Compared with transplanted tumors, autochthonous tumors are difficult to cure using experimental radiation therapy in mice. Here we analyzed differences in immune-related gene expression profiles between mouse fibrosarcomas subcutaneously induced by 3-methylcholanthrene (3MC) and their corresponding transplanted tumors. The immune genes examined were Pd1, Pdl1, Pdl2, Cd3d, Cd8a, Cd8b, Ifnγ, Itga2, Gzmb, and Foxp3. Among 12 tumors, one was non-transplantable and showed a benign phenotype with an abundance of DX5+ natural killer cells and CD8+ T cells together with increased IFNγ expression and

Pancreatic cancer (PC) is among the most lethal malignancies, characterized by late-stage diagnosis, limited treatment options, and high therapeutic resistance. Growing evidence indicates that long noncoding RNAs (lncRNAs) are key regulators of PC pathogenesis, influencing tumor initiation, progression, and resistance mechanisms. This review comprehensively examines the biogenesis and molecular functions of lncRNAs, with a focus on their regulation through critical oncogenic pathways, including PI3K/Akt, JAK/STAT, and Wnt/β-catenin. The roles of lncRNAs in promoting epithelial-mesenchymal tran

Cyclin E1 (CCNE1), a pivotal member of the cyclin family, governs the G1/S phase transition of the cell cycle by binding to and activating cyclin-dependent kinase 2 (CDK2), thereby initiating DNA replication and driving S-phase entry. In a broad spectrum of human malignancies-including breast, ovarian, gastric, and non-small cell lung cancers-amplification or overexpression of CCNE1 disrupts the orderly regulation of cell cycle progression, leading to uncontrolled proliferation, impaired DNA damage repair, and genomic instability. These oncogenic consequences are intimately associated with inc

OBJECTIVE: Individuals with BRCA1/2 pathogenic variants (PV) are recommended risk-reducing salpingo-oophorectomy between 35 and 45 years of age to prevent ovarian cancer. Trials evaluating the oncologic safety of risk-reducing salpingectomy are ongoing. One's decision on the type and timing of surgery has implications for fertility, quality of life, and long-term health. Published literature on patient preferences has focused on White women from North America and Europe; thus, findings are not necessarily generalizable to patients from other backgrounds. To address this gap, we sought to asses

Background: Glycosylation and inflammation are pivotal in tumor progression, yet the specific glycosyltransferases bridging these processes remain poorly defined. This study investigated Exostosin-1 (EXT1), a key enzyme in heparan sulfate (HS) biosynthesis, as a mechanistic bridge connecting inflammation, stromal remodeling, and immune evasion-driven cancers. Methods: We used a multi-omics approach including Least Absolute Shrinkage and Selection Operator (LASSO) Cox regression on The Cancer Genome Atlas (TCGA) pan-cancer cohorts, transcriptomics, survival, single-cell RNA sequencing (scRNA-se

Emerging evidence highlights the critical role of metabolic pathways in breast cancer (BC) progression. Here, we developed a butyrate metabolism-specific gene (BMRG) signature to predict clinical outcomes and immunotherapy responses in BC, providing a novel pathway-focused prognostic tool. Using data from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO), we identified 102 butyrate metabolism-related differentially expressed genes (DEGs) through the intersection of DEGs, WGCNA-derived key module genes, and BMRGs. Univariate Cox followed by least absolute shrinkage and selection

Psychologically Informed Reminder Messages (PIRMs) may promote health-behavior change, but their impact on preventive genetic screening is unclear. Due to low adherence to a screening program, Clalit Health Services initiated an intervention to encourage BRCA1/2 carrier screening in July 2022. This observational study assesses the effectiveness of the intervention in encouraging screening and its spillover effect on related healthcare utilization, focusing on Ashkenazi Jewish females aged 25-50 with no relevant cancer history. Patients were randomly assigned to receive one of five PIRMs. Four

BACKGROUND: Triple-negative breast cancer (TNBC) is an aggressive subtype with high malignancy and poor prognosis. Immunotherapy is a promising treatment for TNBC patient. Although T cell-mediated tumor killing related genes (TTKRGs) play critical roles in antitumor immunity, their prognostic value and potential function in TNBC is still unclear.
METHODS: Transcriptomic data from TCGA-BRCA and TTKRGs were curated to determine the prognostic genes in TNBC and a prognostic model was further established. GSE135565 dataset was used to validate the prognostic model. Furthermore, the differences be

BACKGROUND: The lack of reliable screening biomarkers for prostate cancer (PC) diagnosis makes tissue biopsy the gold-standard strategy. However, its frequent inconclusive results often lead to repeated procedures, increasing patient burden and healthcare costs. In this context, machine learning (ML) presents a novel approach for identifying gene signatures associated with tumor presence, offering a promising avenue for improved PC detection. Therefore, the present study explored the diagnostic potential of an ML-based gene signature and its applicability in tissue and plasma samples. METHODS:

BACKGROUND: Dysregulation of transcription factors potentiates cancer cell proliferation, stemness, cellular plasticity, metastasis, and therapy resistance and also links with diagnosis/prognosis of the diseases. Thus, investigation of uncharacterized transcription factors is a prime aim for designing novel therapeutics. Large family C2H2 type zinc finger proteins (ZNFs) often bind to nucleic acids and also act as transcription factors. However, unregulated expression of these ZNFs was found to cause diverse pathological conditions including cancer.
METHODS: Cancer database and specific GEO a

BACKGROUND: Hepatocellular carcinoma (HCC) features a complex tumor immune microenvironment (TIME) where natural killer cell exhaustion (NKEX) facilitates immune evasion. Its regulatory networks and prognostic value remain insufficiently understood.
METHODS: We integrated single-cell (GSE149614) and bulk RNA-seq (TCGA-LIHC) data. Following Seurat-based clustering, we utilized CellChat and pySCENIC for communication and transcription factor analysis. NKEX-associated modules were identified via gene set variation analysis and weighted gene co-expression network analysis. A prognostic signature

While Wilms tumors commonly arise from renal precursor cells and maintain features of the developing kidney, recent studies have demonstrated substantial genetic, histologic, and molecular heterogeneity. To further investigate tumor variability as well as unifying features in tumor biology, we performed single-nucleus RNA sequencing (snRNA-seq) on treatment-naive, favorable-histology Wilms tumors utilizing a reference atlas established from tumor-adjacent kidney samples and fetal kidney. Transcriptional profiles of blastemal, stromal, and epithelial components were correlated with tumor histol

PURPOSE: A key clinical challenge in prostate cancer is the identification and validation of biomarkers with high specificity for indolent long-term outcomes. We applied a novel statistical method to identify tumor transcriptomic biomarkers that optimally predicted patients with low metastatic potential.
METHODS: Using tumor whole-transcriptome data from the Health Professionals Follow-up Study (HPFS, discovery set) and Physicians' Health Study (PHS, validation set), we compared patients who died of prostate cancer or developed metastases ("lethal," n = 113) and patients with > 8 years of met

A comprehensive understanding of the underlying molecular mechanisms of prostate cancer is essential for the development of precise diagnostic biomarkers. In this study, we applied the unsupervised multi-omics factor analysis framework (MOFA) to integrate DNA methylation, gene expression, and metabolic profiles derived from the same individuals, aiming to characterize the biological landscape of normal, malignant, and aggressive prostate tissue. Our analysis identified distinct molecular pathways associated with aggressive disease, specifically those involved in zinc metabolism, cell cycle reg

Giant cell tumor of bone (GCTB) is an intermediate bone neoplasm defined by recurrent H3F3A mutations and limited systemic treatment options beyond denosumab. Patient-derived cancer cell lines (PDCs) offer a scalable platform for mechanistic studies and therapeutic discovery, yet the extent to which culture dimensionality alters baseline molecular states and drug response in GCTB remains unclear. Here, we performed integrated transcriptomic, proteomic, and pharmacologic profiling of thirteen patient-derived GCTB cell lines cultured under two-dimensional (2D) monolayer and three-dimensional (3D

Cross-strand chimeric RNAs (cscRNAs) represent an emerging class of noncanonical fusion transcripts arising from bidirectional transcription, yet their landscape and functional significance in gastric cancer remain unexplored. Here, we systematically investigated cscRNA expression patterns, prognostic relevance, and tumor immune microenvironment associations in gastric cancer through integrated computational and experimental approaches. Analysis of 372 TCGA-STAD samples revealed tumor-specific cscRNA enrichment that was independent of clinicopathological variables including tumor stage, histol
