Human papillomaviruses (HPVs) infect specific human epithelial tissues. Because viral propagation in cultured cells has not been achieved, studies of HPV genetic activities have been difficult and rely largely on analyses of patient specimens by conventional biochemical methods. HPV type 6 and type 11 infections often result in genital warts (condylomata acuminata). Structural mapping of RNAs from such warts reveals that they use alternative promoters, splice sites, and polyadenylation sites to produce complex families of overlapping mRNAs that span multiple open reading frames. Based on the m

Many parasites are transmitted through trophic interactions, yet the specific pathways connecting intermediate and definitive hosts often remain poorly understood in marine food webs. In the Southwestern Atlantic, the Argentine anchovy (Engraulis anchoita) has been considered a potential vector of larval Anisakis to higher trophic levels, but low infection levels in San Matías Gulf suggest alternative transmission routes in this gulf. We investigated the role of pearlsides (Maurolicus spp.), abundant mesopelagic fishes in the gulf, in the transmission of anisakid nematodes. Examination of 229

Thymidine analogues are widely used as antiviral and anticancer agents; however, improper dosage and prolonged exposure can lead to severe hematological and genetic complications, necessitating reliable methods for their detection and discrimination. We report an array-based fluorescence sensing platform for the discrimination of structurally related thymidine analogues in aqueous media using a Pd-modified ZnO-polymer nanocomposite as a single cross-reactive probe. The nanocomposite was synthesized by functionalizing ZnO nanoparticles with a KG3 polymer matrix followed by palladium ion modific

The global expansion of dengue virus (DENV) poses a major public health threat, yet its circulation dynamics remain poorly understood. By integrating global genetic, epidemiological, and mobility data within a phylodynamic framework, we show that DENV circulation follows a source-sink model, with tropical endemic regions acting as persistent viral reservoirs ('sources') that seed epidemics in temperate areas ('sinks'). Most areas at the fringe of endemic transmission exhibit sink-like characteristics with exception of South China. We identify a low-intensity dispersal route from South China to

Leishmaniasis is a complex disease caused by protozoan parasites of the genus Leishmania, which are transmitted by phlebotomine sand flies. The clinical manifestations of leishmaniasis are diverse, ranging from self-healing cutaneous lesions to fatal systemic disease. Mouse models are instrumental in advancing our understanding of the immune system against infections, yet their limitations in translating findings to humans are increasingly highlighted. The success rate of translating data from mice to humans remains low, largely due to the complexity of diseases and the numerous factors that i

Severe Fever with Thrombocytopenia Syndrome Virus (SFTSV), a tick-borne phlebovirus first identified in China, causes severe illness characterized by high fever, thrombocytopenia, leukopenia, and, in some cases, multi-organ failure and death. With mortality rates ranging from 5% to 30% in endemic regions, SFTSV has emerged as a significant public health threat across East Asia, including South Korea and Japan, with potential for broader outbreaks. This review synthesizes recent advances in SFTSV animal models and candidate vaccines, highlighting their contributions and limitations. Current ani

Pompe disease is an autosomal recessive genetic disease caused by a mutation in the acid α-glucosidase (GAA) gene, which results in dysfunction of the GAA and a buildup of glycogen in lysosomes. Recently, new research has found additional causes of the disease pathology, such as the role of autophagy. Currently, the standard of care is enzyme replacement therapy (ERT). ERT has been proven to increase survival in Pompe disease as well as improve pulmonary and motor tests compared to no treatment. However, some patients do not see improvement from these treatments, and the life expectancy and qu

The development of effective poultry vaccines is crucial for maintaining flock health and productivity. In this study, we developed and evaluated a recombinant chimpanzee adenovirus vaccine (PAD-S1-HN) that simultaneously expresses the infectious bronchitis virus (IBV) spike subunit protein S1 and Newcastle disease virus (NDV) hemagglutinin-neuraminidase HN protein. The recombinant virus was successfully rescued in HEK293 cells, and transmission electron microscopy confirmed its typical adenoviral morphology. The expression of the IBV S1 and NDV HN proteins was validated by indirect immunofluo

Dengue and leptospirosis are prevalent diseases in tropical and subtropical regions, posing significant public health challenges. These illnesses exhibit overlapping symptoms, including fever, muscle pain, and headaches, which complicates diagnosis and delays appropriate treatment. This study explores the use of attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR) combined with multivariate analysis to distinguish between the two infections by analyzing blood plasma in both liquid and dry states. A total of 114 patient samples at varying infection stages (n = 43 for

While substantial amounts of antibiotics and pesticides are applied to maintain orchard yields, their influence on the dissemination and risk of antibiotic resisitome in the orchard food chain remains poorly understood. In this study, we characterized the bacterial and fungal communities and differentiated both antibiotic resistance genes (ARGs) and virulence factor genes (VFGs) in the soil, Chinese bayberry (matured and fallen), and fruit fly gut, collected from five geographic locations. Our results showed that fruit fly guts and soils exhibit a higher abundance of ARGs and VFGs compared wit

Conventional cancer therapies have many limitations. In the last decade, it has been suggested that bacteria-mediated immunotherapy may circumvent the restrictions of traditional treatments. For example, Salmonella enterica is the most promising bacteria for treating cancer due to its intrinsic abilities, such as killing tumor cells, targeting, penetrating, and proliferating into the tumor. S. enterica has been genetically modified to ensure safety and increase its intrinsic antitumor efficacy. This bacterium has been used as a vector for delivering anticancer agents and as a combination thera

Catecholaminergic polymorphic ventricular tachycardia (CPVT) is an inherited channelopathy. In this review, we summarize the epidemiology, pathophysiology, clinical characteristics, diagnostics, genetic mutations, standard treatment, and the emergence of potential gene therapy. This inherited cardiac arrhythmia presents in a bimodal distribution with no association between sex or ethnicity. Six different CPVT genes have been identified, however, most of the cases are related to a heterozygous, gain-of-function mutation on the ryanodine receptor-2 gene (RyR2) and calsequestrin-2 gene (CASQ2) th

Antimicrobial resistance (AMR) is predicted to cause a worldwide annual toll of 10 million deaths by 2050. This looming public health threat has been linked to antibiotic overuse and pollution, which places selective pressures on AMR maintenance and transfer in and between microbial populations. We examined the distribution, diversity and potential mobility of AMR genes in cyanobacteria. While cyanobacteria are not pathogenic, we hypothesised that they could be a major environmental reservoir for AMR genes. Genes encoding AMR to seven antimicrobial drug classes were found in 10% of cyanobacter

Male and female Plasmodium falciparum gametocytes are the parasite lifecycle stage responsible for transmission of malaria from the human host to the mosquito vector. Not only are gametocytes able to survive in radically different host environments, but they are also precursors for male and female gametes that reproduce sexually soon after ingestion by the mosquito. Here, we investigate the sex-specific lipid metabolism of gametocytes within their host red blood cell. Comparison of the male and female lipidome identifies cholesteryl esters and dihydrosphingomyelin enrichment in female gametocy

Novel approaches for malaria prophylaxis remain important. Synthetic DNA-encoded monoclonal antibodies (DMAbs) are a promising approach to generate rapid, direct in vivo host-generated mAbs with potential benefits in production simplicity and distribution coupled with genetic engineering. Here, we explore this approach in a malaria challenge model. We engineered germline-reverted DMAbs based on human mAb clones CIS43, 317, and L9 which target a junctional epitope, major repeat, and minor repeat of the Plasmodium falciparum circumsporozoite protein (CSP) respectively. DMAb variants were encoded

In the current scenario of the coronavirus pandemic caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), considerable efforts have been made to control the pandemic by the development of a strong immune system through massive vaccination. Just after the discovery of the genetic sequences of SARS-CoV-2, the development of vaccines became the prime focus of scientists around the globe. About 200 SARS-CoV-2 candidate vaccines have already been entered into preclinical and clinical trials. Various traditional and novel approaches are being utilized as a broad range of platforms.

Down's syndrome results from trisomy of chromosome 21, a genetic change which also confers a probable 100% risk for the development of Alzheimer's disease neuropathology (amyloid plaque and neurofibrillary tangle formation) in later life. We aimed to assess the effectiveness of diffusion-weighted imaging and connectomic modelling for predicting brain amyloid plaque burden, baseline cognition and longitudinal cognitive change using support vector regression. Ninety-five participants with Down's syndrome successfully completed a full Pittsburgh Compound B (PiB) PET-MR protocol and memory assessm

Adolescence constitutes a period of vulnerability in the emergence of fear-related disorders (FRD), as a massive reorganization occurs in the amygdala-prefrontal cortex network, critical to regulate fear behavior. Genetic and environmental factors during development may predispose to the emergence of FRD at the adult age, but the underlying mechanisms are poorly understood. In the present study, we tested whether a partial knock-down of tuberous sclerosis complex 2 (Tsc2, Tuberin), a risk gene for neurodevelopmental disorders, in the basolateral amygdala (BLA) from adolescence could alter fear

Tetracycline-inducible systems are widely used control elements for mammalian gene expression. Despite multiple iterations to improve inducibility, their use is still compromised by basal promoter activity in the absence of tetracyclines. In a mammalian system, we previously showed that the introduction of the G72V mutation in the rtTA-M2 tetracycline activator lowers the basal level expression and increases the fold-induction of multiple genetic elements in a long chimeric antigen receptor construct. In this study, we confirmed that the G72V mutation was effective in minimising background exp
