Despite the critical role of lipid-mediated signaling in regulating host immunity, endorsed by growing evidence, the interaction between lipid metabolism and immune response remains largely unknown. This review aims to elucidate the immunomodulatory role of a lung-enriched lipid metabolic pathway mediated by the phospholipase A2 (PLA2) family, which comprises a diverse range of lipid-hydrolyzing enzymes. Based on their location, structure, substrate specificity and physiological roles, PLA2s can be classified into secreted PLA2s (sPLA2s), cytosolic PLA2s (cPLA2s), calcium-independent PLA2s (iP

BACKGROUND: Immunotherapy efficacy in colorectal cancer (CRC) is largely restricted to microsatellite instability-high (MSI-H) tumors, highlighting an urgent need to overcome resistance in microsatellite-stable (MSS) CRC. Mitochondrial DNA (mtDNA) leakage activates the cGAS-STING pathway, a potent inducer of antitumor immunity. However, endogenous regulators constraining mtDNA release in CRC, particularly within the inner mitochondrial membrane (IMM), remain poorly defined.
METHODS: In CRC cohorts from TCGA, integrated bioinformatics analysis identified dysregulated mitochondria-associated ge

Pulmonary involvement in cystic fibrosis (CF) is characterised by respiratory infections caused by bacteria, viruses, and fungi, as well as by dysregulated inflammatory and immune responses. Although essential for the host's initial defence against these microorganisms, the innate immune response is altered in its main cellular (airway epithelial cells (AECs), monocytes, macrophages, and neutrophils) and molecular (cytokines, chemokines, signal transduction pathways, and transcription factors) components. MicroRNAs (miRNAs) form a regulatory network at the level of inflammatory and immune resp

Respiratory infections are the fourth leading cause of death globally. Acinetobacter baumannii is a significant cause of ventilator-associated pneumonia and antibiotic-resistant attributable deaths worldwide. Iron is an essential nutrient; vertebrates exploit this by sequestering iron from invading pathogens in a process termed nutritional immunity. The competition for iron is a crucial determinant of A. baumannii infection severity and outcome. Here, we have discovered an A. baumannii acyltransferase that we have named AimS. AimS promotes fitness in low iron environments and contributes to A.

UNLABELLED: Interferon regulatory factor 8 (IRF8) is a master transcription factor of myeloid differentiation, but whether IRF8 intrinsically controls B cell function in tumors remains unknown. Using paired single-cell transcriptomic and chromatin accessibility profiling of tumors from wild-type and Irf8-deficient mice, we identify a B cell-intrinsic IRF8 axis regulating antigen presentation and sustaining anti-tumor CD8⁺ T cell immunity. IRF8 establishes conserved chromatin accessibility programs across myeloid cells and plasmablasts centered on antigen processing and MHC class I presentation

Cisplatin resistance is a major obstacle to effective chemotherapy in patients with cervical cancer and is associated with an immunosuppressive tumour microenvironment. Copper ions are essential trace elements that regulate redox and immune balance. However, the connections among copper ions, cisplatin resistance and immune regulation remain unclear. Here, higher serum copper levels correlated with increased pan-cancer mortality. Integrative transcriptomic and single-cell RNA sequencing analyses revealed lower copper-related transcriptional activity in CD8⁺ T cells from cervical cancer tissues

Metabolic dysfunction-associated steatotic liver disease (MASLD) has emerged as the leading chronic liver disorder worldwide. Its pathological progression typically begins with hepatic steatosis, advances to metabolic dysfunction-associated steatohepatitis (MASH) and fibrosis, and in some cases culminates in hepatocellular carcinoma (HCC). This continuum is influenced not only by metabolic disturbances but also by coordinated local and systemic inflammatory and immune responses. This article systematically summarizes the inflammatory mechanisms underlying MASLD/MASH progression, highlighting t

BACKGROUND: Oysters are known for their rich nutritional profile and medicinal properties documented in traditional medicine. Their bioactive compounds exhibit antioxidant, anti-inflammatory, antimicrobial, and osteogenic activities, leading to widespread applications in food and medicine. Oyster shells—composed of > 90% calcium carbonate and various minerals and amino acids—are traditionally used as antacids in East Asia. Despite these recognized benefits, the mechanisms by which oyster shells modulate immune responses remain unclear. In this study, we elucidated the immunomodulatory effects

Serine proteases are a class of enzymes that orchestrate an immune response. These proteases can be hijacked by viruses to facilitate entry and spread, while simultaneously supporting the innate immune system in neutralizing pathogens. This review highlights the dual roles of exogenous serine proteases, emphasizing neutrophil serine proteases (NSPs) that facilitate viral entry and promote disease progression while also contributing to antiviral defense by degrading viral glycoproteins. Additionally, the potential to modulate serine protease activity to boost host defenses will be discussed, of

Proteins encoded by mRNA vaccines can be expressed by a diversity of transfected cell types but how cell-type-specific expression influences immunity is poorly understood. To investigate this, we incorporated synthetic microRNA target sites (miRT) into lipid nanoparticle (LNP)-delivered mRNA vaccines to silence mRNA expression specifically in professional antigen-presenting cells (pAPCs), hepatocytes or myocytes. We found that mRNA expression in pAPCs was dispensable for priming antigen-specific T cells, whereas mRNA expression in myocytes induced similar or stronger immune responses, includin

Personalized neoantigen mRNA vaccines showed good efficacy in treating metastatic melanoma, pancreatic cancer, and breast cancer in early phase clinical trials. Strategies are needed to further enhance neoantigen mRNA vaccine-induced anti-tumor immunity. This study explored conventional adjuvant co-administration to enhance mRNA vaccine-induced anti-tumor immunity. We found a clinically used CpG 1018 adjuvant was highly effective to enhance ovalbumin (OVA) and neoantigen mRNA-induced T cell responses and anti-tumor immunity in B16F10-OVA melanoma models. Mechanistic studies found CpG 1018 main

Messenger RNA lipid nanoparticles are promising vaccine carriers, yet their movement through the body after intramuscular injection and the resulting tissue-specific immune effects remain poorly defined. Here we show that lipid nanoparticle trafficking from the injection site into systemic organs is a key determinant of localized immunity, particularly the formation of tissue-resident cytotoxic T cells. We compared multiple nanoparticle formulations that differed in lipid composition, including ionizable lipids and helper lipids commonly used in clinical vaccines. These formulations exhibited

Dendritic cells (DCs) can activate T cells to trigger sustained antitumor immune responses, a process in which the continuous migration of antigen-loaded DCs from the tumor microenvironment to tumor-draining lymph nodes is critical. DCs have evolved a complex and dynamic regulatory network to mediate their migration to specific locations, with multiple transporters including amino acid transporters reported to participate in this process. Slc1a2 is highly expressed in the nervous system, where it mediates the clearance of extracellular glutamate, primarily in astrocytes; however, its role in t

Following the COVID-19 pandemic, measles has seen a resurgence across Europe, including Italy. Increased vulnerability among young adults has raised concerns about waning vaccine-derived immunity. This study evaluated measles immunity among adult blood donors in the Apulia region to identify sero-susceptibility patterns and inform targeted public health strategies. A retrospective seroprevalence study was conducted using sera from 1579 healthy blood donors, aged 18 to 65 y, who attended donation centers in Apulia region between November 2023 and February 2024. Anti-measles virus (MV) IgG antib

OBJECTIVES: This study evaluates seroprevalence of diphtheria toxoid immunoglobulin G antibodies among displaced populations in Abyan Governorate, which is to the best of our knowledge the first such assessment in Yemen's conflict setting.
METHODS: A cross-sectional study in 390 displaced individuals (aged 1 month-90 years) assessed anti-diphtheria antibody levels through enzyme-linked immunosorbent assay. Geometric mean titers (GMTs) and seroprotection rates were analyzed by age, gender, and vaccination status.
RESULTS: Overall seroprotection was 76.2%, with 23.8% susceptible (<0.01 IU/ml).

Following severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, the complement pathway becomes activated, generating the stable split product C3d. Although C3d can modulate B cell responses through complement receptor 2 (CR2) in experimental systems, it remains unclear how circulating C3d relates to antibody responses after SARS-CoV-2 infection in humans. Here, we examined plasma C3d levels and their associations with SARS-CoV-2-specific antibody responses and clinical characteristics in convalescent adults and children. In adults, older age, male sex, and a history of severe

Foot-and-mouth disease (FMD) is a major animal infectious disease that has garnered significant international attention. Currently, conventional inactivated vaccines and virus-like particle (VLP) vaccines primarily induce protective immune responses through the generation of neutralizing antibodies. However, these vaccines exhibit limited activation of cellular immunity and offer short-term immune persistence. Furthermore, inactivated vaccines raise certain biosafety concerns. In this study, an FMD mRNA vaccine capable of self-assembling into non-infectious VLP was developed. Immunization of m

Limited understanding of African swine fever (ASF) immunity remains a major barrier to the rational development of safe and effective vaccines. While antibody-mediated protection is still poorly defined, growing evidence highlights a central role for cellular immunity, particularly cytotoxic cells, as components associated with ASF virus (ASFV) infection control. However, the contribution of individual cytotoxic subsets across different virological and immunological contexts is not well characterized. Here, we investigated cytotoxic responses during BA71ΔCD2 live attenuated vaccine-induced pro

Glymphatic transport, meningeal lymphatic drainage, and meningeal immunity have been implicated in metabolic waste clearance, cerebral edema formation, and the regulation of neuroinflammation after stroke. However, the available evidence remains fragmented and has not been systematically integrated. This review was conducted in accordance with PRISMA guidelines. PubMed, Embase, Scopus, the Cochrane Library, and Web of Science were searched from database inception to April 3, 2026, for studies investigating glymphatic pathways, the meningeal lymphatic system, meningeal immune responses, and rel

BACKGROUND: Decorin (DCN) predominantly produced by fibroblasts is a small leucine-rich proteoglycan with tumor-suppressive property. However, whether DCN has a role in shaping the tumor immune microenvironment remains elusive.
METHODS: The TCGA and GEO databases were analyzed to identify fibroblast-specific secretory proteins that are downregulated in most types of human tumors, positively correlate with CD8⁺ T cell infiltration, and associate with improved response to immune checkpoint blockade (ICB) therapy. The function of DCN in vivo was assessed using cell lines with stable DCN overexpr
