Development of human ectocervical tissue models with physiologic endocrine and paracrine signaling†.

There is a shortage of research models that adequately represent the unique mucosal environment of human ectocervix, limiting development of new therapies for treating infertility, infection, or cancer. We developed three microphysiologic human ectocervix models to study hormone action during homeostasis. First, we reconstructed ectocervix using decellularized extracellular matrix scaffolds, which supported cell integration and could be clinically useful. Secondly, we generated organotypic systems consisting of ectocervical explants co-cultured with murine ovaries or cycling exogenous hormones

Crystallization and preliminary X-ray diffraction analysis of kanamycin-binding β-lactamase in complex with its ligand.

TEM-1 β-lactamase is a highly efficient enzyme that is involved in bacterial resistance against β-lactam antibiotics such as penicillin. It is also a robust scaffold protein which can be engineered by molecular-evolution techniques to bind a variety of targets. One such β-lactamase variant (BlaKr) has been constructed to bind kanamycin (kan) and other aminoglycoside antibiotics, which are neither substrates nor ligands of native β-lactamases. In addition to recognizing kan, BlaKr activity is up-regulated by its binding via an activation mechanism which is not yet understood at the molecular le

Exercise-induced bronchoconstriction.

While mainstream therapies for androgenetic alopecia (AGA), including pharmacological treatments, mesotherapy, low-level light therapy, and hair transplantation, often fall short of clinical needs due to limited efficacy, side effects, or invasiveness, regenerative medicine based on stem cells shows promise yet faces challenges related to immunogenicity, standardization, and safety. Against this backdrop, extracellular vesicles (EVs)-nanoscale signaling vesicles enclosed by a lipid bilayer-have rapidly emerged as an innovative "cell-free" therapeutic paradigm. Potentiated Maturation of hPSC-Derived Dopaminergic Neurons via Convergent Genetic and Small Molecule Modulation.

BACKGROUND: Ventral midbrain dopaminergic (vmDA) neurons derived from human pluripotent stem cells provide a powerful platform for modeling Parkinson's disease (PD) and developing therapeutic strategies. Although robust protocols exist for generating vmDA progenitors, maturation into functionally competent neurons typically requires prolonged culture periods and occurs with substantial heterogeneity in maturity, representing major bottlenecks. METHODS: Building on established protocols for vmDA progenitor differentiation, we introduce a sequential strategy that specifically targets the subseq

Emerging CAR-Treg and regulatory T cell therapies for inflammatory bowel diseases: a systematic review of a new era of treatment.

BACKGROUND: Inflammatory bowel disease (IBD) is a chronic immune-mediated disorder characterized by dysregulated mucosal immunity. Despite major advances in therapies, a high proportion of patients do not achieve or sustain remission. Cellular immunotherapies have emerged as a mechanistically distinct therapeutic strategy with potential relevance for refractory disease. In the present paper, the authors aim to evaluate the rationale and current evidence supporting chimeric antigen receptor (CAR)-engineered immune cells and regulatory T (Treg)-cell-based therapies in IBD. Nitric oxide-primed engineered extracellular vesicles restore bioenergetics in acute kidney injury via mitochondrial transfer.

Background: The disruption of mitochondrial homeostasis in acute kidney injury (AKI) is an important factor that drives persistent renal dysfunction. Mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) have shown great therapeutic potential in AKI, but insufficient specificity of targeting the impaired mitochondrial function. Herein, we developed an engineered nitric oxide (NO)-primed MSC-EVs (pEVs) to restore mitochondrial homeostasis for AKI therapy. Methods: A cisplatin-induced AKI model was established to investigate the therapeutic effects of MSC-EVs. Proteomic and Western blot

The cell-assembled extracellular matrix: A focus on the storage stability and terminal sterilization of this human "bio" material.

The Cell-Assembled extracellular Matrix (CAM) is an attractive biomaterial because it provided the backbone of vascular grafts that were successfully implanted in patients, and because it can now be assembled in "human textiles". For future clinical development, it is important to consider key manufacturing questions. In this study, the impact of various storage conditions and sterilization methods were evaluated. After 1 year of dry frozen storage, no change in mechanical nor physicochemical properties were detected. However, storage at 4 °C and room temperature resulted in some mechanical ch

The generation and characterization of novel Col1a1FRT-Cre-ER-T2-FRT and Col1a1FRT-STOP-FRT-Cre-ER-T2 mice for sequential mutagenesis.

Novel genetically engineered mouse models using the Cre-loxP or the Flp-FRT systems have generated useful reagents to manipulate the mouse genome in a temporally-regulated and tissue-specific manner. By incorporating a constitutive Cre driver line into a mouse model in which FRT-regulated genes in other cell types are regulated by Flp-FRT recombinase, gene expression can be manipulated simultaneously in separate tissue compartments. This application of dual recombinase technology can be used to dissect the role of stromal cells in tumor development and cancer therapy. Generating mice in which

Spatial control of PEG-lipid interfaces at cell surface enables on-demand adhesion and enzymatic detachment of suspension cells.

Suspension cells are widely used in biomedical research and cell-based therapies; however, their lack of stable adhesion to substrates limits their efficient handling, including gene delivery and downstream processing. Although surface engineering approaches have been studied to induce cell adhesion, precise control over attachment and detachment, comparable to that of adherent cells, remains challenging. Here, we report a strategy to control cell adhesion and detachment using enzyme-responsive trans-activator of transcription peptide-poly(ethylene glycol)-lipid (Tat-PEG-lipid) conjugates and

PURPOSE: The value of cell-free techniques in the treatment of cartilage defects remains under debate. In this study, cartilage repair of full-thickness chondral defects in the knees of Goettinger minipigs was assessed by treatment with a cell-free collagen type-I gel or a collagen type-I gel seeded with autologous chondrocytes. As a control, abrasion arthroplasty was included. METHODS: In 18 adult Goettinger minipigs, three full-thickness chondral defects were created in one knee of the hind leg. They were either treated with a cell-free collagen gel, a collagen gel seeded with 2 × 10(5)/ml

DUSP6 ablation restores CAR T-cell fitness impaired by tumor CD58 loss through invigoration of AP-1 signaling.

Primary resistance to chimeric antigen receptor (CAR) T-cell therapies has limited their widespread application. Our prior genome-wide CRISPR/Cas9 screening revealed that the loss of CD58, a crucial intrinsic resistance factor in tumors, resulted in insufficient immune synapse formation and impaired CAR T-cell activation and cytotoxicity. However, the specific signaling pathway and transcriptional changes associated with CAR T-cell dysfunction have not been addressed. Here, we revealed that AP-1-mediated activation was attenuated in CAR T cells impaired by tumor CD58 loss, driving a decrease i

Why does siRNA delivery still fail in neurodegenerative diseases? Biological barriers, nanocarrier engineering, and future translational directions.

Neurodegenerative diseases remain largely untreatable at the disease-modifying level, with currently approved therapies providing primarily symptomatic benefits. Small interfering RNA (siRNA) has emerged as a promising therapeutic modality because of its ability to selectively silence disease-associated genes. However, a major obstacle to clinical translation is not the gene-silencing mechanism itself, but the efficient delivery of therapeutic siRNAs to target cells within the central nervous system. This review examines the barriers that limit siRNA-based therapies for neurodegenerative diseas

Molecular mechanisms and therapeutic strategies for the recurrent F9 (c.520 + 13 A > G) variant in hemophilia B.

BACKGROUND: Hemophilia B (HB), an X-linked recessive disorder, results from variants in the coagulation factor IX gene (F9). The F9 c.520 + 13 A > G variant is a recurrent intronic variant in HB patients, accounting for 15.05% of all documented F9 intronic variants. Despite prior predictions of its impact, the molecular mechanism associated with moderate to mild HB remains undissected. MATERIALS AND METHODS: In silico predictions and splicing-competent cDNA constructs were used to assess the impact of F9 c.520 + 13 A > G on mRNA splicing. Factor IX (FIX) variant (p.V174delinsGHNLM) expression

Heterostructure engineering of Au@UiO-67-bpy(Cu)/MoS2 cascade nanozymes for self-fueled and NIR-enhanced chemodynamic/photothermal cancer therapy.

The development of multifunctional nanozymes capable of integrating catalytic cascade reactions with externally controllable therapeutic activation remains an important challenge in cancer nanomedicine. Herein, we report a heterostructure-engineered cascade nanozyme, Au@UiO-67-bpy(Cu)/MoS2, constructed by integrating a catalytically active Au@UiO-67-bpy(Cu) nanozyme with NIR-responsive MoS2 nanosheets. Unlike conventional UiO-67-based nanotherapeutics that commonly rely on post-synthetic incorporation of photoactive complexes or drug/photosensitizer payloads, this strategy introduces NIR funct

Recombinant Expression and Engineering of MDI- and HDI-Specific Monoclonal Antibodies for Proof-of-Concept Detection of Diisocyanate Adducts and Candidate Assay Calibrators.

Methylene-diphenyl diisocyanate (MDI) and hexamethylene diisocyanate (HDI), important chemicals for polyurethane production, can react with self-molecules, trigger immune responses, and cause asthma. To validate molecular determinants of diisocyanate immune recognition and develop renewable immunoassay reagents, we generated MDI- and HDI-specific recombinant monoclonal antibodies (mAbs) from hybridoma-derived variable-region DNA sequences using a human embryonic kidney (HEK) cell line expression system. Three anti-MDI and one anti-HDI mouse IgG1/κ mAbs were produced, purified, and evaluated fo

A synergistic strategy involving reverse-adaptation and engineered MSC exosomes against ferroptosis in osteoarthritis.

Osteoarthritis (OA) progression is driven by persistent oxidative stress and ferroptosis, which erode chondrocyte viability and extracellular matrix integrity. Although mesenchymal stem cell-derived exosomes (MSC-EXO) hold regenerative promise, their native cargo lacks the adaptability to withstand such a hostile microenvironment, limiting therapeutic efficacy. Here, we demonstrated a synergistic strategy involving reverse-adaptation and engineered MSC exosomes against ferroptosis in osteoarthritis. Firstly, the reverse-adaptation strategy in which OA-like oxidative stress was harnessed to pre

Desmoplakin loss leads to PKC-dependent insertion of series sarcomeres and contractile dysfunction in cardiomyocytes.

BACKGROUND: Mutations in DSP, which encodes the protein desmoplakin, lead to cardiomyopathy with unusually high penetrance. Clinical features include ventricular tachyarrhythmias, fibro-fatty infiltration of both ventricles, and ultimately dilated cardiomyopathy. While some data have been gathered to explain the electrophysiological and contractile consequences of desmoplakin cardiomyopathy, a comprehensive mechanism linking DSP mutations to ventricular dilation and heart failure remains elusive. METHODS: We use iPSC-derived engineered heart tissue (EHT) bearing a functional desmoplakin haplo
