Comparative effects of mannanoligosaccharides, fructooligosaccharides, and potato resistant starch on intestinal integrity, antioxidant status, and cecal microbiota in commercial laying hens.

BACKGROUND AND AIM: Functional carbohydrates are increasingly used as alternatives to antibiotic growth promoters because of their ability to modulate intestinal health and microbial populations. However, comparative information regarding the effects of structurally different prebiotic compounds in laying hens remains limited. This study aimed to compare the effects of mannanoligosaccharides (MOS), fructooligosaccharides (FOS), and potato resistant starch (RS) on productive performance, intestinal integrity, antioxidant status, intestinal morphology, and cecal microbiota in commercial laying h

Cobalt modulates methanol turnover of the alcohol dehydrogenase in Desulfofundulus kuznetsovii strain TPOSR.

Desulfofundulus kuznetsovii strain 17T oxidizes methanol via a two-pathway system involving both alcohol dehydrogenases (ADH) and a cobalt-dependent methanol methyltransferase (MT). In contrast, D. kuznetsovii strain TPOSR lacks the MT pathway, relying solely on ADH for growth on methanol. Despite the absence of the MT pathway, cobalt starvation resulted in lower methanol uptake rates and reduced growth rates in strain TPOSR, suggesting a critical role of cobalt in methanol metabolism outside of its role in the MT system. Given the often-crucial role of metal cofactors such as iron, zinc, and

Effect of Substituting Polyether Ionophore Anticoccidial Drugs With 1, 8-Cineole for the Control of Eimeria Infections in Broilers.

BACKGROUND: Coccidiosis is an important parasitic disease of broiler chickens. Drug resistance to the polyether polyether Ionophore anticoccidial drugs  (PACDs) caused by long-term use has emerged as a significant problem in commercial broiler chicken production. OBJECTIVES: This study explored the feasibility of 1,8-cineole (CIN) in replacing PACD with broiler feeding experiments and intestinal microecological experiments. METHODS: This experiment selected 21-day-old Lingnan yellow (LNY) broilers and randomly divided them into 8 groups, with 30 chickens in each group. They were uniformly fe

Detect early TB infection, evaluate contacts for TB (DETEC-TB) - a protocol for a prospective cohort study on active case finding for tuberculosis disease and tuberculosis infection source tracing.

BACKGROUND: Tuberculosis (TB) is a leading cause of death from infectious disease, with 1.25 million deaths in 2023. Among the estimated 10.8 million people who developed TB in 2023, only 75% were diagnosed and reported to national programs and the World Health Organization (WHO). Tanzania has one of the highest burdens of TB in the world, with an estimated 29% of cases unreported. Innovative control strategies, including improved active case finding, are needed to close this gap. This study aims to determine the proportion of persons with TB disease among contacts of adolescents with newly-ac

Comparing noninvasive sampling techniques with standard cannula sampling method for ruminal microbial analysis.

Rumen microbes play an important role in the conversion of indigestible plant material to energy and protein in dairy cows. Sampling for ruminal contents via cannula is considered the gold standard technique for microbial analysis, but the technique requires ruminally cannulated animals and specialized animal facilities. The purpose of this study was to determine whether other sampling methods and locations along the digestive tract may serve as noninvasive proxies to the cannula method for microbial analysis. Six ruminally cannulated lactating Holstein dairy cows were adapted to a standard to

Combinatorial Loss of the Enzymatic Activities of Viral Uracil-DNA Glycosylase and Viral dUTPase Impairs Murine Gammaherpesvirus Pathogenesis and Leads to Increased Recombination-Based Deletion in the Viral Genome.

Misincorporation of uracil or spontaneous cytidine deamination is a common mutagenic insult to DNA. Herpesviruses encode a viral uracil-DNA glycosylase (vUNG) and a viral dUTPase (vDUT), each with enzymatic and nonenzymatic functions. However, the coordinated roles of these enzymatic activities in gammaherpesvirus pathogenesis and viral genomic stability have not been defined. In addition, potential compensation by the host UNG has not been examined in vivo The genetic tractability of the murine gammaherpesvirus 68 (MHV68) system enabled us to delineate the contribution of host and viral facto

11beta-hydroxysteroid dehydrogenase type 1 and obesity.

The metabolic syndrome consists of a constellation of co-associated metabolic abnormalities such as insulin resistance, type 2 diabetes, dyslipidaemia, hypertension and visceral obesity. For many years endocrinologists have noted the striking resemblance between this disease state and that associated with Cushing's syndrome. However, in the metabolic syndrome plasma cortisol levels tend to be normal or lower than in normal individuals. Nevertheless there is strong evidence that glucocorticoid action underlies metabolic disease, largely from rodent obesity models where removing glucocorticoids

Genetic insights into the relationship between anti-inflammatory drug target genes and oral diseases.

BACKGROUND: Glucocorticoids and nonsteroidal anti-inflammatory drugs (NSAIDs) are cornerstones in the management of oral inflammatory pathologies. However, their precise causal effects on a range of oral diseases and the underlying genetic mechanisms remain poorly understood. Concurrently, emerging evidence on the gut-brain axis suggests a potential connection between intestinal inflammation and the pathogenesis of oral diseases. Although preliminary data point to the gut microbiota's role in disease progression, the specific causal pathways and genetic underpinnings remain largely unexplored.

Integrated genetic and functional analyses reveal the pathogenic effects of a novel STXBP3 variant in very-early-onset inflammatory bowel disease with immunodeficiency.

BACKGROUND: Very early onset inflammatory bowel disease (VEOIBD) is frequently driven by monogenic defects. Pathogenic variants in the syntaxin-binding protein 3 (STXBP3) gene have been implicated in VEOIBD; however, the relationship between specific variant types and clinical manifestations remains incompletely understood. This study aimed to identify the genetic cause in a patient with VEOIBD and explore the possible genotype-phenotype correlations of STXBP3-related disorders. METHODS: We performed trio-whole-exome sequencing (trio-WES) on a male infant presenting with VEOIBD, sensorineural

Ozonated Olive Oil Attenuates Hepatic Steatosis in Obese db/db Mice: Evidence for Triacylglycerol-derived Ozonides as Key Bioactive Species.

Ozonated edible oils have garnered increasing attention as potential functional food ingredients for managing metabolic dysfunction-associated steatotic liver disease (MASLD), a condition primarily driven by oxidative stress and chronic inflammation. This study aimed to elucidate (i) which fraction of ozonated olive oil mediates its hepatoprotective effects and (ii) whether the degree of fatty acid unsaturation influences the physiological responses to ozonated lipids. In Experiment 1, olive oil was fractionated into saponifiable (SAP) and unsaponifiable (unSAP) components, which were then sub

Puerarin modulates the gut-liver-ovarian axis to ameliorate reproductive aging in breeder hens.

BACKGROUND: Puerarin (PU), a natural bioactive isoflavone derived from the edible root of Pueraria lobata, exhibits multi-pharmacological activities, including antioxidant, lipid metabolism-regulating, and immunomodulatory properties. The decline in the reproductive performance of hens is primarily attributed to age-related dysbiosis of gut function and reduced function of the liver-ovary axis. However, the systemic mechanisms linking puerarin-induced microbiota changes to improvements in hepatic and ovarian function in aged hens remain poorly defined. In this study, we performed a multi-omics

Residential green space mitigates genetic susceptibility to incident irritable bowel syndrome in a prospective cohort of 376 749 individuals.

BACKGROUND: Environmental exposures may influence irritable bowel syndrome (IBS) pathogenesis, but the interplay between genetic risk, residential green/blue space, and IBS incidence remains unexplored. METHODS: We aimed to study the association between green/blue space and incident IBS, and to assess effect modification by genetic susceptibility. The study included 376 749 UK Biobank participants at baseline. Residential green space was measured using the normalised difference vegetation index (NDVI), and blue space was defined by distance to the nearest water bodies. For time-to-event analy

IFITM3 knockout alleviates neuronal parthanatos by restoring astrocytic glutamate uptake in intracerebral hemorrhage mice.

INTRODUCTION: Intracerebral hemorrhage (ICH) causes severe neurological deficits mainly attributable to secondary brain injury. Parthanatos is a subtype of regulated cell death triggered by glutamate excitotoxicity. Interferon-induced transmembrane protein 3 (IFITM3) is an immune regulatory molecule involved in neuronal death in various neurodegenerative disorders. However, it remains unclear whether and how IFITM3 and parthanatos participate in secondary brain injury after ICH. OBJECTIVES: This study aims to investigate whether IFITM3 aggravates neuronal parthanatos by impairing astrocytic g

Melatonin Improves Sperm Quality in Asthenozoospermic Men by Preserving Acrosome Integrity, Reducing Inflammation, and Regulating Apoptosis-Related Genes and Proteins during Freeze-Thawing.

OBJECTIVE: Sperm freeze-thaw is a crucial technique in assisted reproductive technologies (ART), but it induces oxidative stress in sperm. Melatonin, a powerful antioxidant, reduces oxidative stress by penetrating mitochondria and neutralising free radicals. However, its protective role in the sperm of asthenozoospermic men during freeze-thaw remains poorly understood. This study examines the in vitro effects of melatonin on sperm quality in asthenozoospermic men during the freeze-thaw process. MATERIALS AND METHODS: In this experimental study, semen samples were collected from 30 asthenozoos
