Mothers' explanations of communicating past health and future risks to survivors of childhood cancer.

Mothers of survivors of Retinoblastoma (Rb) experience unique challenges communicating with their child about the condition. Children are mostly diagnosed within their first year but the consequences continue into young adult life. Here 39 mothers of Rb survivors (23 males, mean age = 10.26 years) were interviewed about their experiences. Mothers were asked about communication with their children about Rb, and future health risks. Interviews were analysed using thematic analysis. Mothers reported that they had informed children about past diagnosis and treatment but had spoken less about genet

Neuroprotection by GDNF-secreting stem cells in a Huntington's disease model: optical neuroimage tracking of brain-grafted cells.

The use of stem cells for reconstructive or neuroprotective strategies can benefit from new advances in neuroimaging techniques to track grafted cells. In the present work, we analyze the potential of a neural stem cell (NSC) line, which stably expresses the glial cell line-derived neurotrophic factor (GDNF) and the firefly luciferase gene (GDNF/Luc-NSC), for cell therapy in a Huntington's disease mouse model. Our results show that detection of light photons is an effective method to quantify the proliferation rate and to characterize the migration pathways of transplanted NSCs. Intravenous ad

Association of a duplicated repeat polymorphism in the 5'-untranslated region of the DRD4 gene with novelty seeking.

Novelty Seeking (NS) is a human personality trait in which impulsive, exploratory, and thrill-seeking behaviors are displayed. Dopaminergic genes have been prime candidates in the search for the genetic factors underlying NS because of the central role that dopamine plays in the brain's reward system. We have investigated whether there is an association between a polymorphic 120 base pairs (bp) repeat that is located in the 5'-untranslated region of the dopamine D4 receptor gene (DRD4) and NS. We genotyped four separate groups from psychiatric clinical studies for the repeat polymorphism. Ther

Genetically engineered bacterial magnetosomes as optimized tracers for magnetic particle imaging.

Medical imaging relies on tracer materials to enable accurate visualization and diagnosis of diseases. Magnetic Particle Imaging (MPI) is an innovative tomographic modality that offers exceptional sensitivity and temporal resolution. These characteristics make MPI particularly promising for clinical applications such as real-time vascular and perfusion imaging, tumor detection, and intraoperative guidance. However, MPI performance has so far been limited by the quality of available tracers, as conventional chemical synthesis provides only restricted control over the size, shape, and magnetic p

CHRNA5-A3-B4, CYP2A6, and DBH Genetic Associations With Smoking Cessation Throughout Adulthood Within Two Longitudinal Studies of Women.

INTRODUCTION: Genetic studies of smoking cessation have been limited by short-term follow-up or cross-sectional design. Within seven genes (CHRNA3, CHRNA5, CHRNB2, CHRNB4, DRD2, DBH, and CYP2A6) influencing biological mechanisms relevant to smoking, this study aimed to identify single nucleotide polymorphisms (SNPs) associated with smoking cessation throughout up to 38 years of follow-up. AIMS AND METHODS: Participants were from two all-female cohort studies, Nurses' Health Study (NHS) (n = 10 017) and NHS-2 (n = 2793). For 132 SNPs providing coverage of these genes, genotype associations wit

Genome-wide association studies of calf resilience traits derived from milk intake variability in Holstein calves.

Understanding and genetically improving dairy calf resilience is critical for enhancing health, welfare, and productivity of dairy animals under increasing environmental and disease pressures. In this study, we conducted genome-wide association studies (GWAS) to identify genetic variants and genomic regions associated with calf resilience traits derived from automated milk feeder (AMF) records in American Holstein calves. Data from 14,749 calves, including milk intake, milk drinking behavior, and calf health records, were available for the analyses. The pedigree records included 21,630 animals

Solar cycles and their relationship to human disease and adaptability.

In this paper, we show that 11-year solar cycle peaks predispose humans to disease, but also endow creativity and adaptability. We give several examples of diseases that are modulated by light and present evidence for an effect of intensity and variation in sunlight, primarily ultraviolet radiation (UVR), on the human genome. The birth dates of nearly 237,000 unique clients in the Maine Medicaid database collected from 1995 to 2004, inclusive, were related to solar cycle irradiance for the past seventy-one years, encompassing seven solar cycles. The sample was divided into four general categor

Sustained Relief of Chronic Pain via a Nav1.7-Targeting ASO-siRNA Conjugate in Rats.

Chronic pain affects a large global population, yet safe, long-acting analgesics remain limited. Nav1.7 is a genetically validated pain target, but conventional small-molecule approaches have had limited success. Therapeutic oligonucleotides-antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs)-offer selective and durable gene silencing. Here, we developed N02C0702, an ASO-siRNA conjugate (ASC) that achieves efficient Nav1.7 knockdown and demonstrates greater activity than the individual ASO (N02A114) and siRNA (N02S154) components at both mRNA and protein levels, without requi

A microglial SPP1-ITGB1 feedback loop sustains thalamic hyperexcitability and post-stroke pain.

Central post-stroke pain (CPSP) represents a failure of homeostatic recovery, yet how acute injury transitions to chronic pain remains elusive. While glial reactivity is a hallmark of stroke, the effectors translating this activation into sustained neuronal hyperexcitability are undefined. Here, we identify microglia-derived secreted phosphoprotein 1 (SPP1) as a key chronification switch for CPSP-like behaviors in mice. Hemorrhage-induced ATP release activates P2X7 receptors, triggering robust SPP1 expression in microglia. This engages a self-amplifying, autocrine integrin β1 (ITGB1) signaling

A molecular and spinal circuit basis for the functional segregation of itch and pain.

The dorsal horn is remarkably diverse, yet how this cellular complexity enables discrimination of sensory modalities remains a fundamental question. Neurons expressing the gastrin-releasing peptide receptor (Grpr+) have long been considered dedicated to itch, yet broad activation also evokes pain-related behavior. Whether Grpr+ neurons are required for pain and how itch- and pain-related functions are organized within this population remain unresolved. Here, we show that Grpr+ neurons comprise functionally distinct subpopulations defined by tachykinin-1 (Tac1) and correspond to species-conserv

O-linked glycan-dependent gating of TPC2 controls lysosomal excitability and organelle remodeling.

Two-pore channel 2 (TPC2) is a lysosomal cation channel involved in calcium and sodium signaling and membrane remodeling. Its dysfunction has been linked to diseases including viral infections, neurodegenerative disorders, and cancer. Here, we identify mucin-type O-linked glycosylation at two conserved luminal residues of TPC2, Ser612 and Ser613, as a structural gating brake. Combining structure-guided mutagenesis, lysosomal patch-clamp recordings, calcium nanodomain imaging, molecular dynamics simulations, and small-molecule modulation, we show that genetic, enzymatic, or pharmacological glyc

Do animal models represent the diversity of clinical Parkinson's disease?

Parkinson's disease (PD) is a multifactorial and progressive neurodegenerative disorder characterized by a complex symptomatology, including a broad spectrum of motor and non-motor symptoms. Disease modeling in animals represents a keystone part of the research aimed at understanding molecular bases driving PD onset, and specific mechanisms shaping pathological trajectories along disease progression, sustaining the inter-individual variability in PD manifestation. Multiple experimental approaches using toxins, viral vectors, and genetic perturbations have been developed to reproduce key PD fea

Transcriptomic landscape of microglia in mouse models of social dysfunction and oxytocin-mediated recovery.

Atypical sociability is a hallmark of neurodevelopmental disorders arising from genetic susceptibility and prenatal environmental perturbations that can affect diverse brain cell types. Using single-cell transcriptomics, we previously identified selective vulnerability of parvocellular oxytocin (OT) neurons in the paraventricular hypothalamus (PVH) following embryonic exposure to valproic acid (VPA), a teratogen that induces social deficits. Neonatal chemogenetic activation of OT neurons rescued these behavioral abnormalities and partially restored dysregulated gene expression. However, the ef

Epidemiology of aggression and violence in mental disorders and the approach of TRR 379.

Aggression in mental disorders (AMD) poses a challenge for everyday clinical practice and has far-reaching societal implications. The biological foundations of the different manifestations, developmental trajectories, and transdiagnostic and disorder-specific deficits remain largely unknown. One reason for the apparent lack of understanding of the complex mechanisms underlying AMD is the heterogeneity of both aggression and mental disorders themselves. This, in turn, hinders progress in developing effective prevention and intervention strategies. Therefore, a collaborative approach between cli

Induction and regulation of a reversible form of suspended animation in C. elegans.

Suspended animation, a state of extreme quiescence with microscopically invisible movement and development, is a remarkable yet poorly understood stress resilience strategy in animals. Here, we describe a newly discovered form of suspended animation inducible by high-population density in isosmotic liquids in C. elegans throughout larval development and adulthood. Transcriptomic, metabolomic and live-cell activity reporter imaging analyses reveal striking molecular and cellular landscape changes by such liquid-induced suspended animation (LISA), including remodeling of gene expression programs

Let there be multifunctionality: Uncovering the dynamical zoo of the AC-DC genetic circuit.

Gene regulatory networks (GRNs) govern many cell processes. While multistability and oscillations are common GRN dynamics, they are typically studied and engineered in isolation. Here, we challenge this separation using the AC-DC circuit, a minimal three-gene network that merges the classical toggle switch and repressilator. Using a thermodynamic formalism and Bayesian inference, we show that a single-inducer version of the circuit displays diverse multifunctional dynamics, including coexistence of oscillations and multistability. We explore robustness, classify emergent behaviors, and analyze

Myosin II-independent contraction of actin filaments in membrane nanotubes.

The ability to generate active stresses within filamentous actin matrices is a fundamental and evolutionarily conserved process driving locomotion and morphogenetic changes in cells. The generation of pushing forces by actin polymerization is reasonably well understood, and is known to drive lamellipodia based motility and filopodial extension. Actin filaments decorated with myosin motors can also generate contractile stresses as in the cell cortex or in cytokinetic rings. In this article we use membrane nanotubes pulled out of axonal shaft to investigate actin dynamics and force generation. W

HSF1 controls transcriptional programs that establish thalamostriatal shaft synaptic architecture and preserve cognitive flexibility.

Cognitive flexibility (CF) declines during aging and is further impaired in neurodegenerative diseases such as Huntington's disease (HD), yet the molecular mechanisms underlying these deficits remain poorly understood. Thalamostriatal (T-S) synapses are critical for CF, and we previously identified Heat Shock Factor 1 (HSF1) as a regulator of T-S density in HD. However, how HSF1 regulates T-S synapses and whether it modulates cognitive flexibility (CF) remained unclear. Here, we combined HSF1 ChIP-seq, transcriptomics, synapto-proteomics, targeted genetic manipulations and behavioral analyses
