Identification and characterization of staphylococcal enterotoxin types G and I from Staphylococcus aureus.

Staphylococcal enterotoxins are exotoxins produced by Staphylococcus aureus that possess emetic and superantigenic properties. Prior to this research there were six characterized enterotoxins, staphylococcal enterotoxin types A to E and H (referred to as SEA to SEE and SEH). Two new staphylococcal enterotoxin genes have been identified and designated seg and sei (staphylococcal enterotoxin types G and I, respectively). seg and sei consist of 777 and 729 nucleotides, respectively, encoding precursor proteins of 258 (SEG) and 242 (SEI) deduced amino acids. SEG and SEI have typical bacterial sign

TAK-603 selectively suppresses Th1-type cytokine production and inhibits the progression of adjuvant arthritis.

We have shown that TAK-603, a new anti-rheumatic drug, is more effective in animal models in which cellular immunity plays a central role. Here, we studied the effect of the drug on Th1 cytokines, which are dominantly produced in this type of immune reaction, in an in vitro system and an in vivo model. We established Th1- and Th2-dominant T-cell lines, and studied the effect of TAK-603 on their cytokine production. Th1 cell lines were BALB/c mouse allo-reactive T cells and C57BL mouse mite antigen-reactive T cells, and the Th2 cell line was BALB/c mouse ovalbumin-reactive T cells. TAK-603 supp

Immunological activation markers in the serum of African and European HIV-seropositive and seronegative individuals.

OBJECTIVE: The concentration of type 1 and type 2 cytokines and fibroblast-associated apoptosis-1 soluble receptor (sAPO-1/Fas) was analysed in the sera of Ugandan and Italian HIV-1-seropositive and seronegative individuals. The data were compared to determine whether the immunological status of these groups was different. METHODS: Sixty-seven Ugandan and 30 Italian HIV-positive patients were analysed and stratified according to CD4 counts (group 1, > 500 x 10(6)/l; group 2, 200-500 x 10(6)/l; group 3, < 200 x 10(6)/l). Sera from 15 Ugandan and 11 Italian HIV-negative blood donors were also a

T cell clones from a non-leprosy exposed subject recognize the Mycobacterium leprae 18-kD protein.

Although Mycobacterium leprae shares many protein antigens with other mycobacterial species, there is a degree of specificity in the T cell response to the organism. This is evident in the failure of cross-protection between mycobacterial species and the specific unresponsiveness to M. leprae in lepromatous leprosy patients. The antigenic basis of this specificity is unresolved, but the M. leprae 18-kD protein is one candidate because of its restricted distribution and the isolation of M. leprae-specific T cell clones reactive with the protein from M. leprae-vaccinated subjects. In the course

Expansion and tumour specific cytokine secretion of bryostatin-activated T-cells from cryopreserved axillary lymph nodes of breast cancer patients.

Current adoptive immunotherapy strategies in cancer patients require large numbers of activated T-cells and are limited by the availability of autologous tumour. We describe a novel method of T-cell activation that produced relatively rapid, high-fold expansion of stored, frozen lymphocytes obtained from the lymph nodes of 20 breast cancer patients during axillary dissection but does not require autologous tumour. In vitro exposure of thawed cells to bryostatin-1 (B), a non-tumour promoting protein kinase C activator and ionomycin (I), a calcium ionophore, at day 0 followed by culture in low d

[Lymphokine production in vitro by blood lymphocytes from patients with a defective or hyperactive immune system].

Blood lymphocytes from 8 patients with common variable immunodeficiency (CVID), 5 patients with signs of polyclonal activation of the immune system (1 SLE, 3 AILAP, 1 NHL) and 6 patients with various granulomatous diseases were examined for their capacity to produce lymphokines in vitro. Lymphocytes from 16 healthy individuals served as controls. To induce interleukin-2 (IL-2) and gamma-interferon (gamma-IFN) production lymphocytes were stimulated for 24 hrs with ConA or PHA or for 72 hrs with ConA + PMA + urea. alpha-IFN production was induced with Herpes simplex virus I (HSV) and the mycopla

IL-23 and monocyte interactions redirect human Th17.1 cells to an IL-17AhighIFN-γhigh potentially pathogenic profile.

INTRODUCTION: IL-23-targeted therapies have demonstrated clinical efficacy in psoriatic arthritis (PsA), primarily through the reduction of IL-17A-driven inflammation. Consequently, most studies have focused on classical, high IL-17A-producing, CCR6+CXCR3-CCR4+ Th17 cells. In contrast, the impact of IL‑23 signaling on CCR6+CXCR3+CCR4- Th17.1 cells -traditionally characterized by high IFN-γ and low IL-17A production-remains largely unexplored, despite their recognized pathogenicity in autoimmune disease. The aim of this study was to characterize the effects of IL-23 on human memory Th17.1 cells

Nuclear export block of tyrosine-phosphorylated STAT1 facilitates rapid responsiveness to interferon-γ stimuli at the expense of signal strength.

Interferon-induced STAT1 (signal transducer and activator of transcription 1) signal transduction constitutes a dynamic network of concurrent activation and inactivation steps that directly transmits extracellular information from the plasma membrane to the nucleus. To elucidate how nuclear retention of tyrosine-phosphorylated STAT1 as a fundamental principle of the STAT1 network directs signal transduction, we have developed a mathematical rate equation model for STAT1 nucleocytoplasmic shuttling. Based on an interplay between experimental data on the behavior of transport variants with alter

Challenges of monocyte HLA-DR targeted immunomodulation in sepsis-a prospective observational cohort study.

INTRODUCTION: Reduced monocyte HLA-DR expression, a hallmark of immunosuppression in sepsis, is associated with infectious complications and mortality. Therapeutic strategies, including IFN-γ, have been used to restore monocyte HLA-DR and immune function, but have not consistently improved clinical outcomes. Therefore, we conducted an iterative series of experiments to re-examine the core assumptions and address the key gaps in the current understanding. METHODS: We conducted a prospective cohort study of patients admitted to the intensive care unit (ICU) with sepsis (n = 55, 36% mortality) t

Disease-associated microglia adopt stage-specific phenotypes that regulate T cell fate and immunity in glioma.

Malignant gliomas are lethal brain tumors characterized by profound local immunosuppression and a radically remodeled myeloid landscape. Although these tumors mobilize resident microglia and infiltrating monocyte-derived macrophages, the mechanisms governing their phenotypic convergence and diversification remain elusive. Here, we integrated single-cell profiling and spatial transcriptomics of glioma-associated microglia in the GL261 model. We identified distinct microglial states that aligned with tumor architecture, most notably Cst7-expressing disease-associated microglia (DAMs) that aggreg

Molecular Insights Into TNF-α and IFN-γ-Induced Epithelial Cell Death in Toxic Epidermal Necrolysis.

Rapid and extensive epithelial cell death in skin and mucous membranes is the hallmark of Toxic epidermal necrolysis (TEN). High levels of TNF-α and IFN-γ have been detected in the blister fluids of skin lesions in TEN patients. Available literature shows that TNF-α synergy with IFN-γ (TNF-α/IFN-γ) elicited cell death in diverse cells. Thus, the cytotoxic effect and underlying mechanism of TNF-α and IFN-γ on HaCaT cells were investigated. Confluent HaCaT cells were subjected to serial doses of either TNF-α alone, IFN-γ alone, or combination TNF-α with IFN-γ (TNF-α/IFN-γ) for indicated time int

IFN-γ-induced trained immunity enhances killing of priority pathogens in healthy and genetically vulnerable individuals.

Infectious diseases remain a global health challenge, driven by increasing antimicrobial resistance and the threat of emerging epidemics. Mycobacterium tuberculosis and Staphylococcus aureus are leading causes of mortality worldwide. Trained immunity - a form of innate immune memory - offers a promising approach to enhance pathogen clearance. Here, we demonstrate that IFN-γ induces trained immunity in human monocytes through a mechanism involving mTORC1 activation, glutaminolysis, and epigenetic remodeling. Macrophages derived from IFN-γ-trained monocytes exhibited increased glycolytic activit

Fish-derived lipids directly stimulate NK cell activity and IFN-γ synthesis: A novel dietary intervention strategy to enhance antiviral immunity in humans.

Viral infections are a leading cause of morbidity and mortality worldwide. Clinical studies suggest that a diet rich in marine fatty acids present in fish-derived oil (FDO) may improve antiviral immunity. However, there is limited evidence for FDO antiviral properties, and which components should be considered when designing such a medical food. In this study designed as a clinical trial on healthy volunteers, we demonstrated a direct effect of FDOs on enhancing NK cell cytotoxicity and increase in IFN-γ synthesis, the main components of antiviral immunity. This effect was related to the prese

Immunologic investigations into transgene directed immune-mediated myositis following delandistrogene moxeparvovec gene therapy.

Delandistrogene moxeparvovec is an rAAVrh74 vector-based gene transfer therapy that delivers a transgene encoding delandistrogene moxeparvovec micro-dystrophin, an engineered, functional form of dystrophin shown to stabilize or slow disease progression in DMD. It is approved in the US and in other select countries. Two serious adverse event cases of immune-mediated myositis (IMM) were reported in the phase Ib ENDEAVOR trial (NCT04626674). We hypothesized that immune responses to the micro-dystrophin transgene product may have mediated these IMM events. An interferon-gamma ELISpot assay was use

Diagnostic tests for tuberculosis infection and predictive indicators of disease progression: Utilizing host and pathogen biomarkers to enhance the tuberculosis elimination strategies.

Tuberculosis (TB) remains the leading cause of death worldwide from a single infectious disease. An estimated quarter of the world's population, about 2 billion people, has an immune response to Mycobacterium tuberculosis (Mtb) without clinical, microbiological, or radiological signs of TB disease. This condition is known as TB infection (TBI) and carries a lifelong risk of reactivation with 5%-10% of individuals eventually developing TB disease during their lifetime. Interferon-γ release assay and skin-tests are World Health Organization (WHO)-approved tests for TBI diagnosis and allow to ide
