Gas5A, a putative glucanosyltransferase from Botrytis, functions as cell death inducing protein in plants.

The necrotrophic fungus Botrytis cinerea, releases numerous phytotoxic, cell death inducing proteins (CDIPs) during infection. The precise role of these proteins and their molecular function, however, is still unknown. Here, we report on the identification of a previously unknown CDIP, the glucanosyltransferase Gas5A. Functional characterization revealed that the C-terminal 60 aa of Gas5A are sufficient to induce cell death, independent from its putative enzymatic function. Gas5A localization and functional dependence on the receptor-associated kinase suppressor of BIR1-1 (SOBIR1) and the plan

Double functionalized Sindbis E2 glycoprotein modifies Sindbis tropism favouring infection of α5β1⁺ glioblastoma cells.

Effective cancer therapy hinges on the ability to selectively target malignant cells while sparing healthy tissues. For virotherapy this goal can be achieved by genetic engineering the surface glycoproteins of viruses. This approach, though, often leads to loss of functionality of the engineered proteins, in terms of membrane fusion. Here, we modified the E2 protein of Sindbis virus, preserving the ability of transduction by lentivectors pseudotyped by the Sindbis Env proteins. The modifications introduced allowed to alter the efficiency of transduction of glioblastoma cells expressing integri

Insights into microalgae residue-derived protein-polysaccharide complex as a plant-based egg white foam alternative: chemical characteristics and interfacial behaviors.

Spirulina residues contain various active substances, but is often discarded. In this study, Spirulina residues extract (SWE) demonstrated excellent foaming properties, with a foam overrun of 6.91 L/L, surpassing egg white and other plant proteins. SWE is a polysaccharide-protein complex rich in nonpolar amino acids and has a loose secondary structure dominated by β-sheets and turns. Its high-molecular-weight polysaccharides, primarily composed of glucose, rhamnose, and fructose, form a fibrous network around proteins, promoting rapid adsorption at the air-water interface. The polysaccharides

Integrins and tetraspanins mediate CD36-actin coupling and regulate CD36 organization and signaling.

Submembrane cortical actin (CA) plays a large role in regulating the dynamic organization of cell surface receptors, which in turn regulates … 1-induced CD36 signaling through the Src family kinase Fyn. Given their many interactions with transmembrane proteins, tetraspanins and integrins may provide a ubiquitous mechanism for plasma membrane-CA coupling.

Proteomic analysis of exudates reveals the quality of marinated snakehead fish slice as affected by precooling and freezing treatments.

Proteomic analysis of exudates was carried out to reveal the mechanism underlying the quality of marinated snakehead fish slice as affected by precooling and freezing treatments. The slices treated with fast freezing techniques like liquid refrigerant immersion freezing (LRIF) and liquid nitrogen freezing (LNF) demonstrated superior slice quality and protein integrity, which is mainly due to smaller ice crystals, reduced oxidation, and better moisture retention. Precooling also contributed to preserving the quality. Proteomics exhibited that LNF group enhanced DEPs like heme‑oxygenase, heat sh

Exploration of the bioactive compounds and potential mechanisms of fingerroot (Boesenbergia rotunda) as an anti-breast cancer by bioinformatics.

This research explored the active compounds in Boesenbergia rotunda and predicted its anti-breast cancer mechanism. The Boesenbergia rotunda rhizome was extracted using microwave-assisted extraction (MAE) with ethanol solvent, and the active compound was identified using liquid chromatography-high-resolution mass spectrometry (LC-HRMS). Drug-likeness, bioactivity, and membrane permeability were used to select active compounds with high potential for anti-breast cancer effects. Molecular docking and dynamic simulations were performed to study the interactions between active compounds and target

The SH protein of mumps virus is a druggable pentameric viroporin.

Viral infections are on the rise and drugs targeting viral proteins are needed. Viroporins constitute a growing group of virus-encoded transmembrane oligomeric proteins that allow passage of small molecules across the membrane. Despite sparsity in viroporin structures, recent work has revealed diversity in both the number of transmembrane helices and oligomeric states. Here, we provide evidence that the small hydrophobic protein (SH) from mumps virus is a pentameric viroporin. From extensive biophysical data, a HADDOCK model of full-length SH shows its intracellular C-terminal region to form a

The culmination of multidrug-resistant efflux pumps vs. meager antibiotic arsenal era: Urgent need for an improved new generation of EPIs.

Efflux pumps function as an advanced defense system against antimicrobials by reducing the concentration of drugs inside the bacteria and extruding the substances outside. Various extraneous substances, including antimicrobials, toxic heavy metals, dyes, and detergents, have been removed by this protective barrier composed of diverse transporter proteins found in between the cell membrane and the periplasm within the bacterial cell. In this review, multiple efflux pump families have been analytically and widely outlined, and their potential applications have been discussed in detail. Additiona

Scramblases and virus infection.

The asymmetric distribution of lipids, maintained by flippases/floppases and scramblases, plays a pivotal role in various physiologic processes. Scramblases are proteins that move phospholipids between the leaflets of the lipid bilayer of the cellular membrane in an energy-independent manner. Recent studies have indicated that viral infection is closely related to cellular lipid distribution. The level and distribution of phosphatidylserine (PtdSer) in cells have been demonstrated to be critical regulators of viral infections. Previous studies have supported that the infection of human immunod

Rapsyn as a signaling and scaffolding molecule in neuromuscular junction formation and maintenance.

The acetylcholine receptor (AChR) is highly concentrated at the neuromuscular junction (NMJ), ensuring efficient signal transmission from motoneurons to muscle fibers. This requires the agrin-LRP4-MuSK signaling as well as rapsyn, a peripheral, intracellular protein that is enriched at the NMJ. Mutations of rapsyn have been associated with NMJ diseases including congenital myasthenia syndromes. Rapsyn is a prototype of synaptic adaptor proteins that is thought to bind and anchor neurotransmitter receptors to the postsynaptic membrane. In accord, it interacts with the AChR and a plethora of pro

A lipid-binding protein mediates rhoptry discharge and invasion in Plasmodium falciparum and Toxoplasma gondii parasites.

Members of the Apicomplexa phylum, including Plasmodium and Toxoplasma, have two types of secretory organelles (micronemes and rhoptries) whose sequential release is essential for invasion and the intracellular lifestyle of these eukaryotes. During invasion, rhoptries inject an array of invasion and virulence factors into the cytoplasm of the host cell, but the molecular mechanism mediating rhoptry exocytosis is unknown. Here we identify a set of parasite specific proteins, termed rhoptry apical surface proteins (RASP) that cap the extremity of the rhoptry. Depletion of RASP2 results in loss o

A multidrug ABC transporter with a taste for GTP.

During the evolution of cellular bioenergetics, many protein families have been fashioned to match the availability and replenishment in energy supply. Molecular motors and primary transporters essentially need ATP to function while proteins involved in cell signaling or translation consume GTP. ATP-Binding Cassette (ABC) transporters are one of the largest families of membrane proteins gathering several medically relevant members that are typically powered by ATP hydrolysis. Here, a Streptococcus pneumoniae ABC transporter responsible for fluoroquinolones resistance in clinical settings, PatA…

Borrelia burgdorferi BBI39 Paralogs, Targets of Protective Immunity, Reduce Pathogen Persistence Either in Hosts or in the Vector.

Borrelia burgdorferi genome harbors several paralogous gene families (pgf) that can encode immunogenic proteins of unknown function. Protein-protein interaction assays using a transmission-blocking vaccine candidate, BBA52, as bait identified an interacting partner in spirochetes-a member of pgf 54, annotated as BBI39. We show that BBI39 is a surface-exposed membrane antigen that is immunogenic during spirochete infection, despite the gene being primarily transcribed in the vector with a transient expression in the host only at tick-bite sites. Immunization of rodents with BBI39, or a diverse

The structural basis for CD36 binding by the malaria parasite.

CD36 is a scavenger receptor involved in fatty acid metabolism, innate immunity and angiogenesis. It interacts with lipoprotein particles and facilitates uptake of long chain fatty acids. It is also the most common target of the PfEMP1 proteins of the malaria parasite, Plasmodium falciparum, tethering parasite-infected erythrocytes to endothelial receptors. This prevents their destruction by splenic clearance and allows increased parasitaemia. Here we describe the structure of CD36 in complex with long chain fatty acids and a CD36-binding PfEMP1 protein domain. A conserved hydrophobic pocket a

Platelet actin nodules are podosome-like structures dependent on Wiskott-Aldrich syndrome protein and ARP2/3 complex.

The actin nodule is a novel F-actin structure present in platelets during early spreading. However, only limited detail is known regarding nodule organization and function. Here we use electron microscopy, SIM and dSTORM super-resolution, and live-cell TIRF microscopy to characterize the structural organization and signalling pathways associated with nodule formation. Nodules are composed of up to four actin-rich structures linked together by actin bundles. They are enriched in the adhesion-related proteins talin and vinculin, have a central core of tyrosine phosphorylated proteins and are dep

Analysis of the cross-reactivity of various 56 kDa recombinant protein antigens with serum samples collected after Orientia tsutsugamushi infection by ELISA.

Orientia tsutsugamushi, the etiologic agent of scrub typhus, has a highly expressed and immunodominant 56-kD outer membrane protein. This protein is one of the leading candidates for diagnosis and vaccine development for scrub typhus. Previous studies using recombinant 56-kD protein (r56s) derived from Karp strain (Kpr56) in a mouse model have shown good homologous protection but only moderate to poor heterologous protection. We evaluated the cross-reactivity of recombinant 56-kD proteins from Karp, Kato, Gilliam, TA763, and three chimeric 56-kD proteins. Not all r56s are equally reactive with

Neuroligin-deficient mutants of C. elegans have sensory processing deficits and are hypersensitive to oxidative stress and mercury toxicity.

Neuroligins are postsynaptic cell adhesion proteins that bind specifically to presynaptic membrane proteins called neurexins. Mutations in human neuroligin genes are associated with autism spectrum disorders in some families. The nematode Caenorhabditis elegans has a single neuroligin gene (nlg-1), and approximately a sixth of C. elegans neurons, including some sensory neurons, interneurons and a subset of cholinergic motor neurons, express a neuroligin transcriptional reporter. Neuroligin-deficient mutants of C. elegans are viable, and they do not appear deficient in any major motor functions…

Molecular mechanisms of receptor-mediated endocytosis in the renal proximal tubular epithelium.

Receptor-mediated endocytosis is a pivotal function of renal proximal tubule epithelial cells (PTECs) to reabsorb and metabolize substantial amounts of proteins and other substances in glomerular filtrates. The function accounts for the conservation of nutrients, including carrier-bound vitamins and trace elements, filtered by glomeruli. Impairment of the process results in a loss of such substances and development of proteinuria, an important clinical sign of kidney disease and a risk marker for cardiovascular disease. Megalin is a multiligand endocytic receptor expressed at clathrin-coated p
