Chemogenomic Profiling of the Fungal Pathogen Candida albicans.

There is currently a small number of classes of antifungal drugs, and these drugs are known to target a very limited set of cellular functions. We derived a set of approximately 900 nonessential, transactivator-defective disruption strains from the tetracycline-regulated GRACE collection of strains of the fungal pathogen Candida albicans This strain set was screened against classic antifungal drugs to identify gene inactivations that conferred either enhanced sensitivity or increased resistance to the compounds. We examined two azoles, fluconazole and posaconazole; two echinocandins, caspofung

Optimized Background Regimen for Treatment of Active Tuberculosis with the Next-Generation Benzothiazinone Macozinone (PBTZ169).

The efficacy of the standardized four-drug regimen (comprising isoniazid, rifampin, pyrazinamide, and ethambutol) for the treatment of tuberculosis (TB) is menaced by the emergence of multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains of Mycobacterium tuberculosis Intensive efforts have been made to develop new antibiotics or to repurpose old drugs, and several of these are currently being evaluated in clinical trials for their antitubercular activity. Among the new candidate drugs is macozinone (MCZ), the piperazine-containing benzothiazinone PBTZ169, which is currently be

Drug deposition in coronary arteries with overlapping drug-eluting stents.

Drug-eluting stents are accepted as mainstream endovascular therapy, yet concerns for their safety may be under-appreciated. While failure from restenosis has dropped to below 5%, the risk of stent thrombosis and associated mortality remain relatively high. Further optimization of drug release is required to minimize thrombosis risk while maintaining therapeutic dose. The complex three-dimensional geometry of deployed stents together with the combination of diffusive and advective drug transport render an intuitive understanding of the situation exceedingly difficult. In situations such as thi

High-throughput decoding of antitrypanosomal drug efficacy and resistance.

The concept of disease-specific chemotherapy was developed a century ago. Dyes and arsenical compounds that displayed selectivity against trypanosomes were central to this work, and the drugs that emerged remain in use for treating human African trypanosomiasis (HAT). The importance of understanding the mechanisms underlying selective drug action and resistance for the development of improved HAT therapies has been recognized, but these mechanisms have remained largely unknown. Here we use all five current HAT drugs for genome-scale RNA interference target sequencing (RIT-seq) screens in Trypa

The first case of drug-induced immune hemolytic anemia due to hydrocortisone.

BACKGROUND: Drug-induced immune hemolytic anemia (DIIHA) is a well-known complication of drug treatment. Sensitization can occur, due to interaction of the drug and/or its metabolites with cells of the immune system, after the first drug administration, while the hemolytic crisis generally occurs after repeated administration of a drug. This event occurred in the case described here of acute hemolysis due to the administration of corticosteroids. STUDY DESIGN AND METHODS: To define the etiopathogenesis of the hemolytic crisis, immunohematologic screening and specific tests were performed to i

Comparative safety and effectiveness of perinatal antiretroviral therapies for HIV-infected women and their children: Systematic review and network meta-analysis including different study designs.

BACKGROUND: Nearly all newly infected children acquire Human Immunodeficiency virus (HIV) via mother-to-child transmission (MTCT) during pregnancy, labour or breastfeeding from untreated HIV-positive mothers. Antiretroviral therapy (ART) is the standard care for pregnant women with HIV. However, evidence of ART effectiveness and harms in infants and children of HIV-positive pregnant women exposed to ART has been largely inconclusive. The aim of our systematic review and network meta-analysis (NMA) was to evaluate the comparative safety and effectiveness of ART drugs in children exposed to mate

Novel in vitro and mathematical models for the prediction of chemical toxicity.

The focus of much scientific and medical research is directed towards understanding the disease process and defining therapeutic intervention strategies. The scientific basis of drug safety is very complex and currently remains poorly understood, despite the fact that adverse drug reactions (ADRs) are a major health concern and a serious impediment to development of new medicines. Toxicity issues account for ∼21% drug attrition during drug development and safety testing strategies require considerable animal use. Mechanistic relationships between drug plasma levels and molecular/cellular event

Design and Synthesis of Type-A Proanthocyanidin Derivatives as JAK2 Activators to Protect against Chemotherapy-Induced Myelosuppression.

Chemotherapy-induced myelosuppression (CIM) significantly impairs hematopoiesis. Proanthocyanidin A1 (1) from peanut skin is a JAK2 activator to alleviate CIM, but the SAR and drug-like properties of Type-A proanthocyanidins remain unclear. Here, a series of Type-A proanthocyanidin derivatives (5-35) were designed, synthesized, and evaluated for JAK2 activation, leading to the identification of compound 32 as the most potent activator (EC50 = 64 nM). Target-binding assays demonstrated that 32 directly bound JAK2 and displayed preferential affinity for the JH2 pseudokinase domain over the JH1 c

Bioresorbable Scaffolds: Ready for a Rematch?

PURPOSE OF REVIEW: The bioresorbable vascular scaffold (BVS) is a promising technology with clear theoretical benefits. Large randomized controlled trials (RCTs) of the early generation of this technology, however, demonstrated issues translating these benefits from the abstract into the clinical realm. This review covers the outcomes beginning with the first-generation scaffolds and provides updates on advances made with newer technologies and evolved approaches to platform delivery. RECENT FINDINGS: Pooled individual patient data from the ABSORB studies with 5-year follow-up was published i

2-Arylbenzofurans as Selective Cholinesterase Inhibitors: Design, Synthesis, and Evaluation as Alzheimer's Disease Agents.

New arylbenzofuran derivatives were designed, synthesized, and evaluated as potential inhibitors of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). Five hybrid compounds (31-35) feature a 2-phenylbenzofuran core linked via a heptyloxy spacer to an N-methylbenzylamine moiety, to enhance interactions within the active site of BChE. Biological evaluation revealed that brominated derivatives 34 and 35 showed the highest cholinesterases (ChE) inhibition compared to their chlorinated analogs, with compound 34 showing the highest activity for both AChE (IC50 = 27.7 μM) and BChE (IC50 =

Based on artificial intelligence-assisted generation and in-depth in-silico evaluation of potential inhibitors targeting Stearoyl-CoA desaturase 1 (SCD-1).

Stearoyl-CoA desaturase 1 (SCD-1), a key rate-limiting enzyme in lipid metabolism, catalyzes the conversion of saturated fatty acids to monounsaturated fatty acids. It regulates membrane fluidity and modulates ferroptosis by influencing cellular antioxidant systems. Frequently overexpressed in malignancies, SCD-1 promotes tumor progression by driving metabolic reprogramming, activating proliferative pathways such as PI3K-AKT-mTOR, and suppressing ferroptosis, thereby enhancing tumor survival, invasion, and chemotherapy resistance. Clinical evidence links elevated SCD-1 expression with advanced

Design, synthesis, and anti-fungicidal activities of sulfonyl fluoride functionalized SDHI analogs against plant pathogens.

Succinate dehydrogenase inhibitors (SDHIs) have been widely employed as agricultural fungicides due to their effectiveness and low toxicity. Recently, sulfonyl fluoride (R-OSO2F) has emerged as a promising active moiety in pesticide development. Previously, our team identified aryl sulfonyl fluoride compounds with strong antifungal activity. Expanding on this, we incorporated the sulfonyl fluoride group into the heterocyclic amide scaffold of SDHIs, designing and synthesizing 56 novel aniline-based sulfonyl fluoride derivatives. Their structures were confirmed using 1H NMR, 13C NMR, and high-r
