Investigation of Medium Modifications to ^{12}C Structure Functions in the Resonance Region.

We present results from a high precision experimental study of the nuclear modification of the longitudinal (F_{L}) to transverse (F_{1}) structure function ratio for bound nucleons in the resonance region. The inclusive electron scattering cross sections were measured in Jefferson Lab Experimental Hall C on carbon and deuterium nuclei for a large range of kinematics, allowing for separations of the longitudinal and transverse structure functions to be performed at a range of four-momentum transfer values 0.5≤Q^{2}≤3.75  GeV^{2}. In contrast to the significant body of measurements of the nucle

Deep Learning-Enabled Engineering of a Hyper-Stable and Soluble MPB70-83 Antigen for Sensitive Bovine Tuberculosis Surveillance.

Bovine tuberculosis (bTB) diagnostics are frequently hindered by the structural instability and insolubility of recombinant multi-epitope proteins in prokaryotic expression systems. To address this, we developed an integrated strategy combining immunoinformatics with the ProteinMPNN deep learning framework. We constructed a fusion protein utilizing B-cell epitopes from MPB70 and MPB83, where ProteinMPNN was employed to redesign non-epitope scaffolds for optimized thermodynamic stability. The structural integrity was verified via 200-ns molecular dynamics (MD) simulations. MD analysis revealed

The lack of evidence-based management in electrical storm: a scoping review.

Electrical storm (ES) is associated with a significant risk of morbidity and mortality. Despite this, there has been limited research in ES resulting in uncertainty and inconsistency in the management of this life-threatening condition. The objective of this scoping review was to define the current body of literature evaluating pharmacologic and non-pharmacologic therapies used in the management of ES. A comprehensive search of Medline, CENTRAL and Embase was completed on January 11, 2025. Primary studies on pharmacotherapies in ES were included if they reported therapy-related outcomes and in

Re-evaluating robotic median arcuate ligament release: methodological and generalizability limitations.

This letter critically appraises the study by Simioni A, et al. on median arcuate ligament release (MALR), which reports high rates of symptom resolution and low morbidity in a single-center cohort. While the results are encouraging, several limitations require further consideration. The study population was demographically narrow, with 94% females, with low body mass index (BMI) and selected from a single center using a highly specialized stepwise multidisciplinary algorithm, introducing selection bias that limits generalizability. The absence of a control group prevents the conclusion about

Chronic sarcoid myopathy mimicking facioscapulohumeral muscular dystrophy: a case report.

Chronic sarcoid myopathy is a rare disorder characterized by intramuscular granulomas and generally presents with symmetrical proximal limb-girdle muscle weakness. Here, we present an atypical case of a 68-year-old male with a history of pulmonary sarcoidosis with strikingly asymmetric limb-girdle weakness, progressive >20 years, including periscapular, paraspinal, lower limb and subtle facial involvement, mimicking facioscapulohumeral muscular dystrophy. MR images revealed a striking asymmetric pattern of patchy muscle involvement of paraspinal, lower limb and right periscapular muscles witho

Dual probe ligation in situ hybridization with rolling-circle amplification for high-plex spatial transcriptomics.

New biological insights are increasingly dependent upon a deeper understanding of tissue architectures. Critical to such studies are spatial transcriptomics technologies, especially those amenable to analysis of the most widely available human tissue type, formalin-fixed and paraffin-embedded (FFPE) clinical specimens. Here we build on our previous oligonucleotide probe ligation-based approach to accurately analyze FFPE mRNA, which suffers from variable levels of degradation. Ligation In Situ Hybridization followed by rolling circle amplification (LISH-Lock'n'Roll or LISH-LnR), provides a stre

Rheumatic manifestations of HIV/AIDS.

Rheumatic manifestations of HIV infection encompass a wide spectrum of disorders, arising from direct viral effects, immune dysregulation, opportunistic infections, or antiretroviral therapy (ART)-induced complications. These manifestations can occur at any stage of HIV, and include arthralgia, HIV-associated arthritis, spondyloarthropathies (SpA), inflammatory myopathies, vasculitides, and opportunistic musculoskeletal infections. Arthralgia is common and self-limiting, whereas HIV-associated arthritis mimics rheumatoid arthritis. SpA, particularly reactive arthritis, psoriatic arthritis, and

A Case of Anti-TIF1γ Antibody-Positive Dermatomyositis Associated With Malignancy.

Dermatomyositis (DM) is a rare acquired autoimmune myopathy characterized by proximal muscle weakness, inflammation, and a typical skin rash. It is considered one of the idiopathic inflammatory myopathies (IIM), a group of heterogeneous systemic diseases that include DM, polymyositis, and inclusion body myositis. A significant portion of patients with IIM, particularly adults, can have an association with malignancy, usually preceded by muscle and skin symptoms. We report a case of a 67-year-old man who presented to the accident and emergency department with a six-week history of proximal myop

Age-related hearing loss and dementia risk across the lifespan: mechanisms, equity, and prevention.

Age-related hearing loss (ARHL) has emerged as a significant and potentially modifiable risk factor for neurodegenerative disorders, including Alzheimer's disease. A growing body of evidence links ARHL to structural and functional changes in the brain, with implications for cognitive decline and dementia onset. However, both ARHL and dementia are multifactorial conditions shaped not only by biological mechanisms but also by broader social determinants of health. Inequities in access to hearing care, underrepresentation of marginalised populations in research, and variability in intervention ou

Mutant mice with rod-specific VPS35 deletion exhibit retinal α-synuclein pathology-associated degeneration.

Vacuolar protein sorting 35 (VPS35), the core component of the retromer complex which regulates endosomal trafficking, is genetically linked with Parkinson's disease (PD). Impaired vision is a common non-motor manifestation of PD. Here, we show mouse retinas with VPS35-deficient rods exhibit synapse loss and visual deficit, followed by progressive degeneration concomitant with the emergence of Lewy body-like inclusions and phospho-α-synuclein (P-αSyn) aggregation. Ultrastructural analyses reveal VPS35-deficient rods accumulate aggregates in late endosomes, deposited as lipofuscins bound to P-α

Safe and Orally Bioavailable Inhibitor of Serine Palmitoyltransferase Improves Age-Related Sarcopenia.

The accumulation of ceramides and related metabolites has emerged as a pivotal mechanism contributing to the onset of age-related diseases. However, small molecule inhibitors targeting the ceramide de novo synthesis pathway for clinical use are currently unavailable. We synthesized a safe and orally bioavailable inhibitor, termed ALT-007, targeting the rate-limiting enzyme of ceramide de novo synthesis, serine palmitoyltransferase (SPT). In a mouse model of age-related sarcopenia, ALT-007, administered through the diet, effectively restored muscle mass and function compromised by aging. Mechan

Targeting Protein Misfolding and Aggregation as a Therapeutic Perspective in Neurodegenerative Disorders.

The abnormal deposition and intercellular propagation of disease-specific protein play a central role in the pathogenesis of many neurodegenerative disorders. Recent studies share the common observation that the formation of protein oligomers and subsequent pathological filaments is an essential step for the disease. Synucleinopathies such as Parkinson's disease (PD), dementia with Lewy bodies (DLB) or multiple system atrophy (MSA) are neurodegenerative diseases characterized by the aggregation of the α-synucleinprotein in neurons and/or in oligodendrocytes (glial cytoplasmic inclusions), neur

[Candida albicans: risk factor in covid-19 patients].

UNLABELLED: The Candida Albicans fungus in our body can cause various conditions and will depend directly on the systemic condition of the host. Patients with COVID-19 who have previously presented this fungus can increase the likelihood of morbidity and mortality, since this microorganism can be located in areas that correspond to the respiratory system, generating a functional deficit. If not treated timely, it will proliferate into the blood and digestive system. Many patients with respiratory difficulties on account of this condition require mechanical ventilation to combat it. OBJECTIVE:

Overlap syndrome with antibodies against multiple transfer-RNA components presenting antisynthetase syndrome.

Overlap syndrome is a clinical entity of myositis concomitant with one or more collagen diseases such as systemic lupus erythematosus, systemic sclerosis, and/or rheumatoid arthritis. It is not evident whether the myopathology of overlap syndrome is disease-specific or categorizes one of the four major subsets: inclusion body myositis, immune-mediated necrotizing myopathy, dermatomyositis, and antisynthetase syndrome. We report a patient with overlap syndrome who exhibited autoantibodies against multiple transfer-RNA components by RNA immunoprecipitation, suggesting antisynthetase syndrome. A

Inclusion of high-flavonoid corn in the diet of broiler chickens as a potential approach for the control of necrotic enteritis.

Avian necrotic enteritis (NE) is an infectious disease that impacts poultry worldwide causing economic losses. Discontinued use of antimicrobial growth promoters has been associated with high incidence of the disease, which has led to a necessity for finding new therapeutic alternatives. Flavonoids are polyphenolic compounds that have been studied for their health-promoting properties in animals and humans. This study presents a flavonoid-rich corn (PennHFD), as a potential alternative for ameliorating NE in broiler chickens. The effect of a diet formulated with PennHFD was compared to a diet

First-Trimester Diagnosis of Supernumerary Hemivertebra.

Hemivertebra is a common cause of congenital scoliosis and results from a lack of formation of one-half of the vertebral body. This condition is very rare and can present as solitary or as a syndrome component: i.e., the split notochord syndrome, which often implies vertebral defects, from a bifid vertebra to hemivertebrae, or fused vertebrae. We describe a case of supernumerary lateral hemivertebra detected prenatally at 12 weeks of gestation and the ultrasonography specifics that lead to early and accurate diagnosis, monitoring during pregnancy, and follow-up at the 4-year period. The case i

Ferritinophagy and α-Synuclein: Pharmacological Targeting of Autophagy to Restore Iron Regulation in Parkinson's Disease.

A major hallmark of Parkinson's disease (PD) is the fatal destruction of dopaminergic neurons within the substantia nigra pars compacta. This event is preceded by the formation of Lewy bodies, which are cytoplasmic inclusions composed of α-synuclein protein aggregates. A triad contribution of α-synuclein aggregation, iron accumulation, and mitochondrial dysfunction plague nigral neurons, yet the events underlying iron accumulation are poorly understood. Elevated intracellular iron concentrations up-regulate ferritin expression, an iron storage protein that provides cytoprotection against redox
