OBJECTIVES: In order to clarify the transmission cycle and genetic identity of Borrelia spirochetes in the non-endemic country of Taiwan, the causative agents responsible for human borreliosis were isolated from skin biopsies of patients and their genetic identities were determined.
METHODS: Serum samples and skin biopsy specimens were collected from 95 patients: 85 with suspected Lyme disease and 10 controls. Infection with Borrelia burgdorferi was verified by Western immunoblot analysis and isolation of the Borrelia spirochetes from skin biopsy specimens. The genetic identities of these iso

Hepatocellular carcinoma (HCC) is associated with hepatitis B virus (HBV) as an etiologic agent in 80% of cases, and is the major cause of death among HBV carriers. Family history of HCC is a known risk factor for the development of HCC among chronically HBV infected patients; therefore, genetic factors are likely to modify the risk of HCC. However, the genetic factors that determine progression to HCC remain mostly to be recovered. It is estimated that there are millions of single nucleotide polymorphisms (SNPs) within human genome and they are likely to explain much of the genetic diversity

It is generally accepted that human cancers derive from a mutated single cell. However, the genetic steps characterizing various stages of progression remain unclear. Studying a unique case of metastatic melanoma, we observed that cell lines derived from metachronous metastases arising over a decade retained a central core of genetic stability in spite of divergent phenotypes. In the present study, we expanded our previous observations comparing these autologous cell lines of clonal derivation with allogeneic ones and correlated array comparative genomic hybridization (aCGH) with gene expressi

INTRODUCTION: Inflammatory bowel disease (IBD) comprising ulcerative colitis (UC) and Crohn's disease (CD) is multigenic disorder. Tremendous progress has been achieved in unravelling the genetic background of IBD. It has led to the discovery of mutations in NOD2 associated with ileal CD and numerous other genes have been found to be associated with IBD susceptibility.
METHODS: A review of the literature on the genetic background of IBD was performed.
RESULTS: It is only partially understood how mutations in NOD2 lead to CD. Mouse models, in vitro data and studies in humans offer conflicting

The search for genes in bipolar disorder has provided numerous genetic loci that have been linked to susceptibility to developing the disorder. However, because of the genetic heterogeneity inherent in bipolar disorder, additional strategies may need to be employed to fully dissect the genetic underpinnings. One such strategy involves reducing complex behaviors into their component parts (endophenotypes). Abnormal neurophysiological, biochemical, endocrinological, neuroanatomical, cognitive, and neuropsychological findings are characteristics that often accompany psychiatric illness. It is pos

Bardet-Biedl syndrome (BBS) is a genetically heterogeneous recessive disease characterized primarily by atypical retinitis pigmentosa, obesity, polydactyly, hypogenitalism, and mental retardation. Despite the presence of at least five loci in the human genome, on chromosomes 2q, 3p, 11q, 15q and 16q, as many as 50% of the mutations appear to map to the BBS1 locus on 11q13. The recessive mode of inheritance and the genetic heterogeneity of the syndrome, as well as the inability to distinguish between different genetic loci by phenotypic analyses, have hindered efforts to delineate the 11q13 reg

OBJECTIVES: The study attempts to determine the genetic origin of hydatidiform mole and non-molar abortion using the polymerase chain reaction (PCR) method, and reevaluates this method as a possible diagnostic tool for hydatidiform mole.
MATERIAL AND METHODS: A total of 71 cases which consists of 21 complete hydatidiform moles, three partial hydatidiform moles, and 47 non-molar abortions were investigated. The genetic origin was analyzed by the PCR method applied to the variable number of tandem repeat regions of the Apolipoprotein B gene, the human type-II collagen gene, and the probe YNZ22.

The vitamin B12-binding protein, transcobalamin II, is a trace component of plasma with a rapid turnover. This protein is essential for absorption, transport, cellular uptake and for recycling of vitamin B12 (cobalamin). Congenital transcobalamin II deficiency, an inborn error of metabolism is inherited as a recessive trait. The homozygous form of the deficiency is accompanied by severe clinical, hematological and immunological disturbances in the first months of life. Analytical, genetic, biochemical and clinical aspects of transcobalamin II in man and in vertebrates have been reviewed here.

The placenta is essential for fetal growth and the maintenance of pregnancy. Primary human cytotrophoblasts (CTBs) provide a physiologically relevant model of this transient organ, yet their use has been limited by the difficulty of genetic manipulation. Recombinant adeno-associated virus (AAV) vectors, widely used to modulate gene expression in other systems, offer low immunogenicity, low cytotoxicity, and high efficiency in primary cells. Here, we provide proof of concept of an AAV-mediated gene delivery approach for primary CTBs. This platform enables in-depth, cell-biological studies to el

Although Q fever is an important zoonotic infection with a worldwide distribution, no human isolates of Coxiella burnetii have been identified in Korea. For the first time, we identified the nucleotide sequence of C. burnetii from a 32-year-old man with an acute febrile illness in Korea. Diagnosis of acute Q fever was confirmed by seroconversion using indirect immunofluorescence antibody assays. Phylogenetic analysis demonstrated high sequence similarity (99.6%-100%) with C. burnetii 16S rRNA sequences identified from the reservoir. These results are the first genetic analysis of C. burnetii i

Bioengineered skin has great potential for use in regenerative medicine for treatment of severe wounds such as burns or chronic ulcers. Genetically modified skin substitutes have also been used as cell-based devices or "live bioreactors" to deliver therapeutics locally or systemically. Finally, these tissue constructs are used as realistic models of human skin for toxicological testing, to speed drug development and replace traditional animal-based tests in a variety of industries. Here we describe a method of generating bioengineered skin based on a natural scaffold, namely, decellularized hu

Recent work in mice has suggested that an enriched environment can delay disease onset in models of Huntington's disease (HD). The component of an enriched environment most likely able to be translated to humans is increased exercise. But here I note that a recently reported case of HD in a marathon runner who had, for many years before becoming symptomatic with HD, been running at an advanced amateur/semiprofessional level, suggests that in humans exercise is not able to prevent or even delay onset of HD. In general study of marathon runners may be useful to study if exercise can prevent or d

The epidemiology of cryptosporidiosis, an infection caused by several genotypically and phenotypically diverse Cryptosporidium species, has been dynamically changing over the past decade from that of a rare, largely asymptomatic infection to an acute enteric disease of animals and humans. In this review, the current understanding of factors (biology and epidemiology) contributing to the emergence of cryptosporidiosis in animals, including parasite biology, genetic diversity, environmental spread, livestock production trends, presence of the parasite in livestock and companion animals, and pote

A case of the rare genetic trisomy 22 in a live calf is described. The calf had low blood thyroxine level and low growth rate. It had several defects including brachygnathia superior, strabismus convergence, aortal cusp insufficiency and hypertrophy of clitoris. Chromosome analysis was performed on cultured blood lymphocytes and fibroblast cells. In all counted metaphases 61 chromosomes were present. The extra chromosome was identified as a chromosome 22 by R-banding. The defects of the calf have similarities with cases of partial trisomy 3p25-pter in human. This section of the human chromosom

The postcranial skeleton of a Japanese macaque that had been trained for bipedalism over an 11-year period was studied. Considerable modifications in the hindlimb bones caused by bipedal postural and locomotor behaviour were observed. Changes occurred in joint morphology, articular dimensions and shape-dependent strength of long bones, reflecting the causal relationship between function and morphology. However, the conditions under which the modifications are developed are somewhat different from those in humans, as the monkey's bipedalism is distinct from that of humans. The modifications see

Samples of human osteogenesis imperfecta (OI) bone were analyzed for osteonectin content by SDS gel electrophoresis and immunodetection on Western blots. The OI bone osteonectin content varied from normal to severely depressed. Previously, we showed that two clinically identical but genetically unrelated bovine models of OI were differentiated biochemically by their bone osteonectin content: one OI model had normal bone osteonectin while the other was severely depressed in this parameter. The data in this pilot study suggest that further investigation of bone osteonectin content may prove usef

Advances in long-read sequencing (LRS) and assembly algorithms have made it possible to create highly complete genome assemblies for humans, animals and plants. However, ongoing development is needed to improve accessibility, affordability, and assembly quality and completeness. 'Cornetto' is a new strategy in which we use programmable selective nanopore sequencing to focus LRS data production onto the unsolved regions of a nascent assembly. This improves assembly quality and streamlines the process, both for humans and non-human vertebrates. Cornetto enables us to generate highly complete dip

Gene editing in non-human primates may lead to valuable models for exploring the etiologies and therapeutic strategies of genetically based neurological disorders in humans. However, a monkey model of neurological disorders that closely mimics pathological and behavioral deficits in humans has not yet been successfully generated. Microcephalin 1 (MCPH1) is implicated in the evolution of the human brain, and MCPH1 mutation causes microcephaly accompanied by mental retardation. Here we generated a cynomolgus monkey (Macaca fascicularis) carrying biallelic MCPH1 mutations using transcription acti

Angiotensin II (Ang II) plays a key role in hypertension, renal and cardiovascular pathophysiology via intracellular pathways that involve the activation of a multiplicity of signaling mechanisms. Although experimental and genetic animal models have been developed and used to explore Ang II signaling's role in hypertension, a complete understanding of the processes mediating Ang II signaling in hypertension in humans remains elusive. One impediment is that these animal models do not exhibit all the traits of human hypertension, making it impossible to extrapolate from them to humans. To overco

The use of gene-engineered T cells expressing chimeric single-chain (scFv) receptors capable of codelivering CD28 costimulation and T cell receptor zeta chain (TCR-zeta) activation signals has emerged as a promising treatment regimen for cancer. Using retroviral transduction, primary human T lymphocytes were gene-engineered to express the scFv-CD28-zeta chimeric receptor reactive with the ErbB2 tumor-associated antigen. We demonstrated the ability of these gene-engineered human T cells to produce high levels of cytokines, proliferate vigorously, and mediate lysis of ErbB2(+) tumors in an antig
