Pseudomonas aeruginosa is a multidrug-resistant Gram-negative nosocomial pathogen posing a major healthcare burden due to its intrinsic antibiotic resistance and environmental adaptability, which continue to significantly constrain therapeutic options. This pathogen is a leading cause of hospital-acquired respiratory and urinary tract infections that are often persistent and difficult to treat. The elucidation of metabolic adaptations permitting P. aeruginosa survival and supporting virulence in diverse host environments is essential for informing novel treatments. This study investigates the

Adoptive T cell therapy (ACT) remains limited in solid tumors by poor T cell persistence within the metabolically hostile tumor microenvironment (TME). Although IL-9-producing CD8+ T cells (Tc9) consistently demonstrate superior antitumor efficacy compared with conventional Tc1 cells, the selective pressures that shape their functional advantage remain unclear. Here, we show that effective ACT-mediated tumor control is accompanied by a marked increase in intratumoral extracellular ATP (eATP), representing a common metabolic consequence of tumor cell destruction. Despite comparable ATP accumula

BACKGROUND: Olaparib is a poly(ADP-ribose) polymerase (PARP) inhibitor used to treat several malignancies. Previous studies have reported that olaparib may induce ovarian injury by elevating reactive oxygen species and impairing cellular energy homeostasis. ATP is a central regulator of intracellular energy balance and antioxidant defense, whereas thiamine pyrophosphate (TPP), the active form of vitamin B1, supports ATP biosynthesis and possesses antioxidant and anti-inflammatory properties. This study compared the protective effects of ATP and TPP against olaparib-induced ovarian injury in ra

Polycomb Repressive Complex 1 (PRC1) and its antagonistic H2A deubiquitinase USP16 have been regarded as nuclear proteins acting on chromatin. However, our recent studies revealed that a fraction of PRC1 and USP16 also localizes to mitochondria, where PRC1 plays a critical role in maintaining mitochondrial function. The role of USP16 in this context, however, has not yet been investigated. This study shows that USP16 interacts with and stabilizes TOM40, the core subunit of the TOM complex. USP16 deubiquitinates K48-linked ubiquitin chains from lysine 175, 184, and 309 of TOM40. USP16 knockout

Although metformin (MET), the well-known antidiabetic drug, exhibits clear antineoplastic effects and is reported to target mitochondria, several issues are still open in this regard, thus limiting its utilization as an anticancer drug alone or in combination with other molecules. Here a functional investigation was carried out to reveal how MET impacted on mitochondrial functions and cell energy metabolism in human cultured clear cell renal carcinoma cells (ccRCCs), in which the anticancer effect of MET is already known. The in vitro effect of increasing MET concentrations on cell viability,

Viral NS3 helicases convert the chemical energy of ATP hydrolysis into directional motion along nucleic acids, but the molecular basis of this coupling remains unclear. Here, we combine AI-based structural modeling, molecular dynamics simulations, and hybrid QM/MM free energy calculations to investigate ATP hydrolysis in the Zika virus NS3 helicase (ZIKV NS3h). We first address the structural limitations of the available ternary crystallographic structure, which displays a noncanonical ATP-Mg2+ coordination mode and an RNA-binding arrangement inconsistent with the expected ATP-bound state. Our

Anatid alphaherpesvirus 1 (AnHV-1) is an emerging threat that induces severe disease and death in both domestic and wild waterfowl. Birds have an elevated basal body temperature, and suffer at times from environmental heat stress, yet little are known about the mechanisms involved in avian virus replication at higher temperatures. In this study, we examined the effects of hyperthermia on replication dynamics and host cellular responses to AnHV-1 in chicken embryonic fibroblast cultures and an in ovo model. Hyperthermic conditions dramatically increased AnHV-1 replication, viral gene expression

Cardiac complications after major hepatic surgery are frequent, yet how hepatic ischemia-reperfusion injury (HIRI) damages the remote heart is unknown. In 382 hepatectomy patients, the incidence of myocardial injury after noncardiac surgery (MINS) rose from 20.5% to 50% with increasing postoperative liver damage severity (P < 0.0001). In a murine HIRI model, cardiac dysfunction lagged behind hepatic injury; circulating ATP surged within one hour of reperfusion from the ischemic liver and was both sufficient and necessary to drive remote cardiac damage. Extracellular ATP activated the P2X7 rece

The fundamental intracellular mechanisms underlying exercise intolerance and muscle weakness in patients with systemic sclerosis (SSc) are poorly understood. Thus, in six SSc patients (55 ± 11 yr) and six age-matched healthy controls (50 ± 13 yr), we assessed both skeletal muscle metabolism and intracellular O2 availability in vivo during submaximal contractions. Specifically, during dynamic plantar flexion exercise at 40% of maximal aerobic power, we used a multimodal approach combining phosphorus (31P) and proton (1H) MRS of deoxymyoglobin to examine both skeletal muscle energy pathways and

This study elucidated the role of the XKR8 gene in noise-induced hearing loss (NIHL) through animal experiments, PCR experiments, inner ear morphological experiments, ATP and mitochondrial complex I and II assays, and Western blotting experiments. The results showed that pre-exposure ABR showed no significant difference in baseline hearing thresholds between XKR8-/-and WT mice (P > 0.05). On day 1 post-exposure, ABR results indicated hearing thresholds around 85 dB in both groups. On days 3, 7, and 14, XKR8-110 dB mice had lower hearing thresholds than WT-110 dB mice (P < 0.05). Huntington's disease (HD) is a progressive neurodegenerative disorder in which mitochondrial dysfunction and impaired energy metabolism contribute to disease pathogenesis. Surprisingly, we find that ATP levels are not diminished but instead elevated in the striatum of R6/2 HD mice despite impaired TCA cycle intermediates and mitochondrial deficits. Integrative metabolomics, gene expression profiling, and pharmacological perturbation reveal that increased reactive oxygen species stabilize hypoxia-inducible factor-1α (HIF1α), driving enhanced glucose uptake and glycolytic flux. In vivo dynamic g

Plasma-activated water (PAW) is a promising technology for food disinfection with high efficiency and sustainability. However, there is a risk that PAW can induce the viable but non-culturable (VBNC) state in pathogens such as Salmonella, which brings a challenge to the practical application of PAW. This study suggests that the outer membrane protein PagC plays a critical role in the VBNC state formation in Salmonella Typhimurium under PAW stress. Flow cytometry analysis revealed that the proportion of VBNC cells in the pagC deletion mutant (ΔpagC) was 77.6%, lower than that in the wild type (

Trastuzumab resistance remains a major clinical barrier in the treatment of HER2-positive breast cancer. To identify therapeutic candidates capable of overcoming resistance, we established trastuzumab-resistant derivatives of SKBR3 and BT474 cells and performed a high-throughput screen of a modified ChemDiv Natural Product-Based Library. Myricetin emerged as a top hit with selective anti-survival activity against resistant cells while sparing normal breast epithelial cells. Mechanistic studies revealed that myricetin selectively suppressed mitochondrial complex I activity without affecting com

OBJECTIVE: Polyamine metabolism is known to be dysregulated in colorectal cancer (CRC), contributing to tumor progression, microenvironment acidification, and immune evasion. This study aimed to investigate the impact of polyamine pathway inhibition using α-difluoromethylornithine (DFMO) on extracellular acidification and energy metabolism in early-stage CRC tissues compared to non-cancerous controls.
METHODS: A total of 50 individuals (25 early-stage CRC patients and 25 non-cancer controls) provided colorectal tissue samples, which were subjected to a 24-hour ex vivo incubation with or witho

Temozolomide (TMZ) resistance remains a fundamental obstacle in the treatment of glioblastoma (GBM). While the metabolic sensor AMPK is known to influence cancer cell survival, the specific role of its regulatory γ-subunit isoforms in orchestrating chemoresistance is poorly understood. This study investigates how the dynamic remodeling of the AMPK heterotrimer contributes to TMZ evasion in GBM. We observed significantly low expression of AMPKγ2 in glioma patient samples, but the treatment of GBM cell lines with TMZ led to a robust increase in AMPKγ2 expression with a concomitant decrease in AM

Colorectal cancer (CRC) cells are 'addicted' to glutamine to satisfy energy and biosynthetic needs. Inhibiting glutamine metabolism enzymes, like glutaminase, is a potential cancer therapy strategy. Although the GLS inhibitor CB-839 is being evaluated in clinical trials, a comprehensive assessment of its antitumor activity in CRC cells is crucial. The present study aimed to evaluate the impact of CB-839 treatment on different CRC cell lines in terms of survival and proliferation. Furthermore, metabolic adaptations resulting from CB-839 treatment, particularly in energetic pathways, were invest

Glycogen synthase kinase 3 beta (GSK3β) is a pivotal serine/threonine kinase implicated in diverse pathological conditions, making it a compelling target for therapeutic intervention. In this study, we employed a structure-based drug discovery approach to identify novel ATP-competitive GSK3β inhibitors through a multi-tiered computational framework. Reported inhibitors from various repositories were systematically analysed to establish physicochemical and interaction-based filters, facilitating the rational curation of screening candidates. Toxicity assessment via Derek Nexus further refined t

OBJECTIVE: The blood-brain barrier (BBB) is a semi-permeable microvascular barrier, composed of endothelial cells conjoined by tight junction proteins. Following pathological conditions, i.e., traumatic brain injury (TBI), BBB dysfunction occurs, leading to microvascular hyperpermeability, resulting in cerebral edema formation and elevated intracranial pressure. Recent evidence suggests that the activation of pro-inflammatory signaling pathways is critical to BBB dysfunction. The NLRP3 inflammasome has been implicated as a key component of pro-inflammatory signaling. The aim of this study was
