Incidence of myiasis among humans-a review.

Myiasis, the infestation of tissues of live humans and other vertebrate animals with dipteran larvae (maggots), is a phenomenon of widespread occurrence throughout the tropical regions of the world. It is commonly seen in domestic and wild animals but occurs rarely in man also. The tissue invasion in man by maggots is generally a well-recognized complication of neglected wounds. The condition may be asymptomatic but occasionally results in more or less severe problems and even death when larvae invade body cavities or areas that forbid their direct visual examination. Many cases of myiasis, ho

Mesenchymal stem cells retain their defining stem cell characteristics after exposure to ionizing radiation.

PURPOSE: Mesenchymal stem cells (MSCs) have the ability to migrate to lesion sites and undergo differentiation into functional tissues. Although this function may be important for tissue regeneration after radiation therapy, the influence of ionizing radiation (IR) on cellular survival and the functional aspects of differentiation and stem cell characteristics of MSCs have remained largely unknown. METHODS AND MATERIALS: Radiation sensitivity of human primary MSCs from healthy volunteers and primary human fibroblast cells was examined, and cellular morphology, cell cycle effects, apoptosis, a

A comprehensive analysis of normal variation and disease-causing mutations in the human DSPP gene.

Within nine dentin dysplasia (DD) (type II) and dentinogenesis imperfecta (type II and III) patient/families, seven have 1 of 4 net -1 deletions within the approximately 2-kb coding repeat domain of the DSPP gene while the remaining two patients have splice-site mutations. All frameshift mutations are predicted to change the highly soluble DSPP protein into proteins with long hydrophobic amino acid repeats that could interfere with processing of normal DSPP and/or other secreted matrix proteins. We propose that all previously reported missense, nonsense, and splice-site DSPP mutations (all ass

Neural mediation of the motilin motor effect on the human antrum.

To elucidate the mode of action of motilin on the stimulation of human gastrointestinal motility, we studied the effect of exogenous motilin during muscarinic or serotoninergic pharmacological blockade. Manometric recording of the interdigestive antroduodenal motility was carried out in 27 healthy volunteers until the appearance of a spontaneous antral phase III. The tested blocker was then administered intravenously and was followed 30 min later by a 10-min infusion of synthetic human motilin (50 ng/kg). Motilin administered on a background of saline induced a premature phase III migrating fr

Characterization of a 50-kDa component of epithelial basement membranes using GDA-J/F3 monoclonal antibody.

Using the monoclonal antibody GDA-J/F3, a 50-kDa noncollagenous component of human skin basement membrane zone was identified. Immunofluorescence stainings of normal human skin with the GDA-J/F3 antibody showed a linear fluorescence decorating the basement membrane zone. With immunoelectron microscopy, the epitope was localized to the insertion points of the anchoring fibrils into the lamina densa. The antigen is distinct from collagen VII, from the main structural protein of the anchoring fibrils, and from several other structural molecules of the basement membrane zone, because the GDA-J/F3

A multi-omics approach reveals PFHxS as an environmental driver of gastric cancer via KEAP1 downregulation.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental pollutants increasingly linked to human disease. Perfluorohexanesulfonic acid (PFHxS), a widespread PFAS detected in human serum, has an uncharacterized role in gastric cancer (GC), one of the leading causes of cancer mortality worldwide. Here, we employed an integrated multi-omics approach combining network toxicology, large-scale transcriptomic analyses from TCGA and GEO cohorts, single-cell RNA sequencing, and molecular simulations, followed by in vitro and in vivo validation at environmentally relevant concentrations.

Bio-Inspired Janus All-Nanofiber Self-Powered Electronic Skins for Multifunctional, High-Resolution, Artifact-Free Human-Machine Interface.

Artificial skins combining sustainable tactile sensing and high-precision contextual interaction could transform medical devices and robotics yet developing body-integrated systems that are electrically stable, comfortable, and scalable remain challenging. We develop a heterogeneous Janus e-skin enabled by gel-fiber-mediated interlayer adhesion and co-design of materials, structure, and architecture. An engineered polyurethane ionogel (PUIL) coupled with an elastic SBS scaffold redistributes mechanical stress. Hydrogen bonding and ion-dipole interactions immobilize the liquid metal (LM) conduc

Predicting the Habitat Suitability for Endangered Tree Species Pterocarpus marsupium in Nepal, Using Ensemble Species Distribution Models.

Pterocarpus marsupium Roxb., locally known as Bijaysal, is a high-value tropical deciduous tree with a restricted and fragmented distribution in Nepal. We integrated 129 spatially occurring records with climatic, anthropogenic, edaphic, and topographic variables in an ensemble species distribution modeling framework (biomod2) to project current and future (2050s and 2090s) habitat suitability under four shared socioeconomic pathways (SSP) scenarios. Minimum temperature of the coldest month and precipitation seasonality were the dominant climatic drivers, followed by soil organic carbon, bulk d

M6 Metabolite Contributes to the Efficacy of the Rac/Cdc42 Inhibitor MBQ-167 in Metastatic Breast Cancer.

Metastasis remains a major challenge in cancer treatment because of the lack of effective targeted therapies. MBQ-167, a first-in-class dual Rac/Cdc42 inhibitor currently in a phase I clinical trial, has demonstrated promising activity in preclinical breast cancer models by reducing tumor burden and preventing metastasis. To characterize its metabolism, we conducted liver microsome assays and identified several MBQ-167 metabolites. Of these, M6 was the primary metabolite from dog and human plasma following oral administration of MBQ-167. The M6cpharmacokinetics profile parallels that of MBQ-16

3D HucMSCs derived extracellular vesicles enhanced therapeutic efficacy in treating intrauterine adhesions via BECN1 delivery.

Intrauterine adhesion (IUA) is a significant cause of female infertility and a critical public health concern among women of childbearing age. Emerging evidence has highlighted the therapeutic potential of mesenchymal stem cell-derived extracellular vesicles (EVs) in IUA treatment. However, conventional two-dimensional (2D) culture systems impose constraints on the yield and biological activity of EVs. In this study, we employed a three-dimensional (3D) culture platform based on gelatin methacrylate (GelMA) microspheres to culture human umbilical cord mesenchymal stem cells and isolate MSC-EVs

L-(+)-Ergothioneine ameliorates preeclampsia-associated vascular endothelial dysfunction by modulating the Nrf2-PPARγ-sFlt-1 axis.

BACKGROUND: Preeclampsia (PE) is a serious complication of pregnancy, with vascular endothelial dysfunction being a core pathological feature. This study aimed to investigate whether L-(+)-ergothioneine (LET) ameliorates PE-associated endothelial dysfunction by regulating the Nrf2-PPARγ-sFlt-1 axis. METHODS: A LPS-induced trophoblast dysfunction model was established using lipopolysaccharide (LPS)-induced human trophoblast cells (HTR8/SVneo). Techniques including CCK-8 assay, flow cytometry, wound healing assay, ELISA, qPCR, Western blot, and immunofluorescence were employed to assess the eff

Microplastics and neurotoxicity: could prenatal exposure to microplastics boost congenital enteric neuropathies?

Microplastics (MPs) pollution represents an increasing worldwide problem and a real global challenge for human health, which also affects unborn children. Specifically, during their degradation, they can release a broad range of toxic and hormonally active agents, such as plasticizers. Thus, microplastics alone are pernicious, but they often also carry other harmful chemicals and even problematic bacteria on their surface and within their structure (heavy metals, pesticides, parabens, etc.), which amplifies their toxic potential. Due to their induction of oxidative damage, inflammation, mitoch

Local agricultural transition, crisis and migration in the Southern Andes.

The transition to agriculture was a transformative process in human history with wide-ranging demographic and social consequences1. Across South America, agriculture was adopted at different times and through diverse pathways, resulting in a mosaic of regionally distinct farming histories2,3. The Uspallata Valley, at the southern frontier of Andean farming, offers a unique opportunity to examine a case of late adoption of agriculture. Here we show that agriculture in the Uspallata Valley was adopted by local hunter-gatherers, as evidenced by genetic continuity between pre-farming and farming p

Design, synthesis and biological evaluation of novel artesunate-selenylindole hybrids against colorectal cancer.

Colorectal cancer (CRC) remains a major global public health threat, creating an urgent demand for novel therapeutic agents. Artesunate (ART), a semisynthetic derivative of artemisinin, has demonstrated anti-colorectal cancer effects, but its moderate antitumor potency (IC50 > 50 μM) severely limits further clinical application. To enhance the activity against colorectal cancer (CRC), we adopted a scaffold hybridization strategy to rationally design and synthesize 22 novel artesunate-selenylindole hybrids. All derivatives were screened for their anti-proliferative activity against four human c

SPARC promotes angiogenesis and cell migration via VEGFR2-ERK1/2-SPHK1 axis in pterygium.

Pathological angiogenesis is a primary driver of pterygium progression, yet the mechanistic role of Secreted Protein Acidic and Rich in Cysteine (SPARC) remains to be fully elucidated. This study investigated the clinical significance and molecular function of SPARC in pterygium-associated neovascularization. Expression analysis in human pterygium tissues revealed a significant upregulation of SPARC, which positively correlated with lesion microvascular density as assessed by immunofluorescence and anterior segment optical coherence tomography angiography. Functionally, in vitro assays demonst

Copper metallodendrimers effectively deliver proapoptotic siRNA to human liver carcinoma HepG2 cells.

The delivery of therapeutic nucleic acids to target cells faces many challenges, and novel carriers continue to be investigated. Metallodendrimers present several beneficial features as drugs and nucleic acid carriers, including specificity toward pathological cells, low toxicity to normal cells, and the capacity to cross cellular membranes. In the present work, we investigated the efficacy of synthesized Cu(II) metallodendrimers in delivering pro-apoptotic siRNAs, specifically siBCL2 and siMCL1, to human liver carcinoma HepG2 cells. The resulting siRNA/dendrimer complexes exhibited a strong p

Biomimetic lymph node-like scaffolds for optimized CAR-T cell expansion and potentiated antitumor efficacy.

Chimeric antigen receptor T cell (CAR-T) therapy has shown remarkable promise in treating hematological malignancies. However, the ex vivo expansion of CAR-T cells is time-consuming, potentially impairing CAR-T cell function. Physiologically, T cell activation and proliferation occur within the lymph node (LN) paracortex, a dynamic environment structured by a three-dimensional (3D) reticular network (RN) that promotes cell migration and mediator delivery. Mimicking this physiological niche offers a compelling strategy to improve CAR-T cell expansion. Inspired by the structure of the RN, we dev

Condensation of the Golgi Apparatus Activates YAP1 to Promote Gastric Cancer Progression.

UNLABELLED: Alterations in the structure of the Golgi apparatus play a pivotal role in cancer progression and invasion. A better understanding of how Golgi morphology regulates the metastatic potential of cancer cells could help identify potential treatment strategies. In this study, we investigated how specific structural variations in the Golgi, particularly fragmentation and condensation, influence the malignancy of gastric cancer using human cell lines, xenograft mouse models, and human patient tissue samples. Gastric cancer cells with condensed Golgi structures exhibited increased prolife

Development of novel anti-CDH1/E-cadherin monoclonal antibodies for versatile applications.

Cadherin (CDH)-mediated extracellular homophilic binding is crucial for maintaining tissue homeostasis. The epithelial cell-cell adhesion molecule cadherin 1 (CDH1/E-cadherin) forms the adherens junctions in epithelial cells, and the loss of CDH1 facilitates the migration and invasion of carcinoma cells. Although several anti-CDH1 monoclonal antibodies (mAbs) are available for western blotting and immunohistochemistry (IHC), a highly sensitive anti-CDH1 mAb suitable for flow cytometry has not been developed. We developed anti-CDH1 mAbs through a flow cytometry-based high-throughput screening.

The Traits of the Regenerative Potential of Platelet-Rich Plasma from Donors with Type 1 and Type 2 Diabetes Mellitus.

The low predictability of the effects of autologous platelet-rich plasma (PRP) in regenerative therapy for patients with type 1 and type 2 diabetes mellitus (DM) underscores the need for further research assessing the reparative effects of PRP based on the type of DM. The aim of this study was to evaluate the regenerative potential of PRP from young donors (30-40 years old) with DM1 and DM2 in vitro, specifically its effects on human dermal fibroblast cell culture. The in vitro effects of PRP from patients with type 1 and type 2 DM were investigated using a culture of human dermal fibroblasts
