Net Absorption and Metabolism of β-Hydroxy- β-Methyl Butyrate during Late Gestation in a Pig Model.

The leucine metabolite, β-hydroxy-β-methyl butyrate (HMB), is widely used in human nutrition and animal production as a nutritional supplement. Although the HMB usage during late gestation has been demonstrated to have a positive effect on fetal development, knowledge on net absorption and metabolism of HMB and impact of HMB on branched chain amino acids (BCAAs) metabolism is lacking. To address this, we conducted a study using pigs during the perinatal period as a model organism. Eight-second parity sows were fitted with indwelling catheters in the femoral artery and in the portal, hepatic, f

The cartilage collagens: a review of their structure, organization, and role in the pathogenesis of experimental arthritis in animals and in human rheumatic disease.

This contribution reviews the structure and organization of collagen molecules found in cartilage and the roles that they may play in rheumatic diseases. Cartilage is unique in its physical properties and molecular composition, and contains sufficient amounts of types II, IX, X, and XI collagen to deem these molecules as "cartilage-specific." The vitreous body of the eye, a "cartilage-like" tissue is also rich in the same collagens but is type X deficient. Types VI and XII collagen are present in cartilage as well as noncartilaginous tissues. Types II, IX, and XI collagen are organized into ma

Polystyrene microplastics impact on cardiac and pulmonary physiology and microenvironment in a mouse model: Role of taurine supplementation and molecular docking insights.

Polystyrene microplastics (PS-MPs) have recently gained attention as widespread environmental contaminants posing risks to both human and animal health. In this study, we investigated the potential protective effect of taurine (200 mg/kg b.wt) against cardiopulmonary toxicity induced by PS-MPs (10 mg/kg b.wt) in male Swiss mice following a 60-day oral exposure. Molecular docking investigation for both proteins and mRNA targets was carried out utilizing a global, flexible docking strategy that allowed for full ligand conformational freedom and binding surface exploration. We designed an experim

From comparative oncology to AI-enabled precision medicine: translational biomodels for triple-negative breast cancer.

Triple-negative breast cancer (TNBC), defined by the absence of estrogen receptor, progesterone receptor, and human epidermal growth factor receptor 2 expression or amplification, comprises a biologically heterogeneous group of tumors with aggressive clinical behavior and limited biomarker-guided treatment options. Although chemotherapy, immune-checkpoint inhibition, poly(ADP-ribose) polymerase inhibitors, and antibody-drug conjugates have expanded the therapeutic landscape, durable benefit remains constrained by genomic instability, homologous recombination deficiency, phenotypic plasticity,

Probing Cinnamamides and Benzamides as Anthelmintics: Discovery of Potent Drug-Like Agents Against Angiostrongylus cantonensis.

Emergent helminthiases are increasingly impacting global health in both humans and animals, especially given the limited efficacy of existing drugs against these infections. Neuroangiostrongyliasis, an eosinophilic meningitis caused by Angiostrongylus cantonensis, currently lacks effective treatment, highlighting the need for novel anthelmintics. We previously identified cinnamoyl-benzylpiperazine, a simplified analogue of cinnarizine, as an effective anthelmintic agent against first (L1) and third-stage (L3) larvae of A. cantonensis in vitro. In the present work, structural modifications on t

Emerging applications of non-thermal ultrasound technology in functional beverages processing, a comprehensive review.

Non-thermal ultrasonication (US) is emerging as a promising process-intensification technology for producing functional beverages (FB) as the conventional thermal processing and additive-reliant preservation methods often compromise thermolabile nutrients, sensory profile, and clean-label appeal of FB. However, evidence regarding the application of US to the processing and preservation of FB matrices remains limited and fragmented, and clear, reproducible guidelines for process optimization and commercial translation are still lacking. This review critically summarizes recent research on the a

Crosstalk between liver sinusoidal endothelial cells and hepatocytes via IL-1α-IL1R1 axis exacerbates ischaemia/reperfusion injury in aged livers.

BACKGROUND: With population ageing, elderly patients account for a growing proportion of hepatic surgery recipients. Hepatic ischaemia-reperfusion injury (HIRI) is a major cause of postoperative liver dysfunction, particularly in aged livers, yet its mechanisms remain poorly understood. OBJECTIVE: We aimed to elucidate critical cellular interactions and molecular mechanisms underlying aggravated HIRI in aged livers to uncover therapeutic targets. DESIGN: Single-cell RNA sequencing and spatial transcriptomics were performed on liver tissues from humans, rats and mice across ages to define key

Multi-omics analysis reveals STING activation mediates NLRP3-related pyroptosis and exacerbates myocardial ischemia-reperfusion injury.

BACKGROUND: Myocardial ischemia-reperfusion injury (I/R) significantly exacerbates cardiomyocyte damage post-recanalization therapy in acute myocardial infarction. Pyroptosis and the NLRP3 inflammasome are crucial in I/R, yet the precise mechanism remains unclear. MATERIALS AND METHODS: Transcriptomics, proteomics, and single-cell transcriptomics were employed to examine cellular subtype changes and pyroptosis-associated gene regulation in I/R. Differential pyroptosis-related genes were identified from transcriptomics data and validated with proteomics. Single-cell RNA sequencing assessed pyr

Alpha Lipoic Acid Mitigates TBI-Induced Neuroinflammation by Regulating S100B/STIM Signaling via NF-ƙB Pathway.

Alpha-lipoic acid (ALA) is a natural compound present in plants, animals, and humans. It provides neuroprotection through its antioxidant action, reducing oxidative stress and inflammation via pathways such as the Nrf-2 signalling pathway. It has shown protective effect against neuronal damage in Alzheimer's disease and Parkinson's disease. The traumatic brain injury (TBI) is a major cause of cognitive and motor impairments. TBI primarily initiates a series of secondary injury pathways, with glutamate excitotoxicity being a critical factor in neuronal degeneration. In this study, we investigat

The effect of psychological stress on masticatory muscle electromyography-a scoping review.

Temporomandibular disorders (TMDs) are the second most common reason for orofacial pain with myalgia being the most prevalent diagnosis. Overuse of the masticatory muscles and the presence of stress are both risk factors for myalgia. The extent to which these risk factors interact remains unclear, including whether psychological stress contributes to increased masticatory muscle activity. The purpose of this scoping review was to identify, examine, and map the available literature exploring the relationship between psychological stress and masticatory muscle electromyography (EMG). Pre-clinica

Perinatal exposure to perfluoroalkyl substances impairs maternal care and induces depressive-like behavior in mice.

Postpartum mental health disorders are a critical yet understudied aspect of maternal health. Exposure to environmental toxicants such as per- and poly-fluoroalkyl substances (PFAS) has been associated with adverse health outcomes, including reproductive and neurobehavioral dysfunction, whereas their specific effects on maternal behavior and mental health remain poorly characterized. This study investigated the effects of perinatal exposure to environmentally relevant concentrations of a PFAS mixture comprising 10 individual PFAS (PFHxA, PFPeA, PFHpA, PFBA, perfluorooctanoic acid [PFOA], PFOS,

Functional Characterization of Hsp110 in Drosophila Reveals its Essential and Dosage-Sensitive Role in Nervous System Integrity.

The Hsp70 molecular chaperone system is the front line of defense in maintaining cellular proteostasis. In eukaryotes, ATP/ADP nucleotide exchange in the Hsp70 chaperone cycle is stimulated by Hsp110, a divergent member of the Hsp70 chaperone superfamily and co-chaperone of Hsp70. Hsp110 is also a known modifier of neurodegenerative and other protein misfolding-related disorders. Biochemical aspects of Hsp110 chaperone functions have been characterized in vitro, and pathway interactions have been extensively characterized genetically in yeast model systems; however, a detailed understanding of

Disruption of Extracellular Signal-Regulated Kinase Partially Mediates Neonatal Isoflurane Anesthesia-Induced Changes in Dendritic Spines and Cognitive Function in Juvenile Mice.

There is a growing concern worldwide about the potential harmful effects of anesthesia on brain development, based on studies in both humans and animals. In infants, repeated anesthesia exposure is linked to learning disabilities and attention disorders. Similarly, laboratory studies in mice show that neonates exposed to general anesthesia experience long-term cognitive and behavioral impairments. Inhaled anesthetics affect the postsynaptic density (PSD)-95, discs large homolog, and zona occludens-1 (PDZ) domains. The disruption of the synaptic PSD95-PDZ2 domain-mediated protein interactions l

Pleiotropic Effects on Tachyzoite and Host Cell Proteomes in Knock-Out Clones of the Open Reading Frames 297720 and 319730 Constitutively Expressed in T. gondii ShSp1 Tachyzoites.

Toxoplasma gondii, the causative agent of toxoplasmosis widespread in animals and humans, is an intracellular apicomplexan protozoan parasite infecting a variety of host cells. Gene editing using CRISPR-Cas9 has become a standard tool to investigate the molecular genetics of this interaction. With respect to gene knock-out (KO) studies, the general paradigm implies that the gene of interest is expressed in the wildtype and that only the gene of interest is affected by the knock-out. Consequently, the observed phenotype depends on the presence or absence of genes of interest. To challenge this

Prenatal exposure to phthalates and phthalate replacements in relation to chorionic plate surface vasculature at delivery.

Pregnant people are ubiquitously exposed to endocrine-disrupting phthalates through consumer products and food. The placenta may be particularly vulnerable to the adverse effects of phthalates, with evidence from animal models suggesting impacts on placental development and vascularization. We translate this research to humans, examining gestational exposure to phthalates and phthalate replacements in relation to novel markers of chorionic plate surface vascularization. Phthalate and phthalate replacement metabolites were measured in first trimester urine from pregnant participants in the Unde
