Multicenter performance evaluation of the Simplexa C. auris Direct assay for the detection of Candida auris colonization in bilateral axilla/groin swabs.

UNLABELLED: Candida auris is a multidrug-resistant fungal pathogen associated with healthcare outbreaks and high mortality. Its accurate and timely identification is critical for infection prevention, yet conventional culture-based methods are limited by slow growth, morphological ambiguity, and potential misidentification. This multicenter study evaluated the performance of the Simplexa C. auris Direct assay using 2,020 axilla/groin swab specimens collected from six clinical sites compared to culture followed by matrix-assisted laser desorption ionization-time of flight mass spectrometry (MAL

Evaluation of Adhesion and Co-culture Efficacy Assays of Antifungals and Chlorhexidine Against Acanthamoeba and Candida Species Associated with Severe Infectious Keratitis.

Severe infectious keratitis is a leading cause of corneal transplants and vision loss, primarily caused by microorganisms such as the protozoan genus Acanthamoeba spp. and the yeast Candida species. Treating these infections poses significant challenges, particularly in cases of polymicrobial keratitis. Both Acanthamoeba spp. and Candida species can adhere to contact lenses and the patient cornea, forming complex polymicrobial biofilms. This is the first study to explore polymicrobial biofilm formation involving Acanthamoeba and Candida. This study to evaluate adhesion and the co-culture betwe

Expression of mannanase and glucanases in lettuce chloroplasts and functional evaluation of enzyme cocktail against Candida albicans in oral cancer patient samples.

Candida albicans is a human pathogen responsible for several diseases. C. albicans cell wall contains chitin, glucan and mannan. Therefore, this study evaluated efficacy of enzyme cocktail containing lettuce-expressed chitinases, glucanases and mannanase against C. albicans in cell culture or cancer patient samples. Site-specific integration of the Man1, CelO, Cbh1, Cbh2 expression cassettes and removal of the selectable marker gene from lettuce transplastome was confirmed using three sets of PCR primers. Homoplasmy in transplastomic lines was confirmed in Southern blots by the absence of untr

The effects of fabrication methods and build orientation on Candida albicans adhesion on 3D-printed and conventional denture resin: an in vitro comparative study.

BACKGROUND: In dentistry, interest in computer-aided design and computer-aided manufacturing (CAD/CAM) over traditional denture fabrication systems is increasing. However, few studies have compared the physiochemical and biological properties of different fabrication methods in regards to Candida albicans. C. albicans is the most common oral fungal pathogen, often related to dentures and elderly patients. Although various factors have been reported to influence C. albicans proliferation, conclusive evidence is lacking. Therefore, the aim of this study was to evaluate and compare the physicoche

Phosphoglycerate mutase and methionine synthase act as adhesins of Candida albicans to the corneal epithelium, altering their expression during the tissue adhesion process.

The yeast form of Candida albicans uses glycosaminoglycans (GAGs), primarily heparan sulfate, as adhesion receptors for corneal epithelial cells. However, during the transition to the hyphal form, the fungus shifts to using alternative receptors. This study aims to identify fungal adhesins involved in GAG binding and examine their expression dynamics during tissue adhesion. Using chromatography, three proteins from the C. albicans cell wall with high affinity for heparin were identified: methionine synthase, phosphoglycerate mutase, and cytochrome c. These proteins were overexpressed in Escher

Comparative Antimicrobial and Oxidative Damage of Indocyanine Green, Methylene Blue, and Curcumin on Dual-Species Biofilms of Enterococcus faecalis and Candida albicans.

Failure of the endodontic treatment is often associated with persistent polymicrobial biofilms, particularly those involving Enterococcus faecalis (E. faecalis) and Candida albicans (C. albicans), which display synergistic pathogenicity and resistance to standard disinfection methods. This in vitro study compared the antimicrobial activity and oxidative damage induced by indocyanine green (ICG)-mediated laser ablation (LA) with that produced by antimicrobial photodynamic therapy (aPDT) using methylene blue (MB) or curcumin (CUR) in root canals infected with dual-species biofilms. Two promising Bacillus-derived antifungal lipopeptide leads AF4 and AF5 and their combined effect with fluconazole on the in vitro Candida glabrata biofilms.

Introduction: Candida species are endowed with the ability to produce biofilms, which is one of the causes of pathogenicity, as biofilms protect yeasts from antifungal drugs. Candida glabrata (Nakaseomyces glabrata) is one of the most prevalent pathogenic yeasts in humans and a biofilm producer. Methods: The study was aimed at evaluating the combined effects of two highly promising antifungal biomolecules (AF4 and AF5) lipopeptide in nature, chromatographically purified to homogeneity from Bacillus subtilis (B. subtilis) and the standard antifungal fluconazole (at different concentrations) to

Crystal structure of urethanase from Candida parapsilosis and insights into the substrate-binding through in silico mutagenesis and improves the catalytic activity and stability.

Ethyl carbamate (EC) is classified as a Class 2A carcinogen, and is present in various fermented foods, posing a threat to human health. Urethanase (EC 3.5.1.75) can catalyze EC to produce ethanol, CO2 and NH3. The urethanase (cpUH) from Candida parapsilosis can hydrolyze EC, but its low affinity and poor stability hinder its application. Here, the structure of cpUH from Candida parapsilosis was determined with a resolution of 2.66 Å. Through sequence alignment and site-directed mutagenesis, it was confirmed that cpUH contained the catalytic triad Ser-cisSer-Lys of the amidase family. Then, th

Analogues of the anti-malaria drug mefloquine have broad spectrum antifungal activity and are efficacious in a model of disseminated Candida auris infection.

Only three classes of antifungal drugs are currently in clinical use. Here, we report that derivatives of the malarial drug mefloquine have broad spectrum antifungal activity including difficult to treat molds and endemic fungi. Pharmacokinetic and efficacy studies of NSC-4377 indicate it penetrates the central nervous system and is active against Candida auris in vivo. These data strongly support the further development of mefloquine analogs as a potentially new class of antifungal molecules.

Precise genome editing underlines the distinct contributions of mutations in ERG11, ERG3, MRR1, and TAC1 genes to antifungal resistance in Candida parapsilosis.

Candida parapsilosis has recently emerged as a major threat due to the worldwide emergence of fluconazole-resistant strains causing clonal outbreaks in hospitals and poses a therapeutic challenge due to the limited antifungal armamentarium. Here, we used precise genome editing using CRISPR-Cas9 to gain further insights into the contribution of mutations in ERG11, ERG3, MRR1, and TAC1 genes and the influence of allelic dosage to antifungal resistance in C. parapsilosis. Seven of the most common amino acid substitutions previously reported in fluconazole-resistant clinical isolates (including Y1

Isolation of Candida Species Is Associated with Comorbidities, Prolonged Mechanical Ventilation, and Treatment Outcomes in Surgical ICU Patients, a Cross-Sectional Study.

The isolation of Candida may be related to comorbidity, prolonged mechanical ventilation, and survival during intensive care unit (ICU) stay, especially with non-albicans Candida (NAC). To examine the frequency of Candida isolation, associated comorbidities and outcomes in the surgical ICU in Osijek University Hospital, Croatia, the data from the electronic database from May 2016 to 30 June 2023 were analyzed. In a cross-sectional study examining 15,790 microbiological samples, different strains of Candida were observed in 581 samples from 236 patients. The control group (N = 261) was 130 cons
