OBJECTIVES: Presbyacusis, one of the most common ailments of the elderly, is often treated with hearing aids, which serve to reintroduce some or all of those sounds lost to peripheral hearing loss. However, little is known about the underlying changes to the ear and brain as a result of such experience with sound late in life. The present study attempts to model this process by rearing aged CBA mice in an augmented acoustic environment (AAE).
DESIGN: Aged (22-23 months) male (n = 12) and female (n = 9) CBA/CaJ mice were reared in either 6 weeks of low-level (70 dB SPL) broadband noise stimula

BACKGROUND: Metastatic melanoma is a highly aggressive skin cancer and currently resistant to systemic therapy. Melanomas may involve genetic, epigenetic and metabolic abnormalities. Evidence is emerging that epigenetic changes might play a significant role in tumor cell plasticity and metastatic phenotype of melanoma cells.
PRINCIPAL FINDINGS: In this study, we developed a systematic approach to identify genes implicated in melanoma progression. To do this, we used the Affymetrix GeneChip Arrays to screen 34,000 mouse transcripts in melan-a melanocytes, 4C pre-malignant melanocytes, 4C11- no

Estrogens are synonymous with fertility and infertility in mammals. Our knowledge of the biological actions of estrogens, however, is incomplete. Three recent developments have thrown new light on the actions of estrogens in mammalian reproduction that will lead to a greater understanding of their functions. They are (a) the identification of a second estrogen receptor, called ERbeta, (b) the identification of ligand-specific ER coactivators and (c) mouse models with targeted disruption of the genes encoding both ER and the aromatase enzyme. These models provide for the first time animals whic

Freshly isolated human leukemia cells have been shown in the past to display varying in vitro responses to phorbol diesters, depending on their cell type. Specific receptors for the phorbol diesters have been demonstrated on numerous different cells. This study was designed to characterize the receptors for phorbol diesters on leukemia cells freshly isolated from patients with different kinds of leukemia and to determine if differences in binding characteristics for tritium-labeled phorbol 12,13-dibutyrate (3H-PDBu) accounted for the different cellular responses elicited in vitro by phorbol di

Malignant rhabdoid tumors (MRTs) are highly aggressive pediatric tumors associated with loss of expression of SMARCB1, commonly occurring in the central nervous system [referred to as atypical teratoid/rhabdoid tumors (AT/RTs)] and in the kidney and soft tissues. Histologically, MRTs are characterized by immunohistochemical evidence of primitive neuroectodermal, mesenchymal, and epithelial differentiation. The ability of MRTs to differentiate along multiple lines, as evidenced by both histologic features and polyphenotypic immunohistochemical staining, and the proliferative nature of MRT cells

BACKGROUND: The transformed small-cell lung cancer (T-SCLC) has a dismal prognosis, yet there remains a lack of standardized therapeutic guidelines. While this transformation is predominantly driven by concurrent loss of TP53 and RB1, retaining wild-type RB1 is exceptionally rare. We report a rare case of RB1-wildtype T-SCLC with prolonged survival to highlight its diagnostic challenges, potential biological mechanisms, and therapeutic management.

BACKGROUND: Triggering receptor expressed on myeloid cells 2 (TREM2) is a pivotal lipid-sensing receptor that governs the functional plasticity and metabolic fitness of mononuclear phagocytes, including microglia, osteoclasts, dendritic cells and tissue-resident macrophages. While originally identified as a genetic risk factor for Alzheimer's disease (AD), TREM2 functions as a context-dependent immunometabolic checkpoint rather than a uniform phenotypic driver, exerting divergent biological outputs dictated by distinct tissue microenvironments and ligand repertoires.

BACKGROUND: Chronic exposure to androgen receptor (AR)-targeted therapies can drive lineage plasticity, leading to treatment resistance and neuroendocrine prostate cancer (NEPC), characterized by loss of AR expression and acquisition of neuroendocrine (NE) markers including synaptophysin (SYP). We evaluated concordance between circulating tumor cell (CTC) AR and SYP expression patterns and evidence of NE differentiation defined by clinical, pathologic, and genomic features.
METHODS: CTCs from patients with metastatic castration-resistant prostate cancer (mCRPC) were analyzed using the RareCyt

Candida albicans remains a leading etiological agent of both mucosal and invasive fungal infections. The increasing prevalence of antifungal resistance and limited therapeutic options pose a significant clinical challenge and highlight the need for novel drug targets. The metabolic plasticity of C. albicans is closely linked to its pathogenicity, as the utilization of diverse carbon sources influences cell wall biogenesis, virulence factor development, and immune evasion. The most significant one is the metabolic enzyme α-glucosidase, which connects carbohydrate metabolism to the processing of

Reactive oxygen species (ROS) are important regulators of cancer biology, acting as tumor-promoting signaling mediators and inducers of oxidative cell death. Oncogenic signaling, mitochondrial dysfunction, metabolic rewiring, and microenvironmental stress lead to increased basal ROS levels in cancer cells, resulting in a state of chronic oxidative pressure. Tumors develop adaptive antioxidant programs such as glutathione and thioredoxin, NADPH regeneration pathways, and sustained activation of the Nrf2-Keap1 axis to adapt to these conditions, leading to redox plasticity and "Nrf2 addiction" in

Cancer research has undergone a fundamental paradigm shift, moving beyond a tumor-cell-centric framework toward a systems-level understanding in which the nervous system occupies a central regulatory role. The brain and its peripheral extensions engage in sophisticated, bidirectional crosstalk with tumors across multiple biological scales. This complex relationship, now termed the brain-tumor axis, operates through mechanisms spanning systemic neuroendocrine regulation, local neural-tumor interactions, and the distinctive biology of the central nervous system as both a primary and metastatic t

Background: Homeobox transcription factors regulate developmental programs, cellular plasticity, and tumor progression, yet the role of H2.0-like homeobox (HLX) in prostate cancer (PCa) remains poorly defined. We investigated the clinical significance of HLX and its relationship to the pro-metastatic transcription factor TBX2. Methods: Transcriptomic and clinical datasets from TCGA, MET500, and SU2C/PCF cohorts were analyzed to assess HLX expression, clinicopathologic associations, and its relationship with TBX2. Functional studies in human PCa cell lines included TBX2 gain- and loss-of-functi

UNLABELLED: Diabetic retinopathy (DR) remains a leading cause of blindness, characterized by progressive neurovascular dysfunction. While the enzyme 12/15-lipoxygenase (12/15-LO) and its metabolites are upregulated in DR, their interactions with epigenetic regulators, such as miRNAs, are poorly understood. This study investigates the role of 12/15-LO in miRNA dysregulation and its functional consequences in a type 1 diabetic mouse model. We generated 12/15-LO knockout mice on an (Ins2+/akita) (Akita) background. Retinal miRNA expression was profiled using microarray analysis, and retinal struc

BACKGROUND: Calcitonin gene-related peptide (CGRP) is a key mediator of migraine, but the mechanisms through which meningeal CGRP engages central circuits involved in the sensory and affective dimensions of migraine remain incompletely understood. Here, we tested if supradural CGRP is sufficient to induce migraine-like sensory and affective behaviors and if these effects are associated with sex-dependent changes in parabrachial-central amygdala (PB-CeA) signaling.
METHODS: Adult male and female C57BL/6J mice received supradural CGRP or vehicle. Behavioral testing, whole-cell patch-clamp recor

Peripheral nerve injury (PNI) presents a significant clinical challenge often resulting in long-term functional disability. Following injury, successful regeneration heavily depends on the cellular plasticity of Schwann cells (SCs) to undergo dedifferentiation, proliferate, and guide axonal growth. Recently, the pineal hormone melatonin has emerged as a promising therapeutic candidate due to its potent antioxidant properties and ability to modulate SC biological behavior. However, its exact molecular mechanisms and direct impact on SC proliferation remain fragmented across literature. Followin

Glioblastoma (GBM) remains the most aggressive and therapeutically challenging primary brain tumor and is characterized by strong heterogeneity, rapid progression, and poor clinical outcomes. Accumulating evidence indicates that the immune-angiogenic axis is a major contributor to GBM progression and therapeutic resistance, coordinating dynamic interactions among tumor cells, immune populations, and the vascular network within the tumor microenvironment (TME). Aberrant angiogenesis, driven primarily by vascular endothelial growth factor (VEGF), fibroblast growth factor (FGF), platelet-derived

Seasonal influenza in humans is predominantly caused by influenza A virus (IAV) and influenza B virus (IBV), but they differ markedly in host range, evolutionary dynamics, and pandemic potential. Such phenotypic divergence reflects the distinct molecular strategies employed by the two viruses at key stages of their life cycles. Hemagglutinin (HA) of IAV possesses prominent structural plasticity, which endows it with the capacity to recognize both avian-type α2,3-linked and human-type α2,6-linked sialic acid receptors, and the function of IAV polymerase is highly dependent on host acidic nuclea

BACKGROUND: Endothelial-to-mesenchymal transition (EndoMT) contributes to the pathogenesis and rupture of brain arteriovenous malformations (bAVMs), yet its upstream regulatory mechanisms remain largely unclear.
METHODS: S1PR1 expression was first examined across multiple public databases, including the Human Protein Atlas, and subsequently validated in human bAVM tissues via immunohistochemistry and immunofluorescence. Human umbilical vein endothelial cells (HUVECs) with S1PR1 knockdown or overexpression were used to perform transcriptomic (RNA-seq) and proteomic analyses. Enrichment analyse

BACKGROUND: Sex chromosome abnormalities in cattle are rare, and manifestations of genital anomalies due to such abnormalities are even less frequently reported. Among these, XXX/XY chimerism is particularly uncommon. This report presents a Japanese black calf with complex urogenital malformations linked to XXX/XY chimerism, contributing valuable insights into bovine sex determination and reproductive development.
CASE PRESENTATION: A Japanese black calf of phenotypic indeterminate sex, born co-twin to a phenotypically normal male, presented with hypospadias-like features and ambiguous genita
