Ghrelin, not corticosterone, is associated with transitioning of phenotypic states in a migratory Galliform.

In both captive and free-living birds, the emergence of the migratory phenotype is signalled by rapid and marked increases in food intake and fuelling, as well as changes in amount of nocturnality or migratory restlessness. The metabolic hormone corticosterone and, as more recently suggested, the gut-derived hormone ghrelin have been suggested to play a role in mediating such phenomenal phenotypic flexibility given that they both regulate fuel metabolism and locomotion across vertebrate taxa. Here, using the Common quail (Coturnix coturnix) as our study species, we induced autumn migration fol

The G protein biased serotonin 5-HT2A receptor agonist lisuride exerts anti-depressant drug-like activities in mice.

There is now evidence from multiple Phase II clinical trials that psychedelic drugs can exert long-lasting anxiolytic, anti-depressant, and anti-drug abuse (nicotine and ethanol) effects in patients. Despite these benefits, the hallucinogenic actions of these drugs at the serotonin 2A receptor (5-HT2AR) limit their clinical use in diverse settings. Activation of the 5-HT2AR can stimulate both G protein and β-arrestin (βArr) -mediated signaling. Lisuride is a G protein biased agonist at the 5-HT2AR and, unlike the structurally-related lysergic acid diethylamide (LSD), the drug does not typicall

Network pharmacology and molecular docking-based analysis of protective mechanism of MLIF in ischemic stroke.

OBJECTIVE: This study aimed to evaluate the potential mechanism by which Monocyte locomotion inhibitory factor (MLIF) improves the outcome of ischemic stroke (IS) inflammatory injury. METHODS: Potential MLIF-related targets were predicted using Swiss TargetPrediction and PharmMapper, while IS-related targets were found from GeneCards, PharmGKB, and Therapeutic Target Database (TTD). After obtaining the intersection from these two datasets, the Search Tool for Retrieval of Interacting Genes/Protein (STRING11.0) database was used to analyze the protein-protein interaction (PPI) network of the i

Exploration and validation of metastasis-associated genes for skin cutaneous melanoma.

Skin cutaneous melanoma is a malignant and highly metastatic skin tumor, and its morbidity and mortality are still rising worldwide. However, the molecular mechanisms that promote melanoma metastasis are unclear. Two datasets (GSE15605 and GSE46517) were retrieved to identify the differentially expressed genes (DEGs), including 23 normal skin tissues (N), 77 primary melanoma tissues (T) and 85 metastatic melanoma tissues (M). Gene ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis were performed to explore the functions of the DEGs. We constructed protein-protein interact

S-adenosyl-l-methionine antidepressant-like effects involve activation of 5-HT1A receptors.

S-adenosyl-l-methionine (SAMe), a methyl donor, induces antidepressant effects in preclinical and clinical studies of depression. However, the mechanisms behind these effects have been poorly investigated. Since SAMe is involved in monoamine metabolism, this work aimed at 1) testing the effects induced by systemic treatment with SAMe in mice submitted to the forced swimming test (FST) and tail suspension test (TST); 2) investigating the involvement of serotonergic neurotransmission in the behavioral effects induced by SAMe. To do that, male Swiss mice received systemic injections (1 injection/

How suture networks improve the protective function of natural structures: A multiscale investigation.

Myriad natural protective structures consist of bone plates joined by convoluted unmineralized (soft) collagenous sutures. Examples of such protective structures include: shells of turtles, craniums of almost all animals (including humans), alligator armour, armadillo armour, and others. The function of sutures has been well researched. However, whether, and if so how, sutures improve protective performance during a predator attack has received limited attention. Sutures are ubiquitous in protective structures, and this motivates the question as to whether sutures optimize the protective funct

Predictive equations to estimate peak aerobic capacity and peak heart rate in persons with Down syndrome.

Estimations based on the available equations for predicting oxygen uptake (V̇o2) from treadmill speed of locomotion are not appropriate for individuals with Down syndrome (DS). We aimed at developing prediction models for peak absolute oxygen uptake (V̇o2peak) and peak heart rate (HRpeak) based on retrospective data from a healthy population with and without Down syndrome (DS). A cross-sectional analysis of V̇o2peak and HRpeak was conducted in 196 and 187 persons with and without DS, respectively, aged from 16 to 45 yr. Nonexercise data alone versus combined with HRpeak were used to develop eq

Pharmacological Prevention of Neonatal Opioid Withdrawal in a Pregnant Guinea Pig Model.

Newborns exposed to prenatal opioids often experience intense postnatal withdrawal after cessation of the opioid, called neonatal opioid withdrawal syndrome (NOWS), with limited pre- and postnatal therapeutic options available. In a prior study in pregnant mice we demonstrated that the peripherally selective opioid antagonist, 6β-naltrexol (6BN), is a promising drug candidate for preventive prenatal treatment of NOWS, and a therapeutic mechanism was proposed based on preferential delivery of 6BN to fetal brain with relative exclusion from maternal brain. Here, we have developed methadone (MTD)

Linear elastic and hyperelastic studies of equine hoof mechanical response at different hydration levels.

Most simulation studies on equine hoof biomechanics employed linear elastic (LE) material models. However, the equine hoof wall's stress-strain relationship is nonlinear and varies with hydration level. Therefore, it is essential to investigate the accuracy of the LE model compared to more advanced material models, such as hyperelastic (HE) or viscoelastic models. The current research investigated performances of LE and three HE models (Mooney-Rivlin, Neo-Hookean, and Marlow) in describing equine hoof's mechanical behavior using finite element (FE) analysis. In the first attempt, a rectangular

Effects of lower body positive pressure treadmill on functional improvement in knee osteoarthritis: A randomized clinical trial study.

BACKGROUND: Knee joint pain and stiffness are the two main symptoms of knee osteoarthritis (OA) and thus restrict a patient's activities, such as walking and walking up and downstairs. The lower body positive pressure (LBPP) treadmill as one of the emerging body weight support system devices brings new hope for exercise-related rehabilitation for knee OA patients. AIM: To investigate the biomechanical effects and the subjective clinical assessment of LBPP treadmill walking exercise when compared with conventional therapy in mild to moderate knee OA patients. METHODS: Eighteen patients with m

Neural Correlates of Motor Imagery of Gait in Amyotrophic Lateral Sclerosis.

BACKGROUND: Gait impairment is poorly characterized in amyotrophic lateral sclerosis (ALS), despite increasing evidence of extrapyramidal and cerebellar dysfunction. Gait impairment adds to the considerable motor disability of ALS patients and requires targeted multidisciplinary interventions. PURPOSE: To assess gait imagery-specific networks and functional adaptation in ALS. STUDY TYPE: Prospective. POPULATION: Seventeen ALS patients with lower motor neuron predominant (LMNp) disability, 14 patients with upper motor neurons predominant (UMNp) disease, and 14 healthy controls were included.

Human plantar fascial dimensions and shear wave velocity change in vivo as a function of ankle and metatarsophalangeal joint positions.

The plantar fascia (PF), a primary contributor of the foot arch elasticity, may experience slack, taut, and stretched states depending on the ankle and metatarsophalangeal (MTP) joint positions. Since PF has proximodistal site difference in its dimensions and stiffness, the response to applied tension can also be site specific. Furthermore, PF can contribute to supporting the foot arch while being stretched beyond the slack length, but it has never been quantitatively evaluated in vivo. This study investigated the effects of the ankle and MTP joint positions on PF length and localized thicknes

Acrophobia and visual height intolerance: advances in epidemiology and mechanisms.

Historical descriptions of fear at heights date back to Chinese and Roman antiquity. Current definitions distinguish between three different states of responses to height exposure: a physiological height imbalance that results from an impaired visual control of balance, a more or less distressing visual height intolerance, and acrophobia at the severest end of the spectrum. Epidemiological studies revealed a lifetime prevalence of visual height intolerance including acrophobia in 28% of adults (32% in women; 25% in men) and 34% among prepubertal children aged 8-10 years without gender preponde

Loss of Piccolo Function in Rats Induces Cerebellar Network Dysfunction and Pontocerebellar Hypoplasia Type 3-like Phenotypes.

Piccolo, a presynaptic active zone protein, is best known for its role in the regulated assembly and function of vertebrate synapses. Genetic studies suggest a further link to several psychiatric disorders as well as Pontocerebellar Hypoplasia type 3 (PCH3). We have characterized recently generated Piccolo KO (Pclogt/gt ) rats. Analysis of rats of both sexes revealed a dramatic reduction in brain size compared with WT (Pclowt/wt ) animals, attributed to a decrease in the size of the cerebral cortical, cerebellar, and pontine regions. Analysis of the cerebellum and brainstem revealed a reduced

The changes in systemic monocytes in humans undergoing surgical decompression for degenerative cervical myelopathy may influence clinical neurological recovery.

BACKGROUND: Degenerative cervical myelopathy (DCM) is the most common cause of non-traumatic spinal cord injury worldwide. Surgical decompression is recommended as the preferred treatment strategy for DCM as it halts disease progression and improves neurologic symptoms. We previously demonstrated that neuroinflammation, including monocytes, plays a critical role in the pathobiology of DCM and in ischemic-reperfusion injury (IRI) following surgical decompression. Monocytes are able to enter the spinal cord and brain tissues due to damage to the blood spinal cord and blood brain barrier followin

Mild Neonatal Brain Hypoxia-Ischemia in Very Immature Rats Causes Long-Term Behavioral and Cerebellar Abnormalities at Adulthood.

UNLABELLED: Systemic hypoxia-ischemia (HI) often occurs during preterm birth in human. HI induces injuries to hinder brain cells mainly in the ipsilateral forebrain structures. Such HI injuries may cause lifelong disturbances in the distant regions, such as the contralateral side of the cerebellum. We aimed to evaluate behavior associated with the cerebellum, to acquire cerebellar abundant metabolic alterations using in vivo 1H magnetic resonance spectroscopy (1H MRS), and to determine GFAP, NeuN, and MBP protein expression in the left cerebellum, in adult rats after mild early postnatal HI on

Desktop VR Is Better Than Non-ambulatory HMD VR for Spatial Learning.

Use of virtual reality (VR) technology is proliferating for designing and upgrading entertainment devices, and creating virtual environments that could be used for research and training. VR is becoming a strong research tool by providing a tighter control on the experimental environment and by allowing almost limitless possibilities of creating ecologically valid stimuli. However, the enhanced fidelity between the real and virtual worlds that VR provides does not always benefit human performance. For a better understanding, and increasing VR's usability, we need to identify the relevant consti

Retention and generalizability of balance recovery response adaptations from trip perturbations across the adult life span.

For human locomotion, varying environments require adjustments of the motor system. We asked whether age affects gait balance recovery adaptation, its retention over months, and the transfer of adaptation to an untrained reactive balance task. Healthy adults (26 young, 27 middle-aged, and 25 older; average ages 24, 52, and 72 yr, respectively) completed two tasks. The primary task involved treadmill walking: either unperturbed (control; n = 39) or subject to unexpected trip perturbations (training; n = 39). A single trip perturbation was repeated after a 14-wk retention period. The secondary t

Ascending Lipopolysaccharide-Induced Intrauterine Inflammation in Near-Term Rabbits Leading to Newborn Neurobehavioral Deficits.

BACKGROUND: Chorioamnionitis from ascending bacterial infection through the endocervix is a potential risk factor for cerebral palsy. Tetrahydrobiopterin, an essential cofactor for nitric oxide synthase (NOS) and amino acid hydroxylases, when augmented in the fetal brain, prevents some of the cerebral palsy-like deficits in a rabbit hypoxia-ischemia model. OBJECTIVES: To study the effect of lipopolysaccharide (LPS)-induced intrauterine inflammation in preterm gestation on motor deficits in the newborn, and whether biosynthesis of tetrahydrobiopterin or inflammatory mediators is affected in th

Visual feedback during pedaling allows individuals poststroke to alter inappropriately prolonged paretic vastus medialis activity.

Individuals who have experienced a stroke often demonstrate inappropriate muscle activity phasing in the paretic leg during locomotion. Past research has demonstrated that inappropriate paretic phasing varies between behavioral contexts and is reduced during unilateral pedaling with the nonparetic leg inactive. We investigated whether individuals could voluntarily alter activity in a target muscle of the paretic limb in a consistent behavioral context and whether this voluntary change differed between bilateral and unilateral pedaling. During a fixed-speed motorized pedaling task, participants
