Neuro-visual pathway after mild traumatic brain injury: a systematic scoping review of symptoms, objective testing, and rehabilitation.

Visual complaints are common after mild traumatic brain injury (mTBI), yet the scope, assessment approaches, mechanisms, and rehabilitation strategies reported in the literature remain heterogeneous. To synthesize evidence on post-concussive visual symptoms, diagnostic/assessment approaches, treatment/rehabilitation strategies, and putative mechanisms. Relevant studies published between 1997 and 2025 were identified and screened; 57 studies reporting visual outcomes following mild traumatic brain injury were included. The literature comprised cohort and cross-sectional studies, randomized and

An Adaptive, Time-Informed, Phenotype-Guided Pharmacological Framework for Severe Traumatic Brain Injury : A Hypothesis-Driven Narrative Review.

Severe traumatic brain injury (TBI) remains difficult to treat because successful control of mass lesions, intracranial pressure, cerebral perfusion, oxygenation, and systemic instability does not ensure that secondary cellular injury has stopped. PubMed/MEDLINE and ClinicalTrials.gov were searched through July 28, 2026, and representative candidates were organized across five overlapping pathophysiological nodes. Evidence was described by study source, purpose, and result direction rather than assigned a formal quality grade. Human evidence was heterogeneous. Glibenclamide/glyburide and hyper

Economic burden of adult mild traumatic brain injury in the United States: a scoping review of healthcare-related charges and costs.

Mild traumatic brain injury (mTBI) accounts for 80-90% of all traumatic brain injuries, yet its economic burden remains poorly characterized. We synthesized direct healthcare charges and costs associated with mTBI in the United States (U.S.). A PubMed search through October 14, 2025 identified primary studies reporting direct healthcare-related charges or costs among adults with mTBI. All estimates were standardized to 2025 U.S. dollars. Twenty-one studies (1996-2023) out of 3300 screened (0.64%) met inclusion criteria. Index hospitalization costs ranged from $3984 to $31,316 and remained stab

Deletion of NLRP3 gene blocks traumatic brain injury induced abnormal immune response in mice with a genetic background for AD.

Traumatic brain injury (TBI) is a significant risk factor for the development of Alzheimer's disease (AD) and related dementia. In both TBI and AD, inflammation plays a pivotal role. It is known that the NLRP3 inflammasome plays an important role in AD pathogenesis while TBI triggers activation of the NLRP3 inflammasome in the brain. To evaluate the importance of the NLRP3 inflammasome in mediating the TBI-induced immune response in animals with a genetic background for AD, we have examined immune profiles in the brain using a novel transgenic mouse line at 3xTg background with the NLRP3 gene

Can Oxygen Induced Vasomotion Be Harnessed in Traumatic Brain Injury? A Narrative Review of the Relationship Between Oxygenation and Intracranial Pressure.

The role of cerebral autoregulation in the management of traumatic brain injury (TBI) is of increasing interest. Though oxygen is potently vasoactive, its role in autoregulation is under-appreciated despite the importance of brain oxygen monitoring and pervasive availability of supplemental oxygen in clinical environments. This narrative review intends to provide historical scientific context for oxygen-driven cerebral autoregulation-hereby termed oxygenic autoregulation-and its therapeutic potential, particularly during intracranial pressure (ICP) crises. A review of original research from 19

Heterogenous microglial reactivity contrasts with stable vascular transcriptional programs in mouse models of Alzheimer's, CADASIL, and Traumatic Brain Injury.

The extent to which the cerebrovasculature is affected in various brain disorders is still not well understood. To address this, we established a transcriptomic repository of major vascular cell types and microglia to compare the global transcriptomic response in mouse models of three human brain disorders linked to neuroinflammation and associated vascular reactivity: Alzheimer's disease (AD), traumatic brain injury (TBI), and cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL). Single-cell analysis of >250,000 cells at different disease stages

Retraining dorsal visual pathways improves cognitive skills and executive control networks following mild traumatic brain injury.

BACKGROUND AND OBJECTIVES: Mild traumatic brain injury (mTBI) frequently results in persistent cognitive deficits with limited evidence-based rehabilitation options. Visual timing deficits, potentially linked to dorsal visual pathway dysfunction, represent a promising therapeutic target. This study examined whether a novel intervention targeting dorsal visual pathways (PATH) demonstrates superior efficacy compared to conventional cognitive therapies targeting ventral pathway deficits or working memory impairments. METHODS: Twenty-four participants (aged 23-62 years) with mTBI were randomized

Hepatocyte Growth Factor/MET Activator Rescues Working Memory Deficits after Repeated Mild Traumatic Brain Injury.

Mild traumatic brain injury (mTBI) can produce persistent cognitive and behavioral deficits. These impairments result in a reduced quality of life and difficulty returning to work, school, or other activities. Individuals with repeated injuries show increased risk for greater cognitive impairment and persistence of symptoms. A variety of pharmacological approaches have been tried to limit cognitive symptoms and other aspects of secondary injury following mTBI. However, their efficacy and ability to treat the sequela of mTBI remains disputed and no FDA-approved drug exists for mTBI. However, ne

Dynamic proteomic and phosphoproteomic analysis reveals key pathways and targets in the early stages of high-altitude traumatic brain injury.

Traumatic brain injury (TBI), particularly at high altitudes (HA-TBI), is a leading cause of mortality and disability, yet clear diagnostic and treatment protocols are lacking. This study explores the early pathophysiological changes occurring within 24 h following HA-TBI, with a focus on differentially expressed proteins (DEPs) and phosphorylated proteins (DEPPs). Using a low-pressure hypoxic chamber to simulate high-altitude conditions combined with a controllable cortical impact (CCI) model, we established a rat model of HA-TBI. Neurological function was evaluated using the modified Neurolo

An iterative ROC procedure identifies white matter tracts diagnostic for traumatic brain injury: an exploratory analysis in U.S. Veterans.

OBJECTIVE: Understanding the pathophysiology of traumatic brain injury (TBI) is crucial for effectively managing care. Diffusion tensor imaging (DTI) is an MRI technology that evaluates TBI pathology in brain white matter. However, DTI analysis generates numerous measures. Choosing between them remains an obstacle to clinical translation. In this study, we leveraged an iterative receiver operating characteristic (ROC) analysis to examine white matter tracts in a group of 380 Veterans, consisting of TBI (n = 243) and non-TBI patients (n = 137). METHOD: For each participant, we obtained a whole
