Ruscogenin attenuates osteoarthritis by modulating oxidative stress-mediated macrophage reprogramming via directly targeting Sirt3.

BACKGROUND: Synovial inflammation, Cartilage erosion, and subchondral osteosclerosis, which are collectively referred to as the triad of pathogenesis, contribute to osteoarthritis (OA) progression. Specifically, the M1 macrophage in the synovium worsens the development of the illness and is a significant factor in the deterioration and functioning of cartilage. OBJECTIVE: To investigate whether Ruscogenin attenuates progressive degeneration of articular cartilage in rats with anterior cruciate ligament transection (ACLT)-induced osteoarthritis (OA) by modulating macrophage reprogramming and t

A smart nanoplatform for enhanced photo-ferrotherapy of hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is the third leading cause of cancer-related deaths worldwide. Emerging therapies, such as ferroptosis mediated cancer therapy and phototherapy, offer new opportunities for HCC treatment. The combination of multiple treatments is often more effective than monotherapy, but many of the current treatments are prone to serious side effects, resulting in a serious decline in patients' quality of life. Therefore, the combination therapy of tumor in situ controllable activation will improve the efficacy and reduce side effects for precise treatment of tumor. Herein, we

Counteraction of Myocardial Ferritin Heavy Chain Deficiency by Heme Oxygenase-1.

Given the abundance of heme proteins (cytochromes) in the mitochondrion, it is evident that a meticulously orchestrated iron metabolism is essential for cardiac health. Here, we examined the functional significance of myocardial ferritin heavy chain (FtH) in a model of acute myocardial infarction. We report that FtH deletion did not alter either the mitochondrial regulatory and surveillance pathways (fission and fusion) or mitochondrial bioenergetics in response to injury. Furthermore, deletion of myocardial FtH did not affect cardiac function, assessed by measurement of left ventricular eject

Stage-Dependent Heat-Stress Mechanisms in Chinese Sturgeon (Acipenser sinensis) Early Life-Stages Under Simulated Thermal-Lag Effect.

Hydraulic engineering-driven thermal lag disturbs natural water temperature and induces thermal stress in early-life-stage Chinese sturgeon (Acipenser sinensis), yet stage-specific molecular stress mechanisms remain unclear. This study assessed phenotypic, proteomic and metabolomic responses of endogenous-nutrition embryos and exogenous first-feeding larvae under 16 °C, 19 °C and 22 °C. Phenotypic results showed optimal embryo survival at 16 °C (hatching rate 64.6%), maximum larval growth at 19 °C (specific growth rate 6.79%/d), and severe developmental suppression at 22 °C (with the hatching

CO-mediated restoration of mitochondrial redox homeostasis contributes to alleviating multiple programmed cell deaths in hepatic I/R injury.

Hepatic ischemia-reperfusion injury (HIRI) remains a life-threatening complication in liver surgery and transplantation with no clinically approved therapy. Here we develop a hyaluronic acid-modified, ROS-responsive nanoformulation (CPs@HA) that enables controllable delivery of carbon monoxide (CO) specifically to injured liver tissue. We show that mitochondrial dysfunction is closely associated with three distinct cell death pathways (ferroptosis, cuproptosis, and disulfidptosis) that occur in parallel during HIRI. CPs@HA restores mitochondrial homeostasis through activating the AMPK-SIRT1-PG

The Molecular Signature of Early-Onset Colorectal Cancer Liver Metastases: Distinct Biology and Clinical Challenges.

Early-onset colorectal cancer (EOCRC), defined as diagnosis before the age of 50 years, is increasing globally and is frequently characterized by aggressive biology and a disproportionate burden of liver metastases. This review synthesizes emerging evidence on the distinct molecular, immunologic and clinical features that differentiate EOCRC liver metastases from those arising in older adults. Genomic studies revealed increased chromosomal instability, increased copy number variation burden and unique amplification patterns involving MYC, RAD21, GNAS and MAPK1, alongside altered frequencies of

Biomechanics and Oxidative Stress: An Integrative Review of the Redox-Mechanobiological Axis and the Role of Trace Elements in Disease.

Oxidative stress and mechanobiological signaling are increasingly recognized as interconnected determinants of cellular and systemic homeostasis. Reactive oxygen species (ROS), traditionally associated with molecular damage, are now understood to directly regulate cytoskeletal remodeling, membrane viscoelasticity, mitochondrial dynamics, and mechanotransduction pathways including focal adhesion kinase (FAK), integrins, and Yes-associated protein/transcriptional coactivator with PDZ-binding motif (YAP/TAZ). Conversely, biomechanical forces such as extracellular matrix stiffness, cyclic stretch,

Iron and Other Metal Ions in Human Health and Disease.

Iron is the most abundant trace element, driving both the commonest deficiency and ferroptosis. This review takes iron as the central theme and extends to copper, zinc, and calcium, spanning physiological functions, pathological mechanisms, risk factors, diagnostics, and therapeutics. We classify these ions into redox-active (iron, copper), which engage in catalytic electron transfer, and redox-inert (zinc, calcium), which serve structural and signaling roles without redox cycling. Iron deficiency remains the global leading micronutrient deficit, whereas iron overload triggers ferroptosis-medi

Spatially programmed Fe and Pt electrode-driven electrochemistry reconditions the tumor microenvironment and sensitizes triple-negative breast cancer to PD-1 Blockade.

Excess extracellular acidity is a defining barrier to effective antitumor immunity in solid tumors. Systemic buffers only partially correct tumor acidity and seldom rescue suppressed effector immunity. Here, we developed a spatially programmed electrochemical therapy (ECTFe) integrating a central sacrificial iron anode with peripheral platinum cathodes to engineer the tumor microenvironment in situ. This configuration enables concurrent acidotoxic and ferroptosis-associated tumor debulking in the core, while controlled peritumoral pH relief restores immune cell function. We combined ECTFe with

An engineered ROS-responsive cascade nanoplatform delays Alzheimer's disease progression via Nrf2/GPX4-mediated microglial functional reprogramming.

Alzheimer's disease (AD) is driven by a self-amplifying pathological network in which microglia-mediated neuroinflammation, oxidative stress, and cerebral iron dyshomeostasis are tightly interconnected. Here, we report a ROS-responsive, cascade-targeted nanoplatform (KMAI@NPs) engineered to intervene at this microglia-centered regulatory hub. The nanoplatform integrates an acetylsalicylic acid-modified dextran self-assembly core with PEGylated peptide modules for cascade targeting. By systematically optimizing the dextran molecular weight and acetylsalicylic acid grafting ratio, the self-assem

Targeting the miR-19b/PPP2R5E axis enhances temozolomide response in glioblastoma via ROS-induced DNA damage.

BACKGROUND: Glioblastoma is the most aggressive primary brain tumor. Although temozolomide induces DNA damage, its efficacy is limited by intrinsic resistance mechanisms, leading to tumor relapse. In this study we aimed to identify microRNA-mediated resistance mechanisms. METHODS: MicroRNA screens were performed to identify temozolomide resistance mechanisms. Temozolomide response was evaluated using clonogenic and spheroid assays in glioblastoma cell lines and patient-derived primary cells. DNA damage response was assessed by γH2AX foci formation. Cell cycle distribution, senescence and ferr

Propofol Exerts Cerebroprotective Effects Against Cerebral Infarction by Up-Regulating TRIM67 to Mediate VDAC1 Ubiquitination and Degradation.

Propofol, an intravenous anesthetic, exhibits cerebroprotective effects against cerebral infarction (CI); however, its underlying mechanisms, particularly those involving ferroptosis, remain incompletely understood. This study employed oxygen-glucose deprivation/reperfusion (OGD/R)-treated SK-N-SH cells and middle cerebral artery occlusion (MCAO) rat models to investigate propofol's actions in vitro and in vivo. Propofol alleviated OGD/R-induced neuronal injury by enhancing cell viability, suppressing apoptosis, and reducing the release of inflammatory cytokines interleukin-6 (IL-6) and IL-1β.

Macrophage Iron Metabolism Mediates Immunometabolic Reprogramming and Tissue Homeostasis: From Molecular Mechanisms to Clinical Translation.

BACKGROUND: Macrophages were long regarded as passive executors of erythrophagocytosis responsible for systemic iron recycling. However, increasing evidence has reframed them as immunometabolic hubs that sense diverse environmental cues to modulate systemic iron homeostasis. MAIN BODY: This review examines the molecular architecture underlying macrophage iron metabolism and outlines how iron metabolic processes are dynamically regulated across spatial and temporal scales through the integration of mechanotransductive, mitochondrial, and epigenetic signaling pathways. Across disease contexts,

Effect and Mechanism of Acacetin Against Stress-Induced Gastric Mucosal Damage: Network Pharmacology, Molecular Docking and Experimental Verification.

Stress-induced gastric ulcers (SGU) are categorized as a stress-related mucosal disease (SRMD) that exhibits a high morbidity among ICU patients. However, due to the harmful side effects of proton pump inhibitors (PPIs), there is an urgent need for natural and proactive alternative methods in the treatment of SGU. Acacetin, a natural flavonoid, has demonstrated potential in alleviating gastric ulcers. To elucidate the effects of acacetin in SGU, in this study, gastric ulcers (GU)-related targets were retrieved from GeneCards, DisGeNET, and OMIM. A protein-protein interaction (PPI) network was

Single-Cell Computational Frameworks for Quantifying BET Bromodomain Inhibitor Resistance and Screening Re-Sensitizer Drugs in Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC) is an aggressive subtype characterized by rapid proliferation and a great propensity for metastasis. Therapeutic options for TNBC remain limited due to the absence of targetable hormone receptors. While BET bromodomain inhibitors (BBDIs) exhibit promising anticancer potential, the emergence of drug resistance presents a major challenge. Here, through leveraging single-cell RNA sequencing (scRNA-seq) data across continuous states of BBDI treatment in TNBC, this study conducts an extensive investigation into BBDI resistance, and develops two computational fra

Review Deciphering the Anticancer Efficacy of Bibenzyl Derivatives of Dendrobium Species in Lung Carcinoma.

Metastasis of lung carcinoma is a significant contributor to elevated mortality rates. Globally, it is one of the most fatal carcinomas in terms of mortality and incidence. Despite extensive studies, treatment of lung cancer remains ineffective because of severe side effects and multidrug resistance. Herbal drugs have been documented to augment the effectiveness of conventional therapies, enhance patients' quality of life, and alleviate chemotherapy-related adverse effects. This review describes the therapeutic potential of Dendrobium plants for the management of lung carcinoma. Dendrobium is

Neutrophil Irgm1 ameliorates sepsis-induced myocardial dysfunction by promoting Alox15 degradation.

Sepsis-induced myocardial dysfunction (SIMD), a severe sepsis complication, is characterized by immune dysregulation, with neutrophils playing a central role. While the immunity-related GTPase family M protein (IRGM) in humans and its murine ortholog Irgm1 are key immune regulators, the precise contribution of neutrophil Irgm1 to SIMD pathogenesis remains unclear. This study aims to explore the involvement of neutrophil Irgm1 in SIMD and uncover its mechanisms. This research found that IRGM expression was upregulated in peripheral blood neutrophils from patients with SIMD and inversely correla

Myocardial Ischemia-Reperfusion Injury-Mechanistic Insights and Novel Therapeutics.

Myocardial ischemia-reperfusion (I/R) injury remains a major contributor to infarct expansion and adverse cardiac remodeling despite advances in timely reperfusion therapy. Although restoration of blood flow is essential for myocardial salvage, the abrupt transition from ischemia to reperfusion paradoxically exacerbates cardiomyocyte injury through profound metabolic, ionic, and mitochondrial disturbances. Reperfusion should be viewed not simply as restoration of blood flow, but as a critical biological transition that converts ischemic stress into a self-amplifying injury network. Reperfusion
