Aconserved salt-stress co-expression network in japonica rice reveals partially dissociated hub gene connectivity and transcriptional induction.

INTRODUCTION: Soil salinity severely compromises rice (Oryza sativa L.) growth and grain yield by disrupting ionic and osmotic homeostasis. Although transcriptome profiling has advanced understanding of salt-stress responses in rice, the conservation of co-expression networks across genetic backgrounds and the relationship between network connectivity and transcriptional induction remain poorly characterized. METHODS: We performed integrated RNA sequencing and weighted gene co-expression network analysis (WGCNA) on three japonica rice varieties under control (0 mM), moderate (85.6 mM), and se

Pentalinonsterol alters immune transcriptomic profiles and modulates the Nrf-2 pathway in Leishmania major infected macrophages.

BACKGROUND: Leishmaniasis is a neglected tropical disease caused by protozoan parasites of the genus Leishmania and remains a significant global health burden due to limited therapeutic options and emerging drug resistance, necessitating the search for alternative therapeutic agents. PURPOSE: Our earlier studies have shown that Pentalinonsterol (Pen), isolated from Pentalinon andrieuxii plant, showed promising antileishmanial activities. However, the effects of Pen on the transcriptomic profile and nuclear factor erythroid 2-related factor 2 (Nrf-2) related alterations in host cells remain un

OsDDM1b Controls Grain Size by Influencing Cell Cycling and Regulating Homeostasis and Signaling of Brassinosteroid in Rice.

Snf2 family proteins are the crucial subunits of chromatin-remodeling complexes (CRCs), which contributes to the biological processes of transcription, replication, and DNA repair using ATP as energy. Some CRC subunits have been confirmed to be the critical regulators in various aspects of plant growth and development and in epigenetic mechanisms such as histone modification, DNA methylation, and histone variants. However, the functions of Snf2 family genes in rice were poorly investigated. In this study, the relative expression profile of 40 members of Snf2 family in rice was studied at certa

The important role of glycerophospholipid metabolism in the protective effects of polyphenol-enriched Tartary buckwheat extract against alcoholic liver disease.

Alcoholic liver disease (ALD) is a mounting public health problem with significant medical, economic and social burdens. Tartary buckwheat (F. tataricum (L.) Gaertn, bitter buckwheat) is a kind of healthy and nutritious food, which has been demonstrated to protect against ALD, but the underlying mechanism has not been fully studied. Herein, we aimed to elucidate the beneficial effects of Tartary buckwheat extract (mainly composed of polyphenols including rutin, quercetin, kaempferol and kaempferol-3-O-rutinoside) in terms of lipid metabolism with the aid of lipidomic analysis. In our study, we

Annotation survey and life cycle transcriptomics of transcription factors in rust fungi (Pucciniales) identify a possible role for cold shock proteins in dormancy exit.

Fungi of the order Pucciniales are obligate plant biotrophs causing rust diseases. They exhibit a complex life cycle with the production of up to five spore types, infection of two unrelated hosts and an overwintering stage. Transcription factors (TFs) are key regulators of gene expression in eukaryote cells. In order to better understand genetic programs expressed during major transitions of the rust life cycle, we surveyed the complement of TFs in fungal genomes with an emphasis on Pucciniales. We found that despite their large gene numbers, rust genomes have a reduced repertoire of TFs comp

Lifestyle of Lactobacillus plantarum in the mouse caecum.

Lactobacillus plantarum is a common inhabitant of mammalian gastrointestinal tracts. Strains of L. plantarum are also marketed as probiotics intended to confer beneficial health effects upon delivery to the human gut. To understand how L. plantarum adapts to its gut habitat, we used whole genome transcriptional profiling to characterize the transcriptome of strain WCFS1 during colonization of the caeca of adult germ-free C57Bl/6 J mice fed a standard low-fat rodent chow diet rich in complex plant polysaccharides or a prototypic Western diet high in simple sugars and fat. Integrated transcriptomic and functional trait responses reveal ecological strategy-associated biomass accumulation and allocation under drought in mulberry seedlings.

Drought alters both plant growth and biomass accumulation and allocation, but how molecular responses and functional traits jointly reflect these changes in woody plants remains insufficiently understood. We investigated whether plant ecological strategies, quantified using the competitive-stress tolerant-ruderal (CSR) framework, could provide an integrative description of drought-associated biomass responses in mulberry (Morus atropurpurea) seedlings. In this study, mulberry seedlings were maintained under well-watered, mild-drought, or moderate-drought conditions for 30 days, after which bio

Brown midrib mutants in sorghum and their applications in renewable energy production.

Lignin is a complicated phenolic polymer that is found in the secondary cell walls of plants. It is necessary for plant mechanical strength, vascular integrity, and stress tolerance. In sorghum (Sorghum bicolor L. Moench), the amount and composition of lignin greatly affect forage digestibility, biomass recalcitrance to bioenergy production and biochemical conversion process efficiency. Brown midrib (bmr) mutants, which exhibit reddish-brown pigmentation of the vasculature, have been a classic and potent tool for identifying lignin biosynthesis and engineering cell wall traits. Over the last 6

Network-Guided Identification of Plant-Derived Modulators of Stress-Adaptive Signalling in Neuroblastoma.

Neuroblastoma is characterized by noticeable resistance to chemotherapy, largely driven by the ability of tumour cells to reorganize stress-adaptive signalling networks rather than relying on single oncogenic drivers. We conducted a study to investigate the pharmacological mode of action of doxorubicin in modifying adaptive signalling pathways in SH-SY5Y neuroblastoma cells, and whether the capacity of plant metabolites can exploit emergent biochemical vulnerabilities. Transcriptomic profiling through RNA sequencing conducted 48 h post-doxorubicin exposure unveiled the organized disruption of

Harnessing invasive weed species for fly ash-polluted site remediation: Comparative assessment of organic contaminant degradation and bacterial shifts using GC-MS and 16S rRNA amplicon sequencing.

Phytoremediation of coal fly ash disposal sites is challenging due to the complex mixtures of persistent pollutants in alkaline, nutrient-depleted substrates. A multi-platform evaluation (physicochemical analysis, BCF/TF, GC-MS Van Krevelen type classification, 16S rRNA amplicon sequencing and functional networks) was conducted to assess two natural colonizers, Cannabis sativa L. and Parthenium hysterophorus L., at the NTPC ash disposal site. Weed colonization reduced pH from 9 (NS) to 7.49 (CS) and 7.08 (PS). The large surface depletion of zinc, iron, and manganese caused by these reductions

Positive Regulation of Soybean Seed Vigor and Quality by the Zinc Finger Transcription Factor GmPHD3 Under High-Temperature and High-Humidity Stress.

Field high-temperature and high-humidity (HTH) stress causes soybean seed deterioration, including shrinkage, moldiness, browning, and reduced germination rate, thereby decreasing seed vigor, emergence performance, and commercial value. Clarifying the molecular mechanisms of seed vigor formation under HTH stress is critical for identifying key genes and improving spring soybean seed quality. Plant homeodomain (PHD) proteins are conserved zinc-finger transcription factors involved in chromatin remodeling and stress responses. GmPHD3, a soybean PHD family member, has only been studied via hetero

The role of nitric oxide and nitrogen in mediating copper stress in Brassica juncea L.

Copper (Cu) holds a significant importance in plant metabolism as it serves as an essential micronutrient but becomes toxic at higher concentrations. Nitric oxide (NO), a key signaling molecule, and nitrogen (N) play essential roles in combating toxicity of some metals. This study explores the potential of interactive effects of NO as 100 µM SNP (sodium nitroprusside, NO source) and N (80 mg N kg-1 soil) in mitigating Cu (100 mg Cu kg-1 soil) stress in mustard (Brassica juncea L.) plants. The impaired physio-biochemical changes, photosynthetic efficiency, and the expression level of genes asso

Analysis of plant cell death-inducing proteins of the necrotrophic fungal pathogens Botrytis squamosa and Botrytis elliptica.

Fungal plant pathogens secrete proteins that manipulate the host in order to facilitate colonization. Necrotrophs have evolved specialized proteins that actively induce plant cell death by co-opting the programmed cell death machinery of the host. Besides the broad host range pathogen Botrytis cinerea, most other species within the genus Botrytis are restricted to a single host species or a group of closely related hosts. Here, we focused on Botrytis squamosa and B. elliptica, host specific pathogens of onion (Allium cepa) and lily (Lilium spp.), respectively. Despite their occurrence on diffe

At a supra-physiological concentration, human sexual hormones act as quorum-sensing inhibitors.

N-acylhomoserine lactone (AHL)-mediated quorum-sensing (QS) regulates virulence functions in plant and animal pathogens such as Agrobacterium tumefaciens and Pseudomonas aeruginosa. A chemolibrary of more than 3500 compounds was screened using two bacterial AHL-biosensors to identify QS-inhibitors (QSIs). The purity and structure of 15 QSIs selected through this screening were verified using HPLC MS/MS tools and their activity tested on the A. tumefaciens and P. aeruginosa bacterial models. The IC50 value of the identified QSIs ranged from 2.5 to 90 µg/ml, values that are in the same range as

In-vitro study of the effect of Centella asiatica on cholera toxin production and the gene expression level of ctxA gene in Vibrio cholerae isolates.

ETHNOPHARMACOLOGICAL RELEVANCE: Centella asiatica (L.) Urb or Indian pennywort is a plant of ethnopharmacological relevance, commonly called as Brahmi in South India known for its antimicrobial property in gut and for the treatment of other gut ailments. Natural anti-virulence drugs that disarm pathogens by directly targeting virulence factors or the cell viability and are thus preferred over antibiotics as these drugs impose limited selection pressure for resistance development. In this regard, an in-vitro experimental study was conducted to know the effect of extract of Centella asiatica(L.)
