Differentiation of Variola major and Variola minor variants by MGB-Eclipse probe melt curves and genotyping analysis.

Smallpox, caused by the Variola major virus, is considered to be one of the most lethal of all potential biological weapons and has far-reaching consequences. Real-time polymerase chain reaction (PCR) assays are available as a reliable diagnostic tool to detect members of the genus Orthopoxvirus. In addition real-time PCR assays specific for the variola virus have been developed that distinguish it from other orthopoxviruses. However, a positive identification of variola spp. does not classify the virus as the one that causes smallpox (V. major) or as the variant (Variola minor) that causes a

Progressive multifocal leukoencephalopathy in an immunocompetent Taiwanese patient.

Progressive multifocal leukoencephalopathy (PML) is a deadly demyelinating brain disease caused by JC virus (JCV). Genomic analysis of viral isolates in these cases often shows prototype-like JCV and its variants, which is a virulent strain compared to the latent archetype virions mostly found in the kidney. Here, we report a 57-year-old man who suffered from a subacute course of cognitive impairment and multiple neurologic deficits. Neuroimaging, pathology, and virology studies showed multifocal leukoencephalopathy and the presence of JCV deoxyribonucleic acid in the cerebrospinal fluid. The

Is human herpesvirus 7 the causative agent of pityriasis rosea?--A critical review.

BACKGROUND: Conflicting results on the association of pityriasis rosea and human herpesvirus 7 infection have been reported by different investigators. AIM: To review the level of evidence for such an association. METHODS: Medline was searched with unlimited data entry and 13 reports were retrieved. The data were analyzed for a causative association according to the criteria of Fredericks and Relman, which take into consideration latent infection and the reactivation of viruses characteristic of herpesviruses, and the roles of sequence-based detection methods. RESULTS: None of the criteria

DNA-based immunization against Lyssaviruses.

Rabies is a fatal encephalomyelitis. Most cases occur in developing countries and are transmitted by dogs. Because of their high cost, cell culture vaccines have not totally replaced the unsafe brain-derived vaccines which are still used in many developing countries. Moreover, there will be a need for vaccines against rabies-related viruses against which classical vaccines are not always effective. DNA vaccines would, therefore, be a valuable alternative for the production of cheaper rabies vaccines against a larger spectrum of viruses. In this review we report published data on DNA-based immu

Molecular epidemiology of adenovirus type 7 in Israel: identification of two new genome types, Ad7k and Ad7d2.

The molecular epidemiology of Adenovirus type 7 in Israel was investigated. Fifty-seven adenovirus isolates identified as serotypes 7 or 7a which were recovered from patients in Israel between 1968 and 1995 were analyzed by restriction enzymes digestion using BamHI for primary discrimination and identification of genome types and by six additional enzymes: BstEII, HpaI, BglI, BglII, BclI, and XbaI for confirmation and determination of genomic subtypes. Four digestion patterns were identified with BamHI; one of them was new. Using BstEII, two patterns were obtained, one of them new. Digestion w

[Geotyping of wild poliovirus strains on the territory of Russia and CIS countries in 1987-1995].

33 wild poliovirus strains isolated on the territory of the former USSR were subjected to the comparative sequence analysis of the 150 bp genome fragment VP1/2A on the junction of the genome regions coding basic capsid protein VP1 and viral protease 2A. The dendrograms characterizing the similarity of nucleotide sequences revealed the existence of 3 geographical genotypes (geotypes) of wild poliovirus strains of type 1 (A, G, T) and broad geotype (C) of wild poliovirus strains of type 3. The comparison of the analyzed strains with strains circulating in the neighboring countries at the same pe

DNA diagnosis of HTLV-I.

Human T-lymphotropic virus type I (HTLV-I) was the first retrovirus which was directly associated with adult T-cell leukemia (ATL). Infection with HTLV-I can also lead to various other diseases, e.g. HTLV-I-associated myelopathy/tropical spastic paraparesis (HAM/TSP) and HTLV-I uveitis, possibly via induction of immunodeficiency or hyperreactivity against HTLV-I-infected cells. Epidemiological data have shown that patients who developed these diseases represent a small percentage of HTLV-I-infected individuals living in restricted geographical areas. The identification of HTLV-I-infected indiv

Tree shrew (Tupaia) herpesviruses.

Five Tupaia herpesviruses have been isolated until now: four in our laboratory which were termed THV-2, 3, 4, and 5, whereas THV-1 has been isolated by Melnick and his colleagues. THV-2 was isolated from tumour cell culture of a high-grade malignant lymphoma of a Tupaia, THV-3 was released from a cell culture of another Tupaia lymphoma, THV-4 from a spleen tissue culture of a moribund animal with finely granulated liver cirrhosis, and THV-5 from cultured spleen cells of an apparently healthy tree shrew. THV-1 to 5 were efficiently propagated, plaque-purified and cloned on Tupaia embryonic fibr

Mechanisms of Accelerated Biological Aging and Premature Cardiovascular Events in Young and Middle-Aged Post-COVID-19 Survivors.

This review evaluates the impact of Post-Acute Sequelae of SARS-CoV-2 on adults aged 25 to 55, focusing on the potential for accelerated biological aging and an increased incidence of acute cardiovascular events. By synthesizing evidence from 110 epidemiological and mechanistic studies, we examine the interplay between viral-induced immune dysregulation, endothelial injury, and systemic physiological decline in a demographic previously considered low-risk for severe outcomes. Our analysis indicates that chronic systemic inflammation, potentially maintained by persistent viral reservoirs in imm

The effects of bacteriophage cocktail treatment on healthy gut microbiota: an in vitro human colon model study.

The human gut microbiome is a complex community that plays an important role in health, where perturbations can result in dysbiosis and disease. Bacteriophages (phages) can provide treatment for bacterial gastrointestinal disease, and commercial preparations such as the Intesti bacteriophage cocktail can be taken orally to target bacterial pathogens. However, interactions between these phages and the native gut microbiota are understudied. To investigate the impact of phage treatment, we used simulated gut models seeded with healthy donor microbiota from three individuals, sequenced the DNA an

Antiviral Potential of Momordica charantia: From Traditional Use to Modern Implications.

Background/Objectives: Momordica charantia L. (Cucurbitaceae) (MC), commonly known as bitter melon, is a prominent therapeutic and edible species deeply rooted in traditional medicine for the management of diverse metabolic and infectious pathologies. Increasing evidence suggests that MC is a significant source of antiviral compounds that could act against many different types of viruses in humans. This narrative review summarizes the current knowledge regarding the antiviral properties of MC, with a focus on molecular mechanisms and therapeutic perspectives. Methods: A comprehensive literatur

Viral Mimicry of Alzheimer's Disease: Innate Sensing of Self-Nucleic Acids as a Driver of Glial Senescence.

Alzheimer's disease (AD) is a devastating neurodegenerative disorder defined by progressive memory loss and synaptic failure. For decades, therapeutic development has focused on clearing amyloid-beta plaques, yet the repeated clinical failures of this approach necessitate a fundamental paradigm shift toward the brain's immunometabolic landscape. The "Viral Mimicry" hypothesis posits that AD represents a state of sterile autoimmunity where the innate immune system mistakenly identifies self-nucleic acids as viral pathogens. This "ghost war" is ignited by the convergence of metabolic dysfunction

Progress Towards Elimination: Hepatitis B and C Among Children and Young Adults in Rural Mongolia.

Mongolia is a global hotspot for viral hepatitis, with estimated rates of chronic hepatitis B (HBV) of 11% and hepatitis C (HCV) of 8.5% among adults. Data on infections among children and young adults and the impact of vaccination against HBV are less clear. In 2018 and 2019, 3800 children and young adults aged 25 and younger from all provinces in Mongolia apart from Ulaanbaatar and Tuv were tested for hepatitis B surface antigen (HBsAg), hepatitis B core antibody (HBcAb), hepatitis B surface antibody (HBsAb), hepatitis C antibody (HCVAb) and hepatitis D antibody (HDVAb). Hepatitis B Virus: Epidemiology, Prophylaxis, Therapy, Clinical Outcomes, and Novel Therapeutic Directions.

Hepatitis B virus (HBV) infection is a worldwide health concern that infects nearly 254 million people globally and causes more than 1 million deaths annually. The highest prevalence is seen in sub-Saharan Africa and the Western Pacific region. Cirrhosis, liver failure, and hepatocellular carcinoma (HCC) are reported as leading complications of chronic HBV. The route of transmission of this infection is mainly by exposure to infected blood and bodily fluids. Transmission from mother-to-child remains the predominant route in highly endemic areas. Vaccination has significantly reduced HBV seropr

Charting the research frontier for viral infections, immunomodulation, and dementia: a perspective on synaptic biomarkers as essential clinical trial endpoints.

The viral-inflammatory hypothesis of Alzheimer's disease offers a new paradigm, yet interventions like antivirals and vaccination present a paradox that challenge therapeutic development. This perspective examines the critical research gap concerning cerebrospinal fluid (CSF) synaptic biomarkers in immunomodulatory therapy trials. Following decades of partially successful amyloid-centric trials, focus has shifted to upstream triggers including viral infections like Herpes Simplex Virus Type 1, Varicella Zoster Virus, and Severe Acute Respiratory Syndrome Coronavirus 2. While large observationa
