Semiparametric discovery and estimation of interaction in mixed exposures using stochastic interventions.

Understanding the complex interactions among multiple environmental exposures is critical for assessing their combined impact on health outcomes. This study introduces InterXshift, a novel semiparametric method that provides a nonparametric definition of interaction and facilitates both the discovery and efficient estimation of interaction effects in mixed exposures. Leveraging stochastic shift interventions and ensemble machine learning, InterXshift identifies and quantifies interactions through a model-independent target parameter, estimated using targeted maximum likelihood estimation (TMLE

Cerebrospinal fluid and plasma metabolites in Parkinson's disease: a Mendelian randomization study.

Metabolites in cerebrospinal fluid (CSF) and plasma have been implicated in Parkinson's disease (PD), but their causal roles remain unclear. This study aims to investigate the causal relationships between specific CSF and plasma metabolites and PD risk, using Mendelian Randomization (MR) analysis. We utilized MR with inverse-variance weighting (IVW) as the primary analytical method, supplemented by four additional MR models to validate robustness. Data included 1,400 plasma metabolites from the Canadian Longitudinal Study on Aging, and 338 CSF metabolites from the Wisconsin Alzheimer's Disease

COVID-19 Impacts on Children and Resources for Resilience: What is the Direction of Causation?

COVID-19 has profoundly affected children's well-being. Resilience was often found to be negatively correlated with COVID-19 impacts. However, the role of resilience in directly shaping COVID-19 impacts on children remains unclear, as studies report conflicting evidence regarding the potential causal direction between COVID-19 impacts and resilience. Higher resilience could reduce COVID-19 impacts, while COVID-19 impacts may also disrupt or enhance resilience. Or it could be the case that the two are simply correlated due to shared underlying common factors. This preregistered study uses a twi

Serum urate levels and colorectal cancer risk: a prospective cohort study in of the UK biobank and a Mendelian randomization analysis.

BACKGROUND: Serum urate is the most abundant antioxidant molecule in human blood and may play a role in cancer prevention. However, the association between serum urate levels and colorectal cancer (CRC) risk remains inconclusive, with the underlying causal mechanisms still undefined. METHODS: In the UK Biobank (UKB) cohort, we investigated the prospective association between serum urate levels and the risk of CRC. We used Cox proportional hazards models to estimate the multivariable hazard ratios (HR) for CRC. Subgroup analyses were performed based on anatomical subsite and sex. Additionally,

A genetic exploration of the relationship between posttraumatic stress disorder and cardiovascular diseases.

Experiencing a traumatic event may lead to Posttraumatic Stress Disorder (PTSD), including symptoms such as flashbacks and hyperarousal. Individuals suffering from PTSD are at increased risk of cardiovascular disease (CVD), but it is unclear why. This study assesses shared genetic liability and potential causal pathways between PTSD and CVD. We leveraged summary-level data of genome-wide association studies (PTSD: N = 1,222,882; atrial fibrillation (AF): N = 482,409; coronary artery disease (CAD): N = 1,165,690; hypertension (HT): N = 458,554; heart failure (HF): N = 977,323). Genetic determinants of blood-cell traits and the risk of cardiometabolic Diseases: Triangulating evidence from Mendelian randomization and observational analyses.

BACKGROUND AND AIMS: Blood cell traits (BCTs) are linked to cardiometabolic diseases (CMDs) through mechanisms such as inflammation, thrombosis, endothelial dysfunction, and lipid metabolism. BCTs could serve as readily available, cost-effective biomarkers to enhance CVD risk prediction and, if proven causal, may also emerge as novel therapeutic targets. This study examined the genetic architecture and tested potential causal relations between 29 BCTs and 20 CMDs. METHODS: We employed univariable Mendelian randomization (UVMR) using inverse-variance weighting as the primary method, MR-Egger a

Interactive Effect of Plasma Lipidome on Neuropsychiatric Disorders: A Bidirectional Mendelian Randomization Study.

Lipid metabolism disruptions are implicated in nervous and mental disorders; plasma lipidomics show promise as diagnostic and therapeutic targets. However, clarification of causal relationships between plasma lipids and neuropsychiatric disorders (NPDs) remains unclear. We employed bidirectional Mendelian randomization (MR) to examine causal links between 179 plasma lipids and 17 NPDs. Significant associations in the forward MR were further investigated using linkage disequilibrium score regression (LDSC), Bayesian colocalization (COLOC) analysis, and mediation analysis. Forward MR revealed st

Immune cells and the risk of acne vulgaris: A Mendelian randomization study.

Acne vulgaris is a chronic inflammatory disease affecting the pilosebaceous glands. It primarily manifests on the face, chest, and back, and is prevalent among adolescents of both genders. The mechanism behind it is significantly associated with the inflammatory response triggered by microbial infections and the infiltration of immune cells. However, the precise causal role of immune cells in acne vulgaris is unclear. To this end, we conducted a 2-sample Mendelian randomization analysis to evaluate the relationship between immune cells and the likelihood of developing acne vulgaris. Instrument

Genetic evidence of dietary patterns and diet-derived antioxidants on polyp development through gut microbiota modulation.

Previous studies have shown that dietary habits are associated with the development of several diseases in humans. However, whether these factors contribute to polyps remains unknown. In this study, we employ Mendelian randomization (MR) to investigate the causal relationship between genetically predicted dietary factors, including diet-derived antioxidant intake, and 4 types of polyps: nasal polyps, female genital tract polyps, colorectal polyps, and gastric polyps, as well as to explore the mediating role of the gut microbiota. We identified genetic variants associated with 7 dietary sources

Relation of 91 Circulating Inflammatory Proteins to Nonalcoholic Fatty Liver Disease: A Two-Sample Mendelian Randomisation Study.

Increasingly, emerging research evidence has demonstrated that nonalcoholic fatty liver disease (NAFLD) is a disease closely associated with systemic inflammation. However, the specific upstream inflammatory factors engaged in the pathogenesis of NAFLD remain unclear. Our study aimed to identify the inflammatory regulators causally associated with NAFLD pathogenesis through Mendelian randomisation. A two-sample Mendelian randomisation method was applied to analyse the causal association between 91 circulating inflammatory proteins and NAFLD. Data on circulating inflammatory proteins were deriv
