Increased expression of water channel aquaporin 1 and aquaporin 4 in Creutzfeldt-Jakob disease and in bovine spongiform encephalopathy-infected bovine-PrP transgenic mice.

Spongiform change is a cardinal feature in transmissible spongiform encephalopathies, including Creutzfeldt-Jakob disease (CJD) and bovine spongiform encephalopathy (BSE). It is characterized by swelling of the neuronal processes and vacuolization of the neuropil, leading to increased intraneuronal water content. The present study examines, by gel electrophoresis and Western blotting, the expression levels of the water channels aquaporin 1 (AQP1) and aquaporin 4 (AQP4) in the frontal cortex (area 8) homogenates of sporadic CJD cases (six men, four women; seven cases with methionine/methionine

Cellular basis of steroid neuroprotection in the wobbler mouse, a genetic model of motoneuron disease.

1. The Wobbler mouse suffers an autosomal recessive mutation producing severe motoneuron degeneration and astrogliosis in the spinal cord. It has been considered a suitable model of human motoneuron disease, including the sporadic form of amyotrophic lateral sclerosis (ALS). 2. Evidences exist demonstrating increased oxidative stress in the spinal cord of Wobbler mice, whereas antioxidant therapy delayed neurodegeneration and improved muscle trophism. 21-Aminosteroids are glucocorticoid-derived hydrophobic compounds with antioxidant potency 3 times higher than vitamin E and 100 times higher th

Immunohistochemical localization of capsular polysaccharide antigen in the central nervous system cells in cryptococcal meningoencephalitis.

Cryptococcal meningoencephalitis (CME) is caused by the encapsulated fungus Cryptococcus neoformans (CN) and is a major cause of mortality and morbidity in patients with AIDS. The polysaccharide capsule of CN is important for virulence, and soluble polysaccharide has the potential to cause immune modulation. To better understand the interactions of central nervous system cells and cryptococcal capsular polysaccharide (CNPS) in the pathogenesis of human CME, postmortem brain tissue from 21 patients with CME (13 AIDS and 8 non-AIDS patients) was analyzed. Histopathology and distribution of tissu

Adult primary whole-brain cultures from Thy1-aSyn mice (Line 61) retain Parkinson's disease-relevant phenotypes.

Primary cultures from fetal or postnatal rodent brain are widely used to study alpha-synuclein (aSyn)-associated mechanisms but poorly reflect age-dependent neurodegenerative processes. We established primary whole-brain cultures from adult Thy1-aSyn (Line 61) mice overexpressing human aSyn and compared them with postnatal and wild-type cultures. Cultures were maintained for 21 days and characterized by electrophysiology, immunohistochemistry, gene expression analysis, and protein biochemistry. Adult and postnatal cultures contained all major neural cell types, including neurons, microglia, as

The immunological perspective of major depressive disorder: unveiling the interactions between central and peripheral immune mechanisms.

Major depressive disorder is a prevalent mental disorder, yet its pathogenesis remains poorly understood. Accumulating evidence implicates dysregulated immune mechanisms as key contributors to depressive disorders. This review elucidates the complex interplay between peripheral and central immune components underlying depressive disorder pathology. Peripherally, systemic inflammation, gut immune dysregulation, and immune dysfunction in organs including gut, liver, spleen and adipose tissue influence brain function through neural and molecular pathways. Within the central nervous system, aberra

Alzheimer's Tau seeds-induced pathology enhances hippocampal extracellular diffusion.

The extracellular space (ECS) is a complex, dynamic network occupying about 20% of the brain, filled with a cerebrospinal fluid-like solution rich in extracellular matrix (ECM) molecules. ECS properties regulate molecular diffusion, potentially influencing disease-associated protein spread in neurodegenerative diseases. However, its role in tau propagation remains unexplored. Using wild-type mice injected with tau seeds purified from Alzheimer's disease brain, we applied quantum dot single-particle tracking in live tissue to examine how tau pathology and inflammation influence extracellular di

Cerebrospinal fluid extracellular vesicle-derived miR-9-3p in spinal cord injury with neuroprotective implications and biomarker development.

Spinal cord injury (SCI) often results in severe disability, and early detection of molecular changes is crucial for guiding treatment. In both rat and human samples, we observed a significant increase in cerebrospinal fluid (CSF)-derived extracellular vesicle (EV) miR-9-3p after SCI, prompting further investigation into its role. In a rat model, miR-9-3p levels were significantly lower at the injured spinal levels but higher in the motor cortex, where astrocytes showed the highest expression. Functional analyses revealed that miR-9-3p regulates energy metabolism, immune activity, and oxidativ

Systematic profiling reveals distinct senescence signatures and regulators across human brain cell types.

Cellular senescence contributes to age-related neurodegeneration, yet its manifestation varies across brain cell types and senescence-inducing stressors. Here, we investigated senescence hallmarks in five human brain cell lines - astrocytes, endothelial cells, microglia, oligodendrocytes, and dopaminergic-like neurons - using chronic 5-Bromodeoxyuridine treatment and validated our findings in primary cells and alternative toxin-induced models. Principal component analysis and transcriptional network inference identified both common and cell-type-specific senescence-associated transcriptional r

The dual effects of propofol down-regulating PD-L1 expression and inhibiting autophagy to reduce cerebral ischemia reperfusion injury.

Propofol, an anesthetic, has shown neuroprotective effects in animal and cellular models of cerebral ischemia-reperfusion (I/R) injury, likely by suppressing I/R-induced excessive autophagy. PD-L1 and immune escape are implicated in experimental stroke outcomes, but their mechanisms remain unclear. Here, we demonstrate that oxygen-glucose deprivation/reoxygenation (OGD/R) in astrocytes upregulates PD-L1 expression and autophagy, effects that are attenuated by propofol treatment via the NF-κB pathway. In vivo, propofol protects mice from I/R injury by downregulating PD-L1, inflammatory factors,

[Disseminated intravascular coagulation in critical obstetrical hemorrhage and severe trauma].

Disseminated intravascular coagulation (DIC) associated with critical obstetrical hemorrhage and severe trauma is classified as fibrinolytic DIC in terms of pathology and acute DIC in terms of progression. Obstetrical DIC is triggered by the influx of tissue factors from the placenta, amniotic fluid, and decidua into the maternal circulation. In contrast, trauma-related DIC is caused by vascular endothelial damage and exposure of subendothelial tissue. Specifically, in traumatic brain injury, tissue factors produced in the adventitia of cerebral blood vessels and astrocytes enter the circulati

Role and related mechanisms of non-invasive brain stimulation in the treatment of Tourette syndrome.

Tourette syndrome (TS) is a neurodevelopmental disorder characterized by impaired or delayed functional development. Although the pathology of TS remains to be determined, the continuous development of science and technology has provided new perspectives to understand its pathological mechanism. Research into non-invasive brain stimulation (NIBS) techniques, such as transcranial magnetic stimulation and direct current stimulation, have shown promising therapeutic potential in clinical studies. Furthermore, NIBS has been shown to affect the brain of patients with TS, including synaptic transmis

Pathogenic LRRK2 mutations cause loss of primary cilia and Neurturin in striatal parvalbumin interneurons.

Parkinson's disease-associated, activating mutations in the LRRK2 kinase block primary cilium formation in cell culture and in specific cell types in the brain. In the striatum that is important for movement control, about half of astrocytes and cholinergic interneurons, but not the predominant medium spiny neurons, lose their primary cilia. Here, we show that mouse and human striatal parvalbumin interneurons that are inhibitory regulators of movement also lose primary cilia. Without cilia, these neurons are not able to respond to Sonic hedgehog signals that normally induce the expression of P

Novel insights into Cntnap4 in Alzheimer's disease: Intestinal flora interaction.

Alzheimer's disease (AD) is a neurodegenerative disorder with unclear etiology. This study employs single-cell RNA sequencing (scRNA-seq), high-throughput transcriptome sequencing, 16s rRNA sequencing, and animal experiments to investigate the role of the contactin-associated protein like-4 (Cntnap4) gene in AD and its interaction with intestinal flora. We found that Cntnap4 deficiency in AD mice led to increased Tau protein phosphorylation, amyloid-beta plaque accumulation, and neuronal loss. Astrocytes in Cntnap4-/- mice showed impaired amyloid-beta processing. 16 s rRNA sequencing revealed

Connexin 43 hemichannels and related diseases.

Connexin 43 (Cx43) protein forms hemichannels (connexons) and gap junctions, with hemichannels consisting of six Cx43 molecules and gap junctions formed by two hemichannels. While gap junctions are prevalent in organs like the heart and liver, hemichannels are found in specific cell types, such as astrocytes and osteocytes. They allow the passage of small molecules (<1.5 kDa) between the cytoplasm and extracellular matrix. Cx43 hemichannels have emerged as potential therapeutic targets in various diseases, including central nervous system disorders, bone-related diseases, diabetic complication

The role of interferon beta in neurological diseases and its potential therapeutic relevance.

Interferon beta (IFNβ) is a member of the type-1 interferon family and has various immunomodulatory functions in neuropathological conditions. Although the level of IFNβ is low under healthy conditions, it is increased during inflammatory processes to protect the central nervous system (CNS). In particular, microglia and astrocytes are the main sources of IFNβ upon inflammatory insult in the CNS. The protective effects of IFNβ are well characterized in reducing the progression of multiple sclerosis (MS); however, little is understood about its effects in other neurological/neurodegenerative di

The roles of pyroptosis in the pathogenesis of autoimmune diseases.

The occurrence of autoimmune diseases is a result of the immune system's immune response against healthy components of the body. Pyroptosis is an innovative form of programmed cell death dependent on inflammatory caspases, leading to the release of cytokines. Excessive pyroptosis can lead to a sustained inflammatory response, which may aggravate the development of autoimmune diseases. In rheumatoid arthritis (RA), tumor necrosis factor (TNF) and NLRP3 enhance pyroptosis, exacerbating the disease. In systemic lupus erythematosus (SLE), the release of nuclear antigen promotes the development of

The Two-Way Route between Delirium Disorder and Dementia: Insights from COVID-19.

BACKGROUND: Delirium disorder is a frequent neurological complication of SARS-CoV-2 infection and associated with increased disease severity and mortality. Cognitive impairment is a major risk factor for developing delirium disorder during COVID-19, which, in turn, increases the risk of subsequent neurological complications and cognitive decline. SUMMARY: The bidirectional connection between delirium disorder and dementia likely resides at multiple levels, and its pathophysiological mechanisms during COVID-19 include endothelial damage, blood-brain barrier dysfunction, and local inflammation,

HIV infection of non-classical cells in the brain.

HIV-associated neurological disorders (HAND) affect up to 50% of people living with HIV (PLWH), even in the era of combination antiretroviral therapy (cART). HIV-DNA can be detected in the cerebral spinal fluid (CSF) of approximately half of aviremic ART-suppressed PLWH and its presence is associated with poorer neurocognitive performance. HIV DNA + and HIV RNA + cells have also been observed in postmortem brain tissue of individuals with sustained cART suppression. In this review, we provide an overview of how HIV invades the brain and HIV infection of resident brain glial cells (astrocytes a

Microglial cannabinoid receptor type 1 mediates social memory deficits in mice produced by adolescent THC exposure and 16p11.2 duplication.

Adolescent cannabis use increases the risk for cognitive impairments and psychiatric disorders. Cannabinoid receptor type 1 (Cnr1) is expressed not only in neurons and astrocytes, but also in microglia, which shape synaptic connections during adolescence. However, the role of microglia in mediating the adverse cognitive effects of delta-9-tetrahydrocannabinol (THC), the principal psychoactive constituent of cannabis, is not fully understood. Here, we report that in mice, adolescent THC exposure produces microglial apoptosis in the medial prefrontal cortex (mPFC), which was exacerbated in a mod
