Study on diagnostic models for insomnia and gastralgia with Liver-Spleen Disharmony Syndrome based on machine learning.

BACKGROUND: The prevalence of insomnia and gastralgia has recently increased. Traditional Chinese Medicine (TCM) offers unique advantages in treating these conditions. Accurate syndrome differentiation is essential for effective treatment. However, as a common syndrome in TCM for these conditions, the diagnostic criteria for Liver-Spleen Disharmony Syndrome (LSDS) currently lack uniformity. OBJECTIVE: To develop a machine learning-based diagnostic model for LSDS in patients with insomnia and/or gastralgia. METHODS: In this prospective observational study, clinical data from 575 patients with

BACKGROUND: Syphilis is a sexually transmitted disease (STD) caused by Treponema pallidum (TP) which could progress to neurosyphilis affecting the nervous system. However, the pathogenesis of neurosyphilis remain unknown. In addition, current diagnosis of neurosyphilis is mainly based on serological tests and cerebrospinal fluid (CSF) analysis, with limitations in terms of sensitivity and specificity. METHODS: From April 2021 to April 2023, 129 patients with syphilis were enrolled and divided into the non-neurosyphilis group (common syphilis, CS, n=19), asymptomatic neurosyphilis group (AN, n

Urinary microRNAs for the non-invasive diagnosis of endometriosis identified by next-generation sequencing and machine learning.

BACKGROUND: Endometriosis is a chronic gynecological disease associated with pain, infertility, and delayed diagnosis. Non-invasive biomarkers are urgently needed to facilitate earlier detection and reduce the reliance on diagnostic laparoscopy. MicroRNAs (miRNAs) are stable in body fluids and hold promise as diagnostic tools. METHODS: In this pilot study, urine samples from 34 patients with histologically confirmed endometriosis and 18 control patients (laparoscopically confirmed absence of disease) were analyzed using next-generation miRNA sequencing. Differential expression analysis was pe

A network toxicology and machine learning approach to uncover the molecular machinery of bisphenol A-induced colorectal cancer.

The carcinogenic relevance of environmental contaminant bisphenol A (BPA) to colorectal cancer (CRC) has gained growing attention, yet the molecular networks potentially linking BPA exposure to CRC remain incompletely characterized. This study integrated network toxicology and machine learning to predict candidate molecular targets and putative signaling networks associated with BPA-correlated colorectal carcinogenesis. The study's methodology involved an initial differential expression screening across several CRC transcriptomic datasets to establish a disease-specific gene signature. Subsequ

Machine Learning Prediction of Local Failure in GI Brain Metastases on the Day of Gamma Knife Radiosurgery.

BACKGROUND: Time to local failure after stereotactic radiosurgery (SRS), including Gamma Knife radiosurgery (GKRS), for gastrointestinal (GI) brain metastases is difficult to predict and may vary substantially across tumors and patients. We developed and internally evaluated a survival machine learning framework to estimate tumor-specific time to local failure using pre-SRS features. METHODS: We performed a retrospective study of GI brain metastases treated with GKRS. Local failure was modeled as a time-to-event outcome under right censoring. A Random Survival Forest survival model was traine

Identification of patients at risk for pancreatic cancer in a 3-year timeframe based on machine learning algorithms.

Early detection of pancreatic cancer (PC) remains challenging largely due to the low population incidence and few known risk factors. However, screening in at-risk populations and detection of early cancer has the potential to significantly alter survival. In this study, we aim to develop a predictive model to identify patients at risk for developing new-onset PC at two and a half to three year time frame. We used the Electronic Health Records (EHR) of a large medical system from 2000 to 2021 (N = 537,410). The EHR data analyzed in this work consists of patients' demographic information, diagn

Identification of progression-related genes and construction of prognostic model for chronic kidney disease by machine learning.

BACKGROUND: Early diagnosis and intervention for chronic kidney disease (CKD) can significantly improve patient's quality of life and prognosis. Besides routine laboratory indicators and medical history, risk prediction models can predict CKD outcome. However, there is currently a lack of CKD prognostic prediction models based on transcriptomics and machine learning. METHODS: Utilizing weighted correlation network analysis (WGCNA) and random forest algorithms in GSE137570, three core gene sets of different sizes were constructed, which were externally validated in GSE66494 and GSE180394, and
