OMIP-111: Immune-Profiling of T Helper 1 (Th1), Th2, and Th17 Signatures in Murine Splenocytes by Targeting Intracellular Cytokines.

Functional cytokines shape both innate and adaptive immune responses in the host after infection or immunization. Deep immunophenotyping of the key functional cytokine signatures associated with T cells in murine lymphoid tissue, especially in the spleen, is challenging. Using spectral flow cytometry, we developed a 17-parameter panel to profile major immune cell subsets along with T cells, memory phenotypes, and functional cytokines in murine splenocytes in steady state as well as in stimulated conditions. This panel dissects the memory T cell compartment via CD62L and CD44 expression after m

Lactobacillus lysates protect oral epithelial cells from pathogen-associated damage, increase secretion of pro-inflammatory cytokines and enhance barrier integrity.

Periodontitis is a chronic gum disease characterised by inflammation and the loss of bone. We have explored the potential prophylactic effects of lysates from four Lactobacillus strains against the toxic effects of three periodontal pathogens (Porphyromonas gingivalis, Fusobacterium nucleatum, and Aggregatibacter actinomycetemcomitans). TR146 oral epithelial cells were pre-treated with Lactobacillus lysates (L. rhamnosus - GG, L. rhamnosus - SD11, L. reuteri and L. plantarum) and then challenged with pathogenic material (live cells, lysates, or supernatants). Cytokine analysis was performed on

Immune profiling of human vestibular schwannoma secretions identifies TNF-α and TWEAK as cytokines with synergistic potential to impair hearing.

BACKGROUND: Vestibular schwannoma (VS) is an intracranial tumor arising from the Schwann cells of the vestibular nerve and is an important cause of sensorineural hearing loss (SNHL) in humans. The mechanisms underlying this SNHL are incompletely understood and currently, there are no drugs FDA approved specifically for VS. This knowledge gap significantly limits the development of effective treatments aimed at preventing, stabilizing, or reversing VS-induced SNHL. METHODS: To identify effector molecules involved in VS-induced SNHL, we analyzed 47 immune-related factors secreted by tumor tissu

Profiling blood-based neural biomarkers and cytokines in experimental autoimmune encephalomyelitis model of multiple sclerosis using single-molecule array technology.

Experimental autoimmune encephalomyelitis (EAE) is a preclinical animal model widely used to study multiple sclerosis (MS). Blood-based cytokines and neural biomarkers are predictors of neurodegeneration, disease activity, and disability in patients with MS. However, understudied confounding factors cause variation in reports on EAE across animal strains/studies, limiting the utility of these biomarkers for predicting disease activity. In this study, we investigated blood-based analyte profiles, including neural markers (NFL and GFAP) and cytokines (IL-6, IL-17, IL-12p70, IL-10, and TNF-α), in

The impact of cytokines and tumour-conditioned medium on the properties of murine in vitro generated myeloid-derived suppressor cells.

Myeloid-derived suppressor cells (MDSCs) are a heterogeneous group of immature myeloid cells playing a critical role in immune suppression. In vitro-generated MDSCs are a convenient tool to study the properties of tumour-associated MDSCs. Here, we compared six protocols for in vitro generation of functional mouse MDSCs from bone marrow progenitors. The protocols included granulocyte-macrophage colony-stimulating factor (GM-CSF) alone or in combination with interleukin-6 (IL-6) or granulocyte colony-stimulating factor (G-CSF), with or without a tumour-conditioned medium (TCM) derived from B16-F

Changes in concentrations of cytokines and markers of bone turnover in dairy cows during different stages of a production cycle.

Changes in bone metabolism occur to meet the varying demands for calcium during pregnancy and lactation. These changes are regulated locally and systemically by a variety of factors, including hormones, cytokines, chemokines, and growth factors. The goal of the present experiment was to document the dynamics of bone turnover and metabolic state during a complete lactation cycle in dairy cows and to monitor the plasma profile of 10 cytokines previously known to play roles in bone turnover during lactation. Blood samples were collected sequentially from Holstein-Friesian cows during late pregnan

miR-548aj-3p and miR-3127-3p suppress RANKL-facilitated inflammatory cytokines and catabolic factor in osteoarthritis and rheumatoid arthritis.

Osteoarthritis (OA) and rheumatoid arthritis (RA) are highly prevalent joint diseases globally. The common pathological features include synovial inflammation, swelling, joint destruction, and bone remodeling. Arthritis development is associated with joint inflammation, particularly in inflamed synovial cells. Synovial inflammation contributes to joint destruction. The receptor activator of nuclear factor kappa-B ligand (RANKL) is a vital factor that is linked to the activity of osteoclasts and the erosion of bone. Increased levels of RANKL play a role in the course of arthritis. Adverse effec
