[Clinically relevant drug interactions with new generation antidepressants and antipsychotics].

Because antidepressants and antipsychotics are commonly described in combination with drugs used to treat comorbid psychiatric or somatic disorders (e.g. anxiolytics, mood stabilizers, cardiovascular drugs, antimicrobial agents), they may be involved in drug interactions. Furthermore, agents such as lithium and atypical antipsychotics may be used to augment the antidepressant response in cases of refractory depression. Based on their mechanisms, drug-drug interactions can be classified either as pharmacokinetic or pharmacodynamic in nature. The well-documented risk of potentially harmful pharm

GHK-Cu as a Bioactive Metallopeptide and Drug-Delivery Cargo: Coordination Chemistry, Formulation Science, Therapeutic Evidence, and a Translational Roadmap.

Background/Objectives: Glycyl-L-histidyl-L-lysine (GHK) and its copper(II) complex GHK-Cu have been studied for matrix remodeling, inflammation, redox regulation, angiogenesis, and tissue repair, yet the literature frequently treats GHK-Cu as a single active ingredient despite formulation-dependent variation in coordination state, speciation, stability, pharmacokinetics, and toxicity. We critically evaluate GHK-Cu simultaneously as a bioactive metallopeptide and as a drug-delivery cargo, with explicit separation of apo-GHK, canonical GHK-Cu, GHK-derived copper peptides, and non-GHK copper-pept

[Preparation, characterization, and in vitro anti-liver tumor activity of bufalin nanoparticles with Scrophularia ningpoensis polysaccharide and ursodeoxycholic acid as carriers].

Bufalin(BF)has a significant anti-tumor effect, but its clinical application is severely restricted by its high toxicity and poor water solubility. In this study, Scrophularia ningpoensis polysaccharide(SNP)and ursodeoxycholic acid(UDCA) were synthesized into an SNP-UDCA conjugate. BF was encapsulated to prepare BF/SNP-UDCA nanoparticles(NPs). The amphiphilic compound SNP-UDCA was synthesized via the one-step method, and its structure was characterized by Fourier-transform infrared spectroscopy(FT-IR)and proton nuclear magnetic resonance(~1H-NMR). The preparation process of BF/SNP-UDCA NPs was

Natural and Designed Cyclic Peptides as Potential Antiviral Drugs to Combat Future Coronavirus Outbreaks.

The COVID-19 pandemic has underscored the need for effective and affordable antiviral drugs. Anthropogenic activities have increased interactions among humans, animals, and wildlife, contributing to the emergence of new and re-emerging viral diseases. RNA viruses pose significant challenges due to their rapid mutation rates, high transmissibility, and ability to adapt to host immune responses and antiviral treatments. The World Health Organization has identified several diseases (COVID-19, Ebola, Marburg, Zika, and others), all caused by RNA viruses, designated as being of priority concern as

A three-dimensional (3D) liver-kidney on a chip with a biomimicking circulating system for drug safety evaluation.

The liver and kidney are the major detoxifying organs in the human body and play an important role in pharmacokinetics. Drug-induced hepatotoxicity and nephrotoxicity can cause irreversible damage to the liver and kidney and are a major cause of drug failure in later stages. Both animal models and conventional cell culture have a number of limitations, such as animal ethics and gene mismatching and there is an urgent need to develop a new drug toxicity evaluation approach. In this paper, a 3D liver-kidney on a chip with a biomimicking circulating system (LKOCBCS) was constructed to obtain kidn

Phenotypic Screening Approaches to Develop Aurora Kinase Inhibitors: Drug Discovery Perspectives.

Targeting mitotic regulators as a strategy to fight cancer implies the development of drugs against key proteins, such as Aurora-A and -B. Current drugs, which target mitosis through a general mechanism of action (stabilization/destabilization of microtubules), have several side effects (neutropenia, alopecia, and emesis). Pharmaceutical companies aim at avoiding these unwanted effects by generating improved and selective drugs that increase the quality of life of the patients. However, the development of these drugs is an ambitious task that involves testing thousands of compounds through bio

Understanding the neurobiological consequences of early exposure to psychotropic drugs: linking behavior with molecules.

Children receive significant exposure to psychotropic drugs. Some psychiatric disorders are diagnosed and treated in children as young as 2 years old, resulting in exposure to prescription stimulants, antidepressants, and mood stabilizers during brain development. Difficulties in diagnoses at such young ages increase the likelihood that children who are not affected by these disorders receive drug exposure inadvertently. Additionally, the increased availability of caffeine-containing beverages in schools has facilitated exposure to this stimulant in children. However, the consequences of expos

Clinical prospects for liposomes.

The use of liposomes has recently been the subject of considerable attention as a promising and versatile approach to drug delivery. Particularly intriguing is the possibility of targeting liposomes to specific areas of the body such as tumors or sites of inflammation or parasitic invasion for either local accumulation or release of associated drugs. This review focuses mainly on recent in vivo work having clinical potential. An extensive discussion of liposome preparation and entrapment of drugs for controlled release in vivo is also included. The stability of liposomes in biological fluids i

Diving on the Surface of a Functional Metal Oxide through a Multiscale Exploration of Drug-Nanocrystal Interactions.

While recent advances in nanotechnology offer significant possibilities for improving the development of targeted drug delivery systems (DDSs), the design of efficient nanocarriers remains challenging due to the complex interactions among nanoparticles, their surfaces, and therapeutic agents in biological environments. To shed light on such difficulties and provide an instrumental tool for the refinement of DDSs, this study presents a comprehensive computational and experimental approach for the development of zinc oxide nanocrystals (ZnO NCs), exploited as carriers for a hydrophobic drug used

Preparation and performance of a stimuli-responsive drug delivery system: novel light-triggered temperature-sensitive drug-loaded microcapsules.

Stimuli-responsive/smart drug delivery systems (DDSs), particularly those that use temperature as a stimuli-response factor to activate drug release, are the subject of recent research. A phase change material (PCM) is a popular thermally responsive material that can be used as a drug carrier and only when the system temperature is above the phase change point is the drug released following the phase change material changing from solid to liquid. In this study, a novel NIR light-triggered temperature-sensitive drug delivery system is developed for controllable release of acyclovir (ACV). For t

Solid-State Behavior and Solubilization of Flash Nanoprecipitated Clofazimine Particles during the Dispersion and Digestion of Milk-Based Formulations.

Clofazimine, a drug previously used to treat leprosy, has recently been identified as a potential new drug for the treatment for cryptosporidiosis: a diarrheal disease that contributes to 500 000 infant deaths a year in developing countries. Rapid dissolution and local availability of the drug in the small intestine is considered key to the treatment of the infection. However, the commercially available clofazimine formulation (Lamprene) is not well-suited to pediatric use, and therefore reformulation of clofazimine is desirable. Development of clofazimine nanoparticles through the process of

In silico drug repurposing of beta-blockers against HSV-1 TK using molecular docking and molecular dynamics simulations.

Herpes simplex virus type 1 (HSV-1) is highly prevalent, but emerging drug-resistant strains against nucleoside analogs like acyclovir demand novel therapeutics. This study employed an in silico drug repurposing approach to screen 41 beta-blockers as potential non-nucleoside inhibitors targeting HSV-1 thymidine kinase (TK). Molecular docking, 100 ns molecular dynamics (MD) simulations, MM/GBSA binding free energy calculations, per-residue energy decomposition, and ADMET analysis were performed. Docking results revealed several beta-blockers surpassed acyclovir (-5.20 kcal/mol) in binding energ

AS1411-aptamer-functionalized DNA tetrahedron for targeted delivery of vorinostat to suppress gastric cancer progression via ferroptosis induction and epithelial-mesenchymal transition inhibition.

Gastric cancer (GC) exhibits progression and metastatic spread that is tightly linked to ferroptosis and epithelial-mesenchymal transition (EMT). Developing therapies that target both pathways represents a promising clinical strategy. Herein, we identified the histone deacetylase (HDAC) inhibitor vorinostat as a potent dual-pathway modulator through bioinformatics screening. To address its limitations in aqueous solubility and targeting efficiency, a DNA tetrahedron nanostructure conjugated with the AS1411 aptamer (TAV) was synthesized as a novel drug delivery system. The synthesis of TAV was

Computational insights into fenbendazole encapsulation by Cucurbit[n]urils (n = 7, 8): Implications for drug delivery and nanocarrier design through integrated structural, electronic, thermodynamic, and spectroscopic investigations.

Fenbendazole (fenben), a benzimidazole anthelmintic with emerging anticancer potential, faces significant challenges in clinical applications due to its poor aqueous solubility and potential toxicity at high doses. To address these limitations, this study explores the encapsulation of fenben within cucurbit[7]uril (CB[7]) and cucurbit[8]uril (CB[8]) using computational methods to evaluate host-guest interactions, stability, and electronic properties. Density functional theory (DFT) calculations at the APFD/6-31+G(d) level revealed that CB[7] forms more stable complexes with fenben than CB[8],
