Ferroptosis in health and disease.

Ferroptosis is an iron-dependent, non-apoptotic form of regulated cell death that is driven by lipid peroxidation. In the past decade, it has emerged as a core pathogenic mechanism in a wide range of human diseases, such as neurodegenerative disorders, cardiovascular disease, cancer, and notably, reproductive dysfunction. This review provides a comprehensive overview of the molecular mechanisms underlying ferroptosis with a specific focus on the System Xc⁻-GSH-GPX4 axis, iron metabolism, lipid peroxidation, and the regulatory roles of key modulators such as Nrf2, ACSL4, and CISD2. This review

Crotonylation: A novel layer of the epigenetic landscape in disease.

Crotonylation is a lysine acylation modification that links cellular metabolism to epigenetic regulation. Its rigid planar crotonyl group is specifically recognized by "readers" such as YEATS/DPF and confers strong conformational regulatory capacity on proteins. Crotonyl-CoA, derived from amino acid catabolism, fatty acid β-oxidation, and gut microbial metabolites, directly modulates crotonylation levels, positioning this modification as a key metabolite sensor. Crotonylation extensively modifies both histones and non-histone proteins, participating in transcription, DNA repair, autophagy, fer

Tryptophan metabolism: a critical bridge linking gut microbiota to depression.

Depression is a major global public health challenge with limitations in current therapies. Emerging evidence highlights that gut microbiota (GM) critically regulates the gut-brain axis via tryptophan (Trp) metabolism. Trp is metabolized through host-directed serotonin (5-HT) and kynurenine (KYN) pathways, as well as microbiota-specific indole derivative pathways. In depression, gut dysbiosis shifts Trp metabolism toward the KYN pathway, leading to accumulation of neurotoxic metabolites such as quinolinic acid and reduced 5-HT synthesis. Microbiota-derived indoles, functioning as key aryl hydr
