Study on the Mechanism of CCN2 Promoting Sorafenib Resistance in HCC and Its Combined Intervention Strategy.

BACKGROUND: Since its introduction in 2008, sorafenib has remained the standard first-line systemic treatment for advanced hepatocellular carcinoma (HCC). Nevertheless, its clinical benefits are often compromised by the rapid emergence of drug resistance. This study explores the molecular mechanisms underlying sorafenib resistance, with particular emphasis on the involvement of connective tissue growth factor (CCN2/CTGF) in the regulation of c-Met signaling pathways. METHODS: We began by evaluating CCN2 expression levels in HCC tissue samples via immunohistochemistry and analyzing their corre

Expression of matrix metalloprotease-9 in vascular pericytes in human breast cancer.

Matrix metalloprotease-9 (MMP-9; 92-kd type IV collagenase, gelatinase B) is regarded as important for degradation of the basement membrane and extracellular matrix during cancer invasion and other tissue-remodeling events. Expression of MMP-9 was analyzed in 22 cases of human ductal breast cancer by immunohistochemistry and in 8 of these cases also by in situ hybridization. For immunohistochemistry we used affinity-purified polyclonal antibodies as well as a MMP-9-specific monoclonal antibody (clone 6-6B). Three different stromal cell types with a positive MMP-9 immunoreaction were identified

Single-cell profiling reveals transcriptomic signatures of vascular endothelial cells in non-healing diabetic foot ulcers.

BACKGROUND: The treatment of diabetic foot ulcers (DFUs) poses a challenging medical problem that has long plagued individuals with diabetes. Clinically, wounds that fail to heal for more than 12 weeks after the formation of DFUs are referred to as non-healing/chronic wounds. Among various factors contributing to the non-healing of DFUs, the impairment of skin microvascular endothelial cell function caused by high glucose plays a crucial role. Our study aimed to reveal the transcriptomic signatures of non-healing DFUs endothelial cells, providing novel intervention targets for treatment strate

Nociceptive responses and resting-state functional connectivity of the human midbrain tegmentum.

The rostromedial tegmental nucleus (RMTg) has been shown to inhibit midbrain dopaminergic centers through its GABAergic projections, and to be critical for regulating responses to aversive and rewarding stimuli. This nucleus has primarily been evaluated in rodents and non-human primates, while recent postmortem analysis of human brainstem tissue localized GABAergic cell clusters in the putative RMTg. The objectives of this preliminary investigation in healthy controls (HC) were to (i) define RMTg function during evoked thermal stimulation and the resting-state using 7T functional magnetic reso

Key regions aberrantly connected within cerebello-thalamo-cortical circuit and their genetic mechanism in schizophrenia: an fMRI meta-analysis and transcriptome study.

Recent studies have showed aberrant connectivity of cerebello-thalamo-cortical circuit (CTCC) in schizophrenia (SCZ), which might be a heritable trait. However, these individual studies vary greatly in their methods and findings, and important areas within CTCC and related genetic mechanism are unclear. We searched for consistent regions of circuit dysfunction using a functional magnetic resonance imaging (fMRI) meta-analysis, followed by meta-regression and functional annotation analysis. Gene annotation analysis was performed to identify genes over-expressed in these regions by using the All

Exploring mechanobiology network of bone and dental tissue based on Natural Language Processing.

Bone and cartilage tissues are physiologically dynamic organs that are systematically regulated by mechanical inputs. At cellular level, mechanical stimulation engages an intricate network where mechano-sensors and transmitters cooperate to manipulate downstream signaling. Despite accumulating evidence, there is a notable underutilization of available information, due to limited integration and analysis. In this context, we conceived an interactive web tool named MechanoBone to introduce a new avenue of literature-based discovery. Initially, we compiled a literature database by sourcing conten

Tyrosine-Like Crystalloids Localize to Non-Neoplastic True Vocal Cord and Attachments.

BACKGROUND: Tyrosine-rich or tyrosine-like crystalloids (TC) were initially described in salivary gland pleomorphic adenoma. The presence of TC in non-neoplastic tissues is rare, and it has been reported exclusively in the larynx. This study aims to characterize the frequency and anatomical localization of TC in total laryngectomy specimens. METHODS: Review of consecutive laryngectomy specimens in which the cassette summary documented parasagittal section sampling of the right and left vocal folds and the anterior commissure. Data collected included patient demographics, underlying diagnoses,

Immune checkpoint inhibitor (ICI) genes and aging in malignant melanoma patients: a clinicogenomic TCGA study.

BACKGROUND: Cancer diagnoses and deaths among the elderly (65 +) are expected to increase significantly over the next decade. Immune checkpoint inhibitors specifically target ICI genes and enhance immune system function. However, poor outcomes may be associated with aging. METHODS: We downloaded the Genomic Data Commons from the Cancer Genome Atlas (TCGA) and collected gene expression data from malignant melanoma (MM) tissues, the third level as the primary site. The CKTTD ICI genes database were applied and validated using the GEO database and lab experiments. RESULTS: In 414 patients, 13 I

Insulin-Like Growth Factor Pathway and the Thyroid.

The insulin-like growth factor (IGF) pathway comprises two activating ligands (IGF-I and IGF-II), two cell-surface receptors (IGF-IR and IGF-IIR), six IGF binding proteins (IGFBP) and nine IGFBP related proteins. IGF-I and the IGF-IR share substantial structural and functional similarities to those of insulin and its receptor. IGF-I plays important regulatory roles in the development, growth, and function of many human tissues. Its pathway intersects with those mediating the actions of many cytokines, growth factors and hormones. Among these, IGFs impact the thyroid and the hormones that it ge

The ciliary neurotrophic factor induces Stat3 phosphorylation in distinctive cytotypes of organs involved in body metabolism: An immunohistochemical study.

Administration of ciliary neurotrophic factor (CNTF) reduces food intake and body weight in both humans and experimental animals, where it also ameliorates hyperglycemia, hyperinsulinemia, and dyslipidemia. To exert its anti-obesogenic and anti-diabetogenic effects, CNTF targets brain feeding centers as well as multiple peripheral organs inducing the phosphorylation of the transcription factor signal transducer and activator of transcription 3 (p-STAT3). However, data showing which peripheral cytotypes are specifically targeted by exogenous CNTF in vivo in metabolically relevant organs are cur

Unveiling PAK4 as a Key Biomarker in Adrenocortical Carcinoma: Insights from Bioinformatics and Experimental Evidence.

BACKGROUND: Adrenocortical carcinoma (ACC) is a rare and fatal adrenal cortex cancer with a poor prognosis and high mortality rate. Although surgical resection is the primary treatment for ACC, recurrence is still common. p21-activated kinase 4 (PAK4) is linked to tumour development and progression, being overexpressed in various cancers. However, the role of PAK4 in ACC remains unclear. METHODS: In this study, PAK4 expression in ACC was analysed using sequencing data from the Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) databases, assessing its clinical relevance with Kap

Cilium-generated signaling and cilia-related disorders.

Biologists have long known that humans experience their environment through cilia. Light, odorant, and sound perception depend on these microtubule-filled, complex organelles present on cells in primary sensory tissues. Recently, discoveries on the mechanism of assembly of cilia (flagella) in the lowly, biflagellated, eucaryotic green alga Chlamydomonas have triggered a renaissance of interest in the organelles along with a recognition of their key sensory roles in nonsensory tissues. Chlamydomonas researchers uncovered an entirely new set of cellular machinery essential for transporting the p

In silico and in vitro insights into stigmasterol targeting Keap1/Nrf2, Bcl-2/Bax and IKKβ/IκBα protein-protein interactions in Arsenic-induced toxicity.

Inorganic arsenic is a known neurotoxin that contributes to neurodevelopmental and neurodegenerative disorders by promoting oxidative stress, mitochondrial dysfunction and inflammation. It crosses the blood-brain barrier and accumulates in brain tissue. Arsenic-induced neurotoxicity poses a significant health challenge, driving the need for multi-targeted therapeutic agents. This study investigated the neuroprotective potential of stigmasterol, a plant-derived phytosterol, against arsenic-induced damage in mouse HT-22 hippocampal cells through integrated computational and experimental approach

Targeting obesity and adipokine signaling in the breast cancer microenvironment for enhanced tumor therapy.

Breast cancer (BC) is a malignant breast tumor with high rates of metastasis and recurrence, making it the most common cancer in women and leading to a high mortality rate. Emerging evidence has indicated a close relationship between obesity and BC, as adipocytes, the largest cellular component of breast tissue, readily engage in crosstalk with breast cancer cells. Breast cancer cells can induce adipocytes to transform into cancer-associated adipocytes (CAAs) with a malignant phenotype, which secrete multiple cytokines, including leptin, adiponectin, interleukin-6 (IL-6), chemokine ligand 2 (C

Osteomimetic microcalcification shapes the breast tumor microenvironment and metastatic potential.

Microcalcifications are among the most reliable mammographic signs of breast cancer, particularly of non-palpable and in situ disease, yet for decades they were regarded as inert by-products of cellular degeneration-a view that delayed inquiry into their biology. A growing body of in vitro, ex vivo, and tissue-based evidence now reframes breast microcalcification as a process that can be actively regulated and cell-driven, recapitulating physiological and pathological mineralization elsewhere in the body. In this review, we synthesize the mechanistic basis of this paradigm shift and argue that

Mechanical and Electrical Phenotype of hiPSC-Cardiomyocytes on Fibronectin-Based Hydrogels.

A major challenge in cardiac research is the limited translatability of drug screening and toxicity assays due to the use of in vitro models that poorly mimic the native cardiac environment. Human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) offer a promising route forward, but conventional 2D culture on rigid substrates hinders their functional maturation and predictive accuracy. This study addresses this problem by investigating the effect of hybrid fibronectin-based hydrogels with tunable stiffness on the mechanical and electrical properties of hiPSC-CMs. We engineered h

The overexpression of human amylin in pancreatic β cells facilitate the appearance of amylin aggregates in the kidney contributing to diabetic nephropathy.

Diabetic nephropathy is one of the most frequent complications of diabetic patients and is the leading cause of end-stage renal disease worldwide. The complex physiopathology of this complication raises a challenge in the development of effective medical treatments. Therefore, a better understanding of this disease is necessary for producing more targeted therapies. In this work we propose human amylin as a possible mediator in the development of diabetic nephropathy. Islet amyloid polypeptide or amylin is a hormone co-secreted with insulin. The human isoform has the ability to fold and form a

Fetal growth at term and placental oxidative stress in a tissue micro-array model: a histological and immunohistochemistry study.

This study examines 8-hydroxyguanine (8-oxo-Gua) staining in placental tissue samples based on fetal size at birth as well as its relationships with placental histology and other pregnancy variables. This prospective cohort study included women > 18 years with a singleton pregnancy, a live fetus, fluency in Italian, and delivery at term. A total of 165 pregnancies were included in the study. The nuclear syncytiotrophoblast 8-oxo-Gua staining score in LGA was substantially greater than in late FGR (p < 0.05), although the cytoplasm score was lower in SGA and LGA than in AGA (p < 0.05). GATA2 regulates blood/lymph separation in a platelet-dependent and lymphovenous valve-independent manner.

INTRODUCTION: Lymphatic vessels collect interstitial fluid, immune cells, and digested lipids and return these bodily fluids to blood through two pairs of lymphovenous valves (LVVs). Like other cardiovascular valves LVVs prevent the backflow of blood into the lymphatic vessels. In addition to LVVs, platelets are necessary to prevent the entry of blood into the lymphatic vessels. Platelet thrombi are observed at LVVs suggesting that LVVs and platelets function in synergy to regulate blood/lymphatic separation. OBJECTIVES: The primary objective of this work is to determine whether platelets can

Characterization of Hypothalamic MCH Neuron Development in a 3D Differentiation System of Mouse Embryonic Stem Cells.

Hypothalamic melanin-concentrating hormone (MCH) neurons are important regulators of multiple physiological processes, such as sleep, feeding, and memory. Despite the increasing interest in their neuronal functions, the molecular mechanism underlying MCH neuron development remains poorly understood. We report that a three-dimensional culture of mouse embryonic stem cells (mESCs) can generate hypothalamic-like tissues containing MCH-positive neurons, which reproduce morphologic maturation, neuronal connectivity, and neuropeptide/neurotransmitter phenotype of native MCH neurons. Using this in vi
