Immunogenicity and efficacy of a bivalent DNA vaccine containing LeIF and TSA genes against murine cutaneous leishmaniasis.

There is no effective vaccine for the prevention and elimination of leishmaniasis. For this reason, we assessed the protective effects of DNA vaccines containing LeIF, TSA genes alone, or LeIF-TSA fusion against cutaneous leishmaniasis pEGFP-N1 plasmid (empty vector) and phosphate buffer saline (PBS) were used as control groups. Therefore, cellular and humoral immune responses were evaluated before and after the challenge with Leishmania major. Lesion diameter was also measured 3-12 weeks after challenge. All immunized mice with plasmid DNA encoding Leishmania antigens induced the partial immu

The re-emergence of American visceral leishmaniasis in an old focus in Venezuela. II. Vectors and parasites.

As part of an epidemiological study in an old focus of American Visceral Leishmaniasis (AVL) in Venezuela (Guayabita, Aragua State), a longitudinal entomological survey (January 1993-June 1994) was carried out. A total of 3,239 males and 6,043 females belonging to 11 phlebotomine sandfly species were collected. The two recognised vectors of AVL in the New World, Lutzomyia evansi and Lu. longipalpis were found to be sympatric. Lutzomyia evansi was the dominant species (86.4%), almost ten fold times more abundant than Lu. longipalpis (10.6%). The two species alternated seasonally: Lu evansi peak

PKDL and other dermal lesions in HIV co-infected patients with Leishmaniasis: review of clinical presentation in relation to immune responses.

BACKGROUND: Co-infection of leishmaniasis and HIV is increasingly reported. The clinical presentation of leishmaniasis is determined by the host immune response to the parasite; as a consequence, this presentation will be influenced by HIV-induced immunosuppression. As leishmaniasis commonly affects the skin, increasing immunosuppression changes the clinical presentation, such as in post-kala-azar dermal leishmaniasis (PKDL) and cutaneous leishmaniasis (CL); dermal lesions are also commonly reported in visceral leishmaniasis (VL) and HIV co-infection. METHODS: We reviewed the literature with

Antibodies to basement membrane protein nidogen in Chagas' disease and American cutaneous leishmaniasis.

About 50 to 70% of sera from patients with American cutaneous leishmaniasis and chronic Chagas' disease possessed antibodies which reacted in enzyme and radioimmunoassays with nidogen obtained from a tumor basement membrane. The antibodies were of the immunoglobulin M and G classes in acute American cutaneous leishmaniasis but mainly of the immunoglobulin G class in chronic Chagas' disease. Similar antibodies could not be detected in patients suffering from a variety of other infectious or inflammatory diseases when compared with healthy control groups. Inhibition and immunoadsorption studies

In Vivo Bioluminescent Monitoring of Parasites in BALB/c Mouse Models of Cutaneous Leishmaniasis Drug Discovery.

Confirming the in vivo efficacy of potential antileishmanial compounds that display in vitro potency and good chemical characteristics is one of the most important steps in preclinical research drug discovery before human clinical trials begin. Here we describe the use of the in vivo bioluminescent monitoring of high and low inocula of luciferase-expressing Leishmania major (L. major) parasites in traditional and more innovative rodent models of in vivo cutaneous leishmaniasis (CL) drug discovery.

Efficacy of Apigenin and Miltefosine Combination Therapy against Experimental Cutaneous Leishmaniasis.

Leishmaniasis is a neglected tropical disease caused by several different species of Leishmania. Treatment of leishmaniasis involves a limited drug arsenal that is associated with severe side effects, high costs, and drug resistance. Therefore, combination therapy has emerged as a strategy to improve leishmaniasis treatment. Here, we report the interaction of miltefosine and apigenin in vitro and in vivo. Combination therapy using low doses of these two drugs results in good clinical and parasitological responses.

Paediatric visceral leishmaniasis in Italy: a 'One Health' approach is needed.

Here we describe a case of paediatric visceral leishmaniasis recorded in an infant initially suspected for acute lymphoblastic leukaemia due to the clinical and haematological presentation. Eventually the patient was found positive for Leishmania infantum infection and successfully treated. This case emphasises how pivotal a 'One Health' approach is for diagnosing this zoonotic disease; highlighting the importance of including Visceral Leishmaniasis in the differential diagnosis of leukaemia-like syndromes in infants travelling to, and living in, the Mediterranean region.

A case report of visceral leishmaniasis in the Tharaka District, Kenya.

Leishmaniasis is a parasitic disease that is transmitted through a bite of some species of sandflies. It is caused by obliged intra-cellular protozoa of the genus Leishmania and is responsible for a broad spectrum of clinical syndromes: cutaneous leishmaniasis (CL), visceral leishmaniasis (VL) and mucocutaneous leishmaniasis (ML). The visceral disease (classically known as "kala azar") is the most aggressive form and if undervalued is fatal. Here we describe the first case of visceral leishmaniasis in the Tharaka District reported in literature.

Immunologic memory in cutaneous leishmaniasis.

Leishmania major infections induce solid immunity to reinfection. Experimental studies in mice indicate that the CD4+ T cells responsible for this immunity include two populations: parasite-dependent T effector cells and parasite-independent central memory T (Tcm) cells. While there currently is no vaccine for leishmaniasis, the existence of a long-lived population of Tcm cells that does not require the continued presence of live parasites suggests that a vaccine that expands these cells might be efficacious.

Mucocutaneous leishmaniasis in a patient with the human immunodeficiency virus.

We report a case of mucocutaneous leishmaniasis (MCL) in a patient with the human immunodeficiency virus (HIV), Centers for Disease Control (CDC) Stage A2, with no previous history of cutaneous or systemic leishmaniasis. The patient had not travelled outside the province of Málaga, on the Mediterranean coast of southern Spain, so that it concerns an indigenous case, extremely unusual in this area. The HIV infection may well have influenced the defence against leishmania, but the exact mechanism by which this occurred is unknown.

[Visceral leishmaniasis in children in the province of Florence].

Three cases of visceral leishmaniasis are presented: two children who got the disease in Florence and the imported case of a girl coming from Albania with her disease in act. The diagnosis was made showing Leishmania in bone marrow specimen. Therapy with melglumine antimonate was effective and well borne, leading the three children to a complete healing. In the province of Florence visceral leishmaniasis is very rare, but such protozoa and the sand flies are present as shown by the high number of dog with leishmaniasis.

Visceral leishmaniasis in the hoary zorro Dusicyon vetulus: a case of mistaken identity.

The historical identification of the Brazilian 'north-eastern' zorro as Dusicyon vetulus is questioned in relation to its incrimination as a reservoir of Leishmania chagasi, the agent of American visceral leishmaniasis. Comparative cranial and dental morphology showed that specimens of this north-eastern species more closely resemble the crab-eating zorro Cerdocyon thous, conforming with the documented geographical ranges of the respective species. We conclude that the single 'wild' canid host of L. chagasi in the neotropics in C. thous.

[Visceral leishmaniasis and HIV infection: the diagnostic and therapeutic problems].

We present the case of an HIV-seropositive drug addict patient whose seropositivity was unknown until admission to hospital, where he presented with a clinical picture of fever splenomegaly and weight loss. A diagnosis of visceral leishmaniasis was established. We stress the diagnostic difficulties well expressed in the need to repeat the bone marrow aspirate to detect the presence of leishmanias. The lack of response to conventional antimonial therapy is discussed as well as the role of pentamidine as an alternative therapy.

Visceral Leishmaniasis-Associated Hemophagocytic Lymphohistiocytosis in a Patient With Crohn's Disease in Therapy With Anti-TNFα: A Challenging Diagnostic Case Report.

BACKGROUND: Hemophagocytic lymphohistiocytosis (HLH) is a rare yet life-threatening hyperinflammatory syndrome, increasingly reported in patients with inflammatory bowel disease (IBD) receiving biologic therapy. While Epstein-Barr virus and cytomegalovirus represent the most frequent infectious triggers, visceral leishmaniasis (VL) is extremely uncommon and may be easily ignored outside endemic areas. CASE PRESENTATION: We report the case of a 31-year-old man with Crohn's disease treated with escalated anti-TNFα therapy who presented with persistent fever, pancytopenia, hyperferritinemia, hyp

Case Reports of Visceral Leishmaniasis-Associated Hemophagocytic Lymphohistiocytosis in Adults: A Complex Immune Phenomenon.

Background: Visceral Leishmaniasis (VL), a severe systemic parasitic disease caused by Leishmania species, can be complicated by secondary Hemophagocytic Lymphohistiocytosis (HLH). HLH is a life-threatening hyperinflammatory syndrome characterized by excessive immune activation that results in multiorgan dysfunction. The co-occurrence of VL and HLH in adults is a rare but critical diagnostic and therapeutic challenge, often leading to fatal outcomes if treatment is delayed. Case Presentation: We present two cases of adult males (60 and 72 years old) from Greece, an endemic area for L. infantum

Enhanced Translational Activity Is Linked to Lymphatic Endothelial Cell Activation in Cutaneous Leishmaniasis.

Introduction: Cutaneous leishmaniasis (CL) is a significant public health problem leading to permanently disfiguring skin lesions caused by Leishmania parasites. Lesion severity stems from an excessive host inflammatory response that prevents healing. Materials and Methods: Here, we characterized the transcriptional and translational responses of lymphatic endothelial cells (LECs) during murine CL using single-cell RNA sequencing data combined with flow cytometry and in vivo puromycin incorporation to assess translational activity. Results: We identified upregulation of antigen presentation pa
