New case of mitochondrial HMG-CoA synthase deficiency. Functional analysis of eight mutations.

Mitochondrial HMG-CoA synthase deficiency is a rare inherited metabolic disorder that affects ketone-body synthesis. Acute episodes include vomiting, lethargy, hepatomegaly, hypoglycaemia, dicarboxylic aciduria, and in severe cases, coma. This deficiency may have been under-diagnosed owing to the absence of specific clinical and biochemical markers, limitations in liver biopsy and the lack of an effective method of expression and enzyme assay for verifying the mutations found. To date, eight patients have been reported with nine allelic variants of the HMGCS2 gene. We present a new method of e

Evidence that ecto-nucleoside-triphosphate pyrophosphatase serves in the generation of extracellular inorganic pyrophosphate in human bone and articular cartilage.

Extracellular inorganic pyrophosphate (PPi) is important in the regulation of mineralisation of bone, and in the pathogenesis of chondrocalcinosis, an arthritic disease in which calcium pyrophosphate dihydrate crystals form in articular cartilage. Nucleoside-triphosphate pyrophosphatase, which catalyses the formation of PPi, was previously observed at the surface of human articular chondrocytes in culture. A similar enzyme has been identified in osteoblast-like human bone cells in culture, and is active towards purine and pyrimidine nucleoside triphosphates. The enzyme has high affinity for AT

The metabolism of adenine derivatives in different parts of the brain of the rat, and their release from hypothalamic preparations on excitation.

Five enzymes concerned with the metabolism of adenine derivatives were assayed in seven regions of the rat brain. A region which included the hypothalamus had the highest AMP deaminase and adenosine deaminase activities, while its 5'-nucleotidase activities were relatively low. The enzymes named and also the uptake of [14C]adenine by incubated tissue samples were more active with hypothalamic than with neocortical tissues. On superfusion with glucose-bicarbonate saline after assimilating [14C]adenine, the hypothalamic tissues released about 0.2 per cent of their 14C content per minute. This re

Role of oligandrin in enhancing post-harvest disease resistance in cherry tomato through salicylic acid and jasmonic acid signaling pathways.

Salicylic acid (SA) and jasmonic acid (JA) pathways are crucial components of plant defense mechanisms; however, their roles in post-harvest preservation remain inadequately explored. Cherry tomatoes are highly susceptible to fungal pathogens, such as Alternaria nees, after harvest, leading to significant post-harvest losses. This study investigates the effects of oligandrin, an elicitin protein derived from Pythium oligandrum, on enhancing the resistance of harvested cherry tomatoes to A. nees. Oligandrin treatment significantly reduced disease symptoms, including black spots and decay, durin

Biochemical signal transmitted by Fc receptor for immunoglobulin G2a of a murine macrophage-like cell line, P388D1: mode of activation of adenylate cyclase mediated by immunoglobulin G2a binding proteins.

The effects of immunoglobulin G2a binding proteins isolated from P388D1 cells on adenylate cyclase of cyc- cells were investigated to explore a potential role of Fc gamma 2a receptor in the activation of the adenylate cyclase system. Immunoglobulin G (IgG) binding proteins obtained from the detergent lysate of P388D1 cells by affinity chromatography on IgG-Sepharose were separated into two fractions (denoted as IgG-B1 and IgG-B2) by Sephadex G-100 gel filtration in the presence of 6 M urea. Polyacrylamide gel electrophoretic analysis in the presence of sodium dodecyl sulfate revealed that the

CX3CL1/CX3CR1-Mediated Neuron-Microglia Crosstalk Contributes to Methamphetamine-Induced Neuroinflammation and Cognitive Impairment.

Methamphetamine (METH) exposure induces neuroinflammation and cognitive impairment, yet the molecular signals that convert neuronal stress into microglial inflammatory activation remain unclear. This study investigated whether disruption of the CX3CL1/CX3CR1 signaling contributes to METH-induced injury in the prefrontal cortex (PFC). C57BL/6J mice were exposed to METH and assessed for behavioral changes using Y-maze, novel object recognition, and open-field tests. Neuronal injury was evaluated by Nissl staining, and CX3CL1, CX3CR1, and inflammatory mediators were quantified by Western blotting

Development of bimolecular biosensors to detect chemicals with agonist activity toward rat PXR.

The pregnane X receptor (PXR) exhibits broad ligand specificity and responds to diverse chemicals, making it an attractive target for evaluating the biological effects of exogenous chemicals. This study developed biosensors based on the integration of the PXR activation mechanism and NanoLuc Binary Technology to establish an in vitro assay for evaluating interactions between chemicals and rat PXR (rPXR). The biosensors comprised two molecules: rPXR ligand-binding domain (LBD)-SmBiT, in which the LBD is fused to the N-terminus of SmBiT, and LgBiT-LXXLL, in which a coactivator-derived LXXLL moti

HIV broadly neutralizing antibodies with enhanced Fc-CD16 affinity increase NK cell ADCC even with limited envelope binding.

The persistence of latent HIV-1 reservoirs remains the primary barrier to a cure. Shock and kill strategies aim to reactivate these reservoirs and eliminate them via effector cells, such as Natural Killer (NK) cells. However, chronic infection leaves NK cells exhausted. In this study, we investigated the interplay between cytokine-mediated NK cell activation and antibody-dependent cellular cytotoxicity (ADCC) across diverse HIV-1 subtypes. We demonstrated that while cytokine stimulation enhanced natural cytotoxicity, it simultaneously induces shedding of the Fc receptor CD16 via the metallopro

A mitochondrial insertion directs substrate selection and engagement by the protein unfoldase ClpX.

The force-generating ATPases Associated with diverse cellular Activities (AAA+) ATPase domain of protein unfoldases is specified for many substrates and other functional partners through elaboration with accessory domains. Mitochondrial homologs of the unfoldase ClpX contain an insertion within the AAA+ domain that is absent in bacterial homologs. This mitochondrial insertion (MI) maps to the substrate-encountering face of ClpX, leading us to hypothesize that the MI directs interactions of ClpX with mitochondrial substrates, the best-characterized of which is the first enzyme in heme biosynthe

ZXDB Drives Macrophage Inflammatory Programming in Sepsis-Induced Acute Kidney Injury by Recruiting EIF4A3 to Enhance ACACA Translation.

Macrophage-driven inflammation is central to the pathogenesis of sepsis-induced acute kidney injury (SI-AKI), yet the role of RNA-binding proteins (RBPs) in post-transcriptionally regulating this process remains elusive. Here, we identify the RBP zinc finger X-linked duplicated B (ZXDB) as an upstream contributor to SI-AKI by promoting a pathogenic, pro-inflammatory macrophage activation state. We found that ZXDB expression is consistently elevated in M1-like macrophages within the kidney during SI-AKI, where it stimulates pro-inflammatory cytokine secretion and glycolytic reprogramming. Mecha

Development of Peroxymonosulfate Catalytic System and Photoenzymatic Catalytic System Utilizing Zn-Doped g-C3N4 for the Degradation of Methylene Blue in Water.

The excessive discharge of wastewater containing methylene blue (MB) has caused certain adverse effects on human health and daily life. This highlights the urgent need for the development of safe and environmentally friendly methods of degradation. In this study, two catalytic systems based on Zn-doped g-C3N4, namely, ZCN-1%/PMS and ZCN-1%/LC, were constructed for efficient activation of peroxymonosulfate (PMS) and increased laccase stability. Experimental results show that Zn-doped g-C3N4 shows much better photocatalytic activity, combined with good enzyme immobilization ability. At a catalys

Inhibition of ERK5 activity ameliorates osteoarthritis by suppressing NLRP3 inflammasome-mediated chondrocyte pyroptosis.

Osteoarthritis (OA) is a widespread degenerative joint disorder manifesting as chronic pain, functional impairment, and progressive disability, imposing a substantial burden on global public health. Despite extensive research, the precise molecular pathways underlying OA pathogenesis remain incompletely elucidated. Our data indicated that human OA cartilage and cartilaginous tissues from aged mice exhibited markedly elevated levels of extracellular signal-related kinase 5 (ERK5) enzyme activity. Using Col2a1-CreERT2/Erk5flox/flox mice, we found that Erk5 deletion in cartilage inhibited the pro

Responsive nanomedicine strategies achieve pancreatic cancer precise theranostics.

Pancreatic ductal adenocarcinoma (PDAC), the predominant subtype of pancreatic cancer, ranks among the deadliest malignancies worldwide, with a 5-year survival rate remaining below 13 %. Its poor prognosis stems from complex anatomical barriers, a dense and heterogeneous tumor microenvironment (TME), and intricate molecular regulatory networks that collectively hinder early detection and limit therapeutic efficacy. Nanomedicine offers promising solutions by enhancing drug loading, improving delivery, counteracting drug resistance, and enabling stimuli-responsive control. Notably, stimuli-respo

Co-catalysis of modified metal-organic framework immobilized enzyme with nonionic surfactant for biodiesel preparation.

With the increasing demand for sustainable energy, enzyme-catalyzed biodiesel preparation has garnered significant interest owing to its green and efficient characteristics. However, conventional immobilized lipase (LIP) is susceptible to desorption and inactivation and fails to balance the catalytic efficiency and stability, limitations that seriously restrict the industrial application. To address this, we constructed a carrier through hydrophobically modifying University of Oslo-66-NH2 (UiO-66-NH2), an amino-functionalized metal-organic framework (MOF), with octadecylphosphonic acid (OPA).

11β-HSD1 inhibitor alleviates lipid metabolism disorder by activating the AMPK signaling pathway.

Lipid metabolism disorders, which are closely linked to obesity, are significantly modulated by the enzyme 11β-hydroxysteroid dehydrogenase 1 (11β-HSD1), a key regulator of glucocorticoid (GC) activation. This study explored the therapeutic potential of H8, a curcumin analog and selective inhibitor of 11β-HSD1, in mitigating these disorders. Employing an in vitro model of GC-induced differentiation in 3T3-L1 adipocytes and an in vivo C57BL/6 mice model characterized by 11β-HSD1 overexpression, we demonstrated that treatment with H8 effectively reduced GC levels in both serum and cultured cells

4-Phenyl Butyric Acid (4-PBA) Suppresses Neutrophil Recruitment in a Murine Model of Acute Perinatal Inflammation.

Neutrophils are the first immune cells to be recruited to the site of infection and deregulated activation is linked to adverse outcome, especially in premature neonates. Dampening neutrophil activity may therefore be a means of preventing acute and chronic inflammatory diseases; however, little is known about potential drugs to modulate neutrophil activity. 4-Phenyl butyric acid (4-PBA) is a clinically used drug, which acts as a chemical chaperone to inhibit endoplasmic reticulum (ER) stress and to suppress immune activation. Here, we investigated the potential of 4-PBA to regulate neutrophil

Single-cell transcriptomic analysis reveals a systemic immune dysregulation in intravenous immunoglobulin non-responsive Kawasaki disease.

BACKGROUND: Intravenous immunoglobulin (IVIG) has been established as the first-line treatment for Kawasaki disease (KD). However, 10%-20% of patients are unresponsive, increasing their risk of coronary artery complications. METHODS: To elucidate the pathogenesis of IVIG non-responsiveness, we performed single-cell transcriptomic profiling on 24 peripheral blood mononuclear cell (PBMC) samples from responsive and non-responsive KD patients before and after IVIG treatment. Finally, the expression of key cytokines was validated using public bulk RNA-seq data and enzyme-linked immunosorbent assa

Photocatalysis-fueled algae-bacteria mutualism enables P450 enzyme-induced nitrite-free anammox for sustainable nitrogen removal.

Anaerobic ammonium oxidation (anammox) relies on scarce nitrite in wastewater to produce hydroxylamine, a key substrate for anammox nitrogen removal, severely limiting its sustainable implementation. This study pioneers a photocatalysis-driven algae-bacteria mutualistic platform for activating a microalgal P450 enzyme-induced nitrite-independent anammox pathway. This platform integrates two functional blocks: photocatalysis-induced P450 activation circuit, and P450-mediated algal-anammox nitrogen removal module. First, photocatalysis rewired microalgal carbon flux to foster cooperation with he

Customized MHC Class I & II restricted peptides from clinical isolates of Mycobacterium tuberculosis tweak strong cellular immune response in Healthy individuals and Pulmonary Tuberculosis patients: A potential candidate in vaccine design.

Tuberculosis (TB) remains a global health challenge as annual mortality rate due to drug resistant TB is increasing exponentially. This is mostly associated with the delayed diagnosis of Multidrug-resistant (MDR) or latent TB. Effective management of TB demands development of novel immunological strategies, such as peptide-based/subunit vaccines that can stimulate specific immune responses. In this context, we evaluated the immunogenic potential of two Major Histocompatibility Complex (MHC) Class I/II-restricted peptides from Mycobacterium tuberculosis (M. tuberculosis): Rv2588c and Rv0148.
