Plasma neuronal exosomes serve as biomarkers of cognitive impairment in HIV infection and Alzheimer's disease.

Fluid biomarkers for cognitive impairment have the advantage of being relatively noninvasive and capable of monitoring neuronal and other brain cell health in real time. Biomarkers can predict the onset of dementing illness, but also correlate with cognition in a dynamic way allowing us to follow treatment responses and determine brain recovery. Chronic HIV infection causes cognitive impairment in a subset of individuals suggesting "premature aging." Exosomes are small extracellular vesicles that are shed from all cells. They are important in normal cell-to-cell communication as they contain c

Cognition attenuates the relationship between insufficient subjective energy and lower physical activity differentially across European regions.

BACKGROUND: Previous evidence suggests that higher cognitive performance may buffer the negative effects of insufficient subjective energy availability (SEA) on physical activity (PA) engagement. However, regional differences in opportunities, social norms, and contextual demands across Europe may influence this relationship, indicating that the interplay between SEA, cognition, and PA may not be universal. OBJECTIVES: To investigate whether cognitive performance moderates the association between SEA and PA in Europeans aged 50+, and to explore potential differences across the four European r

Cognitive and social impairments in patients with superficial siderosis.

Superficial siderosis of the CNS is a rare condition, caused by deposition of haemosiderin in the superficial layers of the CNS due to repeated chronic subarachnoid or intraventricular haemorrhage. Typically, the hindbrain structures, especially the cerebellum, are most affected. There is a surprising lack of studies investigating in detail the behavioural functioning of patients with such a condition. In this study, we document for the first time the cognitive, social and emotional processing of six patients with a confirmed clinical diagnosis of superficial siderosis. They were aged between

Cognitive impairment among older adults' pilgrims during Hajj: a cross-sectional study of prevalence and associated factors.

BACKGROUND: Hajj, one of the largest annual religious gatherings, attracts millions of Muslim pilgrims globally, with a significant proportion being older adults. Cognitive impairment, encompassing mild cognitive decline to dementia, is a prevalent condition among older adults and can significantly affect their ability to participate in the physically and mentally demanding rituals of Hajj. Despite its implications, data on the prevalence and associated factors of cognitive impairment among pilgrims remain limited. OBJECTIVE: This study aimed to assess the prevalence of cognitive impairment a

Intraneuronal Abeta causes the onset of early Alzheimer's disease-related cognitive deficits in transgenic mice.

Progressive memory loss and cognitive dysfunction are the hallmark clinical features of Alzheimer's disease (AD). Identifying the molecular triggers for the onset of AD-related cognitive decline presently requires the use of suitable animal models, such as the 3xTg-AD mice, which develop both amyloid and tangle pathology. Here, we characterize the onset of learning and memory deficits in this model. We report that 2-month-old, prepathologic mice are cognitively unimpaired. The earliest cognitive impairment manifests at 4 months as a deficit in long-term retention and correlates with the accumu

[Menopause hormone therapy and cognition. Postmenopausal women management: CNGOF and GEMVi clinical practice guidelines].

The results of the WHI, which reported a doubling of the risk of Alzheimer's disease (AD) and a decline in cognitive function in women who were given menopause hormone therapy (MHT), have raised concerns on the deleterious impact of MHT on the central nervous system. Such as for the cardiovascular system, the very late age of initiation of treatment and the nature of the molecules have led to conclusions that cannot be extended to women in their fifties, at the onset of their menopause which is the usual age of MHT initiation. The molecules, which are used in France, 17-beta estradiol and natu

Lifetime and 10-year absolute risk of cognitive impairment in relation to amyloid PET severity: a retrospective, longitudinal cohort study.

BACKGROUND: A key knowledge gap concerns the lifetime risk of developing cognitive impairment among individuals who are cognitively unimpaired with abnormal Alzheimer's disease biomarkers. Our aim was to compute lifetime and 10-year absolute risk of cognitive impairment as a function of continuous amyloid PET. METHODS: In this retrospective, longitudinal cohort study of data from participants of the population-based Mayo Clinic Study of Aging (Olmsted County, MN, USA), we computed lifetime and 10-year absolute risk of cognitive impairment in participants who were cognitively unimpaired and ag

Brooding rumination predicts lower cognitive functioning in late-life depression: multivariate multiple regression analyses.

INTRODUCTION: Late-life depression (LLD) is associated with cognitive deficits, particularly in memory and executive functions. Rumination, namely brooding, may also negatively impact cognition. Few studies have investigated multivariate relationships between depressive symptoms and different types of rumination on cognition in LLD, which was the focus of the current study. We also explored whether relationships differed by gender and depression age of onset. METHOD: Cognition was measured via five cognitive composites (Attention/Working Memory, Processing Speed, Language, Episodic Recall, an

Active Seizures Predict Worse Instrumental Activities of Daily Living in Individuals With Normal Cognition and Dementia.

OBJECTIVES: Cognitive disorders are common in older persons with seizures (PWS). Cognitive disorders are often associated with impaired Instrumental Activities of Daily Living (IADLs). However, the effects of seizures on IADLs remain unexplored. This study (9/2005-3/2024) from 42 US Alzheimer's Disease Research Centers examined baseline and longitudinal IADL decline in cognitively normal and cognitively impaired participants grouped as active seizures (past 12 months), remote seizures (history but none in 12 months), and controls (no seizures). METHODS: Pfeffer Functional Activities Questionn

The Cerebellar Cognitive Affective Syndrome in Ataxia-Telangiectasia.

Ataxia-telangiectasia (AT) is an autosomal recessive, multisystem disease causing cerebellar ataxia, mucocutaneous telangiectasias, immunodeficiency, and malignancies. A pilot study reported cognitive and behavioral manifestations characteristic of the cerebellar cognitive affective / Schmahmann syndrome (CCAS). We set out to test and further define these observations because a more comprehensive understanding of the spectrum of impairments in AT is essential for optimal management. Twenty patients (12 males; 9.86 ± 5.5 years, range 4.3 to 23.2) were grouped by age: AT-I (toddlers and preschoo

Trajectories of Rumination and Negative Cognitive Style from Late Childhood Through Adolescence: Modeling Normative Growth Patterns and Predicting Cognitive Vulnerabilities.

Negative cognitive style and rumination are prominent cognitive vulnerabilities (CVs) that contribute to development of psychopathology, especially internalizing problems. Yet, little is known about trajectories of CVs across development (age) and gender and what predicts CVs. This study characterized CV trajectories from ages 9-18 and investigated predictors of CV trajectories. Community youth (N=607; M age=12.37; 55% girls) completed repeated assessments of CVs across three years; youth and parents were assessed for many CV predictors at baseline. Measurement invariance supported partial sca

Amnestic and non-amnestic symptoms of dementia: An international study of Alzheimer's disease in people with Down's syndrome.

UNLABELLED: The presence of age-related neuropathology characteristic of Alzheimer's disease (AD) in people with Down syndrome (DS) is well-established. However, the early symptoms of dementia may be atypical and appear related to dysfunction of prefrontal circuitry. OBJECTIVE: To characterize the initial informant reported age-related neuropsychiatric symptoms of dementia in people with DS, and their relationship to AD and frontal lobe function. METHODS: Non-amnestic informant reported symptoms (disinhibition, apathy, and executive dysfunction) and amnestic symptoms from the CAMDEX-DS infor

Prevalence, risk factors and consequences of cerebral small vessel diseases: data from three Asian countries.

BACKGROUND: Cerebral small vessel disease (SVD) has been suggested to be more common in Asians compared with Caucasians. However, data from population-based studies in Asia are lacking. We report on the prevalence, risk factors and consequences of SVD from contemporary studies in three Asian countries using 3-Tesla MRI for the evaluation of SVD. METHODS: Clinical, cognitive and 3-Tesla brain MRI assessments were performed among participants of three studies from Singapore, Hong Kong and Korea. SVD markers include white matter hyperintensities (WMHs) using the modified Fazekas scale, lacunes a

Pain, Genes, and Function in the Post-Hip Fracture Period.

Post-hip fracture generalized pain can lead to a progressive decline in function and greater disability. The purpose of this study was to explore the factors that influence pain among older adults post-hip fracture, including genetic variability, and evaluate whether pain directly or indirectly influenced upper and lower extremity function. This was a secondary data analysis using data from the first 200 participants in a Baltimore Hip Study (BHS), BHS-7. Assessments were done at 2 months post-hip fracture and included age, sex, marital status, education, cognitive status, comorbidities, body

Psychopathology of cognitive disengagement syndrome (CDS): a network analysis based on CBCL scales in 72,106 Chinese school students.

Cognitive Disengagement Syndrome (CDS) is a cluster of behavioral problems that severely affect an individual's functioning. Currently, there is no consensus on the main clinical features of CDS, and further exploration in large samples is needed. Using a cluster-stratified random sampling method, 72,106 children and adolescents were recruited from five provinces in mainland China for this study. Using both the traditional two-factor scoring method and the CBCL DSM-oriented scales, we assessed individual behavioral problems from psychopathological and DSM-oriented perspectives. The balance between cognitive reserve and brain imaging biomarkers of cerebrovascular and Alzheimer's diseases.

The cognitive reserve hypothesis explains the disparity between clinical and pathological phenotypes and why, in two individuals with the same extent of neuropathology, one may be demented while the other remains cognitively intact. We examined the balance between brain magnetic resonance imaging measures of the two most common pathologies associated with brain ageing, cerebrovascular disease and Alzheimer's disease, and parameters of cerebral reserve in well-characterized participants born in 1936, for whom childhood intelligence is known. Brain magnetic resonance imaging was carried out at 1

A randomized double-blind study comparing 25 and 50 mg TC-1734 (AZD3480) with placebo, in older subjects with age-associated memory impairment.

Cognitive decline is a feature of ageing and can be defined as normal (age-associated memory impairment) or pathological (mild cognitive impairment/Alzheimer's disease). Stimulation of selective brain-specific neuronal nicotinic acetylcholine receptors might offer symptomatic treatment for normal ageing. The objective of this study was to assess the safety, tolerability and efficacy of TC-1734 (AZD3480), a selective α4β2 nicotinic agonist, in the treatment of age-associated memory impairment. A randomized placebo-controlled trial was conducted in 16 community-based centers within the USA. Subj
