PD-L1 regulates c-MET phosphorylation and contributes to MET-dependent resistance to osimertinib in EGFR-mutant NSCLC.

BACKGROUND: Programmed death-ligand 1 (PD-L1) is a well-recognized predictive biomarker for immunotherapy in non-oncogene-addicted non-small cell lung cancer (NSCLC). However, its role in epidermal growth factor receptor (EGFR)-mutant NSCLC remains unclear. This study aims to investigate the impact of PD-L1 on the signaling pathways in EGFR-mutant NSCLC. METHODS: The regulatory role of PD-L1 was investigated through in vitro manipulation of PD-L1 expression across several EGFR-mutant cell lines, followed by analysis via human receptor tyrosine kinase (RTK) array, Western blotting, protein tyr

Comprehensive proteomic and pathological profiling identifies PRAS40 as a novel biomarker and mediator of primary immune checkpoint blockade resistance in non-small cell lung cancer.

BACKGROUND: Immune checkpoint blockade (ICB) has revolutionized the treatment landscape of non-small cell lung cancer (NSCLC), yet primary resistance remains a significant clinical challenge. Recent evidence implicates PRAS40 (AKT1S1) in regulating cellular survival and immune responses, but its role in immunotherapy resistance is not fully understood. METHODS: Transcriptomic data from TCGA and GTEx cohorts were analyzed to assess PRAS40 expression. Prognostic value was evaluated using Cox regression. Immune microenvironment features were characterized with CIBERSORT and TIMER. Predictive eff

SLC25A39 regulates Hedgehog signaling to promote tumor progression and sorafenib resistance in hepatocellular carcinoma.

Sorafenib is the standard treatment for advanced hepatocellular carcinoma (HCC), yet resistance limits its efficacy. The Hedgehog (HH) signaling pathway contributes to drug resistance by maintaining HCC stem cell characteristics, but its role at the single-cell level is underexplored. Utilizing single-cell RNA sequencing (scRNA-seq) and machine learning, we identified solute carrier family 25 member 39 (SLC25A39) as a key regulator of the HH pathway. Analyses of various cohorts revealed that elevated SLC25A39 correlates with poor prognosis in HCC patients. SLC25A39 knockdown inhibited cancer s

Isolation and Characterization of Putative Cancer Stem Cells from Oral Squamous Cell Carcinoma Cells.

BACKGROUND/AIM: Oral squamous cell carcinoma is the most common oral malignancy, and still, despite the recent therapeutic advances, has rather poor prognosis with low 5-year survival rates. One possible cause for the treatment failures are cancer stem cells, which may participate in the resistance to chemo- and radiotherapies. The aim of our study was to isolate cancer stem cell subpopulations from an oral squamous cell carcinoma cell line based on CD44 and CD326 CSC markers, and to investigate their phenotypic characteristics. MATERIALS AND METHODS: Cell subpopulations with distinct stainin

System analysis links SMARCD3 regulons to growth signaling and MEK inhibitor response in everolimus-resistant ER+ breast cancer cells.

Estrogen receptor-positive breast cancer (ER+BC) accounts for ∼70% of all breast tumors, and 20%-40% of patients develop metastases. Everolimus is an mammalian target of rapamycin (mTOR) inhibitor used in combination with exemestane for metastatic ER+BC. However, resistance remains common and leads to poor survival outcomes. To uncover resistance mechanisms, we analyze transcriptomic profiles from everolimus-sensitive and -resistant ER+BC cell lines. Our study uncovers persistent activation of a growth-factor signaling meta-phenotype in resistant cells involving IGF1R, ESR1, and mitogen-activa

Distinct roles of lysophosphatidic acid (LPA) receptor signaling in regulating gastric cancer cell functions under chemical versus physiological hypoxia.

Hypoxia plays a crucial role in driving tumor progression by altering cellular signaling pathways. Lysophosphatidic acid (LPA) receptor signaling regulates malignant properties in cancer cells, including motility and chemoresistance. This study aimed to compare the cellular functions of gastric cancer AGS cells under cobalt chloride (CoCl2)-induced hypoxia and true hypoxia (1 % O2), with a focus on the role of LPA receptor signaling in mediating these responses. Treatment with CoCl2 (200 μM) elevated LPAR1 and LPAR3 expression while reducing LPAR2 expression, resulting in enhanced cell motilit

TMEM205 promotes cisplatin resistance in triple-negative breast cancer and induces TAM/M2 polarization.

Transmembrane protein 205 (TMEM205) is aberrantly expressed in various tumors and is closely associated with cisplatin (DDP) resistance. However, its role in tumor progression and drug resistance in triple-negative breast cancer (TNBC) remains unclear. This study systematically investigates the regulatory function of TMEM205 in DDP-resistant TNBC and explores the underlying molecular mechanisms through a series of in vitro and in vivo experiments. Ours results demonstrate that TMEM205 promotes biological functions in TNBC cells by activating the Wnt/β-catenin signaling pathway, which subsequen

Sustained Galiellalactone treatment breaks the metastatic resilience of BRAF-inhibitor-resistant melanoma cells.

STAT3 hyperactivation in metastatic and therapy-resistant melanoma drives tumorigenesis and immunotherapy resistance by promoting tumor-stromal interactions, making its inhibition a crucial therapeutic strategy. Interestingly, synthetic STAT3 inhibitors could not manage to enter clinical trials due to their short half-lives, poor bioavailability and non-specific effects. Here, we have shown that Galiellalactone, a natural, irreversible STAT3 inhibitor, blocks IL-6 and WNT5A-mediated STAT3 Phosphorylation and migration in BRAF-inhibitor (BRAFi)-sensitive melanoma cells. Galiellalactone treatmen

Tumor-intrinsic interferon signaling drives pancreatic cancer resistance to tumor mucin1-targeted CAR T cell therapy.

Pancreatic cancer (PC) remains one of the most challenging cancers and has the worst prognosis. Tumor-associated MUC1 (tMUC1) is overexpressed and aberrantly glycosylated in over 80% of human pancreatic ductal adenocarcinoma (PDA). Chimeric antigen receptor (CAR) engineered T cells are an emerging cancer immunotherapy strategy and recently, we successfully engineered tMUC1-specific human and mouse CAR T cells and demonstrated their effectiveness as monotherapy against PDA in vitro and in vivo. In this study, we observed varying sensitivity among human PDA cell lines in response to tMUC1-target

Loss of cyclin C drives resistance to anti-TIGIT therapy by upregulating CD155-mediated immune evasion.

AIMS: CD155 is an immune checkpoint protein expressed in tumor cells that interacts with its ligand T cell immunoreceptor with immunoglobulin and ITIM domain (TIGIT) on natural killer (NK) cells and T cells, mediating inhibitory regulation on immune cells. Blockade of the CD155-TIGIT interaction has demonstrated clinical benefits in patients with advanced cancers. The transcriptional and post-translational mechanisms governing CD155 expression remain largely unknown. METHODS: To identify regulators of CD155, we conducted a genome-wide CRISPR-Cas9 screen in cancer cells. Surface CD155 protein

Apolipoprotein E promotes primary resistance to AR-targeted therapy via inducing TRIM25-mediated AR ubiquitination and sensitizes immunotherapy in prostate cancer.

Rationale: Prostate cancer (PCa) growth is facilitated by the androgen receptor (AR) and its downstream signaling pathways, making AR-targeted therapy crucial for treating advanced stages. Despite this, the response to AR-targeted therapies is inconsistent, with a significant proportion of patients even exhibiting unresponsiveness to therapy from the outset, known as primary resistance. Therefore, a refined categorization framework is imperative for the timely detection of resistant phenotypes and the exploration of novel therapeutic avenues. Methods: Tissue microarrays and clinical cohorts we

Dose-Dense Docetaxel and Radium-223 in Bone-Dominant Metastatic Castration-Resistant Prostate Cancer.

BACKGROUND: Disease progression in castration-resistant prostate cancer (CRPC) remains bone-dominant and docetaxel-responsive. Docetaxel and radium-223 would be a logical combination but myelosuppression is dose-limiting. Dose-dense schedules of docetaxel have comparable activity to bolus dosing with mitigated myelosuppression. We hypothesized that dose-dense docetaxel with standard radium-223 would be a feasible, safe and effective combination in bone-dominant metastatic CRPC. METHODS: Subjects had progressive bone-predominant CRPC. Design was dose escalation plus expansion with 28-day cycle

Roles for the Nerve Growth Factor-Farnesoid X Receptor-Retinol Dehydrogenase 16 Axis in Hepatocellular Carcinoma Prognosis and Chemosensitivity.

Nerve growth factor (NGF) has been shown to upregulate expression of retinol dehydrogenase 16 (RDH16) and farnesoid X receptor (FXR), thereby conferring hepatoprotection against cholestatic and oxidative injury. This study investigated the expression levels of NGF, RDH16, and FXR in human hepatocellular carcinoma (HCC) tissues and explored their relationships, as well as their roles in tumorigenesis and resistance to antitumor chemotherapy. Transcriptomic data set and Kaplan-Meier survival analyses revealed that HCC patients with higher NGF and RDH16 expression exhibited better survival rates.

Efficacy and toxicity of low-doses versus standard-dose enzalutamide in advanced prostate cancer: a real-world study with implications for cancer prevention/interception.

BACKGROUND: Enzalutamide at standard doses (160 mg/day) is effective in advanced prostate cancer (PCa) but is associated with considerable adverse events (AEs) and high financial costs. Preliminary reports suggest that low/intermediate doses (≤80 mg/day) retain efficacy while reducing toxicity. This study evaluates the efficacy and safety of low/intermediate dose vs standard-dose enzalutamide in a real-world cohort. METHODS: This retrospective observational study included 140 assessable patients treated with enzalutamide for metastatic castration resistant (80%) or sensitive (20%) PCa. Patien

PRMT1-catalyzed NUSAP1 methylation enhances Notch2 signaling and 5-FU resistance in gastric cancer.

5-Fluorouracil (5-FU) resistance remains a significant challenge in the treatment of gastric cancer, limiting its clinical efficacy. Our study identifies NUSAP1, a nucleolar and spindle-associated protein, as a key driver of 5-FU resistance in gastric cancer. Proteomic analyses of 5-FU-resistant gastric cancer cell lines revealed that NUSAP1 is significantly upregulated, and functional studies demonstrated its essential role in promoting resistance, proliferation, migration, invasion, and tumor growth. Mechanistic investigations revealed that NUSAP1 undergoes asymmetric dimethylation (ADMA) at

Metabolic alterations driven by LDHA in CD8 + T cells promote immune evasion and therapy resistance in NSCLC.

Non-small cell lung cancer (NSCLC) is a leading cause of cancer-related deaths worldwide. Despite advancements in treatment, prognosis for patients with advanced stages remains poor. Metabolic reprogramming in the tumor microenvironment, particularly abnormal glycolysis, plays a crucial role in immune evasion and treatment response. We collected nine single-cell datasets to create a single-cell atlas of CD8 + T cells from 89 NSCLC patients, revealing ten distinct states of these cells. We employed a multimodal data analysis approach, integrating bulk transcriptomics, single-cell transcriptomic
