Distribution of extracellular matrix receptors in various forms of glomerulonephritis.

Integrins are heterodimeric transmembrane receptor glycoproteins consisting of alpha and beta subunits that mediate adhesion and interactions between cells and extracellular matrix. Such interactions may be perturbed in various pathologic states, resulting in the altered phenotypic expressions of the integrins in affected tissues. To ascertain the alterations in integrins in various renal diseases, their distribution was investigated in different forms of glomerulonephritis by indirect immunofluorescence and immunoelectron microscopy using specific antibodies directed against beta 1 integrins

A CotB1p-mediated one-step immobilization strategy for constructing GPCR affinity chromatography to screen multi-target natural product ligands.

G protein-coupled receptors (GPCRs) are critical targets for drug discovery, yet the screening of GPCR-binding ligands from complex natural products remains challenging due to the lack of mild and universal immobilization strategies for these fragile membrane proteins. Here, we reported CotB1p-mediated one-step immobilization strategy for the construction of GPCR affinity chromatography columns to screen natural product ligands. Three asthma-related GPCRs β2-adrenergic receptor (β2AR), muscarinic M3 receptor (M3R), and cysteinyl leukotriene receptor 1 (CysLT1R) were genetically fused with the

Near-complete phosphorus valorization from sewage sludge: Cost-effective co-production of vivianite and bioavailable detoxified hydrochar via a recyclable citric acid-mediated hydrothermal process.

Efficient phosphorus recovery from sewage sludge is bottlenecked by stable phosphorus-binding within complex organic-inorganic matrices. Conventional hydrothermal treatments suffer from limited P dissolution, inevitable re-fixation, poor hydrochar P-bioavailability, and heavy metal (HM) risks. This work proposes a novel, recyclable citric acid (CA)-mediated hydrothermal process. Relying on strong CA-mediated coordination and its synergistic effects with hydrothermal conditions and sludge components, this process achieves efficient phosphorus release and targeted speciation control while mitiga

[Influence of ecosystem type on the formation mechanisms of iron oxide-associated organic carbon].

Soil is the largest active carbon reservoir in terrestrial ecosystems. The persistence of soil organic carbon (SOC) plays a central role in regulating global carbon cycle and climate change. Iron oxides, with their unique chemical properties, are among the most important mineral phases controlling SOC stabilization. We synthesized current understanding of the mechanisms and drivers of SOC stabilization mediated by iron oxides. We highlighted the association patterns between iron oxides and organic carbon (Fe-OC) across forests, grasslands, and croplands, and explored how biotic and abiotic fac

Fe(III)-mediated PFAS phototransformation under UV222 irradiation: LMCT initiation chemistry and performance in electroplating wastewater.

Per- and polyfluoroalkyl substances (PFAS) are highly resistant to photochemical degradation due to their weak UV absorption and strong C-F bonds. Fe(III) chelation can enhance PFAS photoreactivity via ligand-to-metal charge transfer (LMCT), yet the wavelength/PFAS structure dependence and underlying mechanisms under environmentally relevant conditions remain unresolved. Here, Fe(III)-mediated photochemical transformation of perfluorooctanoic acid (PFOA, 0.1 μM) was investigated under monochromatic irradiation from 222 to 405 nm. Electrochemical measurement indicates strong Fe(III)-PFOA comple

Plant-Derived Senotherapeutics in Cellular Senescence: A Scoping Review of Preclinical Evidence, Mechanistic Pathways, and Metabolomic-Guided Discovery.

Senotherapeutic agents targeting senescent cell (SnC) accumulation represent a promising frontier in aging research. These agents encompass senolytics that selectively eliminate accumulated SnCs and senomorphics that suppress the pathological persistence of the senescence-associated secretory phenotype (SASP). Concerns regarding off-target effects of synthetic senolytics have intensified interest in plant-derived alternatives that offer multitargeted mechanisms and favorable safety profiles. This scoping review was conducted following Joanna Briggs Institute guidelines and PRISMA-ScR, mapped p

A recombinant IgG1 Fc-domain protein ameliorates inflammatory demyelinating peripheral neuropathy.

INTRODUCTION: Guillain-Barré syndrome (GBS) and chronic inflammatory demyelinating polyneuropathy (CIDP) are immune-mediated inflammatory neuropathies characterized by T cell- and/or autoantibody-driven injury to peripheral nerves. Although intravenous immunoglobulin (IVIg) is effective in both conditions, its use is limited by high dosing requirements, supply constraints, and manufacturing complexity. NVG-2089 is a recombinant IgG1 Fc-domain protein engineered to selectively agonize type II Fc receptors, promoting anti-inflammatory signaling through expansion of regulatory T cells (Tregs) and

Plasma-Driven Monocyte Dysfunction Drives Disease Progression in Alcohol-Related Acute-on-Chronic Liver Failure.

INTRODUCTION: Acute-on-chronic liver failure (ACLF) is associated with immune dysregulation, characterized by simultaneous inflammation and immune suppression. Alcohol-related ACLF (A-ACLF) has particularly high mortality, yet the role of monocyte dysfunction and plasma-derived inflammatory mediators remains poorly understood. METHODS: A prospective study was conducted from July 2019 to May 2020, recruiting A-ACLF patients and healthy controls. Monocyte phenotyping and serum immune markers (sCD163, sCD25, ferritin, and IgG immune complexes) were measured. Ex vivo plasma priming experiments we

Transcranial ultrasound combined with intravenous metformin-loaded oxygenated microbubbles attenuates noise-induced hearing loss in mice.

Noise-induced hearing loss (NIHL) involves a biphasic pathophysiology. Intense noise exposure causes immediate cochlear vasoconstriction and ischemia, leading to transient hypoxia. Subsequent reperfusion triggers excess reactive oxygen species (ROS) production, resulting in oxidative stress and hair cell injury. This study therefore developed two oxygenated albumin microbubble (OMB) formulations-ionic-bond metformin-coated (iMet-OMBs) and covalent-bond metformin-encapsulated (cMet-OMBs)-and combined them with transcranial ultrasound (US) to enhance targeted delivery to the cochlea. This approa

cis-Dihydroxylation by Synthetic Iron(III)-Peroxo Intermediates and Rieske Dioxygenases: Experimental and Theoretical Approaches Reveal the Key O-O Bond Activation Step.

Dioxygen (O2) activation by iron-containing enzymes and biomimetic compounds generates iron-oxygen intermediates, such as iron-superoxo, -peroxo, -hydroperoxo, and -oxo, that mediate oxidative reactions in biological and abiological systems. Among the iron-oxygen intermediates, iron(III)-peroxo species are less frequently implicated as active intermediates in oxidation reactions. In this study, we present the combined experimental and theoretical investigations on cis-dihydroxylation reactions mediated by synthetic mononuclear nonheme iron-peroxo intermediates, demonstrating the importance of

Association between superior longitudinal fasciculus, motor recovery, and motor outcome after stroke: a cohort study.

INTRODUCTION: Parieto-frontal interactions are mediated by the superior longitudinal fasciculus (SLF) and are crucial to integrate visuomotor information and mediate fine motor control. In this study, we aimed to characterize the relation of white matter integrity of both parts of the SLF (SLF I and SLF II) to both motor outcome and recovery and its evolution over time in stroke patients with upper limb motor deficits. MATERIALS AND METHODS: Fractional anisotropy (FA) values over the SLF I, SLF II, and corticospinal tract (CST) and upper limb motor performance evaluated by both the upper limb

TDP-43 is a ubiquitylation substrate of the SCFcyclin F complex.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterised by the loss of upper and lower motor neurons in the brain and spinal cord. ALS and frontotemporal dementia (FTD) are overlapping diseases with shared pathological features. Affected neurons of people with ALS and FTD typically contain ubiquitin-immunoreactive inclusions, of which TDP-43 (Tar DNA-binding protein of 43 kDa) is a major component. However, what triggers the formation of these abnormal TDP-43 inclusions is unclear. Previously, we identified CCNF mutations in cohorts of familial and sp

Potential Molecular Mechanisms of Rare Anti-Tumor Immune Response by SARS-CoV-2 in Isolated Cases of Lymphomas.

Recently, two cases of complete remission of classical Hodgkin lymphoma (cHL) and follicular lymphoma (FL) after SARS-CoV-2 infection were reported. However, the precise molecular mechanism of this rare event is yet to be understood. Here, we hypothesize a potential anti-tumor immune response of SARS-CoV-2 and based on a computational approach show that: (i) SARS-CoV-2 Spike-RBD may bind to the extracellular domains of CD15, CD27, CD45, and CD152 receptors of cHL or FL and may directly inhibit cell proliferation. (ii) Alternately, upon internalization after binding to these CD molecules, the S

Pain after a motor vehicle crash: The role of socio-demographics, crash characteristics and peri-traumatic stress symptoms.

BACKGROUND: The vast majority of individuals who come to the emergency department (ED) for care after a motor vehicle collision (MVC) are diagnosed with musculoskeletal strain only and are discharged to home. A significant subset of this population will still develop persistent pain and posttraumatic psychological sequelae may play an important role in pain persistence. METHODS: We conducted a multisite longitudinal cohort study of adverse post-traumatic neuropsychiatric sequelae among patients seeking ED treatment in the aftermath of a traumatic life experience. We report on a sub-group of p

RSV Reprograms the CDK9•BRD4 Chromatin Remodeling Complex to Couple Innate Inflammation to Airway Remodeling.

Respiratory syncytial virus infection is responsible for seasonal upper and lower respiratory tract infections worldwide, causing substantial morbidity. Self-inoculation of the virus into the nasopharynx results in epithelial replication and distal spread into the lower respiratory tract. Here, respiratory syncytial virus (RSV) activates sentinel cells important in the host inflammatory response, resulting in epithelial-derived cytokine and interferon (IFN) expression resulting in neutrophilia, whose intensity is associated with disease severity. I will synthesize key findings describing how R

Reduced intracellular chloride concentration impairs angiogenesis by inhibiting oxidative stress-mediated VEGFR2 activation.

Chloride (Cl-) homeostasis is of great significance in cardiovascular system. Serum Cl- level is inversely associated with the mortality of patients with heart failure. Considering the importance of angiogenesis in the progress of heart failure, this study aims to investigate whether and how reduced intracellular Cl- concentration ([Cl-]i) affects angiogenesis. Human umbilical endothelial cells (HUVECs) were treated with normal Cl- medium or low Cl- medium. We showed that reduction of [Cl-]i (from 33.2 to 16.18 mM) inhibited HUVEC proliferation, migration, cytoskeleton reorganization, tube for

HLA Class II-Triggered Signaling Cascades Cause Endothelial Cell Proliferation and Migration: Relevance to Antibody-Mediated Transplant Rejection.

Transplant recipients developing donor-specific HLA class II (HLA-II) Abs are at higher risk for Ab-mediated rejection (AMR) and transplant vasculopathy. To understand how HLA-II Abs cause AMR and transplant vasculopathy, we determined the signaling events triggered in vascular endothelial cells (EC) following Ab ligation of HLA-II molecules. HLA-II expression in EC was induced by adenoviral vector expression of CIITA or by pretreatment with TNF-α/IFN-γ. Ab ligation of class II stimulated EC proliferation and migration. Class II Ab also induced activation of key signaling nodes Src, focal adhe

Preferential association of hepatitis C virus with CD19+ B cells is mediated by complement system.

UNLABELLED: Extrahepatic disease manifestations are common in chronic hepatitis C virus (HCV) infection. The mechanism of HCV-related lymphoproliferative disorders is not fully understood. Recent studies have found that HCV in peripheral blood mononuclear cells from chronically infected patients is mainly associated with cluster of differentiation 19-positive (CD19+ ) B cells. To further elucidate this preferential association of HCV with B cells, we used in vitro cultured virus and uninfected peripheral blood mononuclear cells from healthy blood donors to investigate the necessary serum compo

Pim-1L Protects Cell Surface-Resident ABCA1 From Lysosomal Degradation in Hepatocytes and Thereby Regulates Plasma High-Density Lipoprotein Level.

OBJECTIVE: ATP-binding cassette transporter A1 (ABCA1) exerts an atheroprotective action through the biogenesis of high-density lipoprotein in hepatocytes and prevents the formation of foam cells from macrophages. Controlling ABCA1 is a rational approach to improving atherosclerotic cardiovascular disease. Although much is known about the regulatory mechanism of ABCA1 synthesis, the molecular mechanism underpinning its degradation remains to be clearly described. APPROACH AND RESULTS: ABCA1 possesses potential sites of phosphorylation by serine/threonine-protein kinase Pim-1 (Pim-1). Pim-1 de
