Retinoic acid reduces oxidative stress and enhances migration-related signaling in vascular endothelial cells.

Despite the important role of retinoic acid (RA) in vascular development, its effects on the behavior of endothelial cells (ECs) lining blood vessels remain incompletely understood. In this study, we investigated the effects of RA on oxidative stress and migration, both of which are associated with angiogenesis, using EA.hy926 human ECs. RA suppressed hydrogen peroxide (H2O2) production by EA.hy926 cells in a treatment duration- and concentration-dependent manner without altering the expression of NADPH oxidase 4 (Nox4), the primary H2O2-generating enzyme. This suppression of oxidative stress

Inorganic polyphosphate chelates Ca2+ but cannot substitute for ATP as an energy source for SERCA.

Inorganic polyphosphate (polyP) is a high-energy polymer with phosphoanhydride bonds analogous to ATP, but whether it can serve as a phosphoryl donor for mammalian ATP-dependent enzymes remains unclear. Because PMCA was previously reported to utilize polyP in human erythrocytes, we tested whether the closely related sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) can use polyP to drive Ca2+ transport or support polyphosphate kinase-like activity. Protein BLAST identified SERCA as the closest non-PMCA homolog in the human proteome, and docking simulations predicted polyP-14 binding to SERCA wit

MZB1 Modulates Inflammatory Severity in Severe Acute Pancreatitis through an IgA-Associated Intestinal Barrier Axis.

Severe acute pancreatitis (SAP) is characterized by excessive inflammatory responses that critically influence disease severity and systemic complications. Although innate immune activation has been extensively studied in SAP, the contribution of adaptive humoral immunity to inflammatory modulation during disease progression remains incompletely understood. In this study, we performed longitudinal single-cell RNA sequencing of peripheral blood mononuclear cells from patients with mild and severe acute pancreatitis to characterize immune alterations across disease stages. We observed dynamic im

GTPase cycle and ER membrane regulate cargo handover during cotranslational protein targeting.

Signal recognition particle (SRP), a universally conserved protein-targeting machine, cotranslationally recognizes signal sequences of nascent endomembrane proteins and engages the SRP receptor (SR) at the endoplasmic reticulum (ER) membrane to unload the translating ribosome. The mechanism of membrane-associated steps in this pathway remains unclear. Here, we address this question using pre-steady-state and single-molecule fluorescence measurements. SR induces release of the signal sequence from SRP and detachment of the SRP guanosine triphosphatase (GTPase) domain from the ribosome. Despite

Protective effects of resveratrol against perfluorooctane sulfonamide-induced cardiac developmental toxicity in zebrafish larvae.

We have reported that Perfluorooctane sulfonamide (PFOSA), a direct precursor of perfluorooctane sulfonic acid (PFOS), disrupts cardiac development by activating the aryl hydrocarbon receptor (AhR) and inducing oxidative stress. Resveratrol (RSV) is a natural polyphenolic compound with both AhR-inhibitory and antioxidant properties. Therefore, we hypothesized that RSV may exert protective effects against PFOSA-induced cardiac defects. In the present study, we demonstrated that PFOSA exposure led to cardiac malformations and functional impairments in zebrafish embryos, effects that were signifi

[1,3]Oxazine-based NIR molecular switches: Hydrochromic behavior, viscosity sensing, and targeted cell imaging.

Molecular switches based on [1,3] oxazine show distinctive properties between their ring-closed and ring-opened forms (RCF and ROF), differing in conjugation length, charge, hydrophilicity, and optical behavior. These features enable low background interference and organelle-selective targeting, which are highly valuable for intracellular fluorescence imaging. However, related studies remain scarce. In this work, two near-infrared (NIR) molecular switches, BOA-1 and BOA-2, were designed using [1,3] oxazine as the core scaffold. They exhibit excellent hydrochromic behavior with a 300 nm absorpt

IGFBP3 repression driven by inflammation links air pollution to placental and developmental defects.

Air particulate matter (PM2.5 and PM10), can cross the placental barrier, triggering oxidative stress and inflammation that compromise fetal development. These insults lead to placental dysfunction and complications including preterm birth, low birth weight, and preeclampsia. In cell line and placental explant models, urban particulate matter (UPM) increased pro-inflammatory cytokines and oxidative stress pathways, impairing trophoblast invasion, angiogenesis, and nutrient transport, while also altering epigenetic modifications and endoplasmic reticulum function. Rodent studies revealed reduce

Cisplatin-induced renal injury: ROS-mediated oxidative stress - mechanism and antioxidant intervention strategies.

Cisplatin, a widely utilised anti-cancer agent, is severely limited in clinical practice due to its various associated toxicities, among which nephrotoxicity is particularly prominent. Cisplatin-induced acute kidney injury is mainly characterized by proximal tubular epithelial cell injury, involving many complex molecular mechanisms. Recent numerous studies have identified oxidative stress as a core of cisplatin-induced nephrotoxicity, which affects the generation of reactive oxygen species (ROS) and the disruption of the antioxidant defense system. ROS not only directly damages biological mac

Calcium homeostasis imbalance in sepsis: molecular mechanisms and therapeutic perspectives.

Calcium ions (Ca2+) are pleiotropic signaling messengers that regulate diverse cellular processes, and their homeostasis is essential for normal physiology at cellular, tissue, and organismal levels. Accumulating evidence identifies Ca2+ dysregulation as a key driver of sepsis onset and progression. In this review, we systematically delineate the mechanisms governing intracellular Ca2+ homeostasis, focusing on coordinated regulation among the endoplasmic reticulum, mitochondria, and lysosomes. We also discuss the molecular basis of disordered Ca2+ signaling in sepsis-associated pathologies and

PFKM acts as a metabolic switch initiating donut-shaped mitochondrial remodeling to promote osteogenesis.

Osteogenic differentiation requires sophisticated mitochondrial adaptation to meet bioenergetic demands, yet regulatory checkpoints governing this organelle reorganization remain poorly defined. Through single-cell RNA sequencing reanalysis and metabolic intervention, this study unveils a non-canonical signaling role for PFKM, traditionally recognized solely as a glycolytic enzyme, in orchestrating mitochondrial remodeling during bone formation. Beyond its established metabolic function, Pfkm suppression triggers distinctive donut-shaped mitochondria through a novel signaling cascade. Mechanis

Dual-Phase Barrier-Penetrating Nanomedicine for Early Intervention on Hepatic Stellate Cells to Enhance Liver Fibrosis Treatment.

Inhibition of activated hepatic stellate cells (HSCs) is essential for treating fibrotic liver disease, while the capillarized hepatic sinusoidal barrier extremely hinders HSC-targeted drug delivery and early intervention. Herein, we developed a dual-phase barrier-penetrating nanomedicine (SFR-RA@lip) based on the coassembly of silibinin (SIL) and riociguat (RIO), with retinoic acid (RA) for HSCs targeting and a lipid shell to facilitate multipathway transcellular transport. In the initial phase, SFR-RA@lip exploits caveolin-mediated endocytosis and endoplasmic reticulum-Golgi apparatus traffi

Imaging mass cytometry reveals functional and immunological changes during type 1 diabetes progression in human pancreata.

The pathogenesis of type 1 diabetes, particularly at autoantibody-positive preclinical stages, remains poorly understood, in part due to limited sample availability. Here we show imaging mass cytometry data of pancreas samples from 88 organ donors, including 28 single and 10 multiple autoantibody-positive donors. We imaged 16 million single-cells using 79 antibodies to characterize β-cell states and the islet-immune interface, correcting for relevant covariates. We identified interactions between pro-inflammatory macrophages and exhausted-like (PD1+TIM3+) T cells. These interactions were chara

Glycation-Driven Mitochondrial and ER Stress Underlies Iodoacetic Acid-Induced Apoptosis in Porcine Uterus and Oviduct Epithelial Cells.

Iodoacetic acid (IAA), a highly cytotoxic disinfection byproduct commonly detected in drinking water, poses a potential risk to female reproductive health. The direct molecular mechanisms underlying its effects on the reproductive system epithelium remain unclear. This study demonstrates that IAA induces glycational stress in primary porcine uterine (UECs) and oviduct epithelial cells (OECs), representing an early event contributing to extensive cellular toxicity. IAA exposure inhibited Glyceraldehyde-3-Phosphate Dehydrogenase (GAPDH) enzymatic activity and promoted the accumulation of advance

Capmatinib Counteracts LPS-Induced Pulmonary and Testicular Injury Through Attenuating TLR4/NF-κB, PERK/PI3K, and Caspase-1/GSDMD Signaling Pathways.

Lipopolysaccharide (LPS)-mediated systemic inflammatory response accounts for the LPS pathogenicity in multi organs as lung and testis. LPS could cause a dysregulation of host response to infection, which may lead to life-threatening organ dysfunction. Accordingly, in our study, the probable protective impact of capmatinib against the LPS-induced acute inflammatory response was explored and the possible causal mechanisms were examined. Male Albino mice were pretreated with capmatinib (5 or 10 mg/kg, daily, orally) for 3 days then received single intraperitoneal injection of LPS (10 mg/kg) at d

Carfilzomib Induces Cardiotoxicity by Blocking Autophagic Flux Through the cGAS-STING Signaling Pathway.

Carfilzomib (CFZ) is a proteasome inhibitor primarily used to treat relapsed and refractory multiple myeloma. However, its clinical application is limited by significant cardiotoxicity, the underlying mechanisms of which remain incompletely understood. In this study, we aimed to elucidate the pathogenic pathways involved. In vitro, CFZ induced mitochondrial dysfunction and apoptosis in AC16 cardiomyocytes in a concentration- and time-dependent manner. Transcriptomic analysis revealed enrichment in pathways related to autophagy and endoplasmic reticulum stress. Mechanistically, CFZ promoted aut

Biochemical Mechanisms of Cellular Stress Adaptation in the Pathogenesis of Chronic Diseases.

Chronic diseases increasingly reflect a shared biological origin: persistent cellular stress. This review summarizes the biochemical mechanisms that normally preserve cellular homeostasis, namely redox regulation, endoplasmic reticulum proteostasis, mitochondrial quality control, autophagy, and DNA damage response, and explains how they fail under sustained lifestyle-related overload. Repeated exposure to psychological stress, sleep disruption, hypercaloric intake, and physical inactivity shifts adaptive signaling toward maladaptation, promoting oxidative damage, protein misfolding, mitochondr

ERAD-level gene switches for on-demand protein secretion and rapidly controlled gene therapies.

On-demand regulation of therapeutic activities is critical to broaden the clinical utility of gene therapies. Here, we employ an Endoplasmic Reticulum-Associated Degradation (ERAD)-centered strategy to develop trigger-inducible gene switches tailor-designed for rapid protein secretion. By temporarily attaching conditional degron motifs to various therapeutic proteins of interest (POIs), inducible, tunable and reversible control of native POI secretion into the bloodstream was achieved through oral administration of specific clinically licensed small-molecule drugs. This simple and generalizabl

Non-cognate CD8 Binding to MHC I Promotes Positive Selection of an MHC-E Restricted CD8 T Cell Population.

MHC-E-restricted CD8 T cells are emerging as an attractive therapeutic mechanism due to their strong protective capacity and ability to respond to cells with defects in antigen processing; however, their thymic development remains poorly understood. Here, we explore the MHC ligand requirement for thymic development of T cells reactive to a self-peptide (FL9) presented by mouse MHC-E (Qa1b) under conditions of deficiency in the endoplasmic reticulum aminopeptidase associated with antigen processing (ERAAP), called QFL T cells. We show that, while QFL T cells can develop in the absence of the re

Fluoxetine disrupts cholesterol metabolism in endothelial cells via SREBP2 activation.

Fluoxetine is a selective serotonin reuptake inhibitor (SSRI) commonly prescribed for the treatment of depressive disorders. Recent clinical reports and studies in animal models have suggested that fluoxetine increases the risk of cardiovascular diseases, but the underlying mechanisms remain unknown. Here, we uncover that fluoxetine disrupts lipid and cholesterol metabolism in primary human endothelial cells (ECs). Fluoxetine triggered an upregulation of cholesterol metabolism genes, leading to the accumulation of lipid droplets in ECs. We find higher levels of cholesterol esters, ceramides, s
