Protocol of the PANEM trial: A randomized, phase III, parallel-arm, placebo-controlled, double-blind, multi-center superiority trial comparing PANcreatic EnzyMes versus placebo in adults after gastrectomy.

BACKGROUND: Patients after partial or total gastrectomy suffer from malnourishment and diminished quality of life (QOL). Due to altered physiological processes after gastric surgery, coordination between chyme and digestive enzymes produced by the pancreas is affected in many ways, finally leading to maldigestion and malabsorption. These in turn promote various symptoms such as discomfort or steatorrhea. Supplementation of digestive enzymes during food intake may improve digestion and decrease nutrition-related abdominal symptoms, finally leading to higher QOL. To date, trials examining the ro

Very early [18F]FDG-PET-guided targeted therapy in untreated advanced-stage classic Hodgkin lymphoma (EORTC-1537-COBRA): primary results of a single-arm, multicentre, phase 2 trial.

BACKGROUND: Brentuximab vedotin combined with doxorubicin, vinblastine, and dacarbazine (A-AVD) improves outcomes in advanced-stage classic Hodgkin lymphoma, but patients with a positive early interim PET have inferior prognosis. We evaluated whether very early [18F]fluorodeoxyglucose ([18F]FDG)-PET-guided treatment adaptation after one cycle of A-AVD improves activity while limiting exposure to intensive chemotherapy. METHODS: This single-arm, multicentre, phase 2 trial was conducted at 16 centres in seven countries (the Netherlands, Spain, Denmark, Belgium, Portugal, Slovakia, and Poland).

Study protocol for a single-arm phase Ib/II trial of Ivonescimab combined with adapted guided stereotactic body radiotherapy and chemotherapy in patients with locally advanced pancreatic cancer.

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) is an aggressive malignancy with limited effective treatment options, contributing to its high mortality of rate. Radiotherapy has shown effect in converting PDAC from a "cold tumor" to "hot tumor”, thus enhancing its response to immunotherapy. The synergistic effect of radiotherapy and immunotherapy may further improve therapeutic efficacy. Additionally, studies have demonstrated that vascular endothelial growth factor (VEGF) inhibitors can enhance the effectiveness of anti-programmed cell death 1/ligand 1 (anti-PD-1/PD-L1) antibodies by tra

Comparative efficacy and safety of neoadjuvant immunotherapy vs chemotherapy in resectable head and neck squamous cell carcinoma: an umbrella review of randomized controlled trials and single-arm studies.

BACKGROUND: The evolution of neoadjuvant therapies for resectable head and neck squamous cell carcinoma (HNSCC) has accelerated with the advent of immune checkpoint inhibitors. While platinum-based induction chemotherapy (ICT) shows modest benefits, emerging neoadjuvant immunotherapy has demonstrated unprecedented pathological response rates (PRR) in early-phase trials. However, significant controversy persists regarding the survival benefits and optimal integration of these strategies into multimodality paradigms. This umbrella review synthesizes meta-analytical evidence to evaluate whether n
