase 1/2a clinical trial evaluating the safety, tolerability, preliminary eff

OBJECTIVE: Irreversible pulpitis in immature teeth is a serious dental disease which may lead to unclosed apical foramen and even tooth loss. Our study proposed a system of G-CSF/laponite/collagen composite and investigated its effects for stem cells of apical papilla (SCAPs) homing, targeting a potential treatment for immature dental pulp regeneration.
METHODS: In vitro, the effect of G-CSF on SCAPs proliferation, migration and differentiation was investigated. The microporous scaffold structure, sustained-release capability and cytotoxic effects of G-CSF/laponite/collagen composite was eval

INTRODUCTION: The role of osteopontin (OPN) following severe injury remains to be elucidated, especially its relationship with type I collagen (encoded by the Col1a1 gene) secretion by newly-differentiated odontoblast-like cells (OBLCs). In this study, we examined the role of OPN in the process of reparative dentin formation with a focus on reinnervation and revascularization after tooth replantation in Opn knockout (KO) and wild-type (WT) mice.
METHODS: Maxillary first molars of 2- and 3-week-old-Opn KO and WT mice (Opn KO 2W, Opn KO 3W, WT 2W, and WT 3W groups) were replanted, followed by f

Dental pulp stem cell (DPSC) transplantation has shown new prospects in dental pulp regeneration, and is of great significance in the treatment of pulpitis and pulp necrosis. The fate and regenerative potential of stem cells are dependent, to a great extent, on their microenvironment, which is composed of various tissue components, cell populations, and soluble factors. N-cadherin-mediated cell-cell interaction has been implicated as an important factor in controlling the cell-fate commitment of mesenchymal stem cells. In this study, the effect of N-cadherin on odontogenic differentiation of D

Human dental pulp cells (hDPCs) possess the capacity to differentiate into odontoblast-like cells in response to exogenous stimuli. Histone methylation is one of the most robust epigenetic marks and is essential for the regulation of multiple cellular processes. Previous studies have shown that histone methyltransferases (HMTs) and histone demethylases (HDMs) are crucial for the osteogenic differentiation of human bone marrow, adipose tissue, and tooth tissue. However, little is known about the role of histone methylation in hDPC differentiation. Here, the expression levels of HMTs and HDMs we

INTRODUCTION: Dental pulp stem cells (DPSCs) are suspected to be an important part of the innate immune response of dental pulp, which is triggered by microorganisms that progressively invade the human tooth during the formation of caries. This study was performed to elucidate the expression of toll-like receptor 4 (TLR4) in dental pulp of deep caries and to determine whether TLR4 modulates the proliferation and migration of DPSCs.
METHODS: Pulp tissue samples were collected from freshly extracted human wisdom tooth. Immunohistochemistry and immunofluorescence were performed to determine the

Neuro-immune interaction has been suggested to play some modulatory role in the immunodefense of the dentin/pulp complex. In this study, we performed a simultaneous immunohistochemical observation of neural elements and pulpal dendritic cells (PDCs) on human carious teeth, to obtain morphological evidence for neuro-immune interaction in response to dentinal tubule-derived carious stimuli. Human third molars bearing a pulp-exposure-free caries lesion were studied. Immunoperoxidase staining was performed with anti-HLA-DR, anti-coagulation factor XIIIa, and anti-CD14 as PDC markers, and anti-low-

The cellular dynamic pattern of pulpal healing 4, 10, 30, and 60 days after replantation of 47 apicoectomized cat incisors was studied after pulse labeling with 3H-thymidine and 35S-sulfate, autoradiography, and routine histology. In the control teeth the labeling index was less than 0.05%. The apical pulpal cells were capable of ground substance formation and cell proliferation already 4 days after replantation, with a labeling index of 7%, which increased up to 43% within 10 days. A gradual postoperative restitution and reorganization within the pulpal cellular compartment was seen. The maxi
